        Ooencyrtus nezarae lshii (Hymenoptera: Encyrtidae) is a major egg parasitoid of Riptortus pedestris (F.) (Hemiptera: Alydidae). One of the components of the aggregation pheromone of R. pedestris, (E)-2-hexenyl (Z)-3-hexenoate (E2HZ3H), is known to attract O. nezarae female. Response of O. nezarae females to E2HZ3H and its isomers was studied using electro-antennogram (EAG) and field tests. O. nezarae females showed higher antennal response to E2HZ3H and Z2HE3H isomers than E2HE3H and Z2HZ3H. In field tests, O. nezarae females were attracted to E2HZ3H traps, but not to Z2E3 traps. Interestingly, the blend of E2HZ3H and Z2HE3H showed significant antagonistic activities to the attraction of O. nezarae females in the field. We conclude from this study that Z2HE3H is a potent antagonist to the behavioral response of O. nezarae.

        Ethyl formate (EF), a methyl bromide alternative fumigant which is commercially used for quarantine and pre-shipment(QPS) purpose, has a potential to control various insect pests. Based on CTP (concentration × time products), we've firstly evaluated fumigation trials of EF on cucumber plants to eradicate horticultural insect pests in greenhouse. In the preliminary test, EF achieved 100% control of cotton aphid, western flower thrip and tobacco whitefly when CTP of EF was 4.4, 5.6 and 3.3 g h m-3, respectively. In 1300m3 scale cucumber greenhouse trial, EF was applied with 2g m-3 dosage for 15 hrs, and cotton aphid, western flower thrip and tobacco whitefly were completely controlled without phytotoxic symptom. EF fumigation could be suitable for greenhouse cultivation in terms of good efficacy on horticultural insect pests with labor-saving benefit than current insecticide spraying applications.

        A surveillance of chigger mites was performed to monitor the incidence of scrub typhus vectors at 5 environmental collection points of 3 locations (Muju, Namwon and Gimge) from April 2016 in Jeollabuk-Do, Korea. During the surveillance period, 3,292 chigger mites were collected and the predominant species were Leptotrombidium pallidum (42.9%). The high environmental collecting rates were recorded at reservoir bank (31.6%) and grassfield (29.7%). We detected Orientia tsutsugamushi from the samples collected in Jeollabuk-Do using nested polymerase chain reaction. The specific DNA of O. tsutsugamushi were detected in 11 pools among total 119 pools and its minimum positive rate was 0.35%. In order to clarify the relationship between chigger mites and scrub typhus cases, further survey will be needed.

        West Nile Fever is one of most serious disease spreading throughout world since 1990. West Nile Virus (WNV) is transmitted by infected mosquitoes. The possibility of overseas inflow of this virus seems to be high in Korea. To know the WNV infection of mosquitoes, we collected mosquitoes in Jeju, Busan, Gunsan, and Incheon using CDC light trap and BG Sentinel trap from April to August in 2016. Collected mosquitoes were conducted virus detection using real-time PCR method. A total of 22,632 mosquitoes representing 9 genera and 18 species were collected. The most dominant species was Culex pippins complex (36%) followed by Aedes vexans (13%), Ae. albopictus (10%) and Ochlerotatus koreicus (10%). WNV was not detected in any mosquitoes until now.

        Dengue is the most important arboviral human infection worldwide. Approximately 50-100 million cases occur annually and more than 2.5 billion people are at risk of infection. This virus is possibility of overseas inflow to be high in Korea. We collected mosquitoes in Jeju, Busan, Gunsan, and Incheon using BG Sentinel trap from April to August in 2016. Collected mosquitoes were conducted virus detection using real-time PCR method. A total of 15 species comprising 7 genera were identified and 3,696 female mosquitoes collected. The most dominant species ratio (SR) was 49.4% (Culex pippins complex) followed by 21.8% (Aedes albopictus), 13.6% (Ae. vexans) and Ochlerotatus dorsalis (4.8%). Dengue virus was not detected in any mosquitoes until now.

