        CE RCD (Recreational Craft Directive) is a certification for the design and construction of small vessels, including pleasure yachts, which are widely used not only in the countries within the European Union, but also in Japan and Southeast Asia. Recently, South Korean leisure craft shipyards have developed interest in exporting to foreign leisure craft markets such as Europe; however, they have encountered difficulties because of the CE RCD regulations, which are relatively complex and difficult to understand. The requirements for buoyancy and stability, which are essential properties that must be understood within the early stage of ship design, are defined based on ISO 12217. However, preparing this assessment according to ship classification regulations is an exceedingly complex task, even with knowledge of naval architecture. In this research, we have developed design support tools to systematically support assessments and preemptively define design information so that buoyancy and stability assessments based on ISO 12217 can be systematically prepared. Our research results were applied to actual examples of yacht design to confirm validity. We believe that the improved yacht design process presented in this research can act as a foundational reference for enhancing the effectiveness and systematic buoyancy and stability assessments.

        The purpose of this study is to keep the safety of the car ferry passengers and vessels by investigating and analyzing vessel safety management systems in Korea and China. To this end, we investigated Korea-China car ferries and the current status and causes of global marine accidents corresponding to the sizes of the vessels from Korea and China. Furthermore, we investigated car ferries’ crew management and safety management. As a result of the analysis of the ferry accident, the causes of human error and ship’s age were the greatest, but the ship’s companies showed a negative stance regarding the age restriction. It seems that it is necessary to utilize the near-miss accident reporting system and differentiate the management of ship’s aging. Also, it was analyzed that both the ship company and the crew of the ship need to strengthen their awareness of safety management.

        The purpose of this study was to investigate the effect of estrus synchronization to altrenogest regumate (progesterone), PMSG/hCG, and artificial insemination (AI) on galectin-3, FGF-9 gene and protein expression. The morpho-metrical parameters of the endometrium and the number of corpora lutea (CL) were recorded. RNA was isolated from endometrial, oviduct and ovary tissues of non-synchronized (Control; n = 7) and AI synchronized (regumate, PMSG/hCG; n = 7) sows. The total number of CL was higher (P<0.05) in pigs treated with regumate/PMSG/hCG. The content of gelactin-3 and FGF-9 mRNA in pre-embryonic development stages increased on particular days, in control and studied in regumate/PMSG/hCG administered pigs. Gelactin-3 and FGF-9 were affected by regumate/PMSG/hCG treatment in the both pre-embryonic development stages (P<0.001, P<0.05) and encdometrial tissue (P<0.001, P<0.01). The regumate/PMSG/hCG treatment resulted in elevated expression of gelactin-3 (P<0.001) and FGF-9 (P<0.005) in oviduct and ovary tissues in comparison to control sows. Moreover, oviduct amount of gelectin-3 mRNA was higher in regumate/PMSG/hCG sows in comparison to the control group (P<0.05), whereas, expression characteristics of gelactin-3 and FGF-9 were investigated by hematoxylin and eosin stained and immunohistochemical staining. The results showed that galectin-3 and FGF-9 were significantly shown in the endometrium, oviduct and ovary tissues of the regumate/PMSG/ hCG. Presented data show that exogenous hormones administration can affect gene and protein expression in the sow reproductive tract.

        The objective of this study was to investigate the influence of the addition of caffeine (alkaloid family) to the ejaculates of boar sperm quality as well as investigate their optimum concentrations for increasing the movement of sperms. Semen was collected from 9 boars by the gloved-hand technique one week interval. Semen followed by cryopreservation with egg yolk extender freezing medium using freezing protocol. The collected semen were frozen on the same day. Motility was assessed for % motile cell characteristics using computer-assisted semen analysis (CASA; SAIS SI-100, Medical supply, Korea). Frozen boar sperms were thawed in Beltsville Thawing Solution (BTS) with 5, 10, and 15mM caffeine were then incubated at 38 celsius degree for 20 minutes. In experiment 1, semen were diluted BTS and addition of different concentration of caffeine to the pre-freeze semen cryopreservation. In experiment 2, incubation of frozen-thawed sperm in BTS supplemented with different concentration of caffeine for 20 minutes improved (P<0.05) after semen cryopreservation-thawing on sperm quality. After thawing significantly increased progressive and total motility. The addition of 10 mM caffeine to cryopreserved semen after thawing significantly increased progressive and total motility compared with other treatment. These result suggest that caffeine enhanced post-thaw motility of cryopreserved boar sperm when added after thawing.

