        In the last 10 years, porcine somatic cell nuclear transfer to generate transgenic pig has been performed tremendous development with introduction and knockout of many genes. However, efficiency of porcine somatic cell nuclear transfer is still low and embryo transfer (ET) is one of important step for production efficiency. In porcine ET for production of transgenic cloned pig, we can consider many of points to increase production rates. In respect of seasonality and weather, porcine ET usually is not performed in summer and winter. Cloned transgenic embryos must be transferred into reproductive tracts of recipients where embryos are located after natural fertilization with similar estrous cycle. If cloned embryos with 2∼4 cell stage are transferred, they must be transferred into oviducts in periovulatory stage. Number and deposition sites of transferred cloned embryos are important. And we must compare the methods of ET between surgical and non-surgical ones in respect of production efficiency. Sow recipients after natural estrus is most preferred recipients however its cost is must be considered. Here we will review many of current studies about porcine embryo transfer to increase production efficiency of transgenic pigs and strategies for further studies.

        The wind field and precipitable water vapor over the Tibetan Plateau are analyzed using the numerical model WRF. The spatial and vertical distributions of the relevant meteorological factors are summarized, providing evidence for selecting and further evaluating an astronomical site. This study serves as a further demonstration towards astro-climate regionalization, and provides us with an essential database for an astronomical site survey over the Tibetan Plateau.

        A growing body of evidence has been supporting the existence of so-called "dark molecular gas" (DMG), which is invisible in the most common tracer of molecular gas, i.e., CO rotational emission. DMG is be- lieved to be the main gas component of the intermediate extinction region from Av~0.05-2, roughly corresponding to the self-shielding threshold of H2 and 13CO. To quantify DMG relative to HI and CO, we are pursuing three observational techniques; HI self-absorption, OH absorption, and THz C+ emission. In this paper, we focus on preliminary results from a CO and OH absorption survey of DMG candidates. Our analysis shows that the OH excitation temperature is close to that of the Galactic continuum back- ground and that OH is a good DMG tracer co-existing with molecular hydrogen in regions without CO. Through systematic "absorption mapping" by the Square Kilometer Array (SKA) and ALMA, we will have unprecedented, comprehensive knowledge of the ISM components including DMG in terms of their temperature and density, which will impact our understanding of galaxy evolution and star formation profoundly.

        The aim of this research is to expand application areas of the advanced manufacturing concept and technology based on combining the traditional weaving techniques with digital forming technology. Through the man-machine interface of traditional weaving techniques of processing machines and craftsmanship complex data visualization and interactive operation, Virtual Reality Technology can realize computer model images, which is a kind of new form of artistic language and the key of VR art presenting its unique advantages. Combined with digital technology, the production of raw materials can generate 2D graphics, and 3D object can also be generated by using rapid solidification laser material preparation and forming technology (3-Dimension Printing). In one word, the traditional weaving techniques of processing machine and craftsmanship realized the complete inheritance. In this paper, main content are 4 parts including the analysis of ethnic traditional weaving technology in southwest areas of China, the usage of fashion digital simulation forming technology, the combination of traditional weaving technology with digital forming technology, and expanding of digital customized technology. We started our research work using grass linen and its weaving machine. In the end, the conclusion of study will be used to build the digital museum online for Southwest Ethnic Traditional Weaving Technology Museum, which is also bridging around world marketing for traditional culture.

        Benzene was oxidized by binary oxidants composed of nitric acid and hydrogen peroxide at 80℃. The product obtained was analyzed with gas chromatograph-mass spectrometer. Eight high value compounds, 2-nitrophenol, 2-chloro-6-nitrophenol, 4-chloro-2- nitrophenol, 2-chloro-4-nitrophenol, 2,4-dinitrophenol, 4-nitrophenol, 2,6-dinitrophenol and 2-chloro-4,6-dinitro-phenol were found, which they have high contents in the range from 4.28% to 32.52%. These compounds are very widely used in organic synthesis. e.g., synthesizing dye, medicines and chemical reagents, pesticide, explosive, polymer, etc.

