Lytic polysaccharide monooxygenases (LPMOs) catalyze the oxidative cleavage of glycosidic bonds in crystalline polysaccharides including chitin and cellulose. The recent discovery of LPMO family proteins in many insect species suggests that they presumably play a role in chitin degradation in the cuticle/exoskeleton, tracheae and peritrophic matrix during insect development. Insect LPMOs belong to auxiliary activity family 15 (AA15/LPMO15) and have been classified into at least four groups based on phylogenetic analysis. In this study, we identified and investigated the physiological functions of group I LPMO15 (MaLPMO15-1 and PhLPMO15-1) in two longhorn beetle species, Monochamus alternatus and Psacothea hilaris. In both species, depletion of LPMO15-1 transcripts by RNAi resulted in a lethal pupal-adult molting defect. The insects were unable to shed their old pupal cuticle and died entrapped in their exuviae. Furthermore, TEM analysis revealed a failure of degradation of the chitinous procuticle layer of their old cuticle, retaining intact horizontal laminae and vertical pore canals containing perpendicularly oriented chitin fibers (pore canal fiber, PCF) in their core. These results indicate that MaLPMO15-1 and PhLPMO15-1 are required for turnover of the chitinous old cuticle, which is critical for insect molting.

Insect LPMOs belong to auxiliary activity family 15 (AA15/LPMO15) and have been classified into at least four groups based on phylogenetic analysis. In this study, we identified, characterized and investigated the physiological functions of group I LPMO15 (MaLPMO15-1 and PhLPMO15-1) in two longhorn beetle species, Monochamus alternatus and Psacothea hilaris. In both species, depletion of LPMO15-1 transcripts in last instar larvae by RNAi had no effect on subsequent larval-pupal molting and the resulting pupae developed normally. However, adverse effects on their development were observed during the pupal-adult molting period. The pharate adults were unable to shed their old pupal cuticle and died entrapped in their exuviae probably due to a failure of degradation of the chitin in their old cuticle, which is critical for completion of the insect molting and continuous growth.

The purpose of the study was to find grey matter (GM) and white matter (WM) volume reduction in the brain reward system among patients with alcohol dependency. This study investigated regional GM and WM in chronic alcoholic patients, focusing primarily on the reward system, including principal components of the mesocorticolimbic reward circuit as well as cortical areas with modulating and oversight functions. Sixteen abstinent long-term chronic alcoholic men and demographically matched 16 healthy control men participated in the study. Morphometric analysis was performed on magnetic resonance brain scans using voxel-based morphometry (VBM)-diffeomorphic Anatomical Registration through Exponentiated Liealgebra (DARTEL). We derived GM and WM volumes from total brain and cortical and subcortical reward-related structures. Morphometric analyses that revealed the total volume of GM and WM was reduced and cerebrospinal fluid (CSF) was increased in the alcohol group compared to control group. The pronounced volume reduction in the reward system was observed in the GM and WM of the nucleus accumbens (NAc), GM of the amygdala, GM and WM of the hippocampus, WM of the thalamus, GM and WM of the insula, GM of the dorsolateral prefrontal cortex (DLPFC), GM of the orbitofrontal cortex (OFC), GM of the cingulate cortex (CC), GM and WM of the parahippocampal gyrus in the alcohol group. We identified volume reductions in WM as well as GM of reward system in the patients with alcohol dependency. These structural deficits in the reward system elucidate underlying impairment in the emotional and cognitive processing in alcoholism.

A fabrication method to improve the processability of thermoplastic carbon nanotube (CNT) mat composites was investigated by using in-situ polymerizable and low viscous cyclic butylene terephthalate oligomers. The electrical conductivity of the CNT mat composites strongly depended on the compression pressure, and the trend can be explained in terms of two cases, low and high compression pressure, respectively. High CNT mat content in the CNT mat composites and the surface of the CNT mat composites with fully contacted CNTs was achieved under high compression pressure, and direct contact between four probes and the surface of the CNT mat composites with fully contacted CNTs gave resistance of 2.1Ω. In this study the maximum electrical conductivity of the CNT mat composites, obtained under a maximum applied compression pressure of 27 MPa, was 11 904 S m-1, where the weight fraction of the CNT mat was 36.5%.
