        Chili pepper (Capsicum annuum) is one of the most important vegetable crop for human being as a rich source of nutritions and spicy condiments. To make reference genome sequence of pepper, we sequenced the whole genome of Capsicum annuum, CM334 using Illumina/Solexa Genome Analyzer GA2. The genome size of CM334 is estimated as 3.5 Gb. A total of 716 Gb (205.96x coverages of the whole genome) of raw sequences were generated. After filtering out the low quality sequences, a total of 233 Gb (66.7x) of the raw sequences were used for assembly. Total assembled contig length and number were 2.93 Gb and 295,502, respectively. N50 and average length were 25.72 kb and 6,5 kb, respectively. By sequencial scaffolding with mate-pair sequences of 2 kb - 20 kb sizes, a total of 3.04 Gb of scaffold which is approximately 90% of the whole genome was assembled. The total number of scaffolds was 33,876 with N50 length of 1,605 kb. For annotation of the pepper genome, a total of 46 Gb of transcriptome sequences were generated from 12 different tissues using Illumina GA2 and Hi-seq 2000. We are under way of analyzing the characteristic traits of pepper using transcriptome data. The progress of pepper genome sequencing project including gene annotation, gene family analysis, comparative genomics studies on evolution of hot taste, genome expansion and fruit development will be presented in the meeting.

        Regarding carotenoids content, the genetic basis, heritability and combining ability in six red pepper inbred lines were investigated using full diallel crosses. Both additive and non-additive gene actions govern inheritance of carotenoids content. The mean square of array through variance and covariance analysis (Wr-Vr) was insignificant, which suggest that inbred lines involved in diallel cross may have no epistatic effects. The Vr/Wr graph revealed the influence of partial dominant gene action towards low carotenoids content and the absence of non-allelic interaction. The H2 component was smaller than the H1 and the [H2/4H1] component was 0.187 less than 0.25, indicating unequal proportion of positive and negative alleles in the parents. The estimates of broad and narrow sense heritability for carotenoids content were 0.956 and 0.832, respectively. The variance of general combining ability (GCA) was relatively higher than that of specific combining ability (SCA), which implied that the additive gene effects were predominant as compared to both dominant and epistatic effects for the accumulation of carotenoids in this genetic population. The values of GCA of ‘62024L1’ and ‘62067L2’ were higher than those of the other parents. These 2 inbred lines, therefore, can be considered as useful breeding materials to enhance fruit carotenoids content in other red pepper varieties.

        This study was done to characterize the anther derived regenerants (R1) including haploids and spontaneous diploids of minipaprika (cvs.‘Vine sweet-red’ ‘Vine sweet-yellow’ and ‘Vine sweet-orange’) in glasshouse. Eleven haploids (three, seven and one from red, yellow and orange, respectively) and sixteen spontaneous diploids (five, nine and two from red, yellow and orange, respectively) were grown in plastic pot with three (red, yellow and orange) anther donor (R0) F1minipaprika varieties. Regenerants were characterized for their plant and fruit characters as well as their fruit color and shape. The homozygosity of spontaneous diploid plants of each population was assessed using simple-sequence repeat (SSR) marker analysis. Haploid plants were characterized by reduced plant height, small leaves, short petiole and internode and small flower bud and all haploids showed the sterility and vice-versa in spontaneous diploid lines. The fruit biometrical traits exhibited greater variation within the spontaneous diploid plants and average value of quantitative traits is lower than standard varieties. MR-4 gave the highest yield (150.5 g) per plant followed by MY-6 (140.0 g) and MY-8 (130.5 g) and the lowest in MY-5 (31.5 g). Morphological marker such as fruit color further determinedthe microspore origin of androgenic diploids obtained in anther culture of ‘Vine sweet-red’. Of the fifteen spontaneous diploid plants, fourteen plants were identified as doubled haploids using microsatellite markers (SSR), and these homozygous lines are recommended to use in minipaprika breeding program.

        본 연구는 고온에 안정적인 TSWV 저항성 유전자원을 선발하기 위해 수행되었다. 우선, 저항성 검정조건을 마련하고자 8점의 TSWV 저항성 육성계통을 두 가지 온도 조건(주간 25℃ ± 3.1℃, 35℃ ± 2.2℃)에서 검정을 실시하였다. 8점 중 두 계통(11VR35와11VR51)은 25℃에서 이병율이 모두 0.0%이었으나 35℃에서는 각각 100%, 80%의 이병율을 보였다. 아울러11VR32 11VR47 11VR53 11VR54 는 25℃에서 이병율이28.6%, 37.5%, 10.0%, 10.0%를 보여 중도저항성으로 평가되었으나 35℃에서는 이병율이 모두 100%로 나타났다. 이러한 결과는 고온(30℃ 이상)에서 저항성이 변화한다는 기존 보고와 유사한 결과를 확인할 수 있었다. 따라서 TSWV저항성 검정은 고온에서 실시하는 것이 안정적이라고 판단되었다. 이러한 조건으로 저항성검정을 실시한 결과 5점의 TSWV 저항성 유전자원을 선발하였다. 5점의 TSWV 저항성 유전자원은 2 C.annuum (11PM25, 11PM28), 3 C.chinense (11FL34, 11FL38, 11FL44)이다. 그러나 이들은 Tsw에 의한 과민반응(hypersensitivity response; HR)과 같은 증상은 없었다. 따라서 5점의 TSWV 저항성 유전자원은 TSWV 저항성 품종육성에 새로운 저항성 재료로 사용될 것으로 기대된다.

