A comparative phenotypic study between bl2 and spl6 mutant wasperformed to characterize spot formation mechanisms of bl2 mutant. Small spots appeared at the seedling stage in bl2 and later it covered large round areas on the leaves whereas, relatively small red spots in parallel line on both leaf surfaces at late tillering stage were observed in spl6. Vegetative and reproductive growth was reduced due to lesion formation at early age in the mutants. Lower growth habit and agronomic trait value was observed in mutants as compared to wild type plants. Genetic segregation data among F2 population revealed that both mutants are recessive in nature. Mesophyll chloroplast was not found in spotted area which demonstrates the damage of chloroplast cell at spotted area due to cell death. Transmission electron microscopy also confirmed the chloroplast damage. Increased level of total chlorophyll and hydrogen peroxide content were observed till 45 days of growth after transplantation under natural environment and dropped at 60 days. Catalase activity was increased until 45 days and decreased at 60 days whereas very slight level difference in protein content was observed. However, increasing level of total ascorbic acid contents were found in spl6 and bl2 as compared with wild type till 60 days after transplantation. Higher expressions of OsPDI and OsGPX1 in bl2 spotted leaves were found whereas OsTPX expression was very low in the spotted leaf. Rice with purple colored pericarp deposit anthocyanin on the seed coat and color accumulation increased rapidly during seed development. The purple color of rice pericarp is genetically determined by the Prp locus. Inheritance of purple pericarp was studied in Prp/ Kumgangbyeo (indica type Korean variety). Pericarp color of the F1 plants was purple and the F2 population of 274 plants segregated into 3 purple: 1 white ratio indicating dominant nature of the purple color. Comparative proteomic approaches using 2-DE were applied to analyze the protein profiles and molecular mechanism of purple color formation in ricepericarp. Results revealed that approximately 1,500protein spots were reproducibly detected in the gels with silver staining across the two biological replicates. Among them, 46 proteins were expressed differentially between purple color pericarp rice and white color pericarp of the wild type rice, in which 28 and 16 protein spots were more than two fold up regulated in the wild type and purple pericarp, respectively. MALDI-TOF MS analysis of nine spots revealed that putative fructokinase,embryo-specific protein and one unknown proteins were abundant in the wild type, whereas, anthocyanidin synthase, putative chloroplast inner envelope protein, and dihydroflavonol reductase were highly abundant in the Prp rice. Results indicated anthocyanidin synthase and/or dihydroflavonol reductase might be involved in the biosynthetic pathway of the purple color formation in the rice pericarp. Photoperiod sensitive genetic male sterile (PGMS) rice is sterile mutant controlled by photoperiod. A PGMS mutant 920S was sterile grown under long-day (LD) photoperiod (14 h light/10 h dark) but fertile grown under short-day (SD) photoperiod (10 h light/14 h dark). Proteome analysis revealed that 12 protein spots were differentially expressed in the spikelets of 920S plants either treated with LD or SD photoperiod. Among these proteins, three proteins including chlorophyll a/b binding protein, vacuolar ATPase β-subunit,~;α-tubulin and an unknown protein were more than three-fold abundant in the spikelet of the SD-treated plants than those of the LD-treated plants. On the other hand, eight proteins including acetyl transferase, 2, 3- biphosphoglycerate, aminopeptidase N, pyruvate decarboxylase, 60S acidic ribosomal protein and three unknown protein spots were more abundant in the spikelets of the LD-treated plants than those of the SD-treated plants. The results suggest that the observed proteins may be involved in sterile or fertile pollen development under LD or SD photoperiod respectively in the PGMS mutant rice.

In Korea, silicate fertilization (SF) is being practiced every four years to enhance rice production. However, the relationship between nitrogen (N) and SF in view of growth characteristics and grain yield of rice has not been examined after watermelon cropping in plastic film house. This study was carried out to identify useful critical N and Si fertilizer levels to sustain grain yield and to improve N use efficiency for rice. The watermelon-rice cropping system has maintained for three seasons in each year from 1998 to 2001 by farmer before this experiment. Experiments on N and Si fertilization levels were evaluated with Hwayoungbyeo (Oryza sativa L.) in 2002 and 2003 at Uiryeong, Korea. The goal of this experiment was to find out the optimum N and Si levels to sustain rice yield by reducing excessive N fertilizer in watermelon-rice cropping system. Nitrogen fertilization (NF) levels were three (0,~;57,~;114kg~;ha-1;0,~;50,~;100% of conventional NF amount) and five (0, 25, 50, 75, 100%) in 2002 and 2003, respectively, and combined with three SF levels (70,~;130,~;180mg~;kg-1;100,~;150,~;200% which were adjusted with Si fertilizer in soil) were evaluated for the improvement of N and Si fertilization level in both years. Rice yielded 3.98-5.95 and 2.84-4.02 t/ha in 2002 and 2003, respectively. Our results showed the combinations of 50% and 100% of N with 200% level of Si produced the highest grain yield in both years, respectably. The grain yield was greatly improved in plot of N25% level when compared to conventional NF (Nl00%) in 2003. In conclusion, NF amount could be reduced about 50% compared to recommended level by specific fertilization of N and Si combination levels for rice growing and grain yield after cultivation watermelon in paddy field.

