        In contrast to conventional silk fibroin, spider silk's potential as a scaffold material for tissue engineering is examined in this work. The remarkable qualities of spider silk are being researched for use in making films for tissue regeneration. In comparison to silk fibroin films, the study's analysis of orb-web spider Trichonephila clavata films highlights their improved cell adhesion and nanofibrous network structure. Tests for solubility substantiate the durability of spider silk films, while in vitro investigations demonstrate low cytotoxicity and enhance cellular viability. The conclusion highlights the exceptional properties of spider silk, which make it a viable option for tissue engineering applications and a step forward for in vitro cell culture and regenerative bioengineering.

        Bombyx mandarina (Lepidoptera: Bombycidae), the presumed ancestor of the domesticated silkworm B. mori, has long been a subject of study to illustrate the geographic relationships in connection with origin of B. mori. We sequenced 100 mitochondrial genomes of B. mandarina collected from South Korea and Japan and these were combined with public data. Phylogenetic and population genetic analyses showed that all individuals collected in South Korea, except one individual from Inje, formed a strong group together with northern China and some individuals of southern China. This group was placed as the sister group to B. mori strains, suggesting that this group could have been served as an immediate progenitor for B. mori. For further robust inference more analysis is underway.

        Bombyx mandarina (Lepidoptera: Bombycidae), the presumed ancestor of B. mori, has long been a subject of study to illustrate the geographic relationships in connection with origin of B. mori. We report 97 mitochondrial genome (mitogenome) sequences of B. mandarina collected from Korea and Japan. Phylogenetic and population genetic analyses showed that all individuals of B. mandarina collected in Korean localities formed a strong group together with all individuals originated from northern China (mainly north of the Qinling-Huaihe line) and some of southern China. This group was placed as the sister group to B. mori strians suggesting that this group had been served as an immediate progenitor for B. mori.

        Bombyx mandarina (Lepidoptera: Bombycidae) is generally regarded as the ancestor of the domesticated B. mori. Recently, over 40 mitochondrial genomes (mitogenomes) mainly from B. mori strains preserved in China and wild individuals of B. mandarina were sequenced to verify the progenitor of B. mori. At this point, we also were curious about the origin and the relationships of Korean silkworms to foreign B. mandarina and B. mori. As a first step, we sequenced the complete mitogenome of the B. mandarina collected in Korea and compared it to pre-exsiting data (37 strains of B. mori and 14 individuals of B. mandarina). The complete mitogenome of B. mandarina was 15,694-bp long, consisted of 13 protein-coding genes, two rRNA genes, 22 tRNA genes and one non-coding region. The 494-bp long A+T-rich region possessed the highest A/T content (95.3%) than any other region of the genome. Overall, the general mitogenome characteristics of the genus Bombyx species have an identical gene arrangement, similar A/ T content (average 82.3%) and so on. Phylogenetic analysis, however, showed that B. mori and B. mandarina formed a distant group each with the highest nodal support. For more findings of mitogenome characteristics of Bombyx including the Korean B. mandarina and those preserved in Korea more mitogenomes, particularly from Korea, might be needed.

        The Japanese oak silkmoth, Antheraea yamamai Guérin-Méneville 1861 (Lepidoptera: Saturniidae), is one of the important natural resources possessing industrial value for silk fiber production. In this study, ten microsatellite markers and two mitochondrial DNA (mtDNA) gene sequences (COI and ND4) were used to investigate the genetic variation and geographic structure of A. yamamai populations in South Korea. Two mtDNA gene sequences revealed very low total genetic variation and resultant low geographic variation, validating to use further variable molecular markers. Population-based FIS, FST, RST, and global Mantel test consistently support that A. yamamai populations are overall well interconnected with a relatively high gene flow. Nevertheless, STRUCTURE analysis using microsatellite data and mtDNA sequences coincidently indicate the presence of two genetic pools in many populations.

        The differences in adult lifetime among various silkworm strains has been suggested that the adult lifetime may be genetically controlled. In this experiment, using J037 and Daizo strains we investigated genetic factors related to the adult lifetime of silkworm. We constructed the full-length cDNA library from the adult male of the J037 strain. A total of 2,688 clones were randomly selected, and we performed a differential display hybridization with cDNA probes generated from J037 and Daizo adult males. In conclusion, 193 clones were identified as differential expressed genes, and 154 unique genes were generated after the assembly of 193 clones. Of the 154 unique genes, the most abundant genes were cytochrome oxidase subunit-1 gene(9 times) and unknown(clone ID; 1-50) gene(5 times). The functional groups of these unique genes with matches in the AmiGo database were constructed according to their putative molecular functions. Among thirteen functional categories, the largest group was unclassified protein(24%). In addition, We analyzed the nucleotide and deduced amino acid sequences of the most highly occurred gene(1-50, EF434397), which consisted of 240 amino acids. However, it is confirmed yet that these genes really have an affected on the silkworms longevity.

