One of the problems associated with in vitro culture of primordial gern cells (PGCs) is the large loss of cells during the initial period of culture. This study characterized the initial loss and determined the effectiveness of two classes of apoptosis inhibitors, protease inhibitors and antioxidants, on the ability of the porcine PGCs to survive in culture. Results from electron microscopic analysis and in situ DNA fragmentation assay indicated that porcine PGCs rapidly undergo apoptosis when placed in culture. Additionally, \ulcorner2-macroglobulin, a protease inhibitor and cytokine carrier, and N-acetylcysteine, an antioxidant, increased the survival of PGCs in vitro. While other protease inhibitors tested did not affect survival of PGCs, all antioxidants tested improved survival of PGCs (p<0.05). Further results indicated that the beneficial effect of the antioxidants was critical only during the initial period of culture. Finally, it was determined that in short-term culture, in the absence of feeder layer, antioxidants could partially replace the effect(s) of growth factors and reduce apoptosis. Collectively, these results indicate that the addition of \ulcorner2-macroglobulin and antioxidatns can increase the number of PGCs in vitro by suppressing apoptosis.
To identify genes implicated in the control of pluripotency as well as characteristics of stem cells, we analyzed expression profiles of genes derived from mouse morulas, blastocysts, embryonic stem cells, mesenchymal stem cells, and uterus tissue cDNA microarray. Comparative analyses of their expression profiles identified putative clones that expressed specifically in specific samples or not in a specific sample. The expression pattern of these candidate clones was analyzed using RT-PCR and non-radioactive in situ hybridization. Functional annotation of these clones on pluripotency and stem cells and molecular mechanisms underlying many facets of mammalian development and differentiation.
Narcotic drugs generally refer to serious and habitual hidden rash such as opium, heroin, methyphetamin, nabinol, cocaine, and so forth. At present, narcotic drugs are spread unchecked and are causing a big social problem. So many countries and narcotists are making every effort to set up a barricade against narcotic drugs. And there is a limit suitable treatment for them. Thus Tuo Yin Tang Jiang is developed. As indicated by Chinese letters, Tuo Yin Tang Jiang(TYTJ) is a crude drug. It is a traditional chinese medicine developed by the study done from June in 1998 to June in 1999 that Hubeisheng was entrusted with by People's Republic of China. This study is a treatise on etiology and syntomatology of narcotism. TYTJ is a medicine which is in accord with Pharmacopeia of the People's Republic of Chinal) in order to remove from the body the toxic materials resulting from narcotic drugs such as opium and heroin. According to the standard diagnosis on narcotism, 105 cases are studied and treated at the Rehabilitation Center attached to Enshi Autonomous Region Hospital in Hubeisheng. 105 cases are divided into 2 groups by double-blind method. One is the experimental group which has 56 cases. The other is the control group which has 49 ones.13 cases among 105 cases are addicted by intravenously injections. 9 cases are by oral takings. It took 10 days for this experiment to be performed. Two groups didn't show a striking individual variation based on the age, gender, period of taking drugs, withdrawal symptoms, complication, and state of health. The experimental group had a higher effect of treatment than the control group had. TYTJ treats diseases effectively and has no side effect, irrespective of the serious or slight addiction to opium and morphine.
Seven genetic lines of Bupleurum falcatum L. from different geographical regions were analysed for saikosaponin contents and chromosomal numbers. The somatic chromosome numbers of B. falcatum originated from Euisong, Iri, Milyang, Sangnam, Taejon, and Youngchon were 2n=20 while Mishimasaiko showed 2n=26. However, chromosome features were different in plants grown in different geographical regions. Generally, Korean lines had higher saikosaponin contents than Mishimasaiko which is Japanese and Sangnam lines had highest saikosaponin contents compared to other tested lines.
This study was conducted to investigate the occurrence of Soil borne wheat mosaic virus(SbWMV) in barley fields in Korea and to examine the host pathogenicity of SbWMV. By using the ELISA test, SbWMV was detected in the six regions : Suwon, Milyang, Jinju, Youngkwang, Iksan, and Chonju. SbWMV was isolated from the two strains, Albori strain from Jinju and Eunpamil strain from Milyang. SbWMV was collected from leaves showing mosaic, yellowing and necrosis stripes. SbWMV was inoculated mechanically on 1∼1.5 leaf stages with leaf-rubbing to identify the host pathogenicity of 36 Korean barley cultivars, a wheat cultivar, two rye cultivars, three Japanese barley cultivars and Chenopodium amaranticola. Viral sympoms of inoculated leaves appeared on moulted loaves about 4 to 6 weeks of inoculation. Baegdong and Tapgolbori, infected from Albori strain and Eunpamil strain infected from Samdobori showed much higher susceptibility than C. amaranticola and C. quinoa which showed ring spots and chlorotic spots respectively. Virus particles were observed by the electron microscope. They were rod-shapes, which are bipartite, of 142 nm or 281 nm in length with 20 nm diameter on infected leaves. Specific detection and identification of SbWMV was set up using the RT-PCR. PCR fragments of SbWMV(0.5kb) were obtained by using the designed primers for SbWMV RNA 2.
