Background : The root of Angelica gigas Nakai is used as a traditional herbal medicine in Korea for the treatment of many diseases. Poor water solubility of the active compounds from A. gigas Nakai is a major obstacle to bioavailability. Methods and Results : This work aimed to enhance the solubility of active compounds by a chemical (viz. surfactant) and physical (HME, hot melt extrusion) crosslinking method (CPC) in A. gigas Nakai. Infrared fourier transform spectroscopy (FT-IR) revealed multiple peaks in extrudate solids representing new functional groups including carboxylic acid, alkynes and benzene derivatives. Differential scanning calorimetry (DSC) analysis of the extrudate showed lower glass transition temperature (Tg) and lower enthalpy (ΔH) [Tg: 43℃; ΔH: <6 (J/g)] compared to the non-extrudate (Tg 68.5℃; ΔH: 123.2) formulations. X-ray powder diffraction (XRD) analysis revealed amorphization of crystal materials in extrudate solid. In addition, enhanced solubility (53%), nanonization (403 ㎚), and higher amount of extracted phenolic compounds were achieved in the extrudate solid compared to non extrudate (36%, 1499 ㎚, respectively). Among the different extrudates, acetic acid and span 80 mediated formulations showed superior extractions efficiency. Conclusion : We conclude that the HME successfully enhanced the production of amorphous nano dispersions of phenolic compound including decursin from extrudate solid formulations. human dermal fibroblasts, which is, associated with the regulation of procollagen biosynthesis resulting from AMRP-induced TGF-β1 expression and the mitogenic activity in HDF cells, and therefore, is expected to reduce the age-dependent loss of extracellular matrix proteins.

Background : The objective of this study was to make colloidal dispersion of the active compound was characterized nano-composite from radix of Angelica gigas Nakai using hot melt extrusion (HME). Therefore, food grade hydrophilic polymer matrices was used to disperse compound in aqueous media. Methods and Results : Extrudate solid formulations (ESFs) mediated various HPMCs (hydroxypropyl methylcellulose) and Na-Alg polymer from ultrafine powder of radix of Angelica gigas Nakai was developed through physical crosslink method (HME) with ionization agent (treatment of acetic acid) and food grade polymer [HPMCs such as HP55, CN40H, AN6 and sodium alignate (Na-Alg)]. X-ray powder diffraction (XRD) analysis confirmed the amorphization of crystal compounds in extrudate solid formulations mediated HP55 (HP55-ESF). Differential scanning calorimetry (DSC) analysis indicated a lower enthalpy (ΔH = 10.62 J/g) of glass transition temperature (Tg) in HP55-ESF. Infrared fourier transform spectroscopy (FT-IR) reveled new functional groups produced in HP55-ESF. Phenolic compound, flavonoid (including decursin and decursinol angelate) and antioxidant activity increased at 5, 10 and 2 times in HP55-ESF. Water solubility (61.5%) nano size (323 ㎚) particle was achieved in the HP55-ESF. Conclusion : Nano-composites have been developed utilizing melt-extruded solid dispersions technology by food grade polymer enhanced nano dispersion (< 500 ㎚) of active compound from radix of Angelica gigas Nakai with enhanced solubility and bioavailability. This nano-composites of radix of Angelica gigas Nakai developmental and marketed products to enable therapeutic performance.

Background : Scopolamine induces cholinergic dysfunction and oxidative stress, and the impairment of memory function. Therefore, oxidative stress and cholinergic dysfunction are important role of the brain pathology of amnesia. In this study, we investigated the impact of Safflower seed against oxidative stress and cholinergic dysfunction on scopolamine-induced amnesic mice. Methods and Results : Mice were orally pretreated with safflower seed (100 ㎎/㎏ body weight) or vehicle for 7 days, and scopolamine (1 ㎎/㎏ body weight) was injected intraperitoneally, 30 min before the behavior tests such as T-maze and novel objective recognition test on first day. To evaluate learning and memory function, the Morris water maze task was performed for 5 days, consecutively. The results showed that spatial perceptive ability and novel object recognition was significantly increased by preadministration of safflower seed compared with scopolamin-induced control mice in the behavior tests. Consistently, immuno blot revealed the elevated expression of superoxide dismutase 1 in the safflower seed pretreated mice, compared to the control mice. Moreover, protein expression of acetylcholinesterase was decreased in safflower seed pre-treated group. Conclusion : Subsequently, our results suggests that the Safflower seed extract improved memory impairment through inhibition of cholinergic dysfunction and oxidative stress.

