In this study, we analyzed the intraseasonal variability and change of winter temperature over South Korea using long-term observations. The lowest temperature in the wintertime, using daily mean, maximum, and minimum temperature data occurred on January 7th in the period of 2003~2012, while on January 22~25th in the past 30 years(1973~1982, 1983~1992, 1993~2002). Representative seven stations in South Korea also showed consistent result. The strengthening of Siberian High and weakening of Aleutian Low in recent 10 years are found to be closely related with the recent intraseasonal temperature change over South Korea. The baroclinic structure of upper-level low and lower-level high system near Korean peninsula provided good condition for vertical cold-air advection, which resulted in minimum temperature on late January in 1973~1982 and early January in 2003~2012 with strengthened low-level northerly flow.
Adaptive development of early stage embryo is well established and recently it is explored that the mammalian embryos also have adaptive ability to the stressful environment. However, the mechanisms are largely unknown. In this study, to evaluate the possible role of aquaporin in early embryo developmental adaptation, the expression of aquaporin (AQP) 5 gene which is detected during early development were examined by the environmental condition. To compare expression patterns between in vivo and in vitro, we conducted quantitative RT-PCR and analyzed localization of the AQP5 by whole mount immunofluorescence. At in vivo condition, Aqp5 expressed in oocyte and in all the stages of preimplantation embryo. It showed peak at 2-cell stage and decreased continuously until morula stage. At in vitro condition, Aqp5 expression pattern was similar with in vivo embryos. It expressed both at embryonic genome activation phase and second mid-preimplantation gene activation phase, but the fold changes were modified between in vivo embryos and in vitro embryos. During in vivo development, AQP5 was mainly localized in apical membrane of blastomeres of 4-cell and 8-cell stage embryos, and then it was localized in cytoplasm. However, the main localization area of AQP5 was dramatically shifted after 8-cell stage from cytoplasm to nucleus by in vitro development. Those results explore the modification of Aqp5 expression levels and location of its final products by in vitro culture. It suggests that expression of Aqp5 and the roles of AQP5 in homeostasis can be modulated by in vitro culture, and that early stage embryos can develop successfully by themselves adapting to their condition through modulation of the specific gene expression and localization.
Genotyping-by-sequencing (GBS) is a robust and rapid tool to develop SNP markers. Reduced sequencing complexity and multiplex sequencing of GBS has reduced genotyping cost for complex genome. However multiplex sequencing brings low sequencing depth which can lead to reduced number of markers. Therefore to find the appropriate condition for GBS is needed. In this research we demonstrated the use of ApeKI and selective primers for GBS of pepper (Capsicum spp.). Selective primers which amplify the GBS library with one or two flanking sequences to ApeKI site were used to increase sequencing depth. By in silico digestion, we developed six different selective primers amplify 4,000-400,000 regions. We made the GBS library with eight pepper accessions in four species using six selective primers and sequenced. Proper selective primers and pooling rate for each species will be determined. This approach will be useful for genotyping Capsicum breeding lines or populations by developing high quality SNP markers.
sy-2 (Seychelles-2) is a temperature sensitive mutant of Capsicum chinense and native to Seychelles Island in Africa. Previously we showed that sy-2 leaves were irregularly shaped and defective in chlorophyll development at temperatures lower than 24℃. A segregation test revealed that the sy-2 gene is controlled by a single recessive gene. To identify the sy-2 gene, we performed a map-based cloning approach using a total of 1,010 F2 plants derived from crossing sy-2 and the wild type C. chinense ‘No.3341’. sy-2 gene is located on chromosome 1, 0.3 cM and 0.1cM away from cosII markers C2_At4g29120 and C2_At1g09070, respectively. The tomato genome sequence between those two markers was compared with pepper genome sequence. We found three of pepper scaffold sequences in this region. We developed seven ingle nucleotide polymorphism (SNP) markers on the pepper scaffold sequences, among which five SNP markers were co-segregated with sy-2. To fill the gap between the scaffolds which contains co-segregating markers, we screened a bacterial artificial chromosome (BAC) library, and end-sequences of total of 22 AC clones were i. We found that five clones were overlapped to cover the gap. We fully sequenced four AC clones and found that the physical distance between C2_At4g29120 and C2_At1g09070 is 343kb. This region contains 70 putative genes such as HSP90-like ATPase family proteins, lipid-transfer proteins, calmodulin-domain protein kinases, and zinc finger proteins (ZFPs). To identify the sy-2 gene, we performed RT-PCR and found that a ZFP-like gene is differentially expressed between WT and sy-2 leaves. This result suggests that the ZFP-like gene is a strong candidate for the sy-2 gene. We are currently characterizing this candidate gene.