        Ixodid ticks are notorious as the vector of severe fever with thrombocytopenia syndrome virus (SFTSV) in Korea. To confirm the transovarial transmission of SFTSV, we collected blood-fed adult ticks from animals in SFTS case reported area of Jeju island in 2015. A total of 476 ticks were collected from four host animal species, horse (81.3%), dog (9.0%), roe deer (7.1%) and wild boar (2.5%). They were identified as 1 genus and 2 species, Haemaphysalis longicornis (87.8%) and H. flava (12.2%). Among them, 131 adults hatched their eggs successfully. The body weight of females was highly correlated with the number of egg masses laid (P < 0.05). We confirmed that one adult H. flava was infected with SFTSV and its infection rate was 0.77%. The eggs of the SFTSV-infected adults also were infected with SFTSV and its transovarial transmission was confirmed. This result suggests that the parent generation of H. flava could transmit SFTSV transovarially into their F1 progeny and H. flava might be one of major vectors of SFTSV in Korea.

        Bee pollen have been used for centuries as a kind of traditional medicine and food diets additive due to their nutritional and biological properties. The present study was to evaluate the biological features of ethanol extracts of 18 different bee pollen (EEBP) samples collected from 16 locations of South Korea. Our results indicated most of the EEBP samples showed 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, inhibitory activities against human β-amyloid precursor cleavage enzyme (BACE-1), and antiproliferative activities towards five cancer cell lines. However, these EEBP samples did not present human acetylcholinesterase (AChE) inhibitory activities, and intestinal bacteria growth inhibitory activities.

        We evaluated the larvicidal activity of 12 Apiaceae plant essential oils and their components against the Asian tiger mosquito, Aedes albopictus, and the inhibition of acetylcholine esterase with their components. Of the 12 plant essential oils tested, ajowan (Trachyspermum ammi), caraway seed (Carum carvi), carrot seed (Daucus carota), celery (Apium graveolens), cumin (Cuminum cyminum), dill (Anethum graveolens), and parsley (Petroselinum sativum) resulted in >90% larval mortality when used at 0.1 mg/mL. Of the compounds identified, α-phellandrene, α-terpinene, p-cymene, (-)-limonene, (+)-limonene, γ-terpinene, cuminaldehyde, neral, (S)-+-carvone, trans-anethole, thymol, carvacrol, myristicin, apiol, and carotol resulted in >80% larval mortality when used at 0.1 mg/mL.

        Bombolitin is a venom peptide originally isolated from bumblebees and possesses various biological activities, including hemolytic activity. Bombolitins exhibit amphipathic α-helical structure in lipid-membrane-mimicking environments. To investigate their pharmacological and toxicological properties, anti-tumor, anti-microbial and cytotoxic activities of bombolitins from Bombus ardens and Bombus ussurensis were evaluated. Bombolitins of the two species exhibited extremely high anti-tumor activity against ovarian tumor cells SK-OV-3 and NIH; OVCAR-3 at 25-50 μM, which is 2-fold more potent than other wasp venom peptides studied to date (Yoon et al., 2015; Yoon et al., 2016). The two bombolitins showed significantly high antimicrobial activity. However, bombolotin of B. ussurensis showed no antimicrobial activity against Escherichia coli. In addition to their high levels of anti-tumor activity, bombolitins showed considerable levels of hemolytic activity. Thus, to utilize bombolitins as a potential candidate for anti-tumor peptide drugs, further studies for reducing cytotoxic properties of bombolitns is essential.

        The habitat of the cockroach varies by species. The German cockroach (Blattella germanica) lives in human dwellings, while the Japanese field roach (Blattella nipponica) lives in a mountainous region. Based on phylogenetic analysis of mtCO I, the two species are closely related to each other and B. germanica is divergent from wild species such as B. nipponica. Their habitats and walking speed differ even though the two species have similar morphology. We hypothesized that habitats might influence walking speed via changes to appendage morphology and enzyme-based physiological differences. We found that phenotype such as appendage length and esterase isozyme expression were clearly different between the two species. These differences might be responsible for the observed difference in walking speed.

        Caloptilia Hübner, 1825 is one of the largest genera in Gracillariidae. 314 species have been known around the world. To date, 19 species were known from Korea. In the present study, one species, C. acericola (Kumata, 1966), is newly recognized from South Korea. Caloptilia acericola was first described from Japan and it also distribute Russian Far East. The name of this species was derived from the genus name (Acer, Aceraceae) of its host plant. We redescribed with illustrations of adults and genitalic structures to distinguish the species from the congeneric species in South Korea.