        The 15,338-bp long complete mitochondrial genome (mitogenome) of the Japanese oak silkmoth, Antheraeayamamai (Lepidoptera: Saturniidae) was determined. This genome has a gene arrangement identical to those of all other sequenced lepidopteran insects, but differs from the most common type, as the result of the movement of tRNAMet to a position 5’-upstream of tRNAIle. No typical start codon of the A. yamamai COI gene is available. Instead, a tetranucleotide, TTAG, which is found at the beginning context of all sequenced lepidopteran insects was tentatively designated as the start codon for A. yamamai COI gene. Three of the 13 protein-coding genes (PCGs) harbor the incomplete termination codon, T or TA. All tRNAs formed stable stem-and-loop structures, with the exception of tRNASer(AGN), the DHU arm of which formed a simple loop as has been observed in many other metazoan mt tRNASer(AGN). The 334-bp long A+T-rich region is noteworthy in that it harbors tRNA-likestructures, as has also been seen in the A+T-rich regions of other insect mitogenomes. Phylogenetic analyses of the available species of Bombycoidea, Pyraloidea, and Tortricidea bolstered the current morphology-based hypothesis that Bombycoidea and Pyraloidea are monophyletic (Obtectomera). As has been previously suggested, Bombycidae (Bombyxmori and B.mandarina) and Saturniidae (A.yamamai and Caligula boisduvalii) formed a reciprocal monophyletic group.

        We determined the complete mitochondrial genome (mitogenome) of the Japanese Oak Silkmoth, Antheraea yamamai (Lepidoptera: Saturniidae) from two overlapping fragments and subsequent shotgun sequencing. The 15,601-bp long A. yamamai mitogenome contains gene arrangement and content identical to the most common arrangement found in lepidopteran insects. Most individual A. yamamai mitochondrial (mt) genes were well within the range found in the respective genes of other insects, except for small ribosomal RNA (1,037 bp). The 336-bp A+T-rich region is relatively smaller than that of other lepidopteran insects. The region is interesting in that it contains tRNA-like structures as found in the A+T-rich regions of other insect mitogenomes. The start codon of A. yamamai COI gene is unusual in that no typical one (ATN) is available. Three of the 13 protein-coding genes have incomplete termination codon T or TA. All tRNA formed stable stem-and-loop structure, except for tRNASer(AGN), the DHU arm of which formed a simple loop as seen in many other metazoan mt tRNASer(AGN).

        We investigated starvation of hatching larvae, occasional artificial hatching and incubation method to establish year-round rearing of the wild silkmoth, Antheraea yamamai. In the test of starvation of hatching larvae for brushing at a time, the survival rate of the fourth instar of larvae starved for 1 day after hatching in 25℃ and 5℃ was 83.3% and 96.0%, respectively. The result represents that the survival rate is high at low temperature during starvation. In the occasional artificial hatching test for multi-times rearing of A. yamamai, the useful hatchability is high at 5℃ in case of preserving eggs for 2 months from incubation time, and at both 2.5℃ and 0℃ in case of over 6 months. A new incubation method with pre-incubation at 15℃ and 24 D photoperiod showed high hatchability about 80% for only 2 days compared with hatching for 5-6 days in traditional incubation method with the preservation at 25℃.

        멧누에나방(Bombyx mandarina) 난각유전자의 형질발현기구를 규명하기 위한 연구의 일환으로 각형성과정에 있어서 난각단백질의 합성과정을 분석하였다. 난각단백질의 합성과정을 여포세포의 기난관배양에 의한 in vitro합성계에 의하여 분석한 결과, 합성과정은 8단계로 나눌 수 있었으며 SDS-PAGE 분석에 의해 17개 이상의 단백질 band로 분리되었다. 멧누에 난각단백질은 합성시기에 따라 3단계로 구분되며 초기에는 비교적 고분자 성분이, 합성의 중후기에는 비교적 저분자성분이 합성되는 등 난각단백질의 합성은 시기특이적으로 조절되었다.

        한국산 멧누에(Bomdyx mandarina)chorion 유전자의 형질 발현기구를 규명하기 위한 연구의 일환으로 본 실험을 실시했다. 멧누에의 난각구조를 주사형 전자현마경에 의해 관찰한 결과 매우 특이적인 구조가 인정되었다. 즉 원추상의 불규칙 돌기에 의한 돌기구조층과 이 돌기 구조층을 덮고 있는 한 층의 얇은 덮개 구조층이 그것이다. 2차원 전기영동법에 의해 chorion 단백질을 분석한 결과, 난각을 구성하는 주요 단백질 성분은 등전점 4~6, 분자량 6~30 kd로 밝혀졌다. 특히 특이난각구조와 관련된 특이단백질 성분을 검출하였으며 이들의 대부분은 고cysteine 단백질인 것으로 추정했다. 이상의 연구결과에 의해 멧누에 난각의 특이 구조 형성에 따른 유전자발현기구 규명을 위한 기초자료가 얻어졌다.