Tomato yellow leaf curl disease is a devastating disease of tomato (Solanum lycopersicum), which is caused by begomoviruses generally referred to as tomato yellow leaf curl virus (TYLCV). The breeding for TYLCV resistance has been based on the introgression of the Ty-3 resistance locus. Knowledge about the exact location of the Ty-3 on tomato chromosome 6 is needed to understand the genomic organization of the Ty-3 locus. In this study, we conducted a genetic analysis using a segregating population derived from a cross between resistant accession S. lycopersicum “A45” and susceptible accession S. lycopersicum “A39”. The F1 plants showed resistance to TYLCV and F1 was self-pollinated to produce F2 progeny. To screen the TYLCV resistance in 145 F2 plants, a leaf agroinfiltration method was used. F2 plants showed a classical Mendelian seregation (106 resistance : 39 susceptibility) for resistance to TYLCV respectively. SCAR and CAPS markers linked to the Ty-3 were tested for genotyping F2 plants and .genotyping and agroinfiltration results were cosegregated in the newly developed F2 population.
xBrassicoraphanus, a new synthetic intergeneric hybrid between Brassica rapa L. ssp. pekinensis and Raphanus sativus L., also locally known as ‘Baemoochae’, is an interesting subject for studying polyploidy and genome plasticity in the family Brassicaceae, but very few genomic and cytogenetic information. Here, we analysed the chromosome complements and pairing of the most fertile lines, BB1 and BB5, using dual-color fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH) to check their chromosomal segregation stability. The somatic chromosome complement of B. rapa was confirmed to be 2n=20 (2.8~4.8μm), of R.sativus, 2n=18 (2.0~3.3μm), and of xBrassicoraphanus, 2n=38 (2.2~5.0μm). There were eight, eight, and seventeen metacentric pairs and two, one, and two submetacentric pairs in B. rapa, R. sativus, and xBrassicoraphanus, respectively. Additionally, three, two, and five pairs of 5S rDNA and five, three, and eight pairs of 45S rDNA were observed in B. rapa, R. sativus, and xBrassicoraphanus, respectively. This suggests that both B. rapa (AA) and R. sativus (RR) genomes, particularly the rDNA arrays, co-exist in xBrassicoraphanus (AARR) genome. In meiosis I, nineteen bivalents were most frequent, and GISH analysis showed ten bivalents from the A genome. This study would provide a useful information for further genomic study of xBrassicoraphanus and its improvement as a new promising breeding variety.