        Skin is the outermost organ and acts as a barrier between the organism and environment. Skin protects the organism from environmental insults, such as chemicals, pathogens, and UV light. Much of the protective function of skin is dependent on the epidermis, a multi-layered epithelium that is composed of various cell types such as keratinocytes and melanocytes. Keratinocytes produce protective components through a sophisticated differentiation process. Disturbance of keratinocyte differentiation is related to several skin diseases such as psoriasis and atopic dermatitis. In this study, we prepared extract of combined medicinal plants (ECMP) consisting of Taraxacum platycarpum H. Dahlstedt, Heartleaf Houttuynia, Glycyrrhiza uralensis Fischer, and root bark of Ulmus davidiana. We demonstrated that ECMP enhanced keratinocyte differentiation and barrier functionality using an in vitro cell culture system and in vivo animal test. Treatment of cultured keratinocytes with ECMP resulted in induction of keratinocyte differentiation, as evidenced by increased differentiation markers such as involucrin, loricrin, and filaggrin. In line with these results, ECMP decreased proliferation of keratinocytes cultured in vitro. ECMP applied topically to tape-stripped mouse skins accelerated reduction of transepidermal water loss (TEWL), indicating fast recovery of barrier function. Immunohistochemistry showed that ECMP increased the filaggrin level in tape-stripped mouse skins. These results suggest that ECMP may be applicable for keratinocyte differentiation-related skin diseases.

        Crystals of proteinaceous insecticidal proteins, Cry proteins, produced by Bacill us thuringiensis (Bt) have been generally used used to control insect pests. In this st udy, through the 3D structure prediction and accompanying mutagenesis study for the Mod-Cry1Ac, 7 and 16 amino acid residues from domain I and II, respectively, responsible for its insecticidal activity against larvae of Plutella xylostella, Spodopt era exigua and Ostrinia furnacalis were identified. To construct novel cry genes wi th enhanced insecticidal activity, we randomly mutated these 24 amino acid sequen ces by in vitro muti site-directed mutagenesis, resulting in totally 34 mutant cry gen es. For further characterization, these mutant cry genes were expressed as a fusion protein with polyhedrin using baculovirus expression system. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded in to polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activit ies of these mutant Cry proteins against to larvae of P. xylostella, S. exigua, and O. furnacalis were assayed, they showed higher or similar insecticidal activity compar ed to those of Cry1Ac and Cry1C. Especially, among them Mutant-N16 showed th e highest insecticidal activity against to both of P. xylostella, S. exigua and Ostrinia furnacalis. Therefore, Mutant-N16 is estimated to have the potential for the efficac ious bioagent.

        This study examined how Chinese college students react to their writing teachers’ corrective feedback (CF). A total of 1,077 students completed a survey questionnaire consisting of two parts: participants’ general information and their reactions to teachers’ CF. It was found that students had positive feelings towards CF, and that they gave the most attention to the problems at the micro-level. They believed that teacher CF could help them greatly with revision in organization, vocabulary, and grammar, but that it was not so helpful with problems in content. Failure to comprehend or make corrections based on their teacher CF was reported. In such cases, some students tended to employ very limited strategies, or they responded to CF in a passive manner. Accordingly, it is suggested that further qualitative research also be conducted to obtain a fuller and finer picture of students’ reactions to their teacher’s CF on their writings.

