This study was conducted to determine the effect of horse manure compost application on Italian ryegrass (IRG) yield and volcanic ash soil characteristics. Because the number of horses in Korea is growing, the amount of horse manure is growing. Jeju island, where about 55 % of the horses live, is composed of volcanic ash soil. This study was conducted for about 7 months. Sowing was conducted on October 2019. Harvesting was conducted at heading stage(2020.5.). Five treatments were established based on the horse manure compost composition. These were 100 % chemical fertilizer (CF), the combination of 50 % horse manure and 50 % chemical fertilizer (combination), horse manure with 50 % nitrogen (HM 50 %), horse manure with 100 % nitrogen (HM 100 %), and horse manure with 150 % nitrogen (HM 150 %). The plant height and dry matter yield were investigated to determine the forage yield and the soil characteristics of pH, total nitrogen, available phosphate, and organic matter were analyzed. The plant heights in the CF, combination, and HM 150 % treatments were 147.8 cm, 144.3 cm, and 147.1 cm respectively (Table 2). Dry matter yield in the CF treatment was about 23,807 kg/ha, which tended to be the highest dry matter yield. HM 150 % and the combination treatment were about 18,804 and 18,455 kg/ha, respectively, which tended to be the highest dry matter yield of the treatments amended with horse manure compost. The dry matter yield of the HM 100 % and HM 50 % treatments was about 15,801 kg/ha and 14,446 kg/ha, respectively (Table 2). The pH of the surface soil tended to increase after the experiment. The soil pH of the HM 150 % treatment was significantly higher than the soil pH of the other treatments. The pH was affected by the amount of horse manure compost, with a pH of 8.1. The available phosphate in the treatments in which horse manure compost was added was higher than the available phosphate in the CF treatment. And the available phosphate in the HM 150 % treatment was significantly higher than the available phosphate in the other treatments (p < 0.05)(Table 3). These results suggest that 50 % horse manure should be applied to IRG as the basal fertilizer and the remaining 50 % should be chemical fertilizer as the top fertilizer. This can provide the proper IRG dry matter yield with less effect on volcanic ash soil.
This study was conducted to figure out the productivity of Italian ryegrass(IRG) and leaching water characteristics based on horse manure compost level in Jeju. This study was conducted for about six months. Six treatments were established : non-fertilizer(NF), chemical fertilizer 100%(CF), horse manure compost 50% and chemical fertilizer 50% combination(Combination), horse manure compost with 50% of nitrogen (50%), 100% of nitrogen(100%), 150% of nitrogen(150%). The highest amount of dry matter yield of IRG was revealed in CF(11,965± 564 kg/ha), and both 150% and Combination were second(p<0.05). Nitrate leaching tended to increase until the third analysis and then decreased. There were not significantly differences among mean nitrate concentrations. The findings of the study suggest that horse manure compost with 50% of nitrogen be applied for IRG as basal fertilization and then 50% of chemical fertilizer be applied as top fertilization.
Prolactin is an anterior pituitary hormone involved in various physiological phenomenon including reproduction. The prolactin receptor (PRLR) is detected in diverse tissues such as brain, ovary, placenta and uterus in several mammalian species. A total of 227 pigs [Korean native pigs (KNP) 27; Landrace pigs 29; Korean native pigs x Landrace F1 91; Nanchuckmacdon pigs 80] were used to investigate the allele frequency difference of the prolactin receptor (PRLR) gene among the four pig lines. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) with Alu I restriction enzyme was used to determine the genotypes of PRLR. Frequencies of PRLR alleles among the four different pig lines were not significantly different (Chi-square=3.94, DF=3, P=0.27). A total of 40 Nanchuckmacdon pigs were used to investigate the effect of the prolactin receptor (PRLR) gene on total number of piglets born (TNB), number of piglets of alive (NBA) using general linear model implemented in MINITAB software. For TNB, the AB genotype had higher genotypic value (10.61) than the values of AA (9.83) and BB (10.30). Likewise, the AB genotype had higher genotypic value (8.96) than the values of AA (8.18) and BB (8.90) for NBA. However, these associations of the PRLR gene with TNB and NBA were not statistically significant. In conclusion, it is necessary to increase the sample size for investigating the effect of the PRLR gene on TNB and NBA in pigs.
