Effects of elevated CO2 and temperature on nitrogen (N) uptake , leaf N concentration, N partitioning , N use efficiency (NUE) and grain yield of pot and field grown rice (Oryza sativa. L.cv. Chukwangbyeo) under canopy-like conditions were studied over three years. Rice plants were grown in pots and in the field in temperature gradient chambers containing either ambient(350ppm) or elevated CO2 concentrations (690 or 650ppm) in conbination with either four or seven temperature regimes ranging form ambient temperature(AT) to AT plus 3℃. There were three N supplies 94g or 6g m-2 to 20g or 48g m-2.Elevated CO2 increased N uptake in field-grown rice ; the magnitude of this effect was thelargest (+15%) at the highest N level. However, in pot-grown rice, N uptake was suppressed with the effect was the largest at high N levels. Leaf N concentration declined at elevated CO2 mainly due to a decrease in N partitiioning to the leaf blades. Air temperature had little effect on the N parameters mentioned previously, wherease NUE for spikelet production declined rapidly with increased temperature irrespective of CO2 concentration. The response of the biomass to elevated CO2 varied with N level, with the greatest response at 20g N m-2 (+30%) . At AT, where high temperature-induced sterility was generally not observed, elevated CO2 increased yield. However, the magnitude of this effect varied greatly (2-39%) with N level, and was mainly dependent on the magnitude of the increase in spikelet number.

It failed occasionally to take a reasonable emergence rate since tobacco (Nicotiana tabacum L.) seeds were planted on late Jan. showing lowest temperature. This experiment was done to measure the effect of GA3 (concentration ; period) , light quality (red ; white ; dark) during or after its treatment, daily irradiation hours( 0 ; 8 ; 12 ;16) and germination termperature (20 or 10 ℃ ocnstant ; 20/10℃ alternating) on the germination rate. Red and white light given during grmination showed no differences between the other daily irradiation hours except that 8 hours red light delayed germination although their 12 hours irradiation had the gratest rate. The rate was increased with increased concentration to GA3 0.01 mM or increased imbibition period to 3 days although the rate of cv. NC 82 was less than that of cv. Burley 21 in the case of dark imbibition of GA3 but daily 12 hours irradiation during germination. Light quality forced during GA3 imbibition eliminated such effect of GA3 shown in the darkness so that only light quality pretreatment and termperature during germinition were affected on the rate. The germination rate of thecultivars was decreased in the order of red, white light, darkness meaning that it was highly influenced by the light quality during GA3 treatment. Regardless of GA3 or light quality treatment,on the other hand, the rate was greater in 20 ℃ constat than 10℃ constant and 20/10℃ alternating germination temperature having similar germinative patterns.

Cathamus tinctorius, Coptis japonica and Asarum sieboldii were tested as substrate for the production of Flammulina velutipes. Among the C. tinctoris , C. japonica and A. sieboldii , C. tinctoris was the best substrate for the production of fruitbody. The effects of addition of C. tinctoris to sawdust substrate resulted in the increased mycelial growth on inoculum culture, 3.1% in ratio of fully culture and shorted one day in culture period. C. tinctoris was decreased 6.1% in ratio of fully culture, 11.0% in ratio of fruitbody productive culm. The addition of C.tinctoris, C.japonica to sawdust substrate increased 134.6%, 114.1% on the yield of the mushroom fruitbody respectively . But A. sieboldii decreased the mycelial growth and pineheading ratio delayed the production of fruitbody.

To give some information on the harvest and storage of Chinese yam (Dioscorea opposita), the experiment was done to examine the effect of its harvesting date and storage method on tuber weight, tuberrelated characters during the storageor at the end of storage. The yam tubers harvested 4 times at 20 days interval from Oct. 15 were stored with different storage methods(pit, room and cold temperature) and on Mar.15. all the characters were measured. Fresh weight of the tuber harvestedafter Nov. 5 and stored were greater than that done on Oct. 15 while dry tuber weight did not have any difference between the harvesting dates. The fresh weigth done directly from the field on Mar. 15 was higher compared to the other harvesting dates owing to its higher moisture content. The rate of sound tuber stored at room or cold temperature condition was higher than that at the field at which the rate was greater at the harvest after late nov. compared to the earlier harvests. Although the two storages were similar at sound tuber rates and marketable yield, the tuber weight was more reduced when stored at room temperature than when done at cold temperature . Brightness and value 'b' of chromaticaity realted to the tuber coloring and sugar content were higher but moisture content was lowerwhen harvested on Oct. 15 than the other havesting dates. All the characteristics realted to the marketability after storage were nearly same when stored at room and cold temperature. Value 'a' of chromaticity and sugar content were higher when harvest on Mar. 15 than when harvested before Dec. 15 but the 'b' value and mositure content were reverse results.

The experiment was conducted to elucidate the optimum planting data and fertilization level of Aruncus dioicus var. kamtscacus HARA. Plant height was increased as the seeding date was delayed. There was no transparent difference on plant height indifferent fertilization level. Nuber of stems was remarkably increased in the fertilization 14-14-12kg. Fresh leaf yield in the fertilization of 24-18-15kg was incresed about 31% comparing in the fertilization of 14-14-12kg. As a result, the optimum planing date and fertilization level was Jan. 15 and 24-18-15kg per 10a, respectively.