Background : The major active components of ginseng are ginsenosides and their pharmacological effects include anticanser, anti-stress, anti-fatigue, antioxidant and aging inhibitory effects. These ginsenosides components is higher in leaves than roots. Therefore, consumers are increasingly interested in using ginseng sprouts. Methods and Results : Ginseng sprouts were cultivated during 60 days from June to late July in greenhouse. After 60 days of cultivation, the stem was hardened, and the cultivation was done until 60th because of the tendency that the value of ginseng sprouts was lowered. The content of the three indicator components were analysed by high performance liquid chromatography. Total ginsenoside content increased 1.07 times in leaves and decreased 0.67 times in roots according to cultivated period. The contents of ginsenoside Rg1, Re, Rb1, Rc F3 and F4 of leaves were increased and ginsenoside Rg1, Re, Rb1, Rb2 and Rf of roots were decreased on cultivated 60 days, especially. Total free sugar content increased 1.29 times in leaves and decreased 0.68 times in roots according to cultivated period. The total phenolic acid contents of leaves decreased slightly until 40 days and then increased. The major components of ginsenoside, free sugar and phenolic acid in leaves were Re, sucrose and sinapic acid, respectively. Conclusion : From the above results, ginseng sprouts cultivated for 60 days is more effective ingredient than roots, so it can be said that it is good for consumers to use.

Background : Pectin lyase-modified red ginseng extract (GS-E3D) is a newly developed ginsenoside Rd-enriched ginseng extract. This study was designed to investigate the acute oral and dermal toxicity of GS-E3D in rat. Methods and Results : The acute oral toxic effects of GS-E3D in female SD rats were examined at dosages of 300 ㎎/㎏ and 2,000 ㎎/㎏. In acute dermal toxicity study, 500, 1,000 and 2,000 ㎎/㎏ of GS-E3D were applied onto the shaved skin of male and female SD rats. The weights of rats were recorded at 0, 1, 3, 7, and 14 days and clinical observation were checked once a day for a period of 14 days. All rats were scarified on 14th day and complete gross examination was conducted to detect any gross change of organs after necropsy. GS-E3D did not produce orally or dermally treatment-related clinical signs of toxicity or mortality in all rats during the 14-day observation period. The oral and dermal LD50 values of GS-E3D were over 2,000 ㎎/㎏ in rat. The oral and dermal administration of GS-E3D revealed no significant change in body weight and gross pathology examination compared to control group. Conclusion : These results indicate that GS-E3D can be used as a food and cosmetic materials without critically adverse effect.

Background : The human patch test is a method used to evaluate potential skin irritation after contact with a cosmetic materials. Pectin lyase-modified red ginseng extract (GS-E3D) is a newly developed ginsenoside Rd-enriched ginseng extract. This study was designed to investigate the skin safety of GS-E3D in human patch test. Methods and Results : Thirty two female volunteers were tested with GS-E3D. GS-E3D (20 ㎕) was applied to occlusive patch test devices and was then applied onto the back of subject with normal skin for 24 hours. Cutaneous irritation responses were evaluated and graded according to criterion of International Contact Dermatitis Research Group (ICDRG) at 30 min, 24 hours, and 48 hours after removing of GS-E3D patch. The average age of subjects was 47.3 ± 9.3 years. Skin reactivity calculated from irritation score in GS-E3D treated group was 0.51 and skin irritation score of no application group was 0, respectively. Skin irritancy was no response in both GS-E3D treated group and no application group. From above data, GS-E3D was identified as a non-irritant according to ICDRG guideline that skin irritation score of ‘0.00 - 0.75’ is a non-irritant. Conclusion : These results indicate that GS-E3D can be useful as a safe cometic ingredient.

