        식이 영양에 의한 충분한 영양분 섭취는 치매에 의한 뇌 인지 기능의 저하의 위험성을 줄일 수 있는 중요한 수단이다. 설포라판은 뇌 인지 기능 개선효과가 있는 것으로 알려져 있는 영양 성분으로, 설포라판이 다량으로 포함된 브로콜리 추출물은 인지 기능 개선 효과가 좋을 것으로 기대된다. 본 연구에서는 제주도 탈염 용암해수 재배로 제조한 브로콜리 추출물이 뇌 인지 기능 개선 효과를 가지고 있는지 시험하 였다. 제주도 용암해수는 유용 미네랄 (아연, 바나듐, 게르마늄)이 풍부해서, 용암 해수에서 재배한 작물은 유용미네랄 함량이 높은 것으로 알려져 있다. 제주도 탈염 용암 해수를 사용해서 재배한 브로콜리 추출물의 화학 조성 중 설포라판과 글루코라판 성분을 분석하기 위해 LC-Q-orbitrap 질량 분석기를 사용하였고, 설포라판의 정량 분석을 위해 HPLC 를 사용하였다. 브로콜리 추출물의 신경 세포 사멸 억제 효과와 항 염증 효과를 시험하기 위해 SH-SY5Y 세포를 사용한 실험을 수행하였고, 시냅스 가소성 촉진 효과를 시험하기 위해 SH-SY5Y 세포에서 시냅스 가소성 관련 단백질의 발현 변화와 아세틸콜린 분해 효소의 활성 변화를 측정하였다. 이러한 실험들을 수행한 결과 탈염 염지하수로 재배한 브로콜리 추출물은 신경 세포 사멸 억제 효과와 항염증 효과가 있음을 확인 하였고, 시냅스 가소성 관련 단백질 발현을 증가 시키고 아세틸콜린 분해 효소의 활성을 억제해서 시냅스 가소성을 증가 시키는 효과가 있는 것을 확인 하였다. 이 상의 결과들은 제주도 탈염 용암해수로 재배한 브로콜리 추출물이 치매에 의한 뇌 인지 기능 저하를 억제하는 좋은 식이 영양분으로 사용될 수 있는 가능성을 제시하였다.

        The purpose 01' pl'esent study was to examine the molecular events in apoptosis by CoCl2, mimicking hypoxic cond ition and recovering effects by LED ir l'adiation on Human SH-SY5Y neuroblastoma cells The SOUl'ce 0 1' light for ir l'adiation was a continuous-wave LED emitting at a wavelenl양h of 590 nm, and manufactured that ene rgy density was 5 mW!cm2 on sample surface, After ir l'adiation, cell viabi lity was measured with BrdU , cell morphol ogy was examined with Diff- Quik staining, cell signaling was monitored with various apoptosis-related molecules using RNase Pl'otection Assay(RPA) , W11en treated with CoC12, apoptotic induction was found in the SH-SY5Y cells in a concentration-dependent and time-dependent manner , Diff-Quik s taining was revealed that DNA fragmentation re presented apoptosis was examined in CoC12-tl'eated group, Moreover, RPA assay of SH-SY5Y cclls lIs ing val'iolls apoptosis-related molecllles showed that the apoptotic cell population was mcreased J-loweve. there was sorne signifïcant change in LED irradiatied cells aftel' treatement of CoC12 The main mechanism for Lhese a poptosis appearecl to be mito c hondriεt - m ecliated pathway, such as cytochrome- c‘ caspase-9, caspase-3, pro-apototic protein ßax, anti-apototic protein Bcl-2, and death receptor• mediated pathway, such as Fas, cas pase- 8, a ncl TNFRl These results demonstrate that CoCI2 induce apoptosis in SH-SY5Y via different dual apop tosis pathway through death receptor pathway as well as mitochondria- dependent pathway and LED irradiation can recl llces the CoCl2-induced apoptosis by blocking their internal signaling pathway

        Background : Cellular oxidative stress as reactive oxygen species (ROS), whether produced endogenously as a consequence of normal cell functions or derived from external sources, pose a constant threat to cells living in an aerobic environment as they can result in severe damage to DNA, protein, and lipids. The effects of Valeriana fauriei extract and fractions on hydrogen peroxide-induced neuronal cell damage are studied. Methods and Results : Oxidative stress plays an important role in the pathological process of neurodegenerative diseases. Valeriana fauriei extract (VFE) and EA fractions (VFEA) was investigated total phenolic contents using method. VFE of total phenolic contents had 2.54 ± 0.01 mg/g, also, VFEA had a 18.78 ± 0.03 mg/g. High phenolic content of the VFEA is expected to better the inhibition of oxidative stress. VFE and VFEA were experimented to inhibit ROS induced 200 μM 3-morpholinosydnonimine (SIN-1). VFE of inhibit SIN-1 induced-ROS dose dependently and signficantly. In addition, VFEA inhibition was also dose dependant and significant. Moreover, The treatment of SH-SY5Y and SK-N-SH cells with VFEA significantly reduced hydrogen peroxide-induced generation of intercellular ROS. Conclusion : From the above results, we may suggest that VFEA might have useful as a material for functional food and pharmaceutics for the pathological process of neurodegenerative diseases.