Blood types in pigs are divided into two types, A and O type. It is important to select and breed O-type pigs that have no hematologic rejection, because the blood type of the donor is an important factor causing immune rejection in xenotransplantation. Therefore, the gene that determines the blood type is GGTA1(glycoprotein, alpha-1,3-galactosyltransferase 1), which generally belongs to the ABO blood group. This study was carried out to develop a simple and accurate method by analyzing the structure of GGTA1 gene which determines blood type. A primer was designed to allow easy identification of the blood type of the target in the first and second deletion regions of intron 7 to distinguish between the A and O genotypes. That is, for the purpose of identifying the blood type using length difference after the PCR, the forward and reverse primers were designed in the highly conservative region. As a result, a pair of primers were prepared and PCR amplification was performed to distinguish three types of genotypes, AA, AO, and OO, using the length difference by electrophoresis. Using the above primers, the parents and their offspring were compared with each other to confirm the correct genetic pattern. And, in four pig breeds, the genes were amplified and the genotype could be correctly identified. In this study, we could diagnose the blood type of AA, AO, OO genotype of pigs by using primer of INDEL region. Especially, since it is possible to diagnose the genotype by the length difference, it is possible to diagnose it quickly and accurately from the gene amplification to the genotype reading
This study was conducted to determine the effect of pentoxifylline levels on sperm motility, survival rate, sperm membrane integrity of frozen semen and fresh-extended equine semen in Jeju cross-bred horses. As a result of sperm characteristic comparison depending on pentoxifylline levles at 30 minutes post-thaw, the progressive motilities were 53.25±2.87 (4mM pentoxifylline) and 50.28±2.14 (8mM pentoxifylline) and significantly higher compared to the control group(40.09±5.15) and other treatment group (16mM pentoxifylline, 41.27± 2.82). The progressive fast motility were 22.44±1.62 (4mM pentoxifylline,) and 22.74±3.07 (8mM pentoxifylline) and significantly higher compared to the control group (13.47±1.48) and other treatment group (16mM pentoxifylline, 14.66±3.68) (p<0.05). As a result of sperm characteristic comparison depending on pentoxifylline levles at 30 minutes post-thaw were 68.96±1.64 (4mM pentoxifylline) and 67.90±6.72 (8mM pentoxifylline) and significantly higher compared to the control group (53.48±4.84) and other treatment group (16mM pentoxifylline, 58.14±2.65) (p<0.05). In conclusion, these results suggest that treatment groups with 4mM and 8mM pentoxifylline were higher compare to equine seperm mobility and the control group and treatment groups with more than 16mM pentoxifylline has a negative effect on sperm characteristics. After thawing, the total motility in post-thawed equine sperm has increased by 10 percent for 1 hour. these results suggest that pentoxifylline contributes to the improvement of the equine sperm motility and characteristics in post-thawed semen.
This study was conducted to investigate the ovarian cycle changes of the mare according to the season. Twenty four Jeju crossbred horses(Thoroughbred x Jeju horse) raised in Subtropical Livestock Research Institute, National Institute of Animal Science, RDA were used to identify follicles and corpus luteum with ultrasonography once a week(May 2016~June 2017). Blood samples of experimental horses were collected twice a week for analysis of P4 hormone levels. The mares were considered to have resumed ovarian cyclicity on the day of ovulation if they followed by regular ovarian cycles. Only 13 cases(61.9%) of the total 21cases showed normal ovarian cycle, and 8 cases (38.1%) showed delayed ovarian cycle. Three cases(16.7%) in October, 5 cases(27.8%) in November and 5 cases(27.8%) in December(27.8%) ceased the heat and the remaining 5 cases(27.8%) showed that the estrus was maintained in winter. Horses that stopped estrus ceased the heat until March of next year, and 27.8% were continued the heat during non-breeding season. Eleven cases(61.1%) of 18 cases in April and 2 cases(11.1%) of 18 cases in May returned the estrus.