Background : Osteoclasts are differentiated from the monocytes/macrophages of hematopoietic cells, that excessive activities of bone-resorbing giant cells leads to pathological bone diseases such as osteoporosis (contained rheumatoid arthritis and autoimmune arthritis). Therefore, it is very important to suppress loss of bone mass by deactivation of osteoclast differentiation. In this context, we evaluated for the effects of black ginseng (BG) extract on TRAP activity, proliferation and differentiation in RANKL-induced osteoclastic RAW264.7 cells. Methods and Results : The aim of this study is to figure out the potential anti-osteoporosis effects and the underlying mechanism of BG extract in RANKL-induced osteoclastic RAW264.7 cells. The ginsenoside Rg3, Rg5, Rk1 and Rh4 content of BG was increased more than Red ginseng (RG). The extracts of BG markedly reduced the activity of tartrate-resistant acid phosphatase-positive (TRAP+) multinucleated cells from osteoclastic RAW264.7 cells, without cytotoxicity. BG clearly inhibited RANKL-induced osteoclast differentiation by decreased calcitonin and TRAP (p < 0.01). Furthermore, ginseonside Rg5 and Rk1 significantly inhibited TRAP activity in formation of osteoclastic differentiation (p < 0.01). It is also found that Ginseonside Rg5 and Rk1 mixture more inhibits osteoclast differentiation activity. Conclusion : Our results suggest that Black ginseng extract has an anti-osteoporosis effects in bone disease when administered as a food supplement and has potential as a therapeutic agent for osteoporosis.

Background : In this study, we investigated the formation of Abeliophyllum distichum callus which has not been reported until now and the culture optimization. The potential of callus cultures was examined by comparing the components and antioxidant activities of callus and wild planting. Furthermore, it was intended to provide data on the possibility of substitution for the active ingredient production of Abeliophyllum distichum callus extraction. Methods and Results : We tried to induce callus by introducing it in vitro culture by using leaves and stem of Abeliophyllum distichum grown in Goesan Chungbuk. In order to investigate the effect of growth regulators on the callus, TDZ, NAA, 2,4-D, IBA and BAP were treated with 0.25 to 5 ㎎/ℓ of single hormone, and the combination hormones were treated with two concentrations of BAP (0.1, 1.0 ㎎/ℓ) and 2,4-D, NAA, IBA at 0.1 ㎎/ℓ and 1.0 ㎎/ℓ. And investigate the effect of inorganic salt concentration on somatic embryogenesis, the incidence of callus was examined by culturing 1X, 1/2X MS medium for 4 weeks. As a result, the effect of growth regulator and treatment concentration on the callus formation of Abeliophyllum distichum, callus induction was the fastest in the 2,4-D condition, but the callus indection was slow. The highest amount was produced ine the NAA condition. The axillary bud was best grown in TDZ condition, but no root was formed. LC chromatogram was able to compare the contents of leaf, stem extracts and callus extracts. Antioxidant ativity showed excellent antioxidative activity in the extracts of the wild planted stem. Conclusion : The optimum culture conditions for callus were 1/2X MS, 1 ㎎/ℓ BAP and 0.1 ㎎/ℓ of 2,4-D. As a result of LC chromatogram, it was confirmed that callus extract contained a large amount of the same components as the stem and leaf extracts grown on the wild planting. Currently, Studies on mass culture using bioreactor to utilize this are underway, We could confirm the potential possibility of Abeliophyllum distichum cultured products.

Background : The isoflavonoids, triterpene and polysaccharides were know as major components in Astragalus membranaceus Bunge. Especially, isoflavonoids are classified as ononin, calycosin-glucoside of glycon binding and formononetin, calycosin of aglycone binding. In this study, we performed fermentation by a lot of microorganisms which were derived foods. Methods and Results : The A. membranaceus was extracted with 50% Ethanol and it was concentrated using rotary evaporator. In order to change in isoflavonoids, we fermented the A. membranaceus extracts with microorganisms which have β-glucosidase activity. we chose 20 microorganisms using esculin agar method and three strains with the highest activity of β -glucosidase were identified by pNP assay. The extracts of A. membranaceus were fermented during 3 days in sterilized distilled water, as 1 Brix and 3 Brix. The isoflavonoids of fermented extract, respectively days 0, 1, 2 and 3, were analyzed using High Pressure Liquid Chromatography (HPLC). In addition, the fermented A. membranaceus extracts was investigated about whitening effect using tyrosinase and radical scavenging activity by 1,1-diphenyl-2-picrylhydrazyl radical (DPPH). As a result of β-glucosidase activity by pNP assay, we selected two strains, Pediococcus pentosaceus (KACC 81017BP), Saccharomyces cerevisiae (KACC 83014BP). On the third day of fermentation of S. cerevisiae, isoflavone aglycone was the highest. The calycosin-glucoside and ononin in fermented A. membranaceus extract were reduced, calycosin and formononetin was increased compared to control. Also, we confirmed higher activity than control in tyrosinase inhibition rate on the third day of fermentation, as 78.38% and lower IC50 value of radical scavenging activity as 584.68 ㎍/㎖. Conclusion : From the above results, we may suggest that A. membranaceus aboveground parts might have useful as a safe material for functional food and pharmaceutics.