Major characteristics of new Waesungri maize inbred line has multi-tiller and ears: five to six tillers and seven to eight ears per plant and flowering date of Waesungri was delayed about 18 and 24 days compared to Mo17 U.S line and IK4 Korean local lines, respectively. Number of ears, fresh and dry weight per plant were significantly different among all tested hybrids including Waesungri/Sinkihong hybrid under different planting times and densities. Especially, both fresh and dry weight of IK1/FR140//Waesungri F1hybrid were significantly higher at high planting density. In kernel weight per unit area, Wnesungri/Sinkihong hybrid was high at high density and IK1/FR140//Waesungri hybrid was high at low planting density. As results of analysis of variance, flowering date was shown a significantly different both planting times and varieties, while other characters including stem height were shown very variable in interactions with enviromental factors.
This study selects Sunwol local group, Gusang local group and Yongjun local group to examine the appropriate amount of applied fertilizer in cultivating the double crop seed of Alisma plantago by transplantation in the southern area, improve and complements the double cropping techniques and contribute to stable production. While the plant height is 45,47 and 49cm and the number of leaves is 11.7, 12.5 and 14.4 at non-fertilizing plot, the plant height is 67, 72 and 75cm and the number of leaves is 15.8, 17.2 and 19.3 at all fertilizing plot and their growth is active and in the character of plant height and the number of leaves, especially in the groove of N-P2O5-K2O= 30-15-45kg/10a, the mean plant height of Sunwol is 75cm and mean number of its leaves is 19.3, the mean plant height and number of leaves in Gusang are 72cm and 17.2 respectively and those of Yongjun are 67cm and 15.8 respectively. While the number of floral axis is 1,2 in non-fertilizing plot, that of the whole varieties in all fertilizing plot is 3, 4, 5 and the mean floral axis of Sunwol is N-P2O5-K2O = 10-5-15, 20-10-30, 30-15-45 kg/10a and 18-0-18 of complex fertilizer and 21-17-17kg/10a and it is small in all fertilizing plot. The yield of dry root per 10a is high in all fertilizing grooves and especially in the groove of N-P2O5-K2O = 30-15-45 kg/10a, it is 372.6kg, in the groove of 18-0-18kg/10a using complex fertilizer, it is 389.1kg and in that of 21-17-17kg/10a, it is 376.7kg.6.7kg.
The result of this experiment which are conducted, to improve the cultivation technology of Alisma plantago, to increase its quantity and to contribute for stable production with Yongiun local group by examining the optimal planting density and transplanting period of double cropping of Alisma plantago in the southern region. The characters of plant height, leaf width and length tend to be reduced as the seeding period is later by the order of the 10th, 20th and 30th of July. The period required for flowering is reduced as the transplanting period is later and dense planting is applied. Plant height, the number of leaves and yield of dry root have much quantity at the dense planting density of 20× 15cm as they are transplanted later in the 30th of August or the 10th of September, but they are rather less in sparse planting density of 20×25cm or 20×35cm.
This test checked jujube witches'-broom disease, sumac witches'-broom disease, paulonia witches'- broom disease, and mulberry dwarf disease whether or not they were infected by phytoplasma, using universal and specific primers. Upon treatment of DNA amplified by PCR of phytoplasma with Alu I , Hpa II and Sat I restricted enzymes, distinction of phytoplasmas was possible. Particularly, phytoplasma of each host was distinguishable by treatment of Hpa II restricted enzyme. Meanwhile, analysis of restricted enzymes of jujube witches'-broom disease showed a higher infectivity of phytoplasmas of two origins. There were a lot of relations between jujube witches'-broom disease and sumac witches'-broom disease, and between paulonia witches'-broom disease and mulberry dwarf disease.
We compared microstructural features of the ordered cell and disordered leaves in Citrus junos Sieb. by electron microscopy. In the cell of the ordered leaves, many chloroplasts and large vacuoles were particularly observed. Also a lot of vessel, companion cell and big nucleus were presented in vascular bundle regions. The mitochondria and the other organelles were interspersed among the chloroplasts in a thin, peripheral layer of cytoplasm. The chloroplast possessed typical grana and intergranal lamellae, numerous starch grains and a few small osmophilic globules. Besides, microbodies were closely associated with the mitochondria and the chloroplast. The process of the formation of the secondary cell wall from primary cell wall was observed the vessel elements, the tonoplast wall and the secondary cell wall. It was observed that the oil sac with the unique perfume distributed the adjacent cell wall. In the cell of disordered leaves, the all of the organelles were thrust toward the cell wall due to the fusion of vacuoles in the cells. It was observed that a lot of the very small particles spreaded in the cytoplasm. The loss of unique perfume of the leaves was resulted in the destruction of the oil sac. Also, there was not observed grana, lamellae, starch and osmophillic globules in the chloroplast. The small distributed organelles was not observed but the elongation of the cell wall was proceed no longer. Therefore, the plasma membrane diverged from the cell wall. All of organelles in the cell had poor function and deformation. A massive vacuole was fulfilled in single cell and the vacuole contains a lot of large and small particles. The organelles were presented on the side of the cell wall according to the enlargement of vacuole and they were observed to be breakdown.
