Plant regeneration via organogenesis and somatic embryogenesis was investigated in Korean soybean cultivars including Cheongja 3, Jinpumkong 2, Taekwangkong and Uram. Cotyledon, cotyledon+hypocotyl and hypocotyl segments of 7-day-old seedlings were cultured on MS medium containing various concentration (0, 1, 2 and 4 ㎎/L) of BA and TDZ. The results showed that MS medium supplemented with BA 2.0 ㎎/L yielded the highest shoot formation ratio of 83.3%. In 4 cultivars, Taekwangkong showed the highest ratio of shoot formation. When various sizes of immature cotyledons (S: 1∼ 2 ㎜, M: 3∼5 ㎜, L: 6∼8 ㎜) were tested on MS medium containing 2,4-D 40 ㎎/L for somatic embryogenesis, the optimum size for embryogenic callus induction was 3∼5 ㎜ in length of immature cotyledons. In 4 cultivars, Taekwangkong showed the highest percentage of embryogenic callus induction. The results indicate that Taekwangkong is the best soybean cultivar for plant regeneration via organogenesis and embryogenic callus induction among the 4 cultivars.
This study was carried out to investigate seed germination and seedling of cabbage and sugar beet in four treatments of salinity including 0 (control), 0.5, 1.0 and 1.5% NaCl. The results showed that different treatments of salinity had considerable effects on the germination and root and shoot length of cabbage and sugar beet. Percent of germination in both species showed significant decrease with increasing salinity up to 1.5% NaCl. This decrease was more evident in cabbage when compared to sugar beet. The required time for germination increased with high levels of salinity. The seedling growth of both species were inhibited by all salinity levels. Particularly at 1.0 and 1.5% NaCl, no measurable length was observed in cabbage and sugar beet. At 0.5% NaCl root growth of both plant species was more affected as compared to shoot growth by salinity.
This study was conducted to obtain the basic breeding information of Colocasia esculenta Schott. Effect of supplemental plant growth regulators and genotype difference were investigated on dormant bud tissue for proliferation. The plant regeneration ratio, plant height and root length were the best upon mixed treatment of 0.8mg/L IAA and 2.0mg/L zeatin. Both leaf weight and root weight were heavy upon culture in a dark condition. The leaf and root weights were heaviest in 6Pie sucrose concentrations. In several collected area the heaviest one was Binnangxin and then in the order of Suwon, Wanju and Puan. Genotype differences of tuber diameter and tuber weight were found in Suwon. Tuber weight was found in the order of Suwon (862mg) >Wanju(723mg) >Puan(649mg) >Binnangxin (424mg).
Aloe in vitro culture was attempted to induce callus and regeneration ability from different explant sources onto MS medium with 0.5mg/l NAA plus 1.0mg/l BA. Anthers that no developed any callus and plant regeneration, while only four out of 274 filament explants induced calli at cut edge without regenerated plants. Twenty ovary explants regenerated four direct plantlets without via callus from the base of epidermal tissues. Regenerated plants on the root tip gave 2n=14 of chromosome numbers.
To establish direct multipropagation through organogenesis from nodal explants of Anoectochilus formosanus Hayata, the nodes were cultured on LS medium containing various concentrations of 6-benzyladenine(BA). High plant regeneration and adventitious bud formation were obtained from supplemented with 4.0mg/l of BA. Plant height was promoted by adding 0.3% activated charcoal. Plantlet regeneration capacity from nodes was depended on nodal parts on the stem, upper position was the best comparing with intermediate and lower.