Phenotypes of panicle, hull and seed of mutant rice (Oryza sativa L.) were characterized. Panicle mutants were classified in 4 groups with their internode length of main rachis, primary rachis, secondary rachis and pedicel. Hull and seed mutants were grouped into 12 based on their mutant characters in shape, size and color of seeds. These natural and spontaneous mutant collections showed distinct phenotypes to wild type rice. This might be useful for the identification of the functions of genetic factors in the Mendelian inheritance.

Korea, rice cultivars have been changing to 'quality' rice rather than high yielding cultivars. However, more than 10% of paddy field has been changed to greenhouse in winter season for cropping of water-melon, oriental-melon, straw berry and et cetra. This experiment has been made to identify the usefulness of critical N and Si fertilization(SF) level to obtain high grain quality rice with reduced insect pest damage by N and SF combination. Before the experiment, watermelon-rice cropping system was maintained for three seasons by farmer from 1998 to 2001. The experiment of N and Si (silicate) fertilization levels was evaluated with Hwayoung-byeo (Oryza sativa L., medium-maturing variety) in 2002 and 2003 in Uiryeong, Korea. Nitrogen fertilization (NF) levels were three and five in 2002 and 2003, respectively, and three SF levels were compared for getting the valuable N/SF level in both years. TOYO-value was positively affected by Si application in N100% plot but it was negatively related with NF level. Normal grain percentage was positively related with TOYO-value and it was highest in 0N plot and Si plots in N100%. Other appearance qualities like powdered, damaged, and cracked grain, were decreased with increasing N fertilization level. SF improved appearance quality in N100% plots but no effects in other treatments. Leaf sheath related diseases were significantly decreased by SF but it was negatively related with NF. In conclusion, SF could be improve grain quality at the same yield levels of conventional fertilization and it also could be reduce the diseases damages of rice plant in all N treatments. NF treatment reduced grain quality and improved grain yield at N50% level, however NF above N50% could not get any kind of benefits. So, compared with conventional fertilizer, reduced NF level is recommended for high grain quality with reduced insect pest damage.

Mass selection (MS) is an efficient selection method to directly improve highly heritable traits. In the present study, two cycles of MS for ear length were conducted on two sweet corn populations, BC2-l0 and BC1-10~timesSyn-II after introgression of exotic germplasm. The improved populations generated from these selections were evaluated in comparison with the base populations at two locations, to determine the genetic gains and performance of the improved populations. The two base populations showed varied average realized responses to MS. In BC2-l0 derived population, the realized responses were 9.1~% in BC2-l0 C1 and 1.2~% in BC2-l0 C2, whereas inBC1-10~timesSyn-II derived population, the realized responses were 5.6~% in BC1-10~timesSyn-II C1 and 2.9~% in BC1-10~timesSyn-II C2. All the improved populations showed longer ears than their respective base populations and the check varieties. Ear length, which was used as the selection criterion in this study, showed high broad-sense heritability in the BC2-l0 and BC1-10~timesSyn-II derived populations, while fresh ear yield revealed low heritability, indicating that selection for ear length in these populations would be more effective than direct selection for yield. Results of this study indicate that MS conducted on BC210 and BC1-10~timesSyn-II had significantly increased ear length and fresh ear yield in both populations. The improved populations obtained would serve as better germplasm sources and further selection in these populations could offer better responses.