        The present study was conducted to investigate the effect of BHT supplementation on sperm motility, viability, acrosomal integrity and plasma membrane integrity after frozen-thawing. One ejaculate was collected from one fertile Hanwoo bull by using artificial vagina at Hanwoo Research Institute. The ejaculate was transferred to laboratory immediately and diluted with pre-warmed semen extender (Optixcell, France) (1:1). Sperm dilutions were extended to a final concentration of 40 x 106 sperm/ml, and divided into 5 groups according to BHT concentration (0, 0.5, 1.0, 2.0 and 4.0 mM) and cryopreserved in LN2 tank until evaluation. Frozen-thawed semen was transferred to 1.5 ml tube and incubated for 0, 2 and 4h. Sperm motility and motility parameters (total motility, VSL with 25μm≥, VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were evaluated by sperm class analysis (SCA, IVOS, Spain). There were not significant effects of BHT supplementation (0, 0.5, 1.0 and 2.0 mM) on total motile and VSL with 25μm≥ at 0, 2 and 4h. However, 4.0 mM of BHT supplementation showed negative effect on total motile (26.3%), VSL with 25μm≥ (1.3%) at 0 h (p<0.001). The viability and acrosomal integrity of spermatozoa were evaluated by Trypanblue/Giemsa staining method and divided into 4 groups; live and intact acrosome (LIA), live and damaged acrosome (LDA), dead intact acrosome integrity (DIA), dead damaged acrosome (DDA). There were no significant differences of LIA, LDA, DIA and DDA on various BHT concentrations at 0 and 2 h. However, 4.0 mM BHT supplementation showed decreased LIA compared with 0, 0.5, 1.0 and 2.0 mM BHT at 4 h (34.6, 37.1, 43.6, 45.4 and 14.7% vs. 0, 0.5, 1.0, 2.0 and 4.0 mM, irrespectively; p<0.01). Addition of 4.0 mM of BHT showed negative effect on plasma membrane integrity compared with that of 0, 0.5, 1.0 and 2.0 BHT at 2 h (71.9, 64.2, 64.6, 67.5 and 31.7 % vs. 0, 0.5, 1.0, 2.0 and 4.0 mM, irrespectively; p<0.05). In conclusion, various BHT concentrations on optixcell extender showed no improvement on sperm motility, viability and plasma membrane integrity.

        Polo-like kinase 1 (plk1) shows multiple events of somatic cell and mammalian oocyte division. In mice, Plk1 distributes to the centromeres from prophase to anaphase and compose spindle apparatus at different stages of mitosis in spindle organization. Somatic cell nuclear transfer (SCNT) has a number of advantages however it is difficult to apply to basic or translational researches due to its low cloning efficiency. The causes of this low cloning efficiency are unclear. However, they are attributed to the cumulative results of several biological and technical factors. In this study, a biological factor plk1 was investigated. B6D2F1 mice (7–8 weeks old) were superovulated with 10 IU of pregnant mare’s serum gonadotropin and 9 U of human chorionic gonadotropin (HCG) 48 hr later. The oocytes were then collected 14 hr after HCG injection and cultured on potassium simplex optimized medium (KSOM). The plk1-specific inhibitor BI2536 was used to understand the influence of plk1. The 2-cell stage embryos were assessed by fluorescence immunoassay. In consequence, all BI2536-treated embryos failed in the first mitotic division which showed plk1 have critical role in the first mitotic division of the mouse embryo. SCNT requires enucleation of oocyte and injecting a donor cell into the enucleated cytoplast. In this process, a respectable amount of plk1 that co-localize with nucleus may be removed together. Fluorescence immunoassay and qPCR were used to monitor the change of plk1 level during SCNT. There was significant difference between the control and enucleated embryos in the level of plk1. In all division-failure 2-cell embryos, incorrect positioning of plk1 was found. Taken together, this results demonstrate that plk1 is critical for successful mitotic division of mouse SCNT 1-cell embryos.

