        Beauveria bassiana is widely studied for its potential as biopesticide. However, little is known about the factors to influence genetic diversity among isolates. In this work, we aimed to study the gene diversity of 42 isolates to figure out the impact of sequences variability on various biological features. The accumulated data showed no correlation between sequence variability and the fungal geographic localization. Meanwhile, weak to strong correlations have been obtained between the sequence diversity of various genes and the studied biological features. Interestingly, Biotrophy-associated gene 2 gene variability was correlated with all the studied biological features, being an important marker to determine isolates of interest for biological control. Therefore, the obtained data would serve as a database to focus on interesting genes related to improving or screening highly virulent isolates as biological control agents.

        Beauveria comprises entomopathogenic fungi frequently isolated from insect cadavers, among which Beauveria bassiana is the most widely studied species of entomopathogenic fungal genus for its high potential as a biological pesticide. Even though it has been reported that B. bassiana is a heterogeneous assemblage of strains, little is known about the factors that might drive the genetic diversity among various isolates. In this work, we ought to study the gene diversity of 33 isolates in order to figure out the relationship between their gene diversity and biological features. First, we analyzed gene sequences that are involved in developmental and immunological processes of the fungal isolates and host. Now we are checking whether the gene diversity is related to the genes function, the isolates geographic localization, thermo-tolerance and virulence of the isolates. The obtained results so far showed no correlation between the genetic diversity and the fungal geographic localization. But interestingly, a positive correlation has been obtained between the virulence and the genetic diversity based on the internal transcribed spacers (ITS). Further analyses are aiming to confirm the obtained data and to analyze possible correlations between the genetic diversity and morphological and transcriptional data of the studied isolates. Based on the obtained results, we could much strongly focus on interesting genes which might be related to improving or screening highly virulent fungal agents as biological control agents.

        국내 4개 지역으로부터 채집된 배추좀나방(Plutella xylostella)의 미토콘드리아 DNA중 COI 유전자 일부 (438 bp)의 염기서열을 결정, 유전적 다양도 및 유전자 이동정도를 파악함으로써 집단 유전적 구조 및 특성에 대하여 연구하였다. 총 21개체로부터 13개의 mtDNA haplotype을 얻었으며 이들의 변이는 0.3~1.4%로 다른 곤충을 대상으로 한 유사연구와 비슷한 크기를 나타내었으며 haplotype 다양도는 매우 높았다(평균 h=0.81). 지리적으로 먼 제주도의 개체군과 경남 김해 두 지역(11km 거리)의 개체군을 비교한 결과, 통계적으로 유의한 정도의 유전적 격리(p<0.05%)는 전혀 관찰되지 않았으며, 대신 상당한 정도의 세대당 암컷 이동률(Nm=2-30)을 보였다. 또한 GenBank에 등록된 하와이의 배추좀나방 haplotype은 본 연구에서 얻은 것들과 유전적으로 흡사하였다. 종합적으로, 국내 배추좀나방은 전체적으로는 많은 haplotype수에 기인한 적절한 크기의 유전적 분화율을 보유하고 있으며 국지적으로는 상당한 이동력에 의한 장거리 이동으로 개체군내 높은 haplotype 다양도를 보이며 동시에 지역간의 유전적 유사성을 나타낸다고 요약되었다.

        Trees are extraordinary long living, sessile organisms that are faced quite variable environmental conditions and biotic stress factors during their life. They have a high level of genetic diversity to ensure adaptation and adaptability. With the massive advance in next generation DNA-sequencing, our abilities to screen this genetic variation have increased drastically. I will give an overview on the different genomic approaches, main target tree species and examples for application in the area of population genetics. Examples will show how high throughput SNP genotyping clarifies our views on taxonomy, phylogeography and provide us with useful data for different diagnostic applications such as species identification, tracking geographic origin and screening the level of pre-disposition to abiotic and biotic stress factors. Since a few years next generation sequencing and advanced molecular marker development have also been used in genetic association studies and transcriptome sequencing (RNASeq) in order to discover genes that are coding economical and ecological important phenotypic traits of trees.         We investigated the expression patterns of a granule-bound starch synthase I (GBSSI = Waxy) gene at different developmental stages of storage and non-storage organs in Amaranthus cruentus. GBSSI transcripts were strongly expressed in the middle and mid-late stages of seed development and thereafter expression decreased. In addition, this gene was expressed in all non-storage organs tested (the leaf, stem, petiole and root) and showed a tendency to increase during plant development. Therefore, our results indicate that the amaranth GBSSI gene exhibits late expression in the perisperm, and that it is expressed in both storage and non-storage tissues. We also investigated the genetic diversity of GBSSI among 37 strains of amaranth grains originating from New World. A comparison of the GBSSI coding sequence revealed an extremely high level of sequence conservation, and a single nucleotide polymorphism between the sequences of non-waxy (Type I) and waxy (Type II) phenotypes was detected. This indicates that a G–T polymorphism in exon 10 (a nonsense mutation) was a unique event in the evolution of the GBSSI gene in amaranth grains.

