Occurrence of Sclerotium Rot on Wild Dolnamul
2024년 경상남도 함양군 백전면 팥 재배 농가 포장 언덕에 야생으로 생육 중인 돌나물(Sedum sarmentosum)에서 흰비단병 증상이 발생하였다. 병징은 돌나물 지제 부위의 줄기 아랫부분이 수침상으로 물러지고 부패하여 서서히 시들면서 포기 전체가 말라 죽었다. 땅과 맞닿은 부위의 줄기에서 병반부와 토양 표면에 흰색 비단결의 곰팡이가 발생하며 갈색의 작은 둥근 균핵이 많이 형성되었다. 감자한천 배지에서 균총은 흰색의 비단결이고 잘 자라며 배양 기간이 경과함에 따라 갈색의 작은 둥근 균핵이 형성되었다. 균핵의 크기는 1-3㎜이며 균사의 폭은 4-9㎛였다. 분자동정을 위해 돌나물 균주의 ITS rRNA 유전자 영역의 염기서열을 PCR 증폭하였고 염기서열을 분석하였다. 유연관계 분석 결과 돌나물 균주는 Agroathelia rolfsii 분류군에 포함되는 것을 확인하였다. 종특이적 프라이머를 이용한 진단 PCR 결과 종특이적 PCR 산물이 생성되었다. 이러한 결과를 근거로 돌나물 흰비단병 균주를 A. rolfsii로 동정하였다. 코흐의 가설을 만족하기 위해 돌나물에 인공감염을 시킨 결과, 감염 15일 후 포장에서 발견한 병징과 동일한 병이 접종부위에서 발달하였고 재분리를 수행하였다. 본 연구는 A. rolfsii에 의한 돌나물 흰비단병의 최초 보고로 향후 흰비단병의 발생 및 방제를 위한 자료로 활용 가치가 높다고 여겨진다.
In 2024, sclerotium rot occurred in the wild dolnamul (Sedum sarmentosum) on a red bean field hill in Baekjeon-myeon, Hamyang-gun, Gyeongsangnam-do, South Korea. Symptoms of the disease include the lower part of the dolnamul appearing water-soaked, rotting and, gradually withering, until the entire plant dries up and dies. White silky molds appeared on the lesions and the soil surface of the stems in contact with the ground, and many small, brown, round sclerotia were formed. The mycelia were white and silky and grew well on potato agar medium, and many small, brown, round sclerotia were formed. The size of the sclerotia was 1-3㎜, and the width of the mycelia was 4-9㎛. The dolnamul isolate was further identified by analyzing the ribosomal transcribed spacers (ITS) region, amplifying, and sequencing. The results of the phylogenetic analysis confirmed that the dolnamul isolate belongs to the Agroathelia rolfsii taxon. The diagnostic PCR using species-specific primers generated species-specific PCR amplicons. Based on these results, the dolnamul isolate was identified as A. rolfsii. To fulfill Koch's postulates, fungus isolated from dolnmul was artificially infected, and 15 days after inoculation, the same symptoms as those found in the field developed, and re-isolation was performed. This study is the first report of sclerotium rot of dolnamul caused by A. rolfsii, and it is considered to have high value as data for the occurrence and control of sclerotium rot in the future.