        Non-target predatory insects can be indirectly exposed to aerial pesticide spraying and fogging to control Monochmus beetles that transmit pine wood nematode, Bursaphelenchus xylophilus. We evaluated potential lethal or sublethal effects of thiacloprid on survival and behavior of a carpenter ant species, Camponotus japonicus Mayr. Field-collected ant colonies were directly exposed to several food items, such as thiacloprid-addicted Monochmus beetles, 10% sugar watered cotton balls contaminated by thiacloprid concentrations, and 10% sugar water. Dead beetle bodies caused no apparent adverse effect through dietary exposure in general, although a few ants were died with paralysis at colony level experiment. At individual level, most ant workers were died within 10 days compared to control group. In contrast, dietary exposure of ants to thiacloprid concentrations showed significant lethal effect with paralysis and impaired walking, especially at 10 and 50 mg/L thiacloprid concentrations. Some intoxicated ants recovered within a few days in 10 and 50 mg/L thiacloprid concentrations, but intoxicated ants were generally shown to be less responsible to enemy ants with low aggressive behavior. Implications for predicting hazards of thiacloprid to beneficial arthropods in pine forests are discussed.

        During tick infestation, the tick secretes bioactive substances that modify the host’s physiological and immunological reactions. The study of tick saliva is important to understand tick biology as tick saliva plays a special physiological role in pathogen transmission. The average salivary protein concentration was found to be 0.169 μg/μl/tick and saliva secretion decreased with increased time of tick detachment from the host. Saliva secretion volume increased to 3.56 μl in the group of ticks with a body weight between 301–350 mg as compared to higher and lower body weight groups. On-chip-electrophoresis results show 13 distinct bands ranging from 9.9 to 294 kDa. For salivary protein LC-MS/MS was performed. A total of 135 tick salivary proteins were identified of which 30 proteins were found exclusively in fully engorged nymph saliva, 74 in fully engorged adult females, and 31 were detected in both stages. Results of this may help researchers to identify tick proteins as potential candidates for further studies aimed to develop novel tick control strategies to affect both the ticks and the pathogens transmitted by them.

        Divergence time estimation over a robust phylogeny is a prerequisite for understanding their evolutionary biology. Recent empirical studies have found that molecular dating analyses are critically affected by the number and temporal distribution of fossil calibration points (Yang & Yoder, 2003; Hug & Roger, 2007), the interpretation of relationships between fossil and extant taxa (Benton & Ayala, 2003; Donoghue & Benton, 2007), and how fossil constraints are treated (Magallón, 2004; Ho & Phillips, 2009). Confident dating thus requires multiple fossils that are securely identified and of sufficient age. We provide a list of lepidopteran fossils potentially useful for calibration points, and end on the perspective for dating the currently working phylogeny of Lepidoptera (Regier et al., 2013), using these fossils. In addition, we show microtomographic images for some lepidopteran fossils that we are currently working on.

        Vollenhovia emeryi (Formicidae: Myrmicinae) is divided into two morphs based on the wing length of the queen caste: the long-winged with a normal wing and the short-winged with the aberrant short wing. The phylogenetic analysis shows that the short-winged is derived from the long-winged. In Korea, only the long-winged morph is infected with Wolbachia while the short-winged is devoid of the bacterium suggesting that the short-winged evolved the resistance to the bacterial infection. Intriguingly, some Japanese short-winged colonies proved to still be infected with the bacterium. We hypothesized that the infected Japanese short-winged is the intermediate form in the process to complete cure. However, the data of the MLST and the measurement of Wolbachia density did not support our hypothesis. Our further experiment using microsatellite markers shows that the infected Japanese short-winged shows the similar genetic background to the long-winged. In this presentation we will discuss the potential resistance evolution in the Korean short-winged and future research direction at the genome level.

        This study was conducted to review the family Psychidae in the Korean peninsula with taxonomic arrangement, some biological notes and phylogenetic analyses. In this study, totally 20 species of 8 genera belonging to 5 subfamilies in the family were recognized from Korea. Among them, a new subfamily Ceratostiinae subfam. nov. is proposed based on the type genus Ceratosticha Meyrick. Four new species are described as new to science. Also, six newly recorded species are first recorded in this study. In addition, ecological characteristics, distributional ranges, host plants and biological notes including life cycle and parasites on the larva were described. A phylogenetic and a molecular clock analysis were conducted to construct the systematic relationship and evolutionary process within the family Psychidae species.