This study was conducted to investigate the breeding status of farms to improve the production efficiency of Hanwoo calf. The study was conducted on 45 farms divided into two groups. This study was conducted to investigate the breeding size and breeding area of Hanwoo cows. The average age at first delivery of Hanwoo was 28.7 months. The number of artificial insemination per pregnancy was 1.45±0.32, and the number of artificial insemination days after birth was 119.8 days. Conception rates were 75.2±16.93% for small farms and 70.6±17.46% for medium sized farms and 71.4±11.03% for large farms. When we looked at farming methods, ‘the farmers using estrus observation aids’ had 10.42% higher calf production rate than the ‘unused farmers’. The farms vaccinated with IBR and BVDV for breeding cattle showed a 4.41% decrease in abortion, stillbirth and mortality. According to farming conditions, conception rate and delivery rate improved by 3.47% and 18.29%, respectively, when grazing and exercising were performed. Observation, immunization and grazing were found to be important indicators for improving calf production efficiency in Hanwoo farm. This study can be used as a research data to improve the reproductive rate of farmhouse sites through the survey on the breeding status of Hanwoo farmers.
Commercial applications of OPU/IVP were to produce embryos and calves from high genetic cows.The aim of this present study was to compare the number of recovered oocytes and cultured in vitro produced embryos from Ovum Pick-up (OPU). OPU derived embryo production was carried out of oocytes by ultrasonographic guided follicular aspiration and then produced in vitro produced blastocysts by IVP culture system. In result, the rate of recovered oocytes was obtained 612 (57.2%) and 451(73.7) G1+G2 grade oocytes. No difference of recovered rate (51.1~62.1%) was seen in six donor. The rate of cleavage and blastocyst development were obtained 320 (70.9%) and 78 (24.4%) that was 3.3±0.4 cleaved embryo and 0.9±0.2 blastocysts per session. Cleavage rate of OPU oocytes in No. 6 donor was 90.6%, significantly (P<0.05) higher than that in the other donors, However, blastocysts was similar (25.8~30.0%). In conclusion, limited numbers of OPU oocytes had competent development when cultured in SOF culture medium
The present study was conducted to investigate the effect of BHT supplementation on sperm motility, viability, acrosomal integrity and plasma membrane integrity after frozen-thawing. One ejaculate was collected from one fertile Hanwoo bull by using artificial vagina at Hanwoo Research Institute. The ejaculate was transferred to laboratory immediately and diluted with pre-warmed semen extender (Optixcell, France) (1:1). Sperm dilutions were extended to a final concentration of 40 x 106 sperm/ml, and divided into 5 groups according to BHT concentration (0, 0.5, 1.0, 2.0 and 4.0 mM) and cryopreserved in LN2 tank until evaluation. Frozen-thawed semen was transferred to 1.5 ml tube and incubated for 0, 2 and 4h. Sperm motility and motility parameters (total motility, VSL with 25μm≥, VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were evaluated by sperm class analysis (SCA, IVOS, Spain). There were not significant effects of BHT supplementation (0, 0.5, 1.0 and 2.0 mM) on total motile and VSL with 25μm≥ at 0, 2 and 4h. However, 4.0 mM of BHT supplementation showed negative effect on total motile (26.3%), VSL with 25μm≥ (1.3%) at 0 h (p<0.001). The viability and acrosomal integrity of spermatozoa were evaluated by Trypanblue/Giemsa staining method and divided into 4 groups; live and intact acrosome (LIA), live and damaged acrosome (LDA), dead intact acrosome integrity (DIA), dead damaged acrosome (DDA). There were no significant differences of LIA, LDA, DIA and DDA on various BHT concentrations at 0 and 2 h. However, 4.0 mM BHT supplementation showed decreased LIA compared with 0, 0.5, 1.0 and 2.0 mM BHT at 4 h (34.6, 37.1, 43.6, 45.4 and 14.7% vs. 0, 0.5, 1.0, 2.0 and 4.0 mM, irrespectively; p<0.01). Addition of 4.0 mM of BHT showed negative effect on plasma membrane integrity compared with that of 0, 0.5, 1.0 and 2.0 BHT at 2 h (71.9, 64.2, 64.6, 67.5 and 31.7 % vs. 0, 0.5, 1.0, 2.0 and 4.0 mM, irrespectively; p<0.05). In conclusion, various BHT concentrations on optixcell extender showed no improvement on sperm motility, viability and plasma membrane integrity.
