        한우에게 급여하는 사료 내 에너지원의 종류에 따라 반추위 미생물의 아미노산 조성에 차이가 있는지를 조사하고자 본 연구를 수행하였다. 국내 한우 비육우 사육에 주로 이용되는 에너지원 사료인 옥수수(T1), 생미강(T2), 소맥(T3) 그리고 소맥피(T4)로 반추위 환경과 유사한 연속식 배양기를 이용하여 72시간까지 배양을 진행하였다. 배양이 진행되는 동안 6시간 간격으로 배양액의 발효 성상을 확인하였으며 pH, NH3-N이 일정하게 유지되는 것을 확인하였으며, 배양 종료 시점인 72시간대에 미생물체 단백질 합성량(MPS), 미생물의 아미노산 조성 및 미생물 균총 변화를 분석하였다. 배양액의 pH는 모든 처리구에서 배양 기간동안 5.5~7.0을 유지하였다. Total VFA 농도는 T1이 23.13 mM으로 가장 낮았고, T4가 29.93 mM으로 가장 높았다(p<0.05). A (acetate) :P (propionate) 비율은 T1이 1.48로 가장 낮았으며, T4가 2.81로 가장 높게 나타났다 (p<0.05). Butyrate는 T1이 2.37 mM으로 가장 낮았으며, T2, T3 그리고 T4는 4.37~4.58 mM으로 차이가 없었다(p<0.05). MPS는 T2 (332.5 mg/L)와 T3 (320.2 mg/L)가 높았고 T1 (137.5 mg/L)와 T4 (154.2 mg/L)로 낮게 나타났다(p<0.05). DGGE band 분석결과 모든 처리구는 57.5 % 이하의 유사도가 나타났다. 미생물의 총 아미노산 함량은 T1 (31.59 %)과 T3 (31.33 %)가 가장 높았으며, T2 (20.09 %)가 가장 낮았다 (p<0.05). 이는 급여된 사료 내 총 아미노산 함량과 반대되는 결과로 나타났다. 따라서, 본 연구에서 한우 급여 에너지원 사료에 따른 반추위 내 미생물 발효 특성과 미생물체 단백질 합성량이 미생물 군집에 영향을 미치며 이에 따라 미생물의 총 아미노산 함량에 영향을 미친다는 것을 확인하였다.

        The study was conducted to evaluate the effects of microbial culture supplements on ruminal fermentation and fermentative quality of Italian ryegrass silage (IRGS) both in vitro and in situ. Three species of microbes (Lactobacillus casei (LC), Bacillus subtilis (BS), and Saccharomyces cerevisiae (SC)) were used in this study. They were applied to IRGS at 30 days after silage manufacture. Various items were measured using in vitro and in situ incubation technique after each microbial supplement was inoculated into IRGS at 0.5×104 CFU/g. In the first experiment, in vitro ruminal fermentation characteristics of IRGS were evaluated at 0, 12, 24, 48, and 72 hours after microbes were inoculated into IRGS. In the second experiment, in situ fermentation characteristics were investigated at 0, 1, 3, and 5 days after the inoculation of each microbial supplement. In vitro ruminal NH3-N content was significantly (p<0.05) increased in LC-, BS-, and SC-IRGS at 12 hrs post incubation compared to that in control IRGS. In vitro ruminal total VFA concentration and dry matter digestibility (DMD) of IRGS were not significantly difference among LC-, BS-, and SC-IRGS, although they were numerically increased in LC-IRGS than those of the other IRGS. In addition, this study evaluated the fermentation characteristics and in situ DMD of IRGS with the lapse of incubation time up to 5 days. Throughout the incubation times from 1 day to 5 days, the pH value was significantly (p<0.05) lower in BS-, LC-, and SC-IRGS than that in control IRGS. Lactate was significantly (p<0.05) higher, and significantly (p<0.05) butyrate was lower in LC-IRGS than that in other treatments at 0 day. It was higher (p<0.05) in control IRGS than that of BS-, LC-, and SC-IRGS at 1-5 days. In situ DMD tended to increase in BS-, LC-, and SC-IRGS compared to that in control IRGS. Especially, DMD was higher in SC-IRGS than that in other treatments at 0 day. It tended to be higher in LC-IRGS at all incubation time. Taken together, these results suggest that it might be useful to select a microorganism by considering the feeding time of IRGS to ruminants because organic acids and DMD of IRGS were affected by the incubation time of each microorganism with IRG silage, especially for L. casei decreased the content of acetate and butyrate in IRGS.

        This study evaluated the effects of TMC (trace mineral-fortified microbial culture) supplementation on growth performance, carcass characteristics, and meat quality parameters of Hanwoo steers during the last 4 months of finishing period. The TMC was a combination of 0.4% trace minerals, 20.0% Na-bentonite, and 79.6% feedstuffs, which was inoculated with a mixed microbial culture (Enterobacter ludwigii, Bacillus cereus, B. subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae). Twenty-four steers were blocked by initial BW (634 ± 16 kg) and randomly allocated to one of two treatments (control vs. 3.3% TMC). The effect of TMC supplementation on the growth performance was not significant. There was no incidence of urolithiasis in TMCfed steers. However 3 out 12 steers (25%) fed the control diet were observed to have urinary calculi. The carcass yield and meat quality parameters were not affected by TMC supplementation, however marbling score was increased in TMC-fed steers (P = 0.08). There was no effect of TMC treatment on the chemical composition of longissimus dorsi muscle (LM). The TMC supplementation increased concentrations of manganese (P < 0.01), cobalt (P = 0.02), iron, and copper (P = 0.06) in LM. In conclusion, TMC treatment did not negatively affect growth performance and meat quality parameters, and positively affected the trace minerals profile of LM.