A study was carried our to investigate the characteristics of physico-chemical components palatability of cooked rice accoring to different transplanting time. The treatments were consisted of five transplanting time, from May 5 to July 5 at 15 -day intervals, and six cultivars ; two early-maturing, two mid-maturing and two late-maturing cultivars which are clearly identified by evaluation of cumulative temperature and growth duration from seeding to heading of rice plants. In gelatinization characteristics of rice grain, alkali digestion value was low in the earlier transplanting regardlessof maturing types. Initial pasting temeprature maximum viscosity and breakdown as amylogram charcteristics were high at the transplanting of June 5. The palatability of cookedrice was evaluated that early-maturing cultivars were much better in early transplanting than in late transplanting . The optimum transplanting time for palatibility was the periods from May 5 to 20 in early maturing varieties, May 20 to June 5 in mid-maturing and June 5 to June 20 in late-maturing ones at Honam district in Korea.

This experiment was conducted to investigated the characteristics of grain appearance, chemical components. gelatinization, and palatability of cooked rice on each rachis branches as harvested from the transplanting plot of May 20 and June 5 in each two varieties of early-maturing and late-maturing types. In evaluation of rachis branches, the percentage of ripended grain and 1,000 grain weight were found to be high and heavy at upper part of panicle. The factors influencing palatability such as maximum viscosity and breakdown as gelatinization characteristics, and hardness, apringiness, cohesiveness and chewiness as texture characteristics of cooked rice were found to be high at upper part of panicle , indicating the parlability was favored more at upper part than at lower part of panicle due to the genetics and physiological metabolism of rice plants.

This study was conducted to examine the effects of media, crown position, clone ,a nd plant growth substances upon the rootability of cuttings for the establishment of the method of mass production by cutting of Dendropanax morbifera Lev. the artifical soil mixtureof vermiculite, perlite and peatmoss (1 : 1 : 1 v/v/v) with IBA 100mg /l treatment was most effective on the rooting rate (85%) was obtained with semihardwood cuttings, which were collected early-August after the first growing seasons. The effect of crown position on rooting of cuttings were obsreved. Average percentage rooting of cuttings on crown position showed 78% , 72% and 65% in middle , upper and lower part with IBA 100mg/l treatment , respectively. Among the four treatment levels of IBA, the highest rooting rate was observed in 100mg/l IBA treatment.

To produce several high-quality species of Chwinamul (Aster scaber, A. Tataricus, Ligularia fischeri), leaves of the plants grown in different natural habitats were anlayzed. Contents in vitamin C and β-carotene were generally higher in plants grown in the field. However, Aster scaber picked form Yongwol and A. tataricus from Mt.Taryong contained relatively higher vitamin c and β carotene. Crude protein and fat contents varied depending on the growing area. Ca content was markedly higher in the plants grown in the natural havitats.

The morphlogical characteristics and cytotoxic acitivity for14 local varieties fo Codonopsis lanceolata collected from Korea were examined to get basic data for breeding and selection of good cultivars. Plant height ranged from 197.2 cm to 253.2 com, that from Beakunsan(No.11) was the highest and and that form Samcheok(No.5) was the lowest. Generally, plant height of those form northern area of Korea tends to be shorter than those from Southern area. Leaf width and length of that form Husando (No. 12) was the largest While that form chejudo(No.4) was the smallest and those from other areas showed no significant difference. As to the number of branches, that form Yanggu(No.1) was theleast and those from other areas did not show Chejudo(No.14) were the heaviest. Among 14 varieties tested, Heongseong(No.3) and Huksando(No.6) exhibited strong cytotoxicity , which showed 45.0 an d62.0μg. ml respectively.

The feeding effects of Herba hottuynia extracts on the lipids profiles and the content of TAB-reactive substances were evaluated in rats fed a high cholesterol diet. Hot water or ethanol extracts of the dried leave were evaporated and lyophilized . The experimental animals wer edevided to four groups and fed the following diets for 4 weeks : BAsal (cholesterol-free diet), CHOL(cholesterol-enriched diet),CW (cholesterol plus water extract) and CE(cholesterol plus ethanol extract). Dietary cholesterol increased significantly the activities of serum GOT and GPT, but the extracts feeding (0.5% of diet) did not influence the activities induced by dietary cholesterol. Although dietary cholesterol increased significantly the concentrationof serum andliver cholesterol, it tended to decrease the concentation of serum triglycerides. CHolesterol feeding had a lowering effect on the lipid peroxidation value of serum, but not inliver.Furthermore, the extracts feeding, especially water extract, decreased markedly the liver peroxidation value. The results suggest that Houttyunia cordata extracts have an in vivo antioxidant effect, judged from the TBA value in the liver rats fed a high cholesterol diet.