The present study was conducted to investigate the effect of different heights from liquid nitrogen (LN2) vapor on sperm motility and morphology after frozen-thawing. Two ejaculates were collected from 2 fertile Hanwoo bulls (A and B) by using artificial vagina at Hanwoo Research Institute. After collection, ejaculates were transferred to laboratory immediately and diluted with semen extender (Optixcell, France). Sperm dilutions were extended to a final concentration of 40 x 106 sperm/ml, and cooled at 4°C for 4 h and loaded to 0.5 ml straws. The straws were divided into 2 groups. Straws were placed in 3 or 9 cm of LN2 vapor for 14 min and then plunged into LN2 tank and cryopreserved until evaluation. Sperm motility and motility parameters (total motility, VSL with 25μm≥, VCL, VSL, VAP, LIN, STR, WOB, ALH and BCF) were evaluated by sperm class analysis (SCA, IVOS, Spain) after frozen-thawed. In bull A, 3cm group showed higher percentages of total motility, VSL with 25μm and VAP compared those with 9cm group (98.0 vs. 93.4%, 62.4 vs. 54.0% and 98.6 vs. 93.2%, 3 vs. 9 cm, irrespectively; p<0.001). In bull B, frozen-thawed sperm of 3cm group showed higher percentages of VSL with 25μm, VCL, VSL, VAP and BCF compared with those of 9cm group (43.5 vs. 26.0%, 123.8 vs. 111.6 μm, 62.9 vs. 57.3 μm and 81.5 vs. 72.5 μm; 3 vs. 9 cm, irrespectively; p<0.001). The viability and acrosomal integrity of spermatozoa were evaluated by Trypanblue/Giemsa staining method divided into 4 groups; live and intact acrosome (LIA), live and damaged acrosome (LDA), dead intact acrosome integrity (DIA), dead damaged acrosome (DDA). In bull A, frozen-thawed sperm of 3 and 9cm groups showed no significant difference in LIA, LDA, DIA and DDA. In bull B, 3 cm group showed higher LIA and lower DIA compared with those of 9 cm group (73.2 vs. 23.7% and 23.7 vs. 32.2%, 3 vs. 9 cm, irrespectively; p<0.001). We suspected that 3 cm vapor on LN2 vapor might be affected positively spermatozoa viability and acrosomal integrity compared with 9 cm group. In conclusion, semen freezing procedure in the present study will improve sperm quality after frozen thawing.
The study aims to assess viability, acrosomal menbrane, integrity and mitochondria membrane potential of sperm separated using a percoll density gradient(45% and 90%) and swim-up methods using Hanwoo epididymal sperm frozen semen. Briefy, motile sperm separated using a percoll gradient and swim-up. 25 μl of sperm dilution from droplets were transferred to 1.5 mL tube and incubated with fluorescent probes at 39°C in dark as follows. After incubation, 75 μl of 5,5',6,6'-tetrachloro-1,1',3,3'- -JC-1; Mitochondrial Membrane Potential Detection kit, Cell Technology Inc., USA) working solution was mixed with sperm dilution and incubated for 30 min. One μl of Hoechst 33342 (H1339; Molecular Probe, Eugene, OR, USA) stock solution was mixed with sperm dilution and incubated for 10 min. And then, 0.5 μl of propidium iodide (PI) stock solution and 0.5 μl of fluorescein peanut agglutinin FITC conjugate (FITC-PNA; Vector Laboratories, FL-1071) were mixed with sperm dilution and incubated for 8 min. After mixing with fluorescent probes and sperm dilution, 5 μl of stained sperm dilution was mounted on a slide glass and covered with cover glass. More than 200 sperms in a slide glass were counted with × 400 magnification by fluorescent microscope (Eclipse Ci_L, Nikon, JAPAN) and evaluated viability, acrosomal membrane integrity, and mitochondrial membrane potential of sperm with triple band filter (DAPI/FITC/TRITC; Nikon, Tokyo, Japan). Live sperm were stained with Hoechst 33342(blue) and dead sperm were stained with PI (red). Damaged acrosomal membrane of sperm was stained with FITC-PNA (green) and intact acrosomal membrane of sperm was not stained. Both of sperm swim-up method with or without BSA separated to Live intact mitochondria (15.39±4.31 vs, 12.58±3.74, and) without significant difference. and percoll density gradient method also similar (7.29±6.54), swim-up method of sperm preparation appeared to be more efficacious in percentage recovery of motile sperm concentration compared to Density gradient method.
