        A strain of Amphidinium species was established from samples collected from the intertidal zone of a sandy beach of Jeju Island, Korea. Its cells were 13.0-15.0 μm in length and 10.0-13.0 μm in width. Its cell shape was round or oval and dorsoventrally flat. A pyrenoid was located in the center of the cell and a nucleus was posteriorly located. Its epicone was small and left-deflecting. Its cingulum had V-shape on the ventral side, forming a ventral ridge and extending to the sulcus. Polygonal amphiesmal vesicles and ring-shaped body scales not described previous were observed on the surface of the cell. Its morphological features were consistent with those of previously described Amphidinium fijiense. Phylogeny based on ITS region and LSU rDNA sequences revealed that this Amphidinium isolate was clearly clustered with other A. fijiense strains, but separated from other Amphidinium species. These results indicate that this Amphidinium isolate is A. fijiense. This study reports its presence for the first time in the intertidal zone of a sandy beach of Jeju Island, Korea.

        Pretilachlor (PRE)는 전 세계적으로 사용되는 아세트아닐리드계 제초제이며 수생 광영양생물에 미치는 영향에 대해서는 알려진 바가 거의 없다. 본 연구에서 해양 와편모 조류 P. minimum를 대상으로 PRE의 생리적, 분자적 독성 영향을 평가하였다. 그 결과, PRE는 P. minimum의 성장률, 색소, 광합성 지표를 유의하게 감소시켰다. 또한, 광합성 관련 유전자 PmpsbA, PmpsaA 및 항산화 단백질 PmGST의 상대적 유전자 발현율과 세포 내 ROS 증가가 유의하였다. 이는 PRE가 P. minimum의 광합성 효율 저하 및 광계 손상을 야기하며, GST가 세포 내 산화 스트레스 및 PRE의 무독화에 관여함을 보여준다. 본 연구 결과는 PRE를 비롯한 아세트아닐라이드계 제초제가 해양 와편모조류의 세포 독성을 야기하며, 광합성 저해가 스트레스의 주요 원인임을 제시한다. 다른 아세트아닐라이드계 제초제가 비표적 광영양 생물에게 미치는 독성 영향은 아직 연구가 부족하므로, 향후 연구에서 다양한 생물종을 대상으로 추가 연구가 필요하다.

        This study reported on the phytoplankton community and seasonal changes in the Seoul passage section and downstream in the Han River in 2012. Field samples were collected monthly from the upper (Paldang), middle (Cheongdam), and downstream (Seongsan) areas of the Seoul passage section. Water temperature, DO, pH, and conductivity were measured at each station. All environmental factors measured were recorded similarly at the three stations. The water temperature ranged from 2 to 30°C and the dissolved oxygen ranged from 4.8 to 9.1 mg L-1, showing typical patterns of temperate regions. The phytoplankton cell concentrations ranged from 990 cells mL-1 (Paldang, December) to 2.9×104 cells mL-1 (Seongsan, March), and the chlorophyll-a content showed similar patterns to the cell numbers. The phytoplankton community was comprised of 75 genera and 95 species, including 37 diatoms, 29 Chlorophyta, 11 cyanobacteria, and two dinoflagellates. The number of species that appeared seasonally varied greatly, from nine species (Paldang, May) to 35 species (Cheongdam, December). Diatoms were the most dominant in all stations and seasons, except in summer. In contrast, chlorophytes and cyanobacteria showed sporadic high numbers in the summer and fall seasons. Four diatoms Stephanodiscus hantzschii f. tenuis, S. hantzschii, Fragilaria sp., and Aulacoseira spp., a chlorophyte Actinastrum hantzschii, and a cyanobacterium Microcystis sp. were each present in proportions greater than 10%. This study provides fundamental data from phytoplankton communities and environmental factors in the Han River for understanding water quality for long-term environmental monitoring.

        국내 금강수계의 하구역에 위치한 하제항 암벽으로부터 기중성 남세균이 채집되어, 단조류 배양되었다. 2개의 남세균 배양주로부터 형태적 형질과 TEM에 의한 세포 미세구조 특징을 관찰하였다. 또한 16S rDNA 염기서열을 규명하여 BLAST 검색, 염기서열 유사도, 유전거리 및 계통 분석을 실시하였다. 국내 배양주는 Wilmottia murrayi와 계통학적으로 동일한 clade를 형성하였고, 99% 이상의 유사도를 나타냈다. TEM을 통한 세포 미세구조는 틸라코이드가 여러겹으로 세포막 주변에 분포하는 특징을 나타냄으로써 Coleofasciculaceae과의 특징과 일치하였다. 이상의 결과를 통해 W. murrayi를 국내 미기록속 및 미기록종으로 보고하였다.

        The incidence of freshwater algal bloom has been increasing globally in recent years and poses a major threat to environmental health. Cyanobacteria are the major component of the bloom forming community that must be monitored frequently. Their morphological identities, however, have remained elusive, due to their small size in cells and morphological resemblances among species. We have analyzed molecular diversity and seasonal changes of cyanobacteria in Paldang Reservoir, Korea, using morphological and 16S rRNA pyrosequencing methods. Samples were collected at monthly intervals from the reservoir March-December 2012. In total, 40 phylotypes of cyanobacteria were identified after comparing 49,131 pyrosequence reads. Cyanobacterial genera such as Anabaena, Aphanizomenon, Microcystis and Synechocystis were predominantly present in samples. However, the majority of cyanobacterial sequences (65.9%) identified in this study were of uncultured origins, not detected morphologically. Relative abundance of cyanobacterial sequences was observed as high in August, with no occurrence in March and December. These results suggested that pyrosequencing approach may reveal cyanobacterial diversity undetected morphologically, and may be used as reference for studying and monitoring cyanobacterial communities in aquatic environments.