        Phytophthora capsici Leonian causes root rot and stem blight in pepper (Capsicum spp.) and is a serious threat to pepper production because of its ability to infect every root, stem, and leaf at any developmental stage. Recently, pepper F1 cultivars resistant to Phytophthora root rot have been commercially released in Korea. However, despite many studies, the inheritance of resistance remains controversial due to differences in experimental methods, including pepper materials, pathogen isolates, inoculation conditions, and evaluation methods. Our aim was to determine the inheritance of Phytophthora root rot resistance by using three different F2 populations derived from crosses between ‘CM334’ (a resistant male parent) and three Korean landraces, ‘Subicho’ ‘Daehwacho’ and ‘Chilsungcho’ (susceptible female parents), and inoculating them with three different pathogen densities (1 ¯ 10 4 , 1 ¯ 105 , and 1 ¯ 106 zoospores/ml). The distribution patterns were varied, depending upon female parental susceptibility as well as inoculum densities. For example, as the inoculum density increased, pepper survival rates decreased. In all of the inheritance analyses, one common dominant resistant gene was participated in resistance to Phytophthora root rot. In addition, we found that a complementary gene, together with the major dominant gene, was necessary for resistance at a high (10 6 ) inoculum density, based on a 9:7 (R:S) segregation ratio. This study will be helpful in developing molecular markers linked to genes that are resistant to Phytophthora root rot.

        The amount of genetic variability of a species is essential for its survival and adaptation in different environments, and studies of genetic diversity using molecular markers are necessary to understand the genetic structure of a population and to orientate effective strategies of germplasm conservation. The aim of current study was to determine the SSR markers that can be used rapidly and reliably to evaluated the pepper of Bulgaria landraces, and applied the markers to assement of introduce genetic diversity of the pepper germplasm. We used 22 polymorphic microsatellite markers to analysis of genetic diversity within 61 pepper collection of Bulgaria landraces germplasm, all SSR primers pairs produced 80 polymorphic and reproducible amplification fragments. An average value of polymorphic information contents (PIC) were 0.334 with a range of 0.061 to 0.63. The mean values of observed (HO) and expected heterozygosity (HE) were 0.383 and 0.154, respectively, indicating a considerable amount of polymorphism within this collection. A genetic distance-based phylogeny grouped into three distinct groups, which was the landrace, moderate and wilde type, genetic distance (GD) value was 0.540. An average day of flowering time was 53 days with a range of 45 to 60 days. The everage od fruit length and width were 9.38cm with a range 2.1 to 23.6cm, and 3.51cm with a range 0.6 to 8.9cm, respectively. Molecular data were complemented with morphological measurements according to the descriptor list for the pepper collection of Bulgaria landraces germplasm.

        Fifty-five sweet pepper (Capsicum annuum L.) germplasm from the Rural Development Administration (RDA) were analyzed for their fruit yield and fruit quality characters, including fruit weight, fruit size, pericarp thickness, total soluble content, fruit lobe, fruit shape, fruit surface and fruit color. Fruit yield per plot ranged from 200 g to 8150 g (average, 1884 g). Fruit fresh-weight per fruit ranged from 27.3 g to 200.0 g with an average of 97.2 g. Fruit length varied from 4.0 cm to 16.2 cm (average, 9.6 cm). Fruit length-to-width ratio ranged from 0.6 to 3.0 with an average 1.7. The fruit pericarp thickness ranged from 1.8 mm to 10.7 mm (average, 4.8 mm). Total soluble content was the highest (8.5˚ Brix) in K156286 and the lowest (5.3˚ Brix) in K156208 with the average value 6.7˚ Brix. Fruit lobes numbers varied from 2 to 4. Blocky, triangular, elongated, rectangular and round fruit shape identified among the sweet pepper accessions. Fruit surface varied from smooth to wrinkle and fruit color also varied. The presence of variability in fruit yield and quality traits within the sweet pepper genotypes can be utilized to develop high yielding sweet pepper variety with better fruit quality characters.