This study was carried out to obtain the information for growth characteristics of crown gall tumor and hairy root transformed by Agrobacterium spp,. on the media with phytohormones, casein hydrolysate and activated charoal. Crown gall tumors and hairly roots were formed respectively on potato tuber discs infected by tumerfaciens A ch 5 and A.rhizogenes ATCC15834. These tumors and roots could be grown on the phytohormone free media. PCR analysis of Rol C and Vir C gene fragments confirmed that crown gall root was prompted on the medium containing 2,4-D 2mg/l with casein hydrolysate lg/l. The survival ration of crown gall tumor callus derived from potato increased on medium containing the activated charcoal 0.5∼0.2mg/l because of the prevention, on the other hand, hairly roots were necrosis on the same medium. Callus derived from hairly root were excellently grown for a short time by suspension culture on liquid medium containing 2.4-d 2mg/L and casein hydrolysate lg/l.

This experiment was conducted to introduce phosphinothricin acetyl -transferase(PAT) gene, resistant to basta and non-selective herbidide, into tobacco(Nicotiana tabacum cv.BY4). For shoot formation,tobacco leaf disks were placed on the MS medium supplemented with 2.0mg/L BA and 0.1mg/L NAA. In this medium condition, tobacco leaf disces were cocultivated with A. tumefaciens MP90 containing NPT IIand PAT resistant to kanamycin and Basta, respectively. Shoots were obtained in the medium containing antibiotics, and those were transferred to rooting medium supplemented with 0.1mg/L NAA and antibiotics. The plants obtaining roots were transplanted into soil. Phenotype of transgenic tobacco plant was mostly as normal plant. However, about 5% was abnormal plant, which did not set seeds. PCR analysis and southern blot were performed to determine transformation. As the results, it was confirmed that PAT gene was stably integrated into tobacco genome.When herbicide, basta, was sprayed to the plants confirmed by PCR, the transgenic plants showed normal growth, whereas normal plants died. Therefore, the result of this experiment show that tobacco transformation for the resistance to basta, non-selective herbicide, was successful because PAT gene was stably integrated into tobacco.

Some morphological character were surveyed and PR-proteinases were induced and characterized from Lilium formosanum Wallace endeimc to Cheju island . Its flower characters were similar to those of white trumpet lilies(Lilium longiflorum Thunb) although its flowering period was later than that of white trumpet lilies and it hadd a wide range of variation among individuals. Six PR-proteinases(II-2, III-1, III-2, IV-1, IV-2 and V) were induced from bulbs by 2.5mM salicylic acid and almost excreted into the intercellular spaces. These PR-proteinases were strongly activated by Ca 2+, , whereas they were strongly inhibited by Cu2+ Co2+ and Fe2+ . Three PR proteinases(II-2, IV-1 and IV-2) were strongly inhibited by 1, 10 -phenanthroline, indicating that these enzymes are metallo-proteinases. Three PR-proteinases(III-1, III-2 and V) had a high sensitivity to PMSF and required β-mercaptoethanol for their activities. These results indicate that these proteinases are cysteine proteinases.

Through total fatty acid analysis by the Gas chromatography, 17 different fatty acids were identified from the sap extracted from bark of Korean Rhus(Anacardiaceae), six species. Linoleic acid(C18 :2) and oleic acid(C18 : 1) were identified as major fatty acids for the species investigated, but except in R. chinensis.In the fatty acid composition for the species, one or two specific-fatty acid was detected, and its could be utilized as a useful taxonomic character.The result of cluster analysis by the 22 reliable characters in the fatty acid composition suggested first group was R.trichocarpa and R. succedanea, the second group, R.verniciflua, R.ambgua and R. sylvestris, but R.chinensis was a greatly independent species. The possible chemotaxonomic application of the fatty acid composition of bark in the genus Rhus was discussed.

Taxonomic studies in external morphology, principal component analysis and cluster analysis were conducted to understand the intraspecific relationship among three habitats (Jirisan, Odaesan and Ulleungdo) of allium voctorialis var. platyphyllum. External morphology such as bulb color, leaf blade length and width, petiole length, total leaf length, peduncle length, perianth lobe length and width, length of anther and filament were useful characters for identification of poplations in three habitats. The results obtained based on the principal component (Pc) analysis of treated 72 OTUs(included outgroup) were divided into three groups by the PC 1 ,2,3 and the sums of contributions for the total variance were 84.1%(PC1 51.0%, PC2 24.9% and PC3 8.2%, respectively). In cluster analysis by the UPGMA and Ward's methods , there was similarities in the compostion of clustered taxa, and only Ulleungdo population was distinctly identified from population of other two habitats.

This study was to establish the floristic composition and vegetion of resources vascular plants of Mt. Kariwang(1,560.6 m ; 128˚33"57〃E, 37˚27"31″N).This work was conducted during the period of 1996 & 1997. Resources vascular plants were composed of 95 families, 329 genera, 485 species, 78 varieties and 14 formae, totaling 577 taxa. Among the investigated resources plants, 26 taxa(4.5%) were Korean endemic species. The forest were comparatively well conservated by Pteridophyta calculation(PTE-Q), 1.3. Useful of source (13.3%) and timber sources(5.05) respectively. The forest vegetation was classified into 1 order, 1 alliances, 1 association and 4 communities.