Sperm recovery from epididymis in animals considered as important tools to preserve high-value or endangered species. However, there are no appropriate castrating indicators of timing for recovery of sperm which can be available to artificial reproduction technologies such as artificial insemination (AI) and in vitro fertilization (IVF), particularly in young Hanwoo bull. Therefore, this study aimed to investigate semen volume, morphology and motility of sperm in epididymis of young Hanwoo bulls at 8, 13, 14, and 15 months of age. About 2 cm of the epididymal tail only was cut and minced using blades. Minced epididymal tail tissues were mixed with semen extender (OptixCell, France, IMV technologies) and sperm were recovered with a cell strainer (100 μm nylon mesh). The number of sperm at 8 months of age was lower than that at 13, 14, 15 months of age in bulls after collection (33.6±27.2 vs. 352.4±39.2, 320.4±113.6 and 422.8±252.4×107cells respectively; P<0.05). After the frozen-thawed sperm the the percentage of abnormal head, tail and dead damaged acrosome did not differ between the ages 13, 14 and 15 months of age in bulls (P>0.05), however, the dead sperm with intact acrosome (DIA), the numbers showed that more than 15 months in 8, 13, 14 months (8.7±4.1 vs. 47.3±12.2, 34.8±14.0, 28.8±8.5, P<0.05). In addition, frozen-thawed sperm at 8 months of age showed low total motile sperm compared to those at 13, 14 and 15 months of age (26.4±8.2 vs. 45.7±29.5, 62.3±41.0, 70.4±15.9%, respectively; P<0.05). In conclusion, sperm derived from epididymal tail at 8 months of age in Hanwoo bulls showed high abnormal morphology and poor motility, which is not adequate for artificial insemination(AI) and in vitro fertilization(IVF). On the other hand, sperm derived from epididymal tail at 13, 14, 15 months of age in bull showed high normal morphology and motility, which may be available for AI and IVF. Epididymal sperm collected from bulls over 13 months is needed for further study whether to use the actual in vitro fertilization.
The study aims to assess the embryo development and survivability of bovine embryos cultured in vitro by addition of cysteine. The rates of metaphase II formation are not significantly different among the three groups(73.8% for TCM199, 76.9% for TCM199 with 0.3mM cysteine and 83.8% for TCM199 with 0.5mM cysteine). No differences on cleavage rate(70.6~74.6%) was observed among three culture medium(70.6% for TCM199, 71.3% for CR1aa, and 74.6% for SOF) with 0.5mM cysteine. However, significantly(P<0.05) higher development rate was obtained in the blastocyst stage by adding 0.5mM cysteine in SOF medium(35.6%) than in TCM199(27.6%) or CR1aa(26.6%). No significant differences in the cleavage rates were observed among the three culture. After freezing the blastocysts cultured with 0.5M cysteine, the re-expansion rates ranged from 61.3% to 86.4% among groups, and hatching rates are from 26.3% to 46.9% among groups. The rates of re-expansion and hatching are significantly(P<0.05) higher in SOF medium(86.4% and 46.9%, respectively) than those in TCM199(61.3% and 26.3%) and CR1aa medium(87.1 and 44.4%). After thawing, the blastocyst re-expansion rate become significantly(P<0.05) higher in in vivo (87.1%) and in vitro (70.3%) embryos. In conclusion, our results demonstrate that supplementation of IVM and IVC media with 0.5mM cysteine improved the quality of in vitro production of embryo and post-thawed embryo. Future studies comparing these culture systems in well-designed trials should be performed.