        A blast search using SPODOBASE gave 10 chymotrypsin (CHY) candidate genes with complete open reading frames (ORFs) of Spodoptera exigua. Among these candidates, Se-CHY2 was chosen and further analyzed in expression and RNAi. Se-CHY2 was expressed only in larval stage and especially in midgut. Its RNAi was successfully performed by double injection of specific double-stranded RNA (301 bp). qPCR showed a decrease of Se-CHY2 expression level by ~50% at 48 h and ~83% at 72 h. RNAi resulted in significant larval mortality. All treated larvae died at 96 h after dsRNA injection. In contrast, control dsRNA treatment did not give any mortality. Enzyme activity of gut juice was measured with N-succinyl-alanine-alanine-prolin-phenylalanine-p-nitroanilide substrate and significantly decreased in larvae treated with dsRNA. These results indicate that Se-CHY2 is a chymotrypsin that is secreted into midgut lumen and plays a role in nutrient digestion.

        Genus Phytocoris Fallén, 1814 is the largest genus among Heteroptera, around 700 species are described worldwide. In this presentation, the genus Phytocoris from Korean Peninsula is reviewed for the first time with eight species, includes one new record, P. minakatai and one new species, P. goryeonus sp. nov. For further comprehension of this genus, figures of dorsal habitus, genitalia and diagnosis of each Phytocoris species are provided.

        A novel halophilic archaeon designated strain CBA1114T was isolated from solar salt in the Republic of Korea. Strain CBA1114T, which is a coccoid and stained Gram-negative, grew in the presence of 15-30% (w/v) NaCl (optimum, 20%) and at 20-50°C (optimum, 40°C) and pH 7.0-9.0 (optimum, pH 8.0). Strain CBA1114T required Mg2+ for growth. Strain CBA1114T had three 16S rRNA genes, rrnA, rrnB and rrnC; similarities between the sequences were 99.7 and 99.9%. The 16S rRNA gene sequence of strain CBA1114T showed a 91.7% similarity to that of Haloterrigena thermotolerans PR5T. In multilocus sequence analysis (MLSA), five housekeeping genes, atpB, EF-2, radA, rpoB’ and secY, were found to be closely related to those of the members of the genera Halorientalis (89.7% similarity of the atpB gene sequence), Halomicroarcula (91.9 %, EF-2), Haloterrigena (85.4 %, radA), Natronoarchaeum (89.2 %, rpoB’) and Natrinema (75.7 %, secY). A phylogenetic tree generated from the results of 16S rRNA gene and MLSA of the five housekeeping genes showed that strain CBA1114T was closely related to the species of the genus Halorientalis in the family Halobacteriaceae. The major polar lipids were identified as phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester and unidentified lipids. According to the results of phylogenetic, phenotypic and chemotaxonomic analyses, we designate strain CBA1114T as Halostella salina gen. nov., sp. nov., which represents a novel species of a novel genus within the family Halobacteriaceae.

        Effects of dielectric barrier discharge atmospheric cold plasma (DACP) treatment on the inhibition of Salmonella and the storability of grape tomato were investigated. Grape tomatoes, with or without inoculation with a cocktail of three strains of Salmonella (~8 log CFU/g tomato), were packaged in a polyethylene terephthalate commercial clamshell container and cold plasma-treated at 35 kVat 1.1 kHz for 3 min using an DACP treatment system equipped with a pin-type high-voltage electrode. DACP treatment resulted in ~1 log CFU/tomato reduction of Salmonella, irrespectively of the size of container (316, 595vs. 758 cm3), the number of grape tomatoes in the container (3, 7, vs. 11), and the position of the tomato in the container(P > 0.05).Rolling integrated during treatment significantly increased the Salmonella reduction rates to 3.1±0.3 and 3.3±0.8 log CFU/tomato in the single-layer and double-layer configurations of the tomato samples in the container (992 cm3), respectively. Rolling-adopted DACP initially reduced the number of total mesophilic aerobes and yeast and molds in the double layer configuration of tomato samples by 1.3±0.3 and 1.5±0.2 log CFU/tomato, respectively, without altering the color and firmness of the tomatoes. The growth of Salmonella, total aerobes, and yeast and molds on DACP-treated grape tomatoes was effectively prevented during storage at 10 °C. DACP treatment did not influence the tomato color index (a*/b*), firmness, weight loss, pH, total soluble solid content, and lycopene concentration of grape tomatoes at 10 and 25 °C (P> 0.05). DACP treatment holds promise as a post-packaging process for improving microbial safety against Salmonella and storability of fresh grape tomatoes.