        The present study was conducted to investigate the effect of different heights from liquid nitrogen (LN2) vapor on sperm motility and morphology after frozen-thawing. Two ejaculates were collected from 2 fertile Hanwoo bulls (A and B) by using artificial vagina at Hanwoo Research Institute. After collection, ejaculates were transferred to laboratory immediately and diluted with semen extender (Optixcell, France). Sperm dilutions were extended to a final concentration of 40 x 106 sperm/ml, and cooled at 4°C for 4 h and loaded to 0.5 ml straws. The straws were divided into 2 groups. Straws were placed in 3 or 9 cm of LN2 vapor for 14 min and then plunged into LN2 tank and cryopreserved until evaluation. Sperm motility and motility parameters (total motility, VSL with 25μm≥, VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were evaluated by sperm class analysis (SCA, IVOS, Spain) after frozen-thawed. In bull A, 3cm group showed higher percentages of total motility, VSL with 25μm and VAP compared those with 9cm group (98.0 vs. 93.4%, 62.4 vs. 54.0% and 98.6 vs. 93.2%, 3 vs. 9 cm, irrespectively; p<0.001). In bull B, frozen-thawed sperm of 3cm group showed higher percentages of VSL with 25μm, VCL, VSL, VAP and BCF compared with those of 9cm group (43.5 vs. 26.0%, 123.8 vs. 111.6 μm, 62.9 vs. 57.3 μm and 81.5 vs. 72.5 μm; 3 vs. 9 cm, irrespectively; p<0.001). The viability and acrosomal integrity of spermatozoa were evaluated by Trypanblue/Giemsa staining method divided into 4 groups; live and intact acrosome (LIA), live and damaged acrosome (LDA), dead intact acrosome integrity (DIA), dead damaged acrosome (DDA). In bull A, frozen-thawed sperm of 3 and 9cm groups showed no significant difference in LIA, LDA, DIA and DDA. In bull B, 3 cm group showed higher LIA and lower DIA compared with those of 9 cm group (73.2 vs. 23.7% and 23.7 vs. 32.2%, 3 vs. 9 cm, irrespectively; p<0.001). We suspected that 3 cm vapor on LN2 vapor might be affected positively spermatozoa viability and acrosomal integrity compared with 9 cm group. In conclusion, semen freezing procedure in the present study will improve sperm quality after frozen thawing.

        Although assisted reproductive technology (ART) has been developed in many mammalian species including cows, the only embryo preservation technology that is available is cryopreservation. In the present study, small molecules were used to preserve embryos at room temperature. The basic medium for embryo preservation consisted of 1% BSA non-cryopreservation medium (BNC) instead of fetal bovine serum (FBS). To maintain survival and prevent damage during embryo storage, three candidate small molecules were selected—CHIR99021, Y-27632 and Thiazovivin—and their concentrations were optimized. Then, the embryos in the small molecule supplemented preservation medium were stored at room temperature. The viability and hatching rate of embryos stored at 10°C were greater for Y-27632-BNC and CHIR99021+Y-27632-BNC compared to BNC. However, the rate was lower for Thiazovivin-BNC compared to BNC. Although there were no surviving embryos after storage at 20°C, the viability and hatching rate of embryos significantly increased in Y-27632-BNC and CHIR99021+Y-27632-BNC compared to BNC. The mechanism by which small molecules enhance survival of embryos during storage was investigated, and expression of heat shock protein 70 was observed to increase. The findings of this work may be useful in improving ART in the agricultural field.