Background : Cirsium setidens is a perennial wild herb that belongs to the Asteraceae family. It grows in the mountainous region of Gangwon-do in Korea and is also called gondre. The bioconversion technology applied in this study has the same meaning as biosynthesis, biocatalyst, etc., and refers to a technique for producing desired products from precursors using enzymatic functions of microorganisms. Therefore, useful microorganisms having immunological activity were selected and Cirsium setidens fermentation products were prepared by applying bioconversion technology. And fermented product extracts were prepared to consider as a good sources of natural immune enhancement and functional food ingredients. Methods and Results : Lactobacillus fermentum, Lactobacillus plantarum, Saccharomyces cerevisiae, Weissella cibaria, and Lactobacillus plantarum were used as isolates from traditional foods. The fermentation product was set to a condition that the inherent physical properties did not change and did not generate a unique odor during fermentation. Cirsium setidens was fermented at 37℃ for 24 hours. And the fermented material was sterilized at 9 0℃ for 1 hour and then dried at 70℃ and pulverized. The contents of pectolinarin and pectolinarigenin, which are non - glycosides, were analyzed before and after fermentation using HPLC. Also NO production was measured in RAW264.7 cells after extract treatment at various concentrations using Griess reagent kit . The content of pectolinarin was increased in fermented Crisium setidens before fermentation, but the content of pectolinarigenin was increased after fermentation. In addition, the water extract of the fermented material accelerated the NO production compared to the pre - fermented material. Conclusion : As a result, relatively high immunostimulating effect were observed in dried Crisium setidens after fermentation, and it was confirmed that it could be a ingredient material for health functional food.

Background : Gastrodia elata Blume (G. elata) is important medicinal resource in korea. Gastrodin and 4-hydroxybenzyl alcohol (4-HBA) are major active compounds of G. elata, and ρ-cresol is major cause of off-odor like pig slurry from G. elata. The off-odor can decrease the quality of fresh G. elata as well as its products. Therefore, this study was performed to investigate the influence of extraction temperature on bio-active and odorous compounds of G. elata extract. Methods and Results : G. elata was extracted with distilled water at 0, 30, 60, and 90℃ for 20, 40, 60, and 120 min. Gastrodin and 4-HBA contents were analyzed by using a HPLC-UVD, and ρ-cresol content was analyzed by using a SPME-GC-MS. Gastrodin content increased as increasing extraction temperature and time, and showed the highest value in extract at 90℃. 4-HBA content showed the highest value at 60℃, and increased as increasing extraction time. Total content of gastrodin and 4-HBA was higher in extract from G. elata at 60℃ for 120 min than other extracts. ρ-Cresol content was varied according to extraction temperature, and was lower in extract at 30 and 60℃ than 0 and 90℃. Conclusion : These results indicated that the extraction temperature can affect the bio-active components and off-odor of G. elata extract, and 60℃ is appropriate to improve the qualities including bio-active component and off-odor of G. elata extract and its products.