Carotenoid isomerase (CRTISO) catalyzes the isomerization of prolycopene to all-trans-lycopene in the carotenoid biosynthetic pathway. We isolated a full-length promoter region of CuCRTISO from Citrus unshiu. We determined if the promoter encoded organ-specific or developmental-specific expression, and identified possible cis-acting promoter elements. The full-length promoter and two truncated versions were fused to the β-glucuronidase (GUS) gene and transformed into Arabidopsis thaliana. Transgenic lines expressing the full-length promoter (pCiso-Prom1) and truncated promoters (pCiso-Prom2 and pCiso-Prom3) showed the same developmental and organ-specific activity. GUS expression was detected in the cotyledon and root at 5 and 10 days after germination, mature leaf, and anther. The CuCRTISO promoter contained several cis-acting elements involved in hormonal and environmental stress. Drought stress or abscisic acid treatment did not induce GUS expression in any transgenic lines. Heat stress induced GUS expression in the pCiso-Prom1 line; this promoter construct contains the heat-stress responsive element (HSE). Ethylene and cold-stress treatments induced GUS expression only in the pCiso-Prom3 line, although all transgenic lines contained the same cis-acting ethylene and low-temperature response elements. which could indicate the existence of unknown repressor element(s) in the CuCRTISO promoter. These studies indicate that CuCRTISO promoter activity is regulated in a developmental and organ-specific manner that responds to heat, cold, and ethylene. These results provide new insights into the role of cis-acting element(s) in CuCRTISO promoter activity. OsLPS is pollen specific gene that express at late stage of pollen development in rice. Based on microarray database, promoter region of two genes Os03g0106900 and Os03g0106500 were identified. The sequence of 2287bp and 2468bp upstream region of these genes were amplified and designated as OsLPS10 and OsLPS11. These promoters were fused with GUS-GFP reporter gene in a destination vector, pKGWFS7 and introduced into rice (Dongjin cultivar) and Arabidopsis (Col-0). The results of GUS assay showed different pattern of gene expression in pollen of rice and Arabidopsis. In Arabidopsis, the OsLPS10 gene strongly activated in young anther and not expressed in mature pollen. Pollen development analysis revealed GUS expression was detected at unicellular stage and strongest at the bicellular pollen developmental stage. No GUS signal was recorded in mature pollen. In case of OsLPS11, no GUS signal was detected in during pollen development of inflorescent. By contrast, in rice, the GUS expression pattern of OsLPS10 and OsLPS11 exhibited similar. GUS expression was first detectable in the anthers of spikelets at the bicellular stage and intensity increased in tricellular and mature pollen. The GUS signal was not detected in the anthers in unicellular microspores in both genes, OsLPS10 and OsLPS11. The results suggested that these genes were different activity in heterologous plant system, monocot and dicot. Complementation analysis and Cis-regulatory elements will be examined to illuminate the characteristic of these genes
Based on the results of microarray analysis we selected ten candidate genes that express in pollen at the early pollen developmental stage. By PCR amplification, the promoter region of these genes were amplified from rice genomic DNA (Nipponbare) and cloned into the destination pKGWFS7 vector via an entry vector, pDONR201. The characteristic of promoters were evaluated in Arabidopsis thaliana (Col-0) through GUS expression analysis. Fifty T2 plants respectively from each promoter were tested. Whole inflorescence of individual plant was stained with 1mM X-Gluc solution to observe tissue-specific GUS expression patterns. The results showed that all 10 promoters activated in pollen tissues. Among them six promoters expressed at the early developmental stage (unicellular) of pollen and the others expressed at both early (unicellular) and late pollen developmental stage (mature pollen). The results indicated that these promoters would be potential applicable for the studies of pollen function. Currently, we are performing these promoters analysis in rice transgenic plants as well as molecular characterization.
A freesia (Freesia hybrida Hort.) ‘Song of Angel’ was developed for the cut flower in the National Institute of Horticultural Herbal Science in 2013. This hybrid was crossed and selected from a seedling of three-way crossing ‘Golden Flame’ and the seedling of ‘Athene’ and ‘Yvonne’ in 2006 and 2007, respectively. Morphological characteristics of the selected freesia hybrid were investigated for 5 years from 2008 to 2012, and then it was named ‘Song of Angel’ in 2013. ‘Song of Angel’ has single flower and yellow petals (RHS, YG13B) with yellow center color (RHS, YG13A). The average flower width is 5.8 cm and the average yield is 4.3. The growth of the plant shows vigorous and the average height is 95.3cm, and it is higher than about 30cm that of control cultivar ‘Yvonne’. The average number of floret per stalk and stalk length, 14.3, and stalk was 11.1 cm length is higher than the ‘Yvonne’, 9.7 and 8.3cm length, respectively. The average days to first flowering of ‘Song of Angel’, 141 days, was approximately 10 days earlier than the control cultivar. It’s average vase life and yield is 10.7 days and 6.2 cormlets per plant, respectively.