        Sucrose synthase 3 which is a third active gene present in rice, is localized predominantly in rice endosperm. This sucrose synthase 3 may play an important role in the starch filling in the milky stage rice seed, probably involving in the starch physicochemical properties. As the genetic diversity at this locus is little informed, forty three rice consisting of japonica, indica and Oryza rufipogon were targeted to amplify full sequence of sucrose synthase 3 to examine the frequency and distribution of nucleotide polymorphism. Total of 755 all sequence variants detected, 491 single nucleotide polymorphisms (SNPs) and 264 indels were successfully identified in 7618 bp of sequence containing the sucrose synthase 3 transcript, promoter and 3' non-transcribed region. The frequency of nucleotide changes and indels were high, on average one polymorphism per 15.5 bp and one indel per 28.9 bp with 11 sequence-based haplotypes distinguishable among the varieties and lines. Both the frequency of nucleotide changes and indels were frequent in non-coding region, but rare in coding region. Sequencing a polymorphism region in the promoter showed one base change on one of cis-element from T (CATGCATA to A (CATGCACA) that might implicate in seed specificity. The presence of a high number of haplotype shared by a few varieties indicated a little information on linkage disequilibrium.

        우리나라에서 자생하는 야생콩(Glycine soja) 81점과 재래종 콩(G. max) 130점에 대해 7개의 SSR 마커의 다형성을 통해 두 종간의 변이를 조사하고 지리적 유연관계를 분석한 결과를 요약하면 다음과 같다. 1. 전체 두종에서 총 144 개의 대립인자(평균 20.6개)를 확인하였고, 각 유전자좌별 복수 대립인자수는 13~41 개로 나타났다. 각 종별 대립인자수로 야생콩은 총 117개의 대립인자수(평균 16.7개)를 나타냈으며 재래종 콩은 총 69개의 대립인자(평균 9.9개)를 나타냈고, 두종간에 서로 공유된 대립인자수는 총 42개였다. 2. 전체 두종에 대한 유전자좌별 유전자 다양성 값 범위는 0.69(Satt141)~0.96 (Sat074)이었다. 또한 전체 다양성 값은 0.81을 나타냈고 야생콩은 0.88, 재래종 콩은 0.69였다. 3. 야생콩과 재래종 콩의 유전적 변이에서 SSR 분석에 의한 정준판별분석 결과, Canl(84.2%)에 의해 좌측은 G. soja(I군), 우측은 G. max(II군) 그리고 두 종이 서로 중복되는 군(III군)으로 구분되었으며, 유전적 기저가 넓은 야생콩이 재래종에 비해 변이가 크게 나타났다. 4. 야생콩의 지리적 유연관계는 2개의 군으로 구분되었는데 I군은 강원, 경상, 전라, 충청도 지역이, II군에는 경기도 지역이 독립된 군을 형성하였으며, I군내에서는 강원도와 경상도 지역이, 그리고 전라도와 충청도 지역이 각각 같은 군을 형성하였다. 재래종 콩도 2개의 군으로 구분되었는데 I군에는 강원도, 경기도, 경상도 지역이, II군에는 전라도, 충청도 지역이 포함되었다. 또한, I군내에서는 경상도 지역이 독립된 군을 형성하였으며, 강원도와 경기도 지역이 강한 유연관계를 나타냈다.것으로 판단된다.서 조사대상오염물질의 분해효율이 0~50% 로 감소하는 결과가 나타났지만, 일반적으로 산업 배기가스에서 측정되는 황화합물의 오염도 수준보다 다소 낮은 농도에 해당하는 황화메틸 0.1 ppm을 오염물질에 첨가하였을 때는 오염물질의 분해효율에 영향이 나타나지 않았다.of sensibility(감) that explores "1′expression" a la Dufrenne.e and accomplish their duties.
