        Several analytical measurement techniques have been developed over the years for ammonia (NH3). However, the field monitoring of NH3 still remains a significant challenge owing to the wide range of possible environmental conditions and NH3 concentration. In this regard, it is imperative to ensure the quality control of techniques to measure the NH3 emission levels reliably. A present study was conducted to compare the five analytical methods for the measurement of atmospheric NH3 via validation tests under laboratory and field conditions. The analytical instruments applied in the present study were based on wet chemistry, gas detection tube, electrochemical sensor, photoacoustic spectroscopy, and cavity ring-down spectroscopy. The reproducibility and linearity of all the analyzed methods were observed to be high with the relative standard deviation and coefficient of determination (R2) being 10% and > 0.9, respectively. In the case of wet chemistry and high NH3 concentration, the measured NH3 results were found to be close to the actual standard gas levels. Response times of electrochemical sensor showed faster from the instruments utilized more than one year and the high NH3 concentrations. In the field tests, NH3 concentration showed higher in the manure storage tank compared with the pig-pen. In both cases, the NH3 concentration levels measured by gas detection tube were found to be quite different from that of wet chemistry. It was proposed that such differences in NH3 concentration could arise due to the inherent instrumental characteristics and the variations in air velocity during sampling/measurement. The periodic instrumental maintenance, verification, replicate analyses, and suitable consideration of environmental factors should be considered for a more reliable measurement of NH3 concentration under real field conditions.

        Oct4 and Nanog are transcription factors involved in pluripotency of stem cells. In general, Oct4 is up-regulated by Nanog. In previous results, however, Oct4 was differentially regulated by various doses of Nanog in P19 cells. High dose Nanog down-regulated the Oct4 expression. This negative feedback event was performed by DNMT and HDAC through the inhibitor assays (5-AZA-cytidine and trichostatin A). To identify the precise recruited sites for DNMT and HDAC, ChIP assay was performed in differential doses of Nanog. As a result, HDAC1, HDAC2, DNMT3A and Nanog were recruited to CR2, CR3, CR1, and CR4 upon high dose Nanog, respectively. Next, to investigate the differentiation potency of the cells upon high dose Nanog, RT-PCR with specific markers for three germ layers was performed. However, there was no increase for three germ layers in high dose Nanog treated cells except E-cadherin expression. E-cadherin is a specific marker for epithelial cells. Taken together, high dose Nanog induces Oct4 down-regulation and results in differentiating embryonic carcinoma cells to epithelial cell type.         Phosphoprotein Enriched in Astrocytes (PEA15) is a 15kD-sized intracellular signaling protein, highly expressed in astrocytes and constitutively expressed in peripheral tissues. Recently it was found that PEA15 expression was elevated in patients suffering type 2 diabetes and suggested to be involved in the syndrome of insulin resistance. To investigate the functional role of PEA15 for the control of blood glucose level, we produced a transgenic pig over-expressing mouse PEA15 (mPEA15). As a model animal, pig has many advantages. They have a higher fecundity and a short generation time and are physiologically similar to human. Using the transgenic pig, we carried out a series of experiments to establish a link between PEA15 expression and the insulin resistance. Our results suggested that, compared with control pig, mPEA15 pig has, (1) a higher blood resistin level, (2) a lower cell membrane-embeded GLUT4 level, and (3) a lower glucose clearing ability based on oral glucose tolerance test (OGTT). When our results combined, it can be concluded that mPEA15 over-expressing pig has many symptoms of insulin resistance and these pigs will become a useful disease model to investigate diabetes mellitus in the near future.

        The objective of this study was to determine the potential hazardous effects of sorting process by flowcytometry on the quality of boar spermatozoa by flowcytometer. Freshly collected boar semen was diluted and divided into two groups; control none sorted and sorted. Sperms in sorted group were processed with flowcytometer for cell sorting with 100 uM nozzle under the 20 psi pressure. Measurements on each parameter were made at two time points, 0hr (right after sorting) and 24 hr post sorting. Although there was a tendency of lower viability in sorted group than none sorted control group, the percentage of live cells in control(75.83+-6.92 & 59.53+-10.34) was not significantly different from sorted (59.7+-7.34 & 43.97+-3.76) at both 0 and 24 hr post sorting. However, sorted sperm showed significantly lower mitochondrial function compared to the control at both 0 h (79.37+-3.22 vs. 63.50+-10.05) and 24hr(67.27+-3.22 vs. 46.97+-5.37) time points (p<0.007). Sperm DNA fragmentation rate was significantly lower in control (22.0+-7.04) than that of sorted (32.27+-7.49) at 24 hr time point (p<0.0002). Taken together, these data suggested thatsorting process by flowcytometer may have influenced sperm motility rather than viability. Also high speed sperm sorting by flowcytometer has significant effects on DNA fragmentation on elapsed time after sorting.