Genetic resources play a great role in crop breeding because of containing a broad array of useful genes. Currently, the harder are rice breeders trying to develop new rice cultivars with the improved traits, they are more often handicapped by the limited availability of germplasm resources. Thus, a desirable core or heuristic (HS) set of germplasm with maximum genetic diversity can be usefully exploited to breakthrough the present and future challenges of the rice breeding. As such we previously developed the rice HS sets of 166 diverse accessions out of a total 24,368 rice germplasms. Here, we report a large-scale analysis of the patterns of genome-wide genetic variations accumulated in the HS as well as Korean rice over the time. We characterized a total of about 11.8 millions of single nucleotide polymorphisms (SNPs) across the rice genome from resequencing a total of 295 rice genomes including 137 HS and 158 KB rice sets, with an average of approximately 10x depth and > 90% coverage. Using about 460,000 high-quality SNPs (HQSNPs), we specified the population structure, confirming our HS set covers all the rice sub-populations. We further traced the relative nucleotide variabilities of HQSNPs and found the level of the diversity was dynamically changing across the KB genome, which reveals the selection history of KB lines in the past and present. In addition, the results of our genome wide association study (GWAS) suggests that our HS can be also a good reservoir of valuable alleles, pinpointing those alleles underlying the important rice agronomical traits. Overall, the resequencing of our HS set re-illuminates the past, present of the germplasm utilization, which will support the Korean rice breeding in the future.
Climate change is not the only issue yet there are the combinations of many existing factors. Urbanization is one of them and is considering the major criteria. Due to the Rapid Urbanization, land use of the existing watershed is becoming impervious layer and even drainage network system is more complicated. Existing system might not be able to handle the amount of water during the storm events thus, hydrologic impact assessment is necessary for a planned development area in designing detention storage for urban drainage systems to minimize the effect of urbanization. It is advisable to reassess the existing structure to handle and to prevent the threat of flood in the future. For such assessment, single or two hydrologic models can be paired for pre- and post- development conditions. Typical pairings are the use of synthetic hydrograph methods for both conditions or synthetic hydrograph for pre-development and urban hydrology model for post-development condition. This study is aiming to assess the flood impact and suggest the proper technique for future flood prevention. To accomplish, the SWMM was adopted to development areas in Korea. The comparison of results can be done to show the new approach can resolve the irrationalities that can occur with the combination of two different models such as smaller peak flow and longer time to peak for post-development conditions. It is thought that the proposed method will improves the accountability of the flood impact assessment on future development areas
Heterogeneous materials such as rock cannot be expected to have homogeneity so as to be used as structural materials. For example, in order to test large rocks more than one cubic meter, measures need to be taken against weaknesses such as internal cracks and fracture zones that can easily be overlooked in compressive strength tests. The impact-echo method could be available for evaluating strength and detecting internal defects in-situ concrete component with relatively good accuracy. In this study, measurements using the impact-echo method were carried for several types of rock specimens used at automobile crash safety tasting, and, by comparing the results with compression strength tests, the applicability of the impact-echo method for strength estimation of heterogeneous materials was evaluated.This work was supported by JSPS KAKENHI Grant Number 25420462.