Background : Cornus kousa fruit has been used as a medicine for a long time. Therefore, for the purpose of utilizing C. kousa fruit as a health functional food, antioxidant activity was tested by extracting ethanol concentrations to identify optimal extraction conditions. Methods and Results : C. kousa fruits used in this study were washed, dried and pulverized. Then, the samples and solvents by ethanol concentration (0, 20, 40, 60 80 and 100%) were extracted at 80℃ for 90 minutes at 1 : 20 ratio, respectively. Afterwards, it was filtered, concentrated, frozen and dried. Total phenol and flavonoid content, DPPH and ABTS radical scavenging activity, ferric reducing antioxidant power and oxygen radical absorbance capacity were measured. The total phenol and flavonoid content is the highest in 60% ethanol extract at 20.08 ㎎·GAE/g, 0.86 ㎎·QE/g. In addition to all antioxidant studies showed the highest activitiy in 60% ethanol extract. Also, when HepG2 cells were treated with ethanol extract at 200 ㎍/㎖ concentrations, the survival rate was more than 90% and cytotoxicity was not observed. Then, HepG2 cells were treated with tBHP to induce oxidative stress, and the cytoprotective effect and inhibitory activity of ROS production were measured. As a result, 60% ethanol extract showed the best activity. Conclusion : When extracting C. kousa fruits with ethanol as solvent, It is expected best to use 60% ethanol to maximize potential as a material for health functional foods.

Background : Dioscorea bulbifera (DB) contain steroidal saponin which has anticancer and anti-inflammatory. DB is a health-functional material used as edible and medical materials. However, DB has not been studied in detail as industrial purposes of fermentation. Methods and Results : D. bulbifera (DB) was fermented by Rhizopus oryzae CN105 for 3 days. Samples were prepared by adding 10% honey to the D. bulbifera (DBH) and honey free D. bulbifera (DB). The pH, total acidity (%) and reducing sugar (%) were investigated. The free sugar and color values related to taste components were analyzed. The pH level decreased while total acidity increased during fermentation. The free sugar and color values related to taste components were analyzed. The content of free sugar in the DB was the highest after 3 days of fermentation. In case DBH, only glucose was the highest after 3 days of fermentation. The content of fructose, sucrose and lactose were decrease during fermentation. Total polyphenol content of DB and DBH was measured by the Folin-Denis method. Total flavonoid content was measured by the Moreno method. Total polyphenol content and flavonoid content was the highest after 1 days of fermentation in both DB and DBH. Conclusion : The results showed that physicochemical components improved during fermentation in DB. But antioxidant activity was the highest after 1 days of fermentation in DB and DBH. These results are indicated that fermented DB can be used as a functional food.

Background : Exposure to Ultraviolet B (UVB) causes oxidative stress, inflammation, pigmentation and severe skin damage. Astragalus membranaceus (AG) has been used as a traditional medicine and have been studied various physiological activities. During the roasting process, bioactive substances is change including antioxidant substances. The aim is study the antioxidant effects and reactive oxygen species (ROS) inhibitory effect of the roasted A. membranaceus (R-AG). on Human dermal fibroblast (HDF) cells. Methods and Results : To prepare of R-AG samples, roasting machine was used. AG and R-AG were extracted to water and 70% ethanol. AG samples were evaluated the antioxidant potential by measuring the 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2`-azino-bis 3-ethylbenzothiazoline-6-sulphonic acid (ABTS) scavenging activities. Additionally, total phenolic contents and total flavonoid contents was compared with antioxidant ingredients. AG and R-AGs were analyzed with HPLC determine the major compounds such as calycosin, mononetin and glycosides. The antioxidant activities of R-AG increased and changed in major compounds. In UVB exposed HDF cells, AGs did not affect cell viability and R-AG inhibited ROS more effectively than AG. Conclusion : From these results, R-AG can inhibit oxidative stress induced UVB in HDF cells.