The recovery of epididymal sperm in animals is considered as one of the important tools to preserve high value or endangered species. However, there are no appropriate castrating indicators such as months of age in bull, sperm morphology, and motility, particularly in young Korean native bull (Hanwoo). Therefore, this study aimed to investigate sperm number, morphology, and motility of sperm in the epididymis tail of young Hanwoo bulls at 8 and 15 months of age. After castration, epididymal tails were collected and minced with blades to recover sperm. In experiments 1 and 2, sperm number, morphology, and motility were examined. Total number of sperm and percentage of normal sperm from bulls at 8 months of age was lower than that of bulls at 15 months of age after collection (P<0.05). Percentage of abnormal head, tail, proximal cytoplasmic droplet, dead and damaged acrosome of sperm from bulls at 8 months of age were higher than those of bulls at 15 months of age (P<0.05). In experiment 3, sperm motility from bulls at 8 and 15 months of age were examined before freezing and after thawing. Frozen-thawed sperm at 8 months of age showed low total motility and motile sperm with ≥ 25 μm/sec compared to those at 15 months of age and commercially-used sperm (P<0.05). In conclusion, sperm derived from the epididymal tail of bulls at 8 months of age showed high abnormal morphology and poor motility, which are not adequate for AI and IVF. On the other hand, sperm derived from the epididymal tail of bulls at 15 months of age showed high normal morphology and motility.
The aim of the study was to investigate the ability of sperm derived from the epididymis in regard to sperm motility, sperm penetration to oocyte and subsequent development of the embryo. Frozen-thawed sperm from epididymis showed similar percentage of motile sperm (VSL ≥ 25 μm/sec) as compared to that of commercial sperm (control). Sperm penetration of frozen-thawed epididymal and commercial sperm was not significantly different. Moreover, cleavage and blastocyst rates were similar in both epididymal and control. Sperm derived from the epididymis also showed fertilizability and subsequent embryonic development
The objective of this study was to investigate the interrelationship of weight and hormonal changes through the administration of vitamin A to first time pregnant heifers. A total of 28 Korean heifers was used for this study. The heifers were divided into two groups - with vitamin A (n=14) and without vitamin A (control) (n=14) in the feed. Body weight increased in vitamin A treated heifers starting 9 months until 15 months. After pregnancy, vitamin A treated heifers were found to maintain higher body weights than the control group. Pre-pregnancy and post pregnancy progesterone levels were not different between the two groups. Serum estradiol levels of heifers at different growth stages showed relatively higher E2 levels than the control. Also, the control during pregnancy may show higher serum E2 levels than the vitamin A treated heifers. The growth phase serum estradiol levels in heifers may be relatively higher than the control. During pregnancy it showed a similar trend. Serum levels of vitamin A treated heifers did not differ from pregnant heifers at 5 months of age. However, after 5 months from conception until 8 months of treatment it showed a high level. Serum cholesterol in pregnant cows was higher in the control group than the treatment from beginning until the end of pregnancy. This is considered to be related to fetus development during pregnancy, as well as the mother's health.
The present study was performed on farm animals to test the effectiveness of progesterone-releasing intravaginal device (Cue-Mate® 1.56 g) and injection of prostaglandin F2α (PGF2α) for synchronization estrus in Hanwoo cattle. The cattle were at random stage of the estrus cycle. The cows were artificially inseminated at day 7 after Cue-Mate withdrawal, using commercial semen from Korean native bulls. There was a season effect on the estrus synchronization rate. It was higher in spring (94.3%) followed by winter (93.3%), autumn (90.4%) and summer (67.2%). In summary, The results of this study revealed that season has influences on estrus behavior of cattle with no significant effect on pregnancy rate. In summary, we suggest summer reproductive management to alleviate the effects of heat stress. It should be based on intensive cooling combined with hormonal treatment. Given that different subgroups of cows benefit differently from the treatments, selective hormonal administration should be considered.
In order to investigate the effects of antioxidants on the culture of mouse preantral follicles in vitro, we examined the effects of taurine, glutathione and catalase on their growth and maturation. Addition of taurine was not effective on the survival of preantral follicles. However, metaphase II rates of oocytes within preantral follicles were signifi-cantly higher in 1 mM treated group than in control and 10 mM treated group (p<0.05). Glutathione did not improved the rates of survival and metaphase II oocytes. However, metaphase II rates of oocytes progressively decreased with increasing glutathione concentration. Catalase also showed that the rates of survival and metaphase II oocytes pro-gressively decreased with increasing concentration. Especially, all of preantral follicles cultured in medium containing 100 IU/ml catalase were degenerated. These results suggest that low concentraion of taurine, as an antioxidant, have positive effect on the culture of mouse preantral follicles in vitro.