        In vitro maturation (IVM) systems have become indispensable for the production of large numbers of competent oocytes in domestic species. The quality of in vitro matured oocyte is one of the important factors determining the success of assisted reproductive technologies (ARTs) including intracytoplasmic sperm injection (ICSI), in vitro fertilization (IVF), and somatic cell nuclear transfer (SCNT) in human and livestock. Incomplete cytoplasmic maturation of oocytes can lead not only to a failure of fertilization but also to a developmental arrest after ARTs. Thus, establishment of a stable IVM system to produce a large number of high quality oocytes, especially in domestic animals, is essential for improvement of ARTs efficiency by producing high quality embryos. The morphological characteristics are commonly used to predict the developmental potential of oocytes and embryos. Usually, normal oocytes shrink when exposed to a hypertonic medium, and recover their morphology when returned to an isotonic medium. During this process, oocytes show various morphologic changes, such as shrinkage in spherical (SSP) or irregular shapes (SIR). In the first study, we investigated whether the shrinkage pattern of oocytes that was observed after hyperosmotic treatment could be used as a morphologic characteristic to predict the quality of IVM oocytes in pigs. We found that SSP oocytes showed improved developmental competence after PA and SCNT. This improved embryonic development was most likely because of the more advanced nuclear and cytoplasmic maturation in SSP oocytes compared with SIR oocytes. Pig oocytes shows a wide variation in the size of perivitelline space (PVS) after IVM. Based on this finding, we examined in the second study whether or not there was any correlation between the PVS size of IVM oocytes and their developmental competence after PA and SCNT. Our results demonstrated that in vitro developmental competence to the blastocyst stage positively correlated with the size of the PVS of oocytes after IVM. In addition, we observed that mature oocytes with a larger PVS showed higher levels of intracellular GSH content and transcription factor expression. Furthermore, enlargement of the PVS by culturing in reduced NaCl medium improves the embryonic development after PA and SCNT. In the third study, we investigated the effects of a hypotonic medium with reduced NaCl (61.6 mM) compared with an isotonic medium (108.0 mM NaCl) on oocyte maturation and embryonic development after PA and SCNT. In addition, we attempted to optimize our IVM system using a hypotonic maturation medium by examining the effects of hypotonic medium during various stages of IVM on oocyte maturation and subsequent embryonic development. Our results demonstrated that maturation of pig oocytes in hypotonic medium with reduced NaCl during the last 11 hr of IVM increased the developmental competence of oocytes after PA and SCNT. These beneficial effects was also shown in a commercial medium (a minimum essential medium; aMEM) in which the NaCl concentration was reduced to 61.6 mM. In addition, IVM of oocytes in medium with reduced NaCl increases the proportion of SSP oocytes in pigs. In summary, our results demonstrate that IVM of pig oocytes in a hypotonic medium with low-NaCl is better able to support embryonic development after PA and SCNT, most likely by improving the cytoplasmic maturation via increased intraoocyte GSH content and widened PVS. Based on these results, the newly developed IVM system using a hypotonic medium with reduced NaCl can produce high quality oocytes and be considered a new strategy for improving ARTs efficiency in pigs.

        In this study, causes and consequences of bullying were explored, focusing on victim personality and appearance factors. The research model included the relationships among bullying, important causes of bullying (aggression, cleanliness, and obesity of victims) and consequences of bullying (self-esteem and depression) and was tested through survey data from 594 (194 male and 404 female) middle school students. Data were analyzed using descriptive statistics, confirmatory factor analysis, and path analysis with SPSS and AMOS. In the results of the model tests, victims who are more aggressive, less clean, and more overweight are more likely to be bullied. In addition, adolescents bullied by others tend to have lower self-esteem and more depression. Furthermore, the model comparison was assessed by gender and a difference between the two gender models was found. In the check of critical ratio for difference, it was concluded that the cleanliness of the victim is a more important cause of bullying for male adolescents than for female adolescents. It is meaningful to confirm the effects of the victim’s appearance factors on bullying in the literature focusing on personality issues of victims. Educators and counselors may refer to the findings in developing counseling guidelines and manuals to prevent bullying in school.

