        port is the intersection of highway, railway, waterway and other transportation modes, and is the key to realizing integrated transportation. There are many excellent ports around the Tumen River Region. With the obvious location advantages, Tumen River Region is an important part of Tumen River regional cooperation and development, and is the key to realizing the “borrow port to sea”, which is raised in “China Tumen River regional cooperation and development planning outline -- regard Changjitu area as the development and opening leading area” (referred to as “planning outline”). This paper focus on the main ports in the Tumen River region, taking them as the research object. Furthermore, the paper makes the strategic plan for the port cluster in the Tumen River region as well as puts a collaboration scheme is proposed by analyzing the research reviews of the Tumen River region and the present situations of the main ports.

        Bee pollen have been used for centuries as a kind of traditional medicine and food diets additive due to their nutritional and biological properties. The present study was to evaluate the biological features of ethanol extracts of 18 different bee pollen (EEBP) samples collected from 16 locations of South Korea. Our results indicated most of the EEBP samples showed 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, inhibitory activities against human β-amyloid precursor cleavage enzyme (BACE-1), and antiproliferative activities towards five cancer cell lines. However, these EEBP samples did not present human acetylcholinesterase (AChE) inhibitory activities, and intestinal bacteria growth inhibitory activities.

        Skin is the outermost organ and acts as a barrier between the organism and environment. Skin protects the organism from environmental insults, such as chemicals, pathogens, and UV light. Much of the protective function of skin is dependent on the epidermis, a multi-layered epithelium that is composed of various cell types such as keratinocytes and melanocytes. Keratinocytes produce protective components through a sophisticated differentiation process. Disturbance of keratinocyte differentiation is related to several skin diseases such as psoriasis and atopic dermatitis. In this study, we prepared extract of combined medicinal plants (ECMP) consisting of Taraxacum platycarpum H. Dahlstedt, Heartleaf Houttuynia, Glycyrrhiza uralensis Fischer, and root bark of Ulmus davidiana. We demonstrated that ECMP enhanced keratinocyte differentiation and barrier functionality using an in vitro cell culture system and in vivo animal test. Treatment of cultured keratinocytes with ECMP resulted in induction of keratinocyte differentiation, as evidenced by increased differentiation markers such as involucrin, loricrin, and filaggrin. In line with these results, ECMP decreased proliferation of keratinocytes cultured in vitro. ECMP applied topically to tape-stripped mouse skins accelerated reduction of transepidermal water loss (TEWL), indicating fast recovery of barrier function. Immunohistochemistry showed that ECMP increased the filaggrin level in tape-stripped mouse skins. These results suggest that ECMP may be applicable for keratinocyte differentiation-related skin diseases.

        ORF78 (ac78) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a baculovirus core gene of unknown function. To determine the role of ac78 in baculovirus life cycle, an ac78-deleted mutant AcMNPV, Ac78KO, was constructed. Quantitative PCR analysis revealed that ac78 is a late gene in the viral life cycle. After transfection into Spodoptera frugiperda cells, Ac78KO produced a single-cell infection phenotype indicating that no infectious budded viruses (BVs) were produced. The defection in BV production was also confirmed by both viral titration and Western blot. However, viral DNA replication is unaffected. Analysis of BV and occlusion derived virus (ODV) revealed that AC78 is associated with both forms of the virions and is a structural protein located to viral envelope. Electron microscopy showed that ac78 also plays an important role in embedding of ODV into occlusion body. This study therefore demonstrates that AC78 is a late virion associated protein and is essential for the viral life cycle.

To determine the role of ac11 in baculovirus life cycle, an ac11-knockout mutant AcMNPV, Ac11KO, was constructed. qPCR analysis revealed that ac11 is an early gene in the life cycle. After transfection into Spodoptera frugiperda cells, Ac11KO produced a single cell infection phenotype indicating that no infectious budded viruses (BVs) were produced. The defection in BV production was confirmed by both viral titration and Western blot. However, viral DNA replication is unaffected. Electron microscopy showed that ac11 is required for nucleocapsids envelopment to form ODV and their subsequent embedding into OB. This study therefore demonstrates that ac11 is an early gene which is essential for the viral life cycle.

        The baculovirus expression system is one of the most popular methods used for the production of recombinant proteins but has several complex steps which have proved inherently difficult to meet a multi-parellel process. We have developed a novel recombinant bacmid, bEasyBm that enabling easy and fast generation of pure recombinant virus without any purification step. In the bEasyBm, attR recombination sites were introduced to facilitate the generation of recombinant viral genome by in vitro transposition. Moreover, extracellular RNase gene from bacillus amyloliquefaciens, barnase, was expressed under the control of Cotesia plutellae bracovirus early promoter. Therefore, only when the barnase gene was replaced to gene of interest, the bEasyBm could replicate in host insect cells. When the bEasyBm was transposed with pDualBac-EGFP and pDualBac-LUC respectively, there were no non-recombinant backgrounds were detected from unpurified BmEasy-EGFP or BmEasy-LUC stocks. In addition, the resulting recombinant virus, BmEasy-EGFP, showed comparable level of EGFP expression efficiency with the plaque-purified recombinant virus, BmEGFP, which was constructed using bBmGOZA system. Based on these results, high-throughput condition for generation of multiple recombinant viruses in a time was established.

        With escalating economic growth during the last three decades, flower industry of China, especially cut flower is sharply developed. In this paper a brief review of the cut flower current situation of globe and current status of flower industry of China especially of cut flower in the world is presented. The acreage, yield, potential of cut flower in China along with distribution of major cut flower products and constraint of cut flower also was indicated in this paper was also presented.