Background : Obesity is a type of metabolic diseases caused by unbalanced in take and consumption of calories. 3T3-L1 is differentiated into adipocytes by various hormones and transcription factors and accumulates intracellular lipid. Therefore, it is important to inhibit the adipocyte differentiation precess for effective obesity inhibition. Aster yomena and Aster glehui are medicinal plant of Compositea family that grows widely in Korea. Aster genus plants have been used to treat snakebite wound or bruises in oriental. The aim of this study was comparison of inhibitory effect oxidation and adipocyte differentiation with Arial parts of A. yomena (AY) and A. glehni (AG). Methods and Results : AY and AG were cultivated from Pyeongchang in Korea, 2018. AY and AG were extracted by 70% ethanol (-E) and water (-W) at room temperature. AG-W has higher phenolic content (6.92 ± 0.23) and AG-E has higher flavonoid content (8.22 ± 0.19) than other extracts. AG-E has higher radical scavenging activity on DPPH and ABTS assay (IC50 value; 104.88 ± 10.50 and 30.06 ± 0.27). In cytotoxity assay, all extracts concentrations of lower 100 ㎍/㎖ were nontoxic to the cells and can be applied for the next assay. The anti-adipogenic effect of extracts were determined in 3T3-L1 cell by Oli Red-O (ORO) staining. The lipid diplot stained with Oil red O was dissolved to determine by microplate-reader. AG-W significantly reduced the adipocyte differentiation of 3T3-L1 cells (70.49%) compared with other extracts (AG-E, AY-W and AY-E). Conclusion : Theses results reveal that the water extract of AG has utility as a functional food material for preventing obesity.

Background : Bamboo fiber contains 97% of dietary insoluble fiber from mature bamboo shoots. Bamboo contains phytosterols and a high amount of fiber which can be labelled as nutraceuticals or natural medicines that are attracting attention from the aspect of health. This study was conducted for the production of dietary fiber from Sinoarundinaria nigra, S (S. nigra) shoots barks by using α-amylase, alcalase, glucoamylase or Viscozyme and improvement of Large intestinal function in rats. Methods and Results : Yields of enzymatic degradation were 80.5% (322 g/400g) and the dietary fiver content of the enzyme digest was 69.7%. Sprague-Dawley rats were fed on diet containing 100 ㎎/㎏ and 200 ㎎/㎏ S. nigra dietary fibers [dietary fiver of bamboo shoots barks (SNDFB)] for one weeks. As a result of weighing of the fecal pellets, loperamide alone group showed a significant decrease compared with the control group, and it was significantly increased in test group (200 ㎎/㎏-SNDFB) compared to loperamide alone group. In addition, the measurement of gastrointestinal charcoal transit ratio in rats, loperamide alone group showed a significant decrease compared with the control group, and it was significantly increased in all the test substance groups compared to loperamide alone group. The number of remnant fecal pellets in the colon after dissection was significantly increased in the loperamide alone group compared to the control group, and was significantly decreased in the test substance groups compared to loperamide alone group. Conclusion : These results suggest that dietary fiber isolated from S. nigra has a marked effect on the improvement of bowel function in rats with loperamide (4 ㎎/㎏)-induced constipation.

Background : Ginseng (Panax ginseng C. A. meyer) is a medicinal herb which has been used for prevention and treatment of diseases in Korea, China and Japan for a long time. Ginseng is well known to have effects such as immunity enhancement, anticancer, antioxidative effect and blood circulation improvement. These pharmacological effects of ginseng have been reported to be due to various ginsenoside components. Recently, many studies have been actively conducted to identify the major components and functionalities of ginseng berry. Ginsenoside, a representative active ingredient of ginseng, is distributed in various parts of ginseng. However, contents of certain ingredient such as ginsenoside Re which have antidiabetic effect is higher in ginseng berry. This study was conducted to compare ginsenoside content and UV blocking effect according to the color of ginseng berry. Methods and Results : The ginseng berry extracts for the component analysis and UV-blocking effect test were obtained by harvesting the ginseng berry by color in 4-years-old ginseng cultivated, and then removing the seeds and extracting only the flesh of the ginseng berry and the peel with methanol. Ginsenosides analysis was carried out by dissolving 100 ㎎ of the extract in 2 ㎖ of methanol, filtering it with a 0.50 ㎛ filter and using it as a sample for HPLC analysis. Analysis of ginsenoside content showed that the total ginsenoside and ginsenoside Rb2 content of yellow ginseng berry were 23.9 and 2.9 ㎎/g, while those of red berry was 19.6 and 2.3 ㎎/g, respectively. The UV blocking effect of yellow ginseng berry was 17.4%, which was higher than that of red berry. Conclusion : The ginsenoside content and UV blocking effect of yellow ginseng berry were superior to those of red berry. These results suggested that yellow berry is more useful as cosmetic material for UV protection.

