        본 연구에서는 멸종위기종이 서식하는 4개 하천 (금강, 지천, 황지천, 섬진강)에서 환경유전자 (environmental DNA, eDNA)와 보편적 어구를 이용한 조사 방법을 적용하여 지점별 종 다양성을 확인하고, 이를 통해 eDNA의 활용을 고찰 하였다. eDNA 조사를 통해서 확인된 종 수는 지점 평균 (± 표준편차) 19종 (±4.4)이며, 이는 어구를 이용한 정량 조사의 10종 (±4.8)과 비교하여 높게 나타났다. 대부분의 지점에서 eDNA 조사가 어구를 이용한 조사보다 효율이 높게 나타났다. 반면 eDNA 조사 결과 고유종 및 근연종에 대해서 동정의 오류가 확인되어, universal primer (MiFish primer set)에 대한 국내 적용의 한계를 확인하였다. 또한 멸종위기종의 서식 여부도 일부 종에 대해서 eDNA 조사 결과가 현장 조사 및 문헌과의 차이를 보였다. 현재 개발된 universal primer는 국내에서 서식하는 모든 담수종의 서식을 확인 하는 데 있어서 결과의 신뢰성을 담보할 수 없기 때문에 universal primer의 보완 및 개발이 필요하며, 멸종위기종과 같은 특정종의 서식 확인을 위해서는 종특이적 마커 개발을 통한 적용이 고려되어야 한다. 마지막으로 eDNA의 현장 조사 방법에 대한 매뉴얼이 개발될 경우, 수생태계 조사에 대한 활용성이 증대될 수 있을 것이다.

        In this study, we developed 11 microsatellite markers specific to A. crataegi using NGS to investigate the genetic relationships of A. crataegi populations from South Korea to circumferential Asian countries (China, Russia, Mongolia, and Japan). Further, two mitochondrial DNA (mtDNA) gene segments (COI and CytB) were sequenced from the samples. The population- and individual-based Principal Coordinates and STRUCTURE analyses collectively suggested that the South Korean population of A. crataegi is most differentiated from the Japanese population, whereas it was closer to Mongolian and Chinese populations. These results collectively suggest that northern populations, in particular, Mongolian populations can be considered as the most genetically compatible one as donee population, when the reintroduction program is launched. †These authors contributed equally to this paper.

        The black-veined white, Aporia crataegi (Lepidoptera: Pieridae), which is distributed mainly in Eastern Asia is presumed to be extinct in South Korea, only with some numbers of dried specimens left, whereas the species is found casually in circumferential countries. One of the common conservation practices for such species is to launch introduction program, but prior population genetic analysis between donor and donee populations might be essential for long-term conservation. In this study, we developed 11 microsatellite markers specific to A. crataegi using Illumina paired-end sequencing to investigate the genetic relationships of A. crataegi populations from South Korea and circumferential Asian countries (China, Russia, Mongolia, and Japan). Further, two mitochondrial DNA (mtDNA) gene segments (COI and CytB) were sequenced from the samples. The population- and individual-based Principal Coordinates and STRUCTURE analyses collectively suggested that the South Korean population of A. crataegi is most differentiated from the Japanese population, whereas it was closer to Mongolian and Chinese populations. The STRUCTURE analysis based on two concatenated mtDNA gene sequences also supported different genetic composition of Japanese population from the remaining populations including that of South Korea and rather similar genetic composition between the populations of South Korea and Mongolia. These results collectively suggest that northern populations, in particular, Mongolian populations can be considered as the most genetically compatible one as doner population, when reintroduction program is launched.