        Through density functional theory calculations, to provide insight into the origins of the catalytic activity of Au nanoparticles (NPs) toward oxidation reactions, we have scrutinized the oxygen adsorption chemistry of 9 types of small unsupported Au NPs of around 1 nm in size (Au13, Au19, Au20, Au25, Au38, and Au55) looking at several factors (size, shape, and coordination number). We found that these NPs, except for the icosahedral Au13, do not strongly bind to O2 molecules. Energetically most feasible O2 adsorption that potentially provides high CO oxidation activity is observed in the icosahedral Au13, our smallest Au NP. In spite of the chemical inertness of bulk Au, the structural fluxionality of such very small Au NP enables strong O2 adsorption. Our results can support recent experimental findings that the exceptional catalytic activity of Au NPs comes from very small Au species consisting of around 10 atoms each.

        For the development of processed food for hypoglcemia, it is important to construct model system to confirm factors that reduce the glycemic index in real food. This study was to investigate the relationship between the gel model system and real food the high amylose type of segoami using response surface methodology. The independent variables were concentration (X1; 10, 15, 20, 25, and 30%) and steaming time (X2; 30, 35, 40, 45, and 50 min). The predicted glycemic index (Y1) was analyzed as a dependent variable. The regression of pGI was 0.7343, indicating that the model fits the data well in the prediction test. The predict glycemic index of the gel was in the range of 71.38 ~ 83.78, depending on the gel preparation conditions and predicted optimum condition was 23.7% gel concentration and 43.8 minutes of gelatinization. The predict glycemic index of rice gel were decreased with an increase of gel concentration, rather than the steaming time. In order to confirm the effect on in vitro digestibility in real food, the real food (Garraedduk) was prepared by applying the optimal conditions. As a result, there was no significant difference between the predicted value (77.1) and the experiment value (76.6). This result showed that the in vitro digestibility in real food can be predicted by applying the gel model system.

        Recently, additive-free sweet potato has been commercialized as a healthier snack food. However, these products are producing by semi-dried form to extend shelf-life, hence the products also possesses hard texture during chewing. To produce crispy sweet potato snack, this study was attempted to dry the semi-dried sweet potato instantly by a hot-press (HP) process. Mashed sweet potato was reformed and semi-dried at 65°C for varying time to provide various final moisture contents. The semi-dried samples were subjected to HP at the process temperature of 175-180°C for 2 seconds. As quality parameters, moisture content, brix, color, texture and sensorial test of the products were evaluated. Based on the results, optimal quality of the product was obtained by the semi-dried sample with 15~18% moisture at which the products exhibited good crispiness with bright yellow color. When the moisture content of the sample before HP was lower than 15%, the final products were normally broken during the HP process. In addition, extreme browning appearance was generated in this condition. Conversely, the sample with more than 18% of moisture was not completely dried by HP and the final product did not possess the crispy texture, although this product showed bright yellow appearance. Consequently, this study demonstrated the potential application of HP in sweet potato processing and the best condition was greatly affected by moisture content of sample prior to HP process.

        Bourbon is the major coffee variety grown in Rwanda. Fully washed bourbon produced in Rwanda is a nascent specialty coffee and no investigation of its quality characteristics has been reported. Thus, this study aimed at providing information about intrinsic flavor characteristics and identifying the most discriminating aroma compounds of this coffee from the major growing areas in four provinces (considered by this study as geographical sub-regions) in comparison with Arabica coffee from Ethiopia and Brazil using electronic nose and electronic tongue. Samples were discriminated and characterized by performing a principal component analysis (PCA). A discriminant factorial analysis (DFA) model was also used to predict the geographic origin of coffee samples based on their flavor characteristics. A similarity was observed between flavor characteristics of coffee samples from western and southern sub-regions as well as between northern and eastern sub-regions of Rwanda. Twenty one most discriminating aroma compounds were found but the identified and confirmed ones are: Butan-2-one, Acetic acid, n-Butanol, 2, 3- Pentanedione, Ethyl-2-methybutyrate, Dimethylsulfide, 2-Prpanol, 2-Methylfuran, Ethylacetate, Ethyl propionate and Heptanal. A DFA model using e-nose was successful in predicting the geographic origin of coffee samples but not with e-tongue. This means that aroma can reliably be used to predict the geographic origin of coffee samples than their taste characteristics. Further investigation is required to test the possibility of origin recognition based on taste characteristics.