This experiment was conducted to investigate the changes of growth and maturity and to clarify the function of supernodulating characters, excessive nodules and high biological nitrogen fixation rate (BNF), on maturity in response to different planting time in supernodulating soybean mutants. Two supernodulating soybean mutants, Sakukei4 and SS2-2, and their parent cultivars, Enrei and Shinpaldalkong2, were planted on May 24 and June 15, 2004. The degrees of the shortening of growth days by the planting time delay were 18 to 22 days in four cultivar, and there were no significant differences among the cultivars. However, four cultivars showed the different maturity properties. Sakukei4, mutated from Enrei, showed later maturity than that of Enrei, and 882-2, mutated from Shinpaldalkong2, showed earlier maturity than that of Shinpaldalkong2. The plant and nodule dry weights at R6 stage of Sakukei4 showed the smallest decrement and those of SS2-2 was showed the largest decrement by the delay of planting time. The photosynthetic rates of Sakukei4 during the late reproductive growth period were slowly decreased, however those of SS2-2 were steeply decreased in two planting time treatments. Overall, the growth of Sakukei4 was decreased slowly, however the growth of SS2-2 was decreased sharply according to the delay of planting time. The percentage of seed yield of Sakukei4 in June planting plot compared with May planting plot at R8 stage was 92~% , which was the lowest decreasing rate of yield among the cultivars, and in the case of SS2-2, it was in 76~% , the highest one. These results indicated that the responses of supernodulating mutants by the delay of planting time were very similar to the wild types. This means supernodulating characters in supernodulating soybean mutants might not affect to the maturity property. Additionally, the maturity property could be considered as an important characteristics to decide or to select on the developments of supernodulating soybean mutants, which have a low productivity by an excessive nodules, especially.

This experiment was conducted to clarify the functions of supernodulating characters on seed yield determination through the comparison of agricultural traits of supernodulating soybean mutants, Sakukei4, SS2-2, and their parent cultivars, Enrei and Shinpaldalkong2. The plant dry weights of supernodulating mutants, Sakukei4 and SS2-2, were 52~% and 61~% of their wild type parents at full seed stage (R6). However, the relative growth rate (RGR) from the pod set stage (R3) to R6 of Sakukei4 was 0.022 g/g/day and that of SS2-2 was 0.016 g/g/day, which were higher than those of their parents. Nodule number and dry weight were increased in two supernodulating mutants by the R6 stage. The nitrogen concentrations of leaf, petiole and stem of Sakukei4 were higher than those of Enrei. SS2-2 showed higher nitrogen concentration in petiole than Shinpaldalkong2 had. The positive correlations were appeared between nodule dry weight, plant dry weight and pod number, in two supernodulating mutants during the period from R3 to R6 stage. Although all of the yield components and seed yield were lower in two supernodulating mutants than their parents at the stage of full maturity (R8), the harvest index was higher in supernodulating mutants. The increasing rates of pod number to stem dry weight in two supernodulating mutants showed the higher than those of two their parents at R8 stage. In conclusion, the relative growth rates during the early to the middle reproductive growth period were higher in supernodulating mutants than the wild types. This could be resulted in an increase in pod number. The increase of relative growth rate was the result of the successive supplement of nitrogen source from biological nitrogen fixation (BNF) of nodules during the middle reproductive growth period in supernodulating mutants.

This experiment was conducted to investigate the soil nitrogen credit of biological nitrogen fixation (BNF) and the nitrogen balance of soybean in soybean-barley cropping systems. Soybean cultivar, Shinpaldalkong2 and barley cultivar, Olbori, were used in soybean mono-cropping (SM), barley monocropping (BM), and barleysoybean double cropping system. The barley-soybean double cropping system was treated with two different levels of nitrogen fertilizers, 0 nitrogen fertilizer (BS-F0), and standard nitrogen fertilizer (BS-F1). Nitrogen and organic matter concentrations in soil of BS-F1 plot on October, 2001 were increased 4.8~%~;and~;5.9~% , respectively, compared with those on October, 2000. The ranges of BNF rate in soybean were 69.1~~ 88.2~% in two years, and the rate was the highest in BS-F0 plot and the lowest in SM plot. The ranges of nitrogen harvest index (NHI) in all treatments were 83.9~~86.7~% . The yield was 270 kg/10a in BSF1 plot and 215 kg/10a in BS-F0 plot. However, the nitrogen balances were +0.6 kg/10a of gain of soil nitrogen in BS-F0 plot and -0.4 kg/10a of loss of soil nitrogen in BS-F1 plot. In comparisons of SM and BS-F1 plots, although the seed yields were similar in two plots, the loss of soil nitrogen was higher in SM than BS-F1 plot. Overall, our results suggest that barley-soybean double cropping system was more effective in respect to seed productivity and soil nitrogen conservation than soybean monocropping system, and the N credit to following crops by soybean cultivation was identified in soybean double cropping system.
