Growth of Salmonella Enteritidis on Gimbap Ingredients and Their Inactivation by UVC-LED Irradiation
즉석섭취식품의 소비 증가와 함께 김밥과 관련된 식중 독 발생 위험이 증가하고 있으며, 특히 계란지단은 Salmonella 오염의 주요 원인 식재료로 지목되고 있다. 본 연구에서는 김밥 재료에 오염된 S. Enteritidis의 보관 온 도에 따른 생육 특성을 평가하고, 김밥 제조 전 단계에서 적용 가능한 표면 살균 기술로서 275 nm UVC-LED의 저 감 효과를 확인하였다. 동물성 식재료인 계란지단과 햄, 식물성 식재료인 오이와 당근을 대상으로 S. Enteritidis 4 균주를 혼합하여 표면 접종한 후 4oC, 10oC, 25oC에서 9 시간 동안 보관하며 균의 생육 변화를 관찰하였다. 모든 재료에서 4oC와 10oC에서는 유의적인 생육이 나타나지 않았으나, 25oC에서는 계란지단(3시간), 당근(1.5시간), 오이 (4.5시간)에서 균수가 유의적으로 증가하였으며, 햄에서는 유의적인 증가가 나타나지 않았다. UVC-LED의 살균 효과 를 평가한 결과, LED와 시료 간 거리를 10 cm (0.66mW/cm2) 로 설정하고 9.9 mJ/cm2의 선량으로 조사하였을 때 계란 지단에서 1.0 log CFU/g, 햄에서 1.2 log CFU/g, 당근에 서 2.6 log CFU/g, 오이에서 2.3 log CFU/g 감소하였다. UVC-LED와 시료 간 거리를 1 cm (1.90 mW/cm2)로 감소 시켰을 때에도 추가적인 살균 효과의 유의적인 증가는 나 타나지 않았다. 이러한 재료별 살균 효과의 차이는 식품 표면의 굴곡 및 불균일성에 의해 UVC 빛 투과가 제한됨 에 따라 미생물 살균 효과가 감소하였기 때문으로 판단된 다. 본 연구 결과, 김밥 재료는 10oC 이하에서 보관할 경 우 Salmonella의 생육을 억제할 수 있으며, 275 nm UVCLED 처리는 김밥 제조 직전에 재료 표면에 적용 가능한 추가적인 살균 기술로서 김밥의 미생물학적 안전성을 향 상시키는 데 활용될 수 있을 것으로 기대된다.
The increasing consumption of ready-to-eat foods has heightened the risk of foodborne illnesses associated with the food dish gimbap, where egg garnishes are considered a potential source of Salmonella contamination. This study evaluated the growth behavior of S. Enteritidis on representative gimbap ingredients at different storage temperatures and assessed the efficacy of 275 nm ultraviolet-C light-emitting diode (UVC-LED) irradiation as a surface decontamination treatment prior to gimbap preparation. A cocktail containing four strains of S. Enteritidis was inoculated onto egg garnish and ham (animal-based ingredients), and cucumbers and carrots (plant-based ingredients). The inoculated samples were stored at 4oC, 10oC, and 25oC for up to 9 h. No significant growth was observed at 4oC or 10oC for any ingredient. However, at 25oC, there were significant increases in Salmonella populations after 3 h on egg garnish, 1.5 h on carrot, and 4.5 h on cucumber; however, there was no significant increase on ham. The potential use of UVC-LED irradiation for rapid surface decontamination was also evaluated. At a distance of 10 cm (0.66 mW/cm2), a UV dose of 9.9 mJ/cm2 resulted in reductions of 1.0 log CFU/g on egg garnish, 1.2 log CFU/g on ham, 2.6 log CFU/g on carrot, and 2.3 log CFU/g on cucumber. Reducing the distance to 1 cm (1.90 mW/cm2) did not significantly improve the inactivation efficacy. The variations in inactivation efficacies among the ingredients were considered to be associated with differences in the direct exposure to UVC caused by surface irregularities and uneven structures. These results suggest that storage below 10oC is critical to prevent Salmonella growth on gimbap ingredients and that 275 nm UVC-LED treatment could improve microbial safety during gimbap preparation.