        Chinese nationals already make up one quarter of global luxury consumption and therefore represent the biggest group of luxury consumers worldwide (Bain & Co, 2012). It seems that there exists a considerable imbalance between the Chinese consumption and production of luxury. Many Western luxury consumers can hardly name a single Chinese luxury brand. However, although it is not on the radar of many experts and most consumers yet, the luxury business community in China is already emerging in big steps. China became an important manufacturing hub for semi-finished luxury products, which already helped building-up industry-specific know-how. The country is also best representing the entrepreneurial spirit in the luxury segment; there is a variety of Western and Chinese market players setting up China-specific luxury brands. New Chinese luxury brands include, for instance, Longio (watches), Mary Ching (shoes), NE Tiger (wedding dresses), Qeelin (jewelry) and Shang Xia (apparel, interior decoration; Heine and Phan, 2013). For the last decades, consumers and marketers strongly believed in the basic principle: Western stuff is hot – and Chinese stuff is not (O'Cass and Siahtiri, 2013). So are Chinese consumers actually ready for Chinese luxury brands? The objective of this paper is to analyze the challenges and strategies of Chinese luxury start-ups – with reference to the attitudes and preferences of Chinese luxury consumers. As Chinese luxury consumers and brands are still underexplored objects of investigation, which cannot be understood adequately by mere quantitative research, we employ an explorative approach with reference to the Grounded Theory. Based on a selection of (potential) Chinese luxury brands, we derive criteria that help answering the question: What are actually Chinese Luxury Brands? The “Chineseness” of a luxury brand depends, for instance, on the location of its headquarters (Mainland, Hongkong or even Taiwan) and the nationality of key people involved including the founders, designers, artisans and the CEO. Based on an analysis of potential Chinese luxury brands, we identified nine major types of players in the Chinese luxury market and their strategies to create Chinese luxury brands. Major Chinese luxury players include, for instance, Western luxury corporations, Chinese designers and Chinese (state-controlled) corporations. It already became quite common for Western luxury corporations to set-up their own more or less “Chinese” luxury brand. For instance, Hermès created Shang Xia and Estée Lauder Osiao (cosmetics). Other options for Western luxury brands include taking over Chinese luxury ventures (such as Shanghai Tang by Richemont) or acquiring Chinese premium brands or traditional Chinese handicraft brands in order to upgrade them into luxury brands. This is done, for instance, by LVMH group, who acquired a majority share in the time-honored winemaker Changyu. Many well-known Western luxury brands such Chanel or Dior were set up by designers. Accordingly, there exist already some successful Chinese fashion designer brands such as NE Tiger (since 1992 by Zhifeng Zhang), Maryma (by Mary Ma since 1999), Rose Studio (by Guo Pei since 1997), and Xander Zhou (since 2007), who may develop into (international) luxury brands. Similar to entrepreneurs, some big Chinese (state-controlled) corporations corporations aim expanding into the luxury segment because they expect attractive business opportunities, but may also aim to improve their corporate and/or brand image. Examples include Shanghai Vive by Jahwa group and SheJi-Sorgere by China Garments group. The combination of market players and their desire and strategies in creating country-specific luxury brands is truly unique to China. For instance, big luxury houses such LVMH never perceived another market such as Germany or the US as that specific and above all relevant enough to set up a specific luxury brand especially for that market. In the subsequent section, the paper shifts its focus to the Chinese luxury consumers. At the very core of this discussion lie the mobile and multi-faceted (and sometimes ambiguous) associations of China with luxury perception (Rovai 2014). These relate to both consumer behavior (the Chinese market for luxury) and production (the reputation of Chinese producers of luxury goods and services). Based on a content-analysis of current literature and press, we categorize and discuss the factors that may impact the attitudes of Chinese consumers towards Chinese luxury brands with reference to the major luxury consumer segments in China. These factors include, for instance, the increased sophistication of consumers, the international exposure of Chinese consumers, the rejection of “potlatch behaviour”, the empowerment of women in Chinese society, the emergence of a creative class in China, the slow fashion movement, the reversal of intergenerational influences, and the “sinofication” trend – the New Cultural Revolution led by the “young emperors” (or “Sugar Generation”), the generation born after 1978 (Wang and He, 2014). Subsequently, we are analyzing the attitudes and preferences of Chinese luxury consumers with a small-scale qualitative survey. We concentrate on the “young emperors” generation and especially on the segments of white collar workers and the second generation rich. We conducted several in-depth interviews about their luxury preferences and then, following an empathic research approach, we went shopping together with the respondents to develop a better and real-life understanding of these luxury consumer segments. The results are content-analyzed and presented with reference to case studies of Chinese luxury brands. On the one hand, the sophistication and sinofication trends of the Chinese creative class may threaten the big Western luxury brands, but on the other hand, they create a basis for the development of Chinese luxury start-ups and increases the chances of Western luxury SME’s and connoisseur brands in China. Based on our analysis of the Chinese luxury consumer behavior, the paper concludes with some recommendations for the development of luxury brands in China.