        A field experiment was conducted to examine the fruit quality characters in second generation (F2) hybrid cultivar and to compare the fruit characters with original F1 hybrid cultivar of minipaprika (yellow and orange type) at the Research Farm, Hwacheon in July, 2010. Fruit characters varied within F2 population of each minipaprika type. In minipaprika yellow, fruit weight varied from 12.2 g to 50.8 g (average 28.5 g) and fruit length/width varied from 1.4 to 2.8 (average, 2.0). Pericarp thickness ranged from 1.8 mm to 4.1 mm (average, 2.9 mm). Total soluble solid (TSS) varied from 6.2˚Brix to 13.5˚Brix with an average of 8.7˚Brix. Fruit volume varied from 10.3 cc to 46.7 cc with an average of 24.4 cc. In minipaprika orange type, fruit weight ranged from 19.7 g to 42.4 g (average, 29.0 g) and fruit length/width varied from 1.5 to 2.6 (average, 2.0). Pericarp thickness varied from 2.1 mm to 4.1 mm with an average of 3.0 mm. TSS varied from 5.0˚Brix to 12.2˚Brix (average, 7.9˚Brix) and average fruit volume was 24.6 cc ranging from 10.7 cc to 35.0 cc. The average fruit quality characters in F2 population in both yellow and orange minipaprika did not differ from their F1 hybrid parent and F2 seed can be an additional way to supply high yielding hybrid cultivars at lower cost to the minipaprika growers.

        Anther derived double haploids (DHs) from sweet pepper genotypes ('Special', 'Derby', 'Bossanova', 'Fiesta', 'Debora' and 'Minipaprika') were used to study the agronomic variation in 2006. Ninety-nine successful DHs regenerants (32 from 'Special', 25 from 'Derby', 23 from 'Bossanova', 10 from 'Fiesta', 6 from 'Debora' and 3 from 'Minipaprika') were transplanted at plastic house and studied on their agronomic characters. Variation in agronomic characters was observed within the DHs of each genotype. DHs obtained from 'Derby' and 'Fiesta' exhibited wide variation in fruit yield plant-1 whereas averaged fruit yield plant-1 was highest in 'Derby' (1608 g) and less variation was observed in DHs of 'Bossanova'. Based on the agronomic characters expressed in DHs population at this environment, SP55, SP56, SP60, and SP116 from 'Special', SP8, SP10, SP14, SP16, and SP34 from 'Derby', SP115, SP119, SP142, SP143, SP196, and SP199 from 'Bossanova', SP41, SP45, and SP114 from 'Fiesta', SP21 from 'Debora' and SP91 from 'Minipaprika' identified as elite inbred lines and these DH lines could be used for commercial hybrids production in sweet pepper. Genetic relationship among the selected inbred lines using molecular markers and their response to diseases are further recommended to study.

        Anthers of sweet pepper F1 cultivar ‘Special’ were cultured on Dumas De Vaulx (C medium), supplemented with 0.1mgL-1 2, 4-D and 0.1 mg · L-1 kinetin with 3% sucrose, and 0.32% phytagel. The calluses obtained were further sub-cultured on Murashige and Skoog (MS) medium without growth regulators for regeneration. Regenerated plantlets were grown in plastic pots under plastic house and characterized their cytological and morphological characters in spring, 2008. Twenty percent plantlets were identified as haploid plants after chromosome and ploidy analysis. Haploid plants contained 12 chromosomes, high stomatal density with small stomatal length as compared to diploid plants. Stomatal length in haploids was 23.3% smaller than diploids. Haploid plants were characterized as small leaf and petiole size, poor vigor, thin stem and short plant height, short internodes and small flower buds, fruit size and fruit weight as compared to diploid plants and most of the haploid fruits were seedless. SP55, SP62, SP68, SP72 and SP77 are found high yielding double haploids with high total soluble content (8.6, 8.7, 9.2, 9.1 and 9.8 ºBrix, respectively) and desirable fruit shape, and recommended them to exploit as inbred lines for heterosis breeding.

        Sweet pepper inbred lines (KNU1003, KNU1006, KNU1007, KNU1009, KNU1015, KNU1017 and KNU2006) developed at Kangwon National University (KNU) through conventional means, inbred lines (5AVS1, 5AVS2, 5AVS3, 5AVS5, 5AVS7 and 5AVS8) collected at Rural Development Administration (RDA) and inbred lines (SP12, SP27 and SP14) derived from anther culture were used as female parents and anther culture derived homozygous lines (SP9, SP10, SP14, SP24, SP25, SP27, SP30, SP32, SP34, SP38, SP43, SP45 and SP51) were used as male parents to produce F1 hybrids. A total of 37 F1 hybrids were evaluated for fruit yield and quality characters in summer season, 2007. Variation in fruit number, fruit weight, fruit yield per plant and fruit volume was observed among the F1 hybrids. Superiority on yield over standard/commercial varieties were differed among F1 hybrids. Hybrid 5AVS8 x SP45 exhibited highest heterosis over Special (16.5%) and Fiesta (24.7%). Fruit quality characters (fruit length, fruit width, pericarp thickness, total soluble solid, fruit shape and fruit color) were varied among the F1 hybrids. Fruit number, fruit weight and fruit volume per plant were correlated with fruit yield. Based on the standard heterosis expressed by the hybrids and quality characters evaluation, KNU1017 x SP27, 5AVS1 x SP43, 5AVS5 x SP27, 5AVS8 x SP45, SP12 x SP38 and SP27 x SP25 hybrids were found to be superior over commercial cultivars and are selected. Inbred lines of these hybrid combinations can be used to produce F1 hybrid seed for commercial production.