        The purpose of this study was to investigate meteorological factors’ effects on clothing sales based on empirical data from a leading apparel company. The daily sales data were aggregated from “A” company’s store records for the Fall/Winter season from 2012 to 2015. Daily weather data corresponding to sales volume data were collected from the Korea Meteorological Administration. The weekend effect and meteorological factors including temperature, wind, humidity, rainfall, fine dust, sea level pressure, and sunshine hours were selected as independent variables to calculate their effects on A company’s apparel sales volume. The analysis used a SAS program including correlation analysis, t-test, and multiple-regression analysis. The study results were: First, the weekend effect was the most influential factor affecting sales volume, followed by fine dust and temperature. Second, there were significant differences in the independent variables’effects on sales volume according to the garments’ classification. Third, temperature significantly affected outer garments’sales volume, while top garments’ sales volume was not influenced significantly. Fourth, humidity, sea level pressure and sunshine affected sales volume partly according to the garments’ item. This study can provide proof of significant relationships between meteorological factors and the sales volume of garments, which will serve well to establish better inventory strategies.

        에너지 음료는 카페인을 주성분으로 타우린, 비타민 같은 다른 energy-enhancing 성분을 함유하고 있다. 미국과 유럽에서는 글루쿠로노락톤이 에너지 음료에 첨가될수 있으나, 국내에서 의약품으로는 허가되어 있다. 따라서 식품 첨가물로는 그 사용이 금지 되어 있어, 지속적으로 수입 및 유통 음료에서 시험검사를 하여 규제하고 있다. 현재 분석법으로 사용하는 LC-PDA 법은 복잡한 유도체화 과 정을 거치고, 음료 중에 당류들이 위양성 결과를 나타내 기도 한다. 이런 기존 방법의 단점을 개선하기 위해 HILICESI- MS/MS (hydrophilic interaction liquid chromatography coupled to electrospray ionization tandem mass spectrometry) 를 이용한 분석법을 개발하고, 선택성, 직선성, 검출한계, 정량한계, 정밀도, 정확성, 재현성에 대하여 분석법 유효성 검증을 수행했고, AOAC, EURACHEM 가이드라인에 부합되는 결과를 얻었다.

        In the present study, biomass-based lignin was extracted from industrial waste black liquor and the extracted lignin was characterized by means of attenuated total reflectance- Fourier transform infrared spectroscopy and 1H-nuclear magnetic resonance spectroscopy. The extracted lignin was carbonized at different temperatures and then activated with steam at 850oC. The extracted lignin in powder state was transformed into a bulky carbonized lignin due to possible fusion between the lignin particles occurring upon carbonization. The carbonized and then pulverized lignin exhibits brittle surfaces, the increased thermal stability, and the carbon assay with increasing the carbonization temperature. The scanning electron microscopic images and the Brunauer-Emmett-Teller result indicate that the steam-activated carbon has the specific surface area of 1718 m2/g, which is markedly greater than the carbonized lignin. This study reveals that biomassbased activated carbon with highly porous structure can be produced from costless black liquor via steam-activation process.

        Nitrogen-doped carbon nanosheets with a developed porous structure were prepared from polyurethane foams by hydrothermal carbonization following ZnCl2 chemical activation. Scanning electron microscopy, thermogravimetric analysis, Fourier transform infrared spectroscopy, solid state 13C nuclear magnetic resonance (NMR) spectra and X-ray photoelectron spectroscopy were used to characterize the nitrogen-doped carbon nanosheet structure and composition. The removal of Cr(VI) by the N-doped carbon nanosheets was investigated. The results showed that the maximum removal capacity for chromium of 188 mg/g was found at pH=2.0 with PHC-Z-3. pH had an important effect on Cr(VI) removal and the optimal pH was 2.0. Moreover, amino groups and carboxyl groups in the nitrogen-doped carbon nanosheet played important roles in Cr(VI) removal, and promoted the reduction of Cr(VI) to Cr(III).