        In this paper, researchers investigated current fashion brands’ social media micro-blogging marketing strategy and consumers’ word of mouth reactions. More than 5,000 of the micro-blogs posted by fashion brands and 143,000 of customers’ comments were analysed in this study. Researchers firstly investigated the overall micro-blogging marketing structure for each fashion brand and compared them. Then researchers identified the type of expression pattern of each fashion brand currently in the micro-blogging context, negative or positive sentiments. Researchers found that fashion brands are using different micro-blogging marketing strategies. Forever21 used 61% micro-blogs for customer communication. H&M posted very diverse micro-blogs content to their official account. Their main micro-blogs were used for new product promotion (43%) and brand’s live event broadcasting (33%). Luxury brands, such as Burberry, more than 52% of micro-blogs posted last year were used for new product promotion and 36% of their micro-blogs contents included celebrities’ images of wearing Burberry product. Chanel used 60% of their micro-blogs to broadcast and introduce their brand events. There was no sale information posted on Chanel and Burberry’s micro-blogs account. Through sentiment analysis, researchers also found the brands have very positive electronic word of mouth (e-WOM). Particular, luxury fashion brands are having better e-WOM than fast fashion brands.

        Many advanced ground-based solar telescopes improve the spatial resolution of observation images using an adaptive optics (AO) system. As any AO correction remains only partial, it is necessary to use post-processing image reconstruction techniques such as speckle masking or shift-and-add (SAA) to reconstruct a high-spatial-resolution image from atmospherically degraded solar images. In the New Vacuum Solar Telescope (NVST), the spatial resolution in solar images is improved by frame selection and SAA. In order to overcome the burden of massive speckle data processing, we investigate the possibility of using the speckle reconstruction program in a real-time application at the telescope site. The code has been written in the C programming language and optimized for parallel processing in a multi-processor environment. We analyze the scalability of the code to identify possible bottlenecks, and we conclude that the presented code is capable of being run in real-time reconstruction applications at NVST and future large aperture solar telescopes if care is taken that the multi-processor environment has low latencies between the computation nodes.

        The present study assessed the effect of FSH and LH on oocyte meiotic, cytoplasmic maturation and on the expression level and polyadenylation status of several maternal genes. Cumulus-oocyte complexes were cultured in the presence of FSH, LH, or the combination of FSH and LH. Significant cumulus expansion and nuclear maturation was observed upon exposure to FSH alone and to the combination of FSH and LH. The combination of FSH and LH during entire IVM increased the mRNA level of four maternal genes, C-mos, Cyclin B1, Gdf9 and Bmp15, at 28 h. Supplemented with FSH or LH significantly enhanced the polyadenylation of Gdf9 and Bmp15; and altered the expression level of Gdf9 and Bmp15. Following parthenogenesis, the exposure of oocytes to combination of FSH and LH during IVM significantly increased cleavage rate, blastocyst formation rate and total cell number, and decreased apoptosis. In addition, FSH and LH down-regulated the autophagy gene Atg6 and upregulated the apoptosis gene Bcl-xL at the mRNA level in blastocysts. These data suggest that the FSH and LH enhance meiotic and cytoplasmic maturation, possibly through the regulation of maternal gene expression and polyadenylation. Overall, we show here that FSH and LH inhibit apoptosis and autophagy and improve parthenogenetic embryo competence and development.

        The objective of this study was to examine the effects of high concentrations of glucose on porcine parthenotes developing in vitro. Addition of 55 mM glucose to the culture medium of embryos at the four-cell-stage significantly inhibited blastocyst formation, resulting in fewer cells in blastocyst-stage embryos and increased levels of apoptosis and autophagy compared to control. Quantitative reverse transcriptase (RT) PCR analysis revealed that the expression of pro-apoptotic genes (Caspase 3, Bax and Bak) and autophagy genes (Atg6 and Atg8/Lc3) were increased significantly by the addition of 55 mM glucose to the culture medium compared to control. MitoTracker Green fluorescence revealed a decrease in the overall mitochondrial mass compared to control. However, the addition of 55 mM glucose had no effect on mRNA expression of the nuclear DNA-encoded mitochondrial-related genes, cytochrome oxidase (Cox) 5a, Cox5b and Cox6b1. These results suggest that hyperglycemia reduced the mitochondrial content of porcine embryos developing in vitro and that this may hinder embryonic development to the blastocyst stage and embryo quality by increasing apoptosis and autophagy in these embryos.

