        The aim of this study was to select the abiotic tolerant sorghum mutants using chlorophyll a transient OJIP analysis of PSⅠ and PSⅡ so called Kautsky’s effect within 1 second. It was clearly identified that wwt-and drought tolerant sorghum mutants could be classified by wet factor index(WFI). On the basis of WFI, wet tolerant sorghum matants were classified as follows; Ⅰ group, MUT534 bmr/new, MUT525 bmr; Ⅱ group, M2P1207 bmr, 25M2-0404 bmr, MUT371 bmr24, unknown bmr22, 10M2-0775 bmr, MUT135 bmr23; Ⅲ group, M2P0411 bmr, MUT641 bmr, M2P1064 bmr36, MUT855 bmr, 25M2-0137 bmr/new, MUT436 bmr, M2P0929 bmr, 25M2-0026 bmr, 10M2-0387 bmr, 25M2-0173 bmr/new; Ⅳ group, 25M2-0698 bmr. In conclusion, for the selection of wet tolerance, four photochemical parameters such as Electron transport flux until PSI acceptors per PSII(RE1o/RC), Performance index for energy conservation from photons absorbed by PSII antenna, until the reduction of PSI acceptors(PI_total ABS), Driving force on absorption basis(DF_total ABS) and Electron transport flux from QA to QB per PSII(ETo/RC) were important photochemical parameters deduced from maximum quantum yield and electron transport efficiency.

        Development of transgenic plant with desirable traits to cultivated plant is one of the important procedures in plant molecular breeding. However, applicable assessment of transgenic plant in laboratorial scale is not much except cultivating transgenic plant for a whole life in field condition. Here, we analyzed chlorophyll fluorescence in three transgenic soybean lines with AtMYB44 transcription factor for assessment of photosynthetic activity under abiotic stresses such as drought. Soybean varieties used in this study were ‘Bert’ and ‘Bert’ derived three transgenic soybeans, ‘AtMYB44 CM35101’, ‘AtMYB44 CM2471’, and ‘AtMYB44 CM4481’. Analyzed five different chlorophyll fluorescence variables are maximum PSII quantum yield (QY_max), steady state PSII quantum yield (QY_Lss), steady state non-photochemical quenching (NPQ_Lss), coefficient of photochemical quenching in steady-state (Qp_Lss), and fluorescence declineratio in steady-state (Rfd_Lss). To determine main chlorophyll fluorescence variable affected by abiotic stress, principal component analysis (PCA) was conducted with five chlorophyll fluorescence variables measured from four varieties. QY_Lss and NPQ_Lss were main chlorophyll fluorescence variables to evaluate abiotic stress, particularly in drought stress. In comparison with transgenic soybean lines based on chlorophyll fluorescence variables, ‘AtMYB44 CM2471’ and ‘AtMYB44 CM4481’ are more tolerant to drought than the others. Interestingly, three transgenic soybean lines which have a same AtMYB44 gene with different regions of chromosome revealed the quite different responses of chlorophyll fluorescence to drought treatment.

        Seashore Paspalum (Paspalum vaginatum Swartz) is a warm season grass and indigenous to tropical and subtropical regions of coastal areas worldwide. The species is used as feed for cattle and horses and has been very successful for golf courses worldwide. One of the most outstanding characteristics of seashore paspalum is its tolerance to saline soils compared to other warm season turfgrasses. The development of new seashore paspalum cultivars with improved traits could be facilitated through the application of biotechnological strategies. The purpose of this study was to product for herbicide resistant seashore paspalum using Arobacterium-mediated transformation and this study is the first report on transformation and herbicideresistant transgenic plants in seashore paspalum. Embryogenic calli were induced from the seeded variety of pseashore paspalum. Embryogenic calli were transformed with Agrobacterium tumefaciens strain EHA105 carrying the binary vector pCAMBIA3301 with two genes encoding gusA and bar. Transformed calli and plants were selected on medium containing 3 mg/l PPT. PCR detected the presence of the gusA and bar gene, indicating both genes are integrated into the genome of seashore paspalum. A chlorophenol red assay was used to confirm that the bar gene was expressed. By application of herbicide BASTA, the herbicide resistance in the transgenic seashore paspalum plants was confirmed.

        The average ratio of the daily UV-B to total solar (75) irradiance at Busan (35.23˚N, 129.07˚E) in Korea is found as 0.11%. There is also a high exponential relationship between hourly UV-B and total solar irradiance: UV-B=exp (a× (75-b))(R2=0.9 0.93). The daily variation of total ozone is compared with the UV-B irradiance at Pohang (36.03˚N, 129.40˚E) in Korea using the Total Ozone Mapping Spectrometer (TOMS) data during the period of May to July in 2005. The total ozone (TO) has been maintained to a decreasing trend since 1979, which leading to a negative correlation with the ground-level UV-B irradiance doting the given period of cloudless day: UV-B=239.23-0.056 TO (R2=0.5 0.52). The statistical predictions of daily total ozone are analyzed by using the data of the Brewer spectrophotometer and TOMS in East Asia including the Korean peninsula. The long-term monthly averages of total ozone using the multiplicative seasonal AutoRegressive Integrated Moving Average (ARIMA) model are used to predict the hourly mean UV-B irradiance by interpolating the daily mean total ozone far the predicting period. We also can predict the next day's total ozone by using regression models based on the present day's total ozone by TOMS and the next day's predicted maximum air temperature by the Meteorological Mesoscale Model 5 (MM5). These predicted and observed total ozone amounts are used to input data of the parameterization model (PM) of hourly UV-B irradiance. The PM of UV-B irradiance is based on the main parameters such as cloudiness, solar zenith angle, total ozone, opacity of aerosols, altitude, and surface albedo. The input data for the model requires daily total ozone, hourly amount and type of cloud, visibility and air pressure. To simplify cloud effects in the model, the constant cloud transmittance are used. For example, the correlation coefficient of the PM using these cloud transmissivities is shown high in more than 0.91 for cloudy days in Busan, and the relative mean bias error (RMBE) and the relative root mean square error (RRMSE) are less than 21% and 27%, respectively. In this study, the daily variations of calculated and predicted UV-B irradiance are presented in high correlation coefficients of more than 0.86 at each monitoring site of the Korean peninsula as well as East Asia. The RMBE is within 10% of the mean measured hourly irradiance, and the RRMSE is within 15% for hourly irradiance, respectively. Although errors are present in cloud amounts and total ozone, the results are still acceptable.
