        Chrysanthemum stunt viroid (CSVd) is a serious pathogen affecting chrysanthemum that has caused significant economic losses to Chrysanthemum flower production worldwide. Control of CSVd disease is difficult due to its contagious nature and long latent period in the field. As chrysanthemum is most often produced by implanting seedlings, it is necessary to diagnose CSVd infection before cultivation. In this study, we screened CSVd infection in seedlings from 30 varieties including 5 domestic, 6 Japanese, and 19 European varieties. Molecular diagnosis of the combination of RT-PCR and nested PCR showed that CSVd was not detected by the first RT-PCR but detected by the second nested PCR analysis in 10 varieties, including 1 domestic, 2 Japanese, and 7 European varieties. Further comparison of 10 identified CSVd nucleotide sequences showed that those are highly conserved (99-100%) and the most similar to an isolate (AB006737) identified in Hokkaido, Japan. Our study suggests that the combination of RT-PCR and nested PCR analysis is successful for the CSVd diagnosis of seedlings and the molecular diagnosis is necessary to prevent the introduction and propagation of viroid disease into the fields.

        The high temperature due to climate change may result in the intensification of several drought and heat stress on crops including potato. These abiotic stress affect on potato development staages; sprout development, tuber initiation and maturation. Potatoes need moderate amounts of nitrogen and cool night for good tuber growth. Especially, high temperature in soil will delay tuber initiation and induce malformation. Therefore, to identify quickly heat tolerant lines and breeding potato lines adapt to high temperature in the field are needed. The objectives of this study were as follows; To apply in vitro screening method for identifying potato lines adapted to high temperature conditions. To verify these results under field assays carried out under natural high temperature field conditions. We used in vitro screening methods with breeding lines from Intranational Potato Center(CIP) under three temperature regime, 18℃, 25℃ and 30℃. All breeding liens had some genotype that produced microtubers at 18℃ and 25℃, with a clear tendency for lower percentage of tuberization at the high temperature. To verify in vitro screening methods for heat tolerance lines, we carried out natural high temperature filed evaluation at Tacna, La Molina and Sanramon in Peru. The results of both the in vitro test and the field assay showed clear relationship and similar expression of tuberization percentage. This finding supports the use of the in vitro assay as a rapid screening methods that represents performance at the field level. But the correlation between performance of the breeding lines under the in vitro and field condions was low. This could be due to differential response to breeding lines to characteristics of the field environment, such as soil salinity, drought, which were not represented in the in vitro assay.

        Low temperature is one of the most severe abiotic stress factors limiting growth, productivity and distribution of winter cereals. Reliable field screening method, which can detect small differences in winter survival, is important for the effective selection and development of plants to identity superior cold tolerant winter cereal genotypes. This study was undertaken to provide improved screening method of winter hardiness in the field by increasing the accuracy in evaluating winter hardiness of barley (Hordeum vulgare L.). We introduced furrow in field screening of winter survival. By sowing the plants at the ridge and base, we could minimize the effect of topographic variation in a field by giving higher and lower level of stress at the same time. This method could be used by breeders to conduct accurate evaluation of winter hardiness by selecting the better treatment, which shows close to normal distribution, among the winter survival rate from the ridge, base and mean survival rate of the two in a screening field.

        The purpose of this study was to evaluate the effect of pesticides on insect pest control of Rhophalosiphum nymphaeae, growth characteristies and dry root yield from the cultivated after early maturing rice cropping. All pesticides treated had no effect on the growh period, flowering rate of Alisma plantago. The major pesticides were methomyl-Lf, 24.1 %, imidacloprid-Wp, 10%, carbosulfan-Wp, 20%, and methomyl-Wp, 45%. Dry yield of root were increased largely with imidacloprid-Wp, 10% (10g/20~ell ), pesticide than the other pesticides and control. All pesticides were had no injury with recommended concentration. On the other hand all pesticides were slightly harmful in the double dosage level for the Alisma plantago.