Curve number (CN), originally developed, compiled by ‘The Natural Resources Conservation Service (NRCS)’, and has been widely used throughout the world. However, there is the uncertainty of CN derived from the use of antecedent moisture condition (AMC)/Antecedent Runoff Condition (ARC). As in Korea where nearly 70% covered by mountainous area, it is still not sufficient handbook precedent to guide or support the estimation of AMC/ARC. The failure to develop formal criteria of applying AMC/ ARC will be a gaping profession and results not only in uncertainty of CN estimation in particular, but also in designing appropriate structures in Korea as a whole. This paper is aiming at presenting a critical review of AMC/ARC and deriving a procedure to deal more realistically with event rainfall-runoff over wider variety of initial conditions. Proposed methods have been developed. It is based on modifying estimated runoff to observed runoff with coefficient of determination and then applying different algebraic expression with the verification of AMC by antecedent rainfall table of NEH-1964. Clearly indicated from 12 years period analysis and 2 years period of verification, normalized root mean square error, RE and E are best described by the application of algebraic expression by Arnold et al. at the value of 0.8950, 0.9460, and 0.0244 respectively for AMC condition and 1.3799, 0.9642 and -0.0197 respectively for ARC condition. Therefore, Arnold et al. algebraic equation is the best representation criteria of AMC and ARC. This algebraic expression might be applied properly considering South Korea condition.
service advertising (PSA) and commercial advertisements on consumers' social and emotional values. Specifically, we include source credibility on advertising models as a mediating variable to compare the path difference between the social and emotional value of consumers, and to test the appropriateness of advertising expenses. Research design, data, and methodology – A total of 219 copies of effective questionnaires were used and methodologies such as Cronbach’s alpha, paired samples t-test, and structural equation modeling (SEM) were conducted. Results – The results showed that the consumer model was effective at PSA, while celebrities and CEOs were effective at commercial advertising. Further, only credibility (among source credibility) made a significant difference in the test of PSA models of both PSA and commercial advertising that consumers preferred. Conclusions – The advertising model types varied depending on advertising types (for example, PSA and commercial advertising). Therefore, celebrities and CEOs appearing in commercial advertising should have no ethical or moral defect by self-control, and marketers should elect the model considering such a fact.
Among twentieth-century Korean writings in English, those of Professor Insoo Lee are singularly impressive for their particular combination of wit, well-reasoned argument, and mastery of subtitles of English idiom at all levels. The importance of T. S. Eliot as perhaps the most influential English or American poet of the twentieth century is established. Professor Lee is, of course, well known as the translator of Eliot’s The Waste Land into Korean. His rendering of the defining poem of the first half of the twentieth century stands today as a monument in the history of Korean translation. Eliot had influenced the works contained in the volume Inside Cloud Cuckoo Land: The Voice of Korea, containing the collected essays, translations, and journalistic articles produced by Professor Lee. Each also was a force for assuming personal moral responsibility in an age defined by warfare, by the trauma resulting from warfare and destruction: the period of World War I and II and the challenges of the subsequent so-called Cold War, and by a general moral lassitude in the era. The English language writings of Professor Lee attest not only the general influence of the era in which Eliot wrote, they also reveal some of the themes and turns of phrase that characterize the writings of Eliot.
Purpose - This study attempted to identify the value promotion clues that may operate as a consumer’s motive, by shedding new light on consumer value and by reconstructing each variable analyzed through the means–end chain (MEC) theory. Research design, data, and methodology - In this study, 202 copies of effective questionnaires using the data of Yang and Ju (2012) were subjected to correlation, regression, and SEM. Results - All store selection attributes were verified as having a positive influence on the relationship quality. Although the store selection attributes were verified as exerting a positive influence on the relationship quality, according to the verification result of the mediating effect, consumer value was verified to be influenced only by the relationship quality instead of by the store selection attributes. Conclusion - As a result of path analysis on the proposed model after modification, it was verified that only product factor had a statistically significant positive influence and that social value was completely mediating between relationship quality and emotional value. It may be highlighted that the MEC theory concept would be applicable to the cause-and-effect relationship model.