        Developmental potential of cloned embryos is related closely to epigenetic modification of somatic cell genome. The present study was to investigate the effects of applying histone deacetylation inhibitor, trichostatin A (TSA) to activated porcine embryos on subsequent development of porcine parthenogenetic and nuclear transfer embryos. Electrically activated oocytes were treated with 5 nM TSA for different exposure times (0, 1, 2 and 4 hr) and then the activated embryos were cultured for 7 days. The reconstructed embryos were treated with different concentrations of 0, 5, 10 and 25 nM TSA for 1 hr. Also 5 nM TSA was tested with different exposure times of 0, 0.5, 1, 2 and 4 hr. And fetal fibroblast cells were treated with 50 nM TSA for 1, 2 or 4 hr and with 5 nM TSA for 1 hr. Cumulus-free oocytes were enucleated and reconstructed by TSA-treated donor cells and electrically fused and cultured for 6 days. In parthenogenetic activation experiments, 5 nM TSA treatment for 1 hr significantly improved the percentage of blastocyst developmental rates than the other groups. Total cell number of blastocysts in 1 hr group was significantly higher than other groups or control. Similarly, blastocyst developmental rates of porcine NT embryos following 5 nM TSA treatment for 1 hr were highest. And the reconstructed embryos from donor cells treated by 50 nM TSA for 1 hr improved the percentage of blastocyst developmental rates than the control group. In conclusion, TSA treatment could improve the subsequent blastocyst development of porcine parthenogenetic and nuclear transfer embryos.

        Plasmids are crucial for determining the pathogenicity and host range of organisms of the Bacillus thuringiensis strains. In this research, a novel serogroup of B. thuringiensis serovar mogi (H3a3b3d), which showed mosquitocidal activity against Anopheles sinensis and Culex pipiens pallens, was isolated from fallen leaves in Mungyeong city, Republic of Korea. In contrast to the complicated plasmid profiles of B. thuringiensis H3 serotype strains, the B. thuringiensis serovar mogi contained two megaplasmids (> 30 MDa) on which the toxin genes were occasionally located. Sequence analysis using 454-pyrosequencing revealed that there are 7 putative cry genes, cry19Bb1, cry73Aa, cry40orf2, cry20Bb1, cry27Ab1, cry56Ba1 and cry39orf2, distributed on the two different megaplasmids, respectively. These cry genes were cloned to the Escherichia coli-B. thuringiensis shuttle vector, pHT1K under the control of its own promoter and p1KSD, which is a recombinant expression vector containing cyt1Aa promoter combined with the STAB-SD sequence, and then introduced into an acrystalliferous B. thuringiensis Cry-B strain for further molecular characterization. To investigate the role of these genes in crystal production, the expression profiles of these toxin genes were analyzed by quantitative PCR (qPCR) from the wild type strain. These results clearly indicate that the cry39orf2 was the dominant ingredient in the crystal. This novel 3a3b3d type strain, B. thuringiensis serovar mogi, could be used as a good resource for studying unknown mosquitocidal cry genes.

        ORF78 (ac78) of Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is a baculovirus core gene of unknown function. To determine the role of ac78 in baculovirus life cycle, an ac78-deleted mutant AcMNPV, Ac78KO, was constructed. Quantitative PCR analysis revealed that ac78 is a late gene in the viral life cycle. After transfection into Spodoptera frugiperda cells, Ac78KO produced a single-cell infection phenotype indicating that no infectious budded viruses (BVs) were produced. The defection in BV production was also confirmed by both viral titration and Western blot. However, viral DNA replication is unaffected. Analysis of BV and occlusion derived virus (ODV) revealed that AC78 is associated with both forms of the virions and is a structural protein located to viral envelope. Electron microscopy showed that ac78 also plays an important role in embedding of ODV into occlusion body. This study therefore demonstrates that AC78 is a late virion associated protein and is essential for the viral life cycle.