        ORF11 (ac11) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a highly conserved baculovirus gene whose homologs are found in all lepidoteran Group I NPV, but its function is unknown so far. To determine the role of ac11 in baculovirus life cycle, ac11 knock-out mutant, Ac11KO, was constructed using the plasmid capture system (PCS). Real-Time PCR analysis showed that ac11 transcript was first detected at 6 h post-infection (p.i.) and accumulated to maximum at 48 h p.i., indicating that ac11 is belong to late gene. When the genomic DNA of Ac11KO was transfected into Sf9 cells, viral replication was restricted to a cell transfected originally. While viral transmission of the Ac11KO was not observed in Sf9 cells, production of budded virus (BV) in Sf9 cells transfected with Ac11KO was observed by transmission electron microscopy (TEM). These results suggest that the ac11 is essential for AcMNPV to produce infective BV.

        Among 154 putative ORFs of Autographa californica multiple nucleopolyhedrovirus (AcMNPV), ac78 and ac79 are highly conserved genes in baculovirus, but their functions in the virus life cycle have been unknown so far. To determine their roles in AcMNPV replication, knockout mutants, ac78KO and ac79KO, were constructed using the plasmid capture system (PCS). Real-Time PCR analysis showed that both of ac78 and ac79 transcripts were first detected at 6 hours post-infection, and accumulated to maximum at 24 hours post-infection, suggesting that both of ac78 and ac79 are belong to late gene. When the genomic DNA of ac78KO was transfected into Sf9 cells, viral replication was restricted to a single cell infection. These results demonstrated that the ac78 play an important role in BV production, and therefore is essential for AcMNPV to mount a successful infection. Whereas Sf9 cells infected with the ac79KO showed normal viral symptoms such as rounding and swelling, OBs were not observed from majority of infected cells. These results suggested that the ac79 might play an important role in OB production.

        Autographa californica nucleopolyhedrovirus (AcMNPV) has a large doublestrand DNA genome of approximately 134 kbp and harbors 156 open reading frames (ORFs). To elucidate DNA replication cascade of AcMNPV, we developed a novel baculovirus genome that can be maintained in Escherichia coli as a plasmid and can infect susceptible lepidopteran insect cells. This genome, named bAc-MK, contains a mini-F replicon and a kanamycin resistance marker. Using a convenient Tn7 transposon-based system, pPCS-S, 55 single ORF-truncated mutants were generated by random insertion into bAc-MK genome. These single ORF-truncated mutants were independently transfected into Sf9 cells, 16 of them were found affecting viral replication since they defected in producing polyhedra. Furthermore, to verify the pathogenicity of the single ORF-truncated mutants, the remaining 39 mutants were subjected to bioassay to Spodoptera exigua 3rd instar larvae. Among them, ac9-, ac49-, ac103- and ac105-knockout mutants showed higher mortality compared to that of bAc-MK. These results suggested that these ORFs could be related to pathogenicity of AcMNPV.

These results suggest that these single ORF-truncated mutants are useful for elucidation of viral replication cascade.

        Suzhou Taohuawu Year Painting is one of the four major Year Paintings in China, which is a symbol of Jiangnan urban Year Painting. During the past years, it experienced fire in Taiping Heavenly Kingdom, Cultural Revolution and the impact when offset printing replaced traditional woodcut Year Painting, covered all over with scars. Trying to start from the history of Year Painting, this article analyzes the difficulties in inheritance and innovation of Taohuawu, interprets the evolution of Taohuawu Year Painting from style, theme, skills, etc. Taohuawu Year Painting is a development hi-story of Year Painting, also a reflection of cultural development in Suzhou.

        As an important carrier of cultural output, film and television can clearly show a country's ideology and regional characteristics. Over the years, south Korean films have been playing an increasingly important role in the world film and television due to their unique cultural background and ideological connotation.Inspired by culture and relying on film as a form of cultural communication, it has exerted a significant influence in the world. Based on the Confucian culture in the application of the south Korean film and television, the research systematically analyzes the theme and core values of Korean film works, and interprets the successful experience of Korean film works under the influence of Confucian culture. The author summarizes the problems in the expression of the core values of society in Chinese films and comes up with straegies to promote the development and dissemination of the film and television industry and the core values of society.

        The Sogdian Merchants were the undertakers of international trade on the Land Silk Road in the Middle Ages, and their business footprints spread all over Eurasia. The ancient trading currency of Sogdian Merchants is a key to study the trading history of the region and the entire Silk Road. Although the economic strength of Sogdiana was strong, the country was very weak and had been controlled by the powerful forces around it. Sogdian Merchants wanted to use large amounts of currency for exchanging activities in countries along the Silk Road, they must imitate the currency of the powerful countries that controlled the Silk Road or even use their currencies directly to meet the needs of long-distance transshipment trade. Most scholars agreed that the Persian Sassanian silver coins used to be the currency of circulation along the Silk Road. Whether the main purpose of the unearthed Byzantine gold coins was to act as circulating currencies on the Silk Road at that time, there is still a great controversy.

        We investigated genetic diversity among and within the populations of cultivated ginseng (Panax ginseng C. A. Meyer ) using SRAP profiles. A total of 24 ginseng plants were sampled from the three populations (two from China, one from Korea). Since all these populations are previously shown closely related to each other assister groups, we used Panax quinquefolium L. and wild ginseng as a reference species, which is not "within the sister group". All individuals from the three populations were screened with a total of 36 primer pairs with 26 primers generated from 328 SRAP bands of DNA gels. The mean gene diversity (HE) was estimated to be 0.057 within populations (range 0.032-0.067), and 0.086 at the species level. The genetic differentiation (Gst=0.31) indicates that genetic variation apportioned 30% among populations and 70% within populations. Generally, the result of this study indicates that ginseng contains high molecular variation in its populations.
