Background : Angelica gigas is a monocarpic perennial plant. A. gigas, also called DangGui or Korean Angelica, is a major medicinal herb used in Asian countries such as Korea, Japan and China. In Korea, we are using the roots of A. gigas. but, Chinese using Angelica sinensis and Japanese using Angelica acutiloba with the same name 'DangGui'. The biggest problem in the use of A. gigas is the confusion with A. acutiloba or A. sinensis. This confusion can cause an medical accident or lack of pharmacological ingredients. In this study, we developed chloroplast InDel markers that can distinguish A. gigas, A. acutiloba or A. sinensis. Methods and Results : We collected 14 Angelica plant samples including A. gigas, A. acutiloba and A. sinensis and extrated DNA using CTAB method. The DNA was diluted to 10 ng/㎕ and kept -20℃. We designed the primer sets using CLC Main Workbench based on chloroplast DNA InDel region of between A. gigas and A. acutiloba. PCR were performed on the 14 Angelica plant samples including A. gigas, A. acutiloba and A. sinensis (5 repeats each). Electrophoresis was performed using fragment analyzer automated CE system. We designed 6 InDel primer sets and the primer sets amplified the amplicons effectively. Three of the 6 primer sets showed polymorphism. Conclusion : We could distinguish A. gigas, A. acutiloba, and A. sinensis using 2 newly developed InDel markers.
This study was carried out to obtain basic information for optimum storage method of leaf, stem and root of Angelica acutiloba Radix, which is being utilized as major cultivated herbal medicine materials, root of Angelica acutiloba was stored for 3 months in different packing materials and sealing methods, such as punched packing, sealing and vacuum packing. The results are summarized as follows. The loss in weight as influenced by packing materials and sealing degree was the lowest at vacuum packing and complete sealing condition with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between polyethylene films, but it was significantly different between sealing conditions. Therefore, vacuum packing and complete sealing with polyethylene film are desirable storage condition for Angelica acutiloba Radix.
This study was conducted to determine feasibility of utilization of Angelica acutiloba. Especially, the quality characteristics of bread prepared with the addition of Angelica acutiloba powder were investigated. Sensory evaluation and spoilage test were conducted for preparation of functional breads which added with ground plant matters (leaves and stems) from Angelica acutiloba. The result showed that the functional breads had high score of overall liking as well as low spoilage rate when added with 0.5 to 1.0% ground plant matters of Angelica acutiloba. Consumer acceptability evaluation showed a significant preference when added 0.5 to 1.0% ground leaves and stems of Angelica acutiloba into breads. Functional breads which added powder of Angelica acutiloba inhibited the growth of fungi. The more addition of Angelica acutiloba powder, the higher the degrees of this inhibited. These results suggested that the shelf-lives of the breads were extended by the addition of Angelica acutiloba powder. Further studies were required for improvement of functionality and diversity of bread products using medicinal plant materials as an additive.
This study was conducted to develop effective production system in greenhouse for leaves and stems of Angelica acutiloba by using connect pots in 2003 and 2004. Seed germination rate and plant biomass of Angelica acutiloba collected in 2004 were higher than those harvested in 2003. Germination rate of Angelica acutiloba seeds collected in 2003 was 10%, while germination rate of seeds collected in 2004 was above 90%. Especially, plant growth and yield of Angelica acutiloba grown in connect pots sized with 4 × 4 × 5cm(length × width × height) were the highest. These results indicate that leaf and stem production of Angelica acutiloba can be improved by using connect pots and optimizing seed collecting time in greenhouse.
참당귀의 전초 (꽃봉우리, 줄기, 잎)와 뿌리의 정유성분을 분석한 결과 전초의 정유 함량은 0.063%로 뿌리의 0.389%에 비해서 아주 적은 함량을 나타내었다. 주요 성분에 있어서 전초와 뿌리의 정성적 차이는 없었으나, 함량에 있어서는 큰 차이를 보였다. 주요 휘발성 성분은 nonane, α-pinene, limonene + β-phellandrene이였으며, 이 중 nonane과 α-pinene의 경우 전초에서는 각각 7.5%와 14.6%이었으나, 뿌리에서는 각각 24.5%와 31.7%로 많은 양을 나타내었다. 이 외에 γ-terpinene, germacrene-d, (E,E)-α-farnesene, β- eudesmol의 경우 전초에서는 5%에서 8% 사이로 나타났으나, 뿌리 에서는 1% 안팎의 적은 함량을 보였다. 참당귀와 일당귀를 분석한 결과 줄기와 잎에서 각각 0.068%, 0.127%와 0.153%, 0.243%로 정유의 함량은 일당귀가 더 높았고, 부위별로는 잎이 줄기의 약 2배 정도 높았다. 참당귀 줄기와 잎의 성분으로는 각각 18개, 32개로 잎에서 더 다양한 향기성분이 분포되어 있었고, 주요 휘발성 성분으로는 α-pinene, myrcene, limonene, germacrene-d, eudesmol and butylphthalide로 잎에서 germacrene-d가 높았고, butylphthalide가 낮은 차이를 보였다. 일당귀줄기와 잎의 휘발성 정유 성분으로는 각각 21개, 32개로 참당귀와 같이 잎에서 더 다양했고, 성분은 γ-terpinene과 butylphthalide로 butylphthalide가 70%에 육박하는 주성분을 차지했다. 그러므로 Angelica gigas와 Angelica acutiloba는 관능적으로 확연히 다른 차이를 느낄 수 있지만, 분석결과로도 전혀 다른 향기성분임을 알수 있었다.
1. Ge는 명일엽(明日葉)이 일당귀에 비하여 2.1배 함유량이 더 높았다. 2. 분해 온도를 높이면 분해를 단시간에 할 수 있었으나 Ge 함량에 있어서는 낮은 온도로 분해할 때보다 낮았다 3. Ge 처리시 callus 형성은 일당귀가 명일엽(明日葉)보다 좋았으나 두 식물 모두 pH 5.7, 5.4, 6.0 순으로 좋았다. 명일엽(明日葉)에서는 일당귀와는 달리 pH 5.7에서 callus증식 중 신초형성이 많았다. 4. Callus형성에는 명일엽(明日葉)과 일당귀 모두 무기 Ge의 5 ppm까지 대체로 좋았으나 10 ppm부터 저해되다가 100 ppm에서는 거의 형성되지 않았고 유기Ge의 처리에서는 10 ppm까지 양호했고, 50 ppm부터 저해하지만 100 ppm에서도 다소 형성되었다.
