        반추위 pH 조정제로서 일반적으로 중조가 많이 사용되고 있으나 사료에 중조의 첨가량을 증가시키는 경우에 기호성을 저하시키는 경향이 있다. 따라서 본 연구에서는 사료의 기호성에 영향을 미치지 않으면서 반추위 완충효과를 나타내도록 개발된 액상형 완충제의 젖소사료 첨가제로서의 이용효과를 구명하기 위하여 수행되었다. 시험축으로 착유 중기 젖소 27두를 공시하였으며, 처리는 대조구와 중조 첨가구, 그리고 액상형 완충제 첨가구로 하여 처리별 9두씩 배치하였다. 대조구는 착유우용 자가배합 TMR 사료를 급여하였으며, 중조 첨가구는 대조 사료에 중조를 두당 1일 150g 추가하여 급여하였고, 액상형 완충제 첨가구는 같은 방식으로 액상형 완충제를 두당 15㎖ 추가하여 사양시험을 수행하였다. 시험결과 산유량은 대조구, 중조 및 액상형 완충제 첨가구의 경우에 각각 23.82, 23.91, 24.91kg으로 대조구에 비하여 액상형 완충제 처리구에서 다소 증가하는 경향이었으나 처리 간에 뚜렷한 유의 있는 결과를 보여주지는 않았다. 처리에 따른 유성분 변화에서는 유단백질 및 무지고형분에서 액상형 완충제를 첨가한 경우에 대조구에 비해 유의 있는 증가(p < 0.05)를 보여주었고, 유지율에서도 대조구에 비하여 처리구가 다소 증가하는 경향을 보였다. 혈액성분 중 포도당과 콜레스테롤은 대조구에 비해 액상형 완충제의 첨가구에서 다소 증가하였으나, 처리 간에 유의 있는 차이를 나타내지는 않았다. 따라서 개발된 액상형 완충제의 추가급여 시 생산성 증대, 특히 유성분 증가 효과가 있어 반추위 pH 조정을 위하여 일반적으로 이용되고 있는 중조를 대체하여 반추동물의 사료첨가제로 이용이 가능할 것으로 평가되었다.

        This study investigates the effect of sodium bicarbonate (NaHCO3) on growth of S. dimorphus. NaHCO3 concentration was varied from 0 to 2 g-C/L. As a result, the increase in concentration of NaHCO3 up to 1.5 g-C/L increased dry weight of algae. The highest specific growth rate of S. dimorphus was 0.36 day-1 which was obtained at concentration of 0.5 g-C/L NaHCO3. pH showed a large variation range at the concentrations lower than 0.5 g-C/L NaHCO3 whereas inorganic carbon, nitrate and phosphorus removal rates were almost same at the concentrations higher than 0.5 g-C/L NaHCO3 (0.75, 1, 1.25, 1.5, 2 g-C/L NaHCO3). Their average inorganic carbon, nitrate and phosphorus removal rate were 70 mg-C/L/d, 11.3 mg-N/L/d, and 1.6 mg-P/L/d, respectively. Thus, NaHCO3 didn’t effect on inorganic carbon, nitrate and phosphorus removal rate of S. dimorphus.

        The sodium bicarbonate cotransporter (NBC) protein is functionally expressed in salivary glands. In this experiment, we examined the role of NBC in HCO₃-formation in human parotid gland acinar cells. Intracellular pH (pHi) was measured in 2'-7'-bis(carboxyethyl)-5(6)-carboxyfluorescein (BCECF)-loaded cells. Acetazolamide (0.1 mM) and 4,4'-diisothio cyanatostilbene-2,2'-disulphonic acid (DIDS, 0.5 mM) were used as specific inhibitors of carbonic anhydrase and NBC, respectively. The degree of inhibition was assessed by measuring the pHi recovery rate (△pHi/min) after cell acidification using an ammonium prepulse technique. In control experiments, △pHi/min was 1.40±0.06. Treatment of cells with 0.5 mM DIDS or 0.1 mM acetazolamide significantly reduced △pHi/min to 1.14±0.14 and 0.74±0.15, respectively. Simultaneous application of DIDS and acetazolamide further reduced △pHi/min to 0.47±0.10. Therefore, DIDS and acetazolamide reduced △pHi/min by 19% and 47%, respectively, while simultaneous application of both DIDS and acetazolamide caused a reduction in △pHi/min of 67%. These results suggest that in addition to carbonic anhydrase, NBC also partially contributes to HCO₃- formation in human parotid gland acinar cells.

        The objective of this study was to examine the effect of caffeine and sodium bicarbonate in a fertilization medium on the fertilizability of boar spermatozoa that were frozen in straws. Boar spermatozoa were extended with Beltsville F5 extender and frozen in 0.25‐ml straws. In vitro matured porcine oocytes were fertilized in vitro (IVF) with frozen‐thawed boar spermatozoa for 6 h in a modified tris‐buffered medium (mTBM) or in its modified medium by substituting the tris with 25 mM sodium bicarbonate (modified bicarbonate‐buffered medium; mBBM). Some of inseminated oocytes were fixed and stained for examination of sperm penetration. IVF embryos were cultured in a North Carolina State University‐23 medium for embryo development. The percentage of live sperm was 47±4% and morphological abnormality of acrosome was found in 14±3% of spermatozoa. Optimal sperm concentration for IVF was 0.75~1.0×106 sperms/ml when mTBM containing 5 mM caffeine was used as the fertilization medium. Sperm penetration was significantly (p<0.05) stimulated by increasing caffeine concentration in the IVF medium. In addition, mBBM significantly (p<0.05) increased sperm penetration (92%) compared to mTBM (65%). More (p<0.05) blastocysts (22% vs. 32%) developed from the oocytes that were fertilized in mBBM containing 1 mM caffeine than from those fertilized in mTBM with 5 mM caffeine. Our results indicate that boar spermatozoa can be frozen successfully in straws with holding their normal fertilizability and that caffeine and sodium bicarbonate stimulates sperm penetration in vitro.

        Extravasation is the accidental injection or leakage of fluid into the subcutaneous or perivascular tissues. Some drugs can cause serious injury such as severe tissue injury, necrosis, and etc. Here we report a case of chemical burn by sodium bicarbonate extravasation due to accidental venous puncture during arterial cannulation. A 42-years-old woman has taken emergency laparotomy surgery due to a stab wound to the abdomen. Massive blood loss has developed, and consequently vital signs were unstable and metabolic acidosis has developed. Sodium bicarbonate has administered via a peripheral intravenous line on the dorsal vein of a right hand that runs to the cephalic vein. However, the cephalic vein that runs by the side of the radial artery has punctured accidentally during the attempt of right radial artery cannulation. Second degree superficial and deep chemical burn by sodium bicarbonate extravasation has developed. Skin lesion about 3 × 4 cm2 with erythema and bullae formation has developed. There were no necrotic changes and the digital sensation was intact. Wet dressing and silicon foam dressing were prescribed. After two weeks, she was discharged. Until then, dermis exposure about 1 × 1 cm2 remained although the skin lesions became getting well.
