Amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers were utilized for evaluation of genetic diversity of 50 strawberry cultivars. Twenty-seven selective AFLP primer pairs generated a total of 407 polymorphic amplification products and 13 SSR primers produced 42 alleles. From UPGMA (unweighted pair-group method arithmetic average) cluster analysis by using 449 polymorphic bands, the strawberry cultivars were divided into six clusters by similarity index of 0.654. Most of Korean and Japanese cultivars were included in the sixth (VI) cluster. AC1466 and Harunoka were not clustered into any group, and the principal component analysis confirmed the pattern of the cluster analysis for those two varieties. Similarity values among the tested strawberry cultivars ranged from 0.531 to 0.951, and the average similarity value L was 0.658. This indicated that the 50 strawberry cultivars were closely related even though they developed from independent breeding programs, respectively. It was confirmed that there were a few genetic variations among the strawberry cultivars used for this study.
