목 적: 조절마비제로 CyclogylⓇ 과 AtropineⓇ 이 사용되고 있으며, 조절마비제를 점안했을 때 결막세포 에 미치는 영향을 알아보고자 하였다. 방 법: 결막세포(clone 1-5c-4)를 배양하여 CyclogylⓇ, cyclopentolate HCl, AtropineⓇ, atropine sulfate를 각각 1%, 0.75%, 0.50%, 0.25% 농도로, 보존제인 benzalkonium chloride(BAC)는 0.01%, 0.0075%, 0.005%, 0.0025% 농도로 15분 처리하여 MTT assay 방법을 사용하여 세포 생존율을 측정하였다. 형태적 변화를 조사하기 위해 hematoxylin-eosin(H-E) 염색 후 광학현미경으로 관찰하고, 유세포분석기를 이용하여 세포자연사를 정도를 측정하였다.
결 과: Cyclopentolate HCl 0.25% 와 atropine sulfate 0.25% 에서만 세포 생존율이 70%를 넘는 것으로 나타났으며, H-E 염색한 결과 CyclogylⓇ 1% 와 AtropineⓇ 1% 에서 세포수가 감소하고 핵의 분절화를 관찰할 수 있었다. CyclogylⓇ 1% 와 AtropineⓇ 1% 에서는 대조군에 비해 A0 수치가 52.92% 와 31.36% 로 세포자연사가 나타났다.
결 론: 조절마비제인 CyclogylⓇ 와 AtropineⓇ 1% 에서는 세포 생존율이 30% 를 넘지 않았으며, 세포자 연사를 일으키는 것으로 나타났다.
Purpose: CyclogylⓇ and AtropineⓇ are in use as cycloplegic agents. This study aimed to investigate the effects of the cycloplegic agents on the conjunctival cell.
Methods: A conjunctival cell line was treated for 15 minutes with CyclogylⓇ, cyclopentolate HCl, AtropineⓇ and atropine sulfate at their concentrations of 1%, 0.75%, 0.50%, and 0.25%, respectively, and the preservative of benzalkonium chloride at the concentrations of 0.01%, 0.0075%, 0.005%, and 0.0025%; and then the cell viability was measured, using the MTT assay. The morphological changes were investigated with an optical microscope after hematoxylin-eosin staining, and apoptosis was measured, using flow cytometry.
Result: It was found that the cell viability exceeded 70% only in 0.25% cyclopentolate HCl and 0.25% atropine sulfate. And as a result of the H-E staining, the cell count decreased and fragmentation of neuclei was observed in 1% CyclogylⓇ and 1% AtropineⓇ. In 1% CyclogylⓇ 1% and 1% AtropineⓇ, the A0 level was found to be 52.92% and 31.36%, compared with the contrast group, and apoptosis occurred.
Conclusion: It was found that in the cycloplegic agents of 1% CyclogylⓇ and 1% AtropineⓇ, the cell viability does not exceed 30% and apoptosis occurs.