        This study aims to offer basic data to effectively preserve and manage pine forests using more precise pine forests’ distribution status. In this regard, this study predicts the geographical distribution change of pine forests growing in South Korea, due to climate change, and evaluates the spatial distribution characteristics of pine forests by age. To this end, this study predicts the potential distribution change of pine forests by applying the MaxEnt model useful for species distribution change to the present and future climate change scenarios, and analyzes the effects of bioclimatic variables on the distribution area and change by age. Concerning the potential distribution regions of pine forests, the pine forests, aged 10 to 30 years in South Korea, relatively decreased more. As the area of the region suitable for pine forest by age was bigger, the decreased regions tend to become bigger, and the expanded regions tend to become smaller. Such phenomena is conjectured to be derived from changing of the interaction of pine forests by age from mutual promotional relations to competitive relations in the similar climate environment, while the regions suitable for pine forests’ growth are mostly overlap regions. This study has found that precipitation affects more on the distribution of pine forests, compared to temperature change, and that pine trees’ geographical distribution change is more affected by climate’s extremities including precipitation of driest season and temperature of the coldest season than average climate characteristics. Especially, the effects of precipitation during the driest season on the distribution change of pine forests are irrelevant of pine forest’s age class. Such results are expected to result in a reduction of the pine forest as the regions with the increase of moisture deficiency, where climate environment influencing growth and physiological responses related with drought is shaped, gradually increase according to future temperature rise. The findings in this study can be applied as a useful method for the prediction of geographical change according to climate change by using various biological resources information already accumulated. In addition, those findings are expected to be utilized as basic data for the establishment of climate change adaptation policies related to forest vegetation preservation in the natural ecosystem field.

        Kentrochrysalis streckeri Staudinger, 1880 is distributed in Russia, Mongolia, China, and North Korea, whereas K. consimilis Rothschild & Jordan, 1903 has been known to occur South Korea and Japan. In the present study we found that the specimens from South Korea were K. streckeri, rather than K. consimilis based on morphology, DNA barcode, and nuclear elongation factor 1 alpha (EF-1α) sequences. The major morphological differences between K. streckeri and K. consimilis include the shape of wing pattern elements of fore- and hindwings and male and female genitalia. A DNA barcode analysis of the South Korean specimens and K. streckeri originated from Russia showed a maximum sequence divergence of only 0.659% (four bp), whereas the barcoding sequence of K. consimilis sequenced in this study and GenBank-registered sequence (JN678086), both of which originated from Japan showed the minimum sequence divergence of 2.965% (18 bp), indicating that the Korean specimens are, in fact, K. streckeri, instead of wrongly known K. consimilis. Phylogenetic analyses both by Bayesian Inference and maximum likelihood methods well supported the monophyly of South Korean specimens and Russian K. streckeri, excluding K. consimilis. The EF-1α-based sequence and phylogenetic analyses of the two species also supported the data from DNA barcode, indicating distribution of K. streckeri in South Korea, instead of K. consimilis.

        Environmental changes such as land-use change including reclamation cause effects on the ecosystem seriously. Present study investigated community structure of hemipterans in several reclaimed lands from western coast of South Korea in 2010, because among arthropods, hemipterans are more influenced by land-use change which caused the change of dominant plants. Six reclaimed lands were selected for our study based on the ages of reclaimed land (0, 5, 12, 16, 20, and 31 years). Dominant plant species of reclaimed lands were belonging to Poaceae, Phragmites communis and Oryza sativa in Yeongsangan II, Sabkyocheon, Geumgan I and Gyehwado, Secale cereale in Seokmun, and Imperata cylindrica and Calamagrostis epigeios in Sihwa. A total of 31 species in 10 families were identified from 4475 collected hemipterans. In Sihwa, Shannon’s diversity was very low compared to other 5 reclaimed lands due to dominance of Paromius exiguus. Because I. cylindrica and C. epigeios were 1st and 2nd host plants of P. exiguus (2824 individuals only collected from Sihwa). In multivariate analysis, 6 reclaimed lands grouped into 2 major groups showing 49.8% in Bray-Curtis similarity between 2 groups. From these results, land-use change such as reclamation project may cause the outbreak of insect pests by destruction of ecosystem functions and simplification of plant community, although community structure of hemipterans may be stable over age of land reclamation.

