        The purpose of this monitoring is to survey the geographical distribution of tick species using dry ice bait traps and flagging methods at each ten provinces (GangwonⅠ,GangwonⅡ, Gyeonggi, Chungbuk, Chungnam, Gyeongbuk, Gyeongnam, Jeonbuk, Jeonnam and Jeju area) and one Metropolitan area in the Republic of Korea for eight months from April through November, 2016. A total of 65,339 ixodid ticks (8,200 females, 1,988 males, 31,453 nymphs and 23,698 larvae) was collected, belonging to three genera (Haemaphysalis, Ixodes and Amblyomma). Haemaphysalis longicornis was the most commonly collected species, which is represented for 96.88% of all the collected ticks and followed by H. flava (2.69%), I. nipponensis (0.35%), A. testudinarium (0.05%) and H. japonica (0.03%) in the Republic of Korea for the study period. Haemaphysalis longicornis was a dominant species observed in these eleven areas.

        We performed a survey for flavivirus infection and distribution of Aedes albopictus that known as Zika and Dengue virus vector using black–light trap and BG-sentinel trap around urban area in Korea. Mosquitoes were collected in 27 cities during March to November (twice a month) year 2016. Total numbers of mosquitoes collected 102,102 including 19 species 8 genera during collecting period. Total 21,467 Ae. albopictus was collected that 20,961(24.3%) by BG-sentinel trap and 506 (3.2%) by Black-light trap in urban area. Trap index(trap/night) of Ae. albopictus was showed highest in Hamyang (TI:992.3) and lowest in Taebaek (TI:0.3) there was only collected by Black-light trap. A total of 894 pools from all collecting Ae. albopictus were performed a Flavivirus detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases (include Zika virus) and vectors in Korea.

        The Ixodid ticks has been suspected to be the vector of severe fever with thrombocytopenia syndrome virus (SFTSV) in Korea. In order to confirm transovarial transmission of Ixodid ticks, a total of blood fed 284 ticks were collected from six host animals. They were identified as 3 genera and 4 species, Haemaphysalis longicornis (97.2%), H. flava (0.7%), Ixodes nipponensis (1.8%) and Amblyomma testudinarium (0.4%). Among them, 192 female adults hatched their eggs successfully. Our results showed a high prevalence of SFTSV among collected ticks (3.1%, 6/192 ticks), and we detected SFTSV from their larvae. This result suggests that the parent generation of H. longicornis can transmit SFTSV transovarially into their F1 progeny and H. longicornis might be one of major vectors of SFTSV in Korea.

        Haemaphysalis longicornis is a major vector for Severe fever with thrombocytopenia syndrome (SFTS) throughout Republic of Korea (ROK). The ticks distribute at vast open fields and various kind of habitate. Control of the ticks with pesticides is almost impossible and personal protection such as use of commercial repellents is very critical for control. However occasional toxic effects of the commercial repellents have been reported that include central nervous system depression and urticaria. In our studies, repellency of three plant essential oils was assessed and was compared with two commercial repellents. Lavendual angustifolia demonstrated the highest repellency and higher repellency than DEET when was individually tested, but binary mixture of Cinnamomum zeylinicam and Juniperus verginiana showed significantly higher repellency with synergic effects than mixture of Lavendula angustifolia and Juniperus verginiana. Further study on repellency of the C. zelinicam in field need.

        Previous laboratory repellent assessment with three plant essential oils against Haemaphysalis longicornis, a major vector for Severe fever with thrombocytopenia syndrome, revealed high repellency of Lavendual angustifolia and high synergic effects of Cinnamomum zeylinicam and Juniperus verginiana. Based on the previous laboratory repellent assessment field studies on repellency of the three plant oils were carried out with formulations such as cream type and spray type. 10% spray type of Cinnamomum zeylinicam demonstrated > 70% of repellency against H. longicornis. 10% spray type of Lavendual angustifolia showed > 80% of repellency. 10% spray type of binary mixture containing Cinnnamomum zeylinicam and Juniperus verginiana showed > 98% repellency against H. longicornis. Further studies on repellency of cream type of the plant oils might be carried out and compared with two commercial repellents.

