        Vitamin K1 (VK1) has been widely used as a coumarin antagonist and for the treatment of hemorrhagic disease in veterinary practice. However, the potential mechanism of adverse reaction after VK1 injection has been not fully elucidated. In this study, two cases of anaphylactic reactions after subcutaneous VK1 injection were presented, and then an experimental study was performed to further characterize the anaphylactic reactions. Two dogs developed anaphylactic reactions after subcutaneous VK1 injections and were promptly treated with antihistamines and glucocorticoids, after which abnormal signs related to anaphylaxis disappeared. Subsequently, a study was undertaken to ascertain the nature of the adverse reactions to subcutaneous VK1 injection. Six healthy laboratory beagle dogs received subcutaneous VK1 administrations once daily for eight days. They were monitored for clinical signs after each injection, and blood samples were collected for the measurement of plasma histamine and immunoglobulin E concentrations using enzyme-linked immunosorbent assay. All six dogs showed mild angioedema after the VK1 injections. The dogs also displayed clinical signs including sneezing, coughing, skin reddening, excess salivation, pawing the ground, and somnolence on days 4, 6, and 8. Plasma histamine and immunoglobulin E concentrations were significantly increased by the repeated injections. In summary, this study describes anaphylactic reactions resulting from subcutaneous VK1 administration in dogs. Clinicians should be aware that the repeated subcutaneous injection of VK1 can trigger an anaphylactic reaction in dogs.

        Enamel knot (EK)—a signaling center—refers to a transient morphological structure comprising epithelial tissue. EK is believed to regulate tooth development in early organogenesis without its own cellular alterations, including proliferation and differentiation. EKs show a very simple but conserved structure and share functions with teeth of recently evolved vertebrates, suggesting conserved signaling in certain organs, such as functional teeth, through the course of evolution. In this study, we examined the expression patterns of key EK-specific genes including Dusp26 , Fat4, Meis2, Sln , and Zpld1 during mice embryogenesis. Expression patterns of these genes may reveal putative differentiation mechanisms underlying tooth morphogenesis.

        Acute vascular rejection has been known as a main barrier occurring in a xenograted tissue of alpha 1,3-galactosyltransferase knock-out (GalT KO) pig into a non-human primate (NHP). Adenosine which is a final metabolite following sequential hydrolysis of nucleotide by ecto-nucleotidases such as CD39 and CD73, act as a regulator of coagulation, and inflammation. Thus xenotransplantation of CD39 and CD73 expressing pig under the GalT KO background could lead to enhanced survival of recipient NHP. We constructed a human CD39 and CD73 expression cassette designed for endothelial cell-specific expression using porcine Icam2 promoter (pIcam2-hCD39/hCD73). We performed isolation of endothelial cells (pAEC) from aorta of 4 week-old GalT KO and membrane cofactor protein expressing pig (GalT-MCP/-MCP). We were able to verify that isolated cells were endothelial-like cells using immunofluorescence staining analysis with von Willebrand factor antibody, which is well known as an endothelial maker, and tubal formation assay. To find optimal condition for efficient transfection into pAEC, we performed transfection with GFP expression vector using four programs of nucleofection, M-003, U-023, W-023 and Y-022. We were able find that the program W-023 was optimal for pAEC with regard to viability and transfection efficiency by flow cytometry and fluorescent microscopy analyses. Finally, we were able to obtain GalT-MCP/-MCP/CD39/CD73 pAEC expressing CD39 and CD73 at levels of 33.3% and 26.8%, respectively. We suggested that pACE isolated from GalT-MCP/-MCP pig might be provided as a basic resource to understand biochemical and molecular mechanisms of the rejections and as an alternative donor cells to generate GalT-MCP/-MCP/CD39/CD73 pig expressing CD39 and CD73 at endothelial cells.

        Runx2 and Osterix, the transcription factors for osteoblast differentiation, are known as fundamental factors to regulate the development of calcified tissues. However, the biological functions of these factors in the development of the periodontal tissues remain unclear. In this study, we investigated the distribution of Runx2 and Osterix during periodontal tissue development of the mice. Mandibles from 14-day-old mice were prepared for paraffin section. Serial sections of the mandible containing 1 st molar tooth germs were obtained as a thickness of 7 μm. Some sections were stained with hematoxylin and eosin. Others were used for immunohistochemistry for PCNA, Runx2, and Osterix. Epithelial cells in growing end of Hertwig’s epithelial root sheath (HERS) and mesenchymal cells adjacent to the growing end of HERS expressed PCNA. Undifferentiated mesenchymal cells and hard tissue forming cells like cementoblasts and osteoblasts in early stage of differentiation expressed Runx2. Fully differentiated cementoblasts and osteoblasts secreting matrix proteins expressed Osterix. However, the cells terminated the matrix formation did not express Osterix. Periodontal ligament cells expressed Runx2 and Osterix. Pulp cells expressed Runx2 only.These results suggest that Runx2 and Osterix might regulate the differentiation of cementoblasts in the same manner as osteoblasts. Runx2 might participate in the process of cementoblast differentiation in early stage, whether Osterix might regulate the maturation and matrix synthesis of the cells.

        본 연구의 목적은 시간 분해능이 향상된 비지역적 평균 (fast non local means, FNLM) 노이즈 제거 알고리 즘을 모델링하여 광학 현미경 영상에서의 적용 가능성을 확인하는 것이다. 이를 위해 실제 흰쥐 (mouse)의 첫째어금니 치아를 사용하여 영상을 획득한 후 기존에 널리 사용되고 있는 노이즈 제거 알고리즘과 제안 하는 FNLM 알고리즘을 각각 적용하여 비교하였다. 정량적 평가는 대조도 대 잡음비 (contrast to noise ratio, CNR), 변동계수 (coefficient of variation, COV), 그리고 최근에 개발된 no reference 기반의 방법인 natural ima ge quality evaluator (NIQE)와 Blind/referenceless image spatial quality evaluator (BRISQUE)를 사용하였다. 결과적으로 모든 정량적 평가 인자에서 제안하는 FNLM 노이즈 제거 알고리즘이 가장 우수한 값을 나타내었다. 특히나 치아의 전체적인 형태학적 영상을 분석할 수 있는 NIQE와 BRISQUE 인자는 원본영상에 비하여 각각 1.14와 1.12배 향상됨을 확인할 수 있었다. 결론적으로 소동물 치아 광학 현미경 영상에서의 FNLM 노이즈 제거 알고리즘의 유용성 및 가능성을 증명하였다.

        This study aimed to investigate the dilapidated Fine View Square located in the city of Gangneung and analyze problems for building it into a sustainable space under an effective management plan. Further to creating an ecological base for restoring the natural circulation, a restorative method for the damaged area, a spatial assignment by the UNESCO’s Man and Biosphere Programme (MAB), and an urban ecological park, we not only wanted to provide an environment that is citizen friendly, but also a practical and realizable project to connect with the various methods and programs mentioned above, while utilizing the Ministry of the Environment 's ecosystem conservation fund. In conclusion, we found that it would be possible to contribute to building an urban ecological park the Ministry of Environment proposed while restoring the lost natural circulation in Gyodong Square in the city of Gangneung and ecological soundness of the city along with the reduced heat island effect and the increased biological diversity and Ecological Soundness.