Background : Wild-simulated ginseng (WSG, Panax ginseng C. A. Meyer) in Korea which depends on an artificial forest growth method. Methods and Results : WSG samples were collected from 8 different regions in Korea in October, 2017, and these were analyzed by ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF MS) using optimized analytical methods to compare the metabolite patterns to cultivated regions. Furthermore, their metabolite compositions differed according to individual plant samples. The metabolite profiling data were processed by multi-variate statistical analyses such as PCA and PLS-DA to determine the differences among geographical origins. To identify significant metabolites according to regional variances of WSG, more sophisticated multi-variate statistical analyses such as metabolite selection should be performed. Conclusion : This metabolomics approach can also be applied to evaluate the overall quality of WSG, as well as to discriminate the cultivars for the medicinal plant industry.

Background : Acetylcholine is related with various functions, including cognition and behavior, and increased activity of cholinesterase has been reported in the brains of people suffering from Alzheimer’s disease (AD). As such, the inhibition of cholinesterase activity could be a means of ameliorating neuronal degenerative diseases such as AD. Reactive oxygen species (ROS) can cause neuronal cell damage. The AchE inhibitory effects of Sorghum bicolor (SB) have been revealed by research. This study was conducted to compare the cholinesterase inhibitory effects and anti-oxidative effects of SB extracts according to their extraction conditions. Methods and results : Eight extracts were prepared from SB seed, which was extracted using three different methods including room temperature extraction, reflux extraction at 85℃, and accelerate solvent extraction (ASE) at 50℃ by using distilled water and/or ethanol as a solvent. AchE and BuchE inhibition activities of the extracts were measured in vitro, and their inhibitory activities on ROS, nitric oxide (NO) production and cell proliferation were analyzed in lipoppolysacchride–treated BV2 mouse microglia cells. According to the results of the experiments, the 50% ethanol extract obtained by room temperature extraction showed a BuchE inhibitory effect of 40% at the final concentration of 100 ㎍/㎖, while the other 50% ethanol extracts showed a BuchE inhibitory effect of around 20%. The 100% Ethanol extract obtained from reflux extraction at 50 ㎍/㎖ showed the highest inhibitory effect on NO generation as 58.3%, whereas the 50% ethanol extract obtained from ASE extraction at 50 ㎍/ ㎖ showed the highest inhibitory effect on ROS generation as 56.0%. Conclusion : The results of the experiments show that the 50% and 100% ethanol extracts prepared under different temperature, pressure and solvent conditions have more effective on strong cholinesterase inhibition, anti-oxidative and anti-inflammatory effects..

Background : The health-promoting effects of green tea are mainly attributed to its polyphenol content, particularly flavanols and flavonols, which account for 30% of a fresh leaf’s dry weight, but the ingredients of the polyphenol content vary depending on the species. This study was conducted to select some candidates with superior anti-oxidative and anti-allergy effects from among seven cultivars of green tea. Methods and Results : Green tea extracts were prepared by extraction with ethanol and by evaporation of the solvent at low pressure. To evaluate their anti-allergy effect and cell viability, the samples were tested for ß-hexosaminidase inhibitory activity and MTT assay of the RBL-2H3 cells, respectively. The anti-oxidation effects of the samples were analyzed with a DPPH radical scavenging activity. According to the results of the experiment, four extracts including Camelia sinensis var. Kemsull, C. sinensis var. Beachwisull, C. sinensis var. Chamnok and C. sinensis var. Fushun showed effective ß-hexosaminidase inhibitory activity at 12.5, 25, 50, 100 ㎍/㎖. At 50 ㎍/㎖, C. sinensis var. Saemidori had the highest cell viability as 86.1%, and all of the samples showed cell proliferation above 70% at 25 ㎍/㎖. The extract of C. sinensis var. Kemsull showed a 60 - 70% inhibitory effect on the DPPH radical at all of the tested concentrations, whereas the extracts of C. sinensis var. Ryohu, C. sinensis var. Saemidori, C. sinensis var. Yabukita showed lower DPPH inhibition effects at around 10 - 30%. Conclusion : The results of this study indicate that the extracts of C. sinensis var. Kemsull, C. sinensis var. Beachwisull, and C. sinensis var. Chamnok have more prominent anti-oxidation and anti-allergy effects than other cultivars, and thus could be utilized as resources for improving health.
