The necessity of conditional gene expression in pigs for transgenic models is raised. Thus, in this study, Cre-loxP conditional expression in porcine fetal fibroblasts was investigated and the transformed fibroblasts were reprogrammed in enucleated oocytes for further early embryonic development. Fetal fibroblasts from miniature pigs were used for transfection with pCALNL-DsRed including floxed neomycin resistant gene and selected with 750 ug/mL neomycin for two weeks. The transfected cells did not express DsRed under fluorescence microscope. After transient transfection of plasmid DNA expressing Cre, the fibroblasts began to express DsRed. The cells expressing Ds- Red were employed into somatic cell nuclear transfer (SCNT). A total of 121 oocytes were used for SCNT and 76 cloned embryos (62.8%)were cleaved. Six blastocysts were grown up after SCNT and expressed DsRed. Deletion of floxed neomycin resistant gene was confirmed by RT-PCR in cloned blastocysts. Taken together, this study demonstrated that Cre-loxP recombination in miniature pig fibroblasts were successfully worked and those sequential transformed cells were developed into pre-implantation stage via SCNT.
본 연구의 목적은 교사의 창의적 교수행동에 영향을 미치는 동기적 요인인 자기결정성, 조화 열정, 교수효능감, 교수몰입간의 구조적 관계를 검증하고, 각 변수의 영향력을 밝히는데 있다. 연구대상은 충북에 소재한 중등학교에 재직 중인 교사 429명이며, 구조방정식모형 분석을 실시 한 연구결과는 다음과 같다. 첫째, 교사의 자기결정성, 조화열정, 교수효능감, 교수몰입 그리고 창의적 교수행동 간의 관 계를 설명하는 최종모형의 적합도는 ²=151.913, df=26, =.000, RMSEA는 .074, GFI는 .940, AGFI는 .904, RMR은 .019, TLI는 .923, CFI는 .954, NFI는 .946으로 모두 기준에 적합한 것으 로 나타났다. 둘째, 교사의 창의적 교수행동에 영향을 미치는 구체적인 경로를 살펴보면, 자기 결정성, 조화열정, 교수효능감, 교수몰입 모두 유의한 직접적인 영향력이 있는 것으로 나타났으 며, 그 중에서도 교수효능감의 직접적인 영향력이 가장 큰 것으로 나타났다. 또한 창의적 교수 행동에 영향을 미치는 간접효과를 상세 분해한 결과, 유의한 경로 중 조화열정이 교수효능감을 통해서 창의적 교수행동에 이르는 경로의 간접적인 영향력이 가장 큰 것으로 나타났다. 따라서 교사의 창의적 교수행동 수준을 높이기 위해서는 동기적 요인인 자기결정성, 조화열정, 교수효 능감, 교수몰입을 통합적으로 증진시킬 수 있는 방안을 고려할 필요할 것이다.
Until now many strategies have been used to produce marker-free transgenic plants such as co-transformation with negative selectable markers, site-specific recombination system, transposable elements mediated transformation, and etc. In this research, embryogenic calli induced from japonica rices, Ilmibyeo and Dongjinbyeo, were tranformed with the vector which simultaneously constructed with cre/loxP and argE genes in T-DNA. Transformation efficiencies were comparably lower than those of our previous studies, since the constructed genome size was relatively big (>10Kb). For eliminate the transformed tissues which contained positive selectable marker gene, tunicamycin was treated at regeneration and selection stages, since cre recombinase gene is expressed under the presence of this antibiotics. The plants were selected first under 50 mg․L-1 hygromycin at 28℃ for 2 weeks after the Agrobacterium-infection at 25℃ for 7 days. And then, the regeneration plants were successfully obtained on MS basal regeneration medium containing 0.1 mg․L-1 tunicamycin. The regenerated plants are now acclimatizing in greenhouse and molecular analysis are currently accomplished with these plants.