        Hipparchia autonoe belongs to the family Nymphalidae (Lepidoptera) and is designated as an endangered insect and national monument in Korea. It only inhabits a very restricted area on Mt. Halla but is widely distributed in several Asian countries including Mongolia. A previous study conducted to understand the genetic relationship between Mt. Halla and Mongolian H. autonoe for conservation purposes suffered from a limited number of samples. Therefore, we sequenced the DNA barcode region of an additional 36 H. autonoe individuals, combined them with previous data from 19 individuals, and performed phylogenetic and population genetic analyses. Furthermore, the internal transcribed spacer 2 (ITS2) region was also sequenced from the 36 samples as a nuclear DNA marker. The existence of independent haplotypes, sequence types, and significant FST estimates (p < 0.05) between Mt. Halla and Mongolian populations indicated hampered gene flow between the populations. Nevertheless, an absence of a reciprocal monophyletic group in Mt. Halla and Mongolian populations by cytochrome oxidase subunit I gene- and ITS2-based phylogeny suggests that the genetic isolation of the Mt. Halla population from the Mongolian populations seemed not large enough to consider them independent genetic entities.

        The black-veined white, Aporia crataegi (Lepidoptera: Papilionoidea), is nearly extinct in South Korea, although substantial numbers of dried specimens are available. One of the common practices for such species is to launch re-introduction program after proper amount of genetic information are analyzed from donor and donee populations. In this study, we sequenced complete mitochondrial genome (mitogenome) of A. crataegi to design species-specific primers for subsequent population works and to further understand the mitogenome evolution in lepodiopteran Papilionoidea. The 15,140-bp long A. crataegi mitogenome that has typical sets of 37 genes is smallest among true butterfly species with overall slightly smaller size in genes and regions throughout the genome. Arrangement of the genome is identical to those of other lepidopteran mitogenomes, in which tRNA cluster located between the A+T-rich region and ND2 gene is translocated into tRNAMet, tRNAIle, and tRNAGln from ancestral arrangement, tRNAIle, and tRNAGln, tRNAMet. The A/T content of the genome at 81.3% is the highest in Pieridae, but lower than that of lycaenid species (81.7% ~ 82.7%) The high A/T content in the genome is also reflected in codon usage, accounting for 41.69% of A/T-composed codons (TTA, ATT, TTT, and ATA). Unlikely the diversified or modified usage of anticodon for tRNASer(AGN) the species of Pieridae including A. crataegi all unanimously have GCT that has been hypothesized as ancestral for Lepidoptera. A total of 111 bp of non-coding sequences are dispersed in 13 regions, ranging in size from 1–49 bp. Among them relatively longer ones (≥ 16 bp) all have relatively higher sequence identity to other regions of the genome, suggesting partial duplication of the sequences during A. crataegi evolution. As has been reported in some species of Lepidoptera, the A. crataegi A+T-region also has typically found conserved sequences (e.g., poly-T stretch, ATAGA motif, ATTTA element, microsatellite-like A/T sequence, and poly-A stretch) and one tRNA-like sequence, and this feature was commonly found in true butterfly species.

        고에너지 X-ray를 사용하는 방사선 이용 시설에서는 고에너지 광자에 의한 광중성자 등의 발생에 대한 방 사선방호 조치가 방사선 안전의 관점에서 매우 중요한 이슈 중의 하나이다. 이러한 광중성자들은 시설 외부로 누설될 경우 skyshine effect에 의해 작업종사자 및 일반 대중에게 예기기 않은 방사선 피폭을 야기할 수 있다. 이 논문에서는 9MeV X-ray를 이용한 화물검색시설을 대상으로 몬테카를로법을 이용하여 광중성자 생성량을 계산하고 이를 통해 시설 외부에서의 중성자 skyshine 방사선량률을 평가하였다.