Stabilization of landfill gas (LFG) generation is recognized as the critical indicator to evaluate the future possibility of environmental impact from the waste landfill. In comparison with leachate quality, the amount of LFG generation is considered more difficult to integrate the sequential monitoring results. Spatially and temporal high variation of the LFG generation and the emission would be influenced by the micrometeorological condition. One of the helpful information to predict the behavior of LFG generation is to estimate the remaining of LFG source in the waste. Biological degradation should decrease the amount of component that should be transformed LFG in the waste. Hence, the LFG generation potential of waste in landfill must be gradually decreased as time goes on. In order to support the assessment of the landfill stability from the viewpoint of LFG, the estimation of the potential of LFG generation of the landfilled waste has been investigated at the landfills that was received the waste incineration ash, slag, C&D inert residue, dredged soil, and so on. The LFG emission behavior has been predicted by using the remaining LFG potential, and it was validated by the investigation of surface LFG emission. Degraded organics by anaerobic incubation had been calculated by Buswell's theoretical equation (Bockreis, et al. 2007). Objected samples that were excavated from 10-15 years old waste layer have shown the little potential of LFG generation (Table 1). A highest content of gasified organics was observed for 2.0m depth of C10 though it was less than 1% of the total weight of sample (dry weight). It would be strongly attributed to intensive pretreatment of waste before the landfilling. Since the landfill operator required the strict quality control for the waste to be disposed of, the content of organics in the waste should be enough low at the initial phase of landfill management. In addition, the effort of the landfill management to promote the biodegradation, such as the lowering of the water level in landfill layer, or ventilation of LFG, had contributed to reduce the biodegradable organics. Fig.1 shows the prediction of methane emission from the landfill. It also exhibited results of investigation of surface LFG emission. The prediction of landfill methane emission was developed by using the parameter that was obtained from excavated waste.
Prediction method for the long-term chemical leaching amount from by-product/recycled materials such as waste concrete and steel slag and so on is necessary to widely promote their effective utilization and evaluate their environmental safety. Although there are the batch leaching tests and the column leaching test as the testing methods for evaluating the long-term leaching behavior, the leaching mechanism and the testing result compatibility in both tests has insufficiently been clarified yet. Thus, the prediction of the leaching behavior from the by-product/recycled materials used in actual civil works and their environmental safety evaluation are by no means certain. This paper shows the difference between the batch leaching tests and the column leaching tests in the chemical leaching behavior of Cu-slag. The batch leaching tests were conducted under liquid/solid ratio = 10, liquid = distilled water, stirring strength = 0, 30, or 120 rpm. After a certain elapsed time, the leaching solution was exchanged with the pure distilled water and then the stirring was restarted. The elapsed time was set at 1, 2, 4, 8, 16, 32 days. The column leaching tests were also conducted under the same conditions as those of the batch leaching tests in order to evaluate the effects of the pore distribution and the pore flow velocity in the Cu-slag column on the leaching behavior. In the column leaching tests, the effluent passing through the column was circulated as the influent (Fig. 1). The leaching duration in the column tests can be equivalent as that in the batch tests, so that the difference in the leaching behavior between the batch leaching tests and the column leaching tests may be dependent on the pore-scale heterogeneous flow and path generated in porous materials. Figure 2 shows the leaching rate evaluated from the batch leaching tests and the column leaching tests. In the same fluid velocity levels, the leaching rate in the column tests was larger than that in the batch tests. The leaching rate has been considered large with the fluid velocity. Although the fluid velocity generated by the stirring was the same as the flushing velocity on the surface of the Cu-slag in the batch tests, the fluid velocity in the column tests was enhanced because the permeant liquid was concentrated into the limited pore space in the Cu-slag column. Thus, the pore-scale heterogeneous flow and path generated in porous materials should be evaluated in order to clarify the compatibility between the batch leaching tests and the column leaching tests.