        Control of the vector mosquitoes is critical for implementation of patient’s decrease. Vector mosquito control has been mainly carried out using pesticides in Republic of Korea (ROK), which has developed the resistance against selected insecticides. Voltage-gated sodium channel (VGSC) and Acethylcholine esterase (AchE) are the target of several classes of insecticides. Genetic mutations of VGSC and AchE have been documented to confer resistances to pyrethroids and organophosphates in mosquitoes, respectively. In our studies, the insecticidal resistable level of An. sinensis from four malaria risk areas near Demilitarized Zone (DMZ) was measured. The partial sequence of An. sinensis voltage gated sodium channel (VGSC) IIS6 domain for Kdr gene and ace1 gene for AchE was sequenced. An. sinensis from two areas demonstrated significantly higher resistance to pyrethroids and mutation rates of Kdr gene were comparatively higher. In all tested areas An. sinensis showed high insecticidal resistance to organophosphates and > 70% of mutation rates of ace1 gene ware higher. Understanding the mechanisms of insecticide resistance among vector mosquitoes populations is very important to efficient and effective vector control.

        Population density of Culex tritaeniorhynchus was annually monitored to predict the possibility of a Japanese encephalitis (JE) outbreak at 10 collection sites throughout Republic of Korea (ROK) during mosquito season from 2011 to 2015. Prevalence of Cx. tritaeniorhynchus in ROK was spatially and timely very variable and was significantly highest at Busan city during August. Monthly average population density of Cx. tritaeniorhychus showed high correlation to the monthly average daily average temperature and monthly average precipitation. Two models for the estimation of occurrence of Cx. tritaeniorhynchus based on annual monthly daily average temperature and monthly precipitation are shown with linear regression equations of exp(0.413×temperature-0.949) and exp(0.01258×precipitation+3.777). JE vector surveillance and vector control is warranted as part of an effective JE management program at ROK.

        The geographical distribution of ixodid ticks in were surveyed at 10 areas from April to November 2015. We investigated tick distribution using dry ice bait trap and flagging method as a part of disease vector monitoring in Korea. As a result, ticks collected 29,992 ixodid ticks with 3 genera, 7 species. Among them, Genus Haemaphysalis, Ixodes and Amblyomma accounted for 29,751 (99.2%), 223 (0.7%) and 18 (0.1%). Haemaphysalis longicornis was accounted for 88.9%, which has become the dominant species. Some species, such as H. japonica and Ixodes persulcatus were only collected Gangwon area. Metropolitan area had the highest population of ticks 6,540 (21.8%), Chungbuk area was next 5,049 (16.8%). In environmental distribution, Genus Haemaphysalis was highly collected in grassland site (32.1%) and Genus Ixodes was highly collected in coniferous forest site (43.5%). Upon analysis of the monthly distribution, H. longicornis, the dominant species, was highly collected at nymph stage in May, and its larva was highly collected in September. These studies may provide important information for control and management of tick in Korea.

        We conducted a investigation for distribution and flavivirus infection of mosquitoes using black –light trap around Incheon international airport there was possible overseas inflow caused by global warming in Korea. Mosquitoes were collected once a week April to October during 6 years (2009~2015). The numbers of mosquitoes collected in Oseong and Eurwang mountain were 8,969 and 7,978 including 12 species 7 genera respectively during collecting period. Culex pipiens complex was dominant species in two collecting area as 4,621(51.5%) in Oseong and 3,761(47.1%) in Eurwang mountain. A total of 16 pools from 88 Aedes albopictus were performed a Dengue virus (DENV) detection and total 628 pools from 11,146 other mosquitoes performed a West nile virus (WNV) detection. Flavivirus was not detected during study period. This study may provide basic information for surveillance of imported diseases and vectors in Korea.

        As the malaria surveillance agency, the Korean Center for Control and Prevention (KCDC) has been monitoring malaria vector mosquito density and Plasmodium vivax infection since 2009. Anopheline mosquitoes were collected at 35 sites in Incheon, northern Gyeonggi and northern Gangwon Province using black light trap daily from April to October 2015. P. vivax infection of malaria vector mosquitoes tested by polymerase chain reaction (PCR). In 2015, a total of 57,926 malaria vector mosquitoes were collected among 138,119 mosquitoes. Three P. vivax positive pools were detected among 1,556 pools (13,745 individuals) and its minimum infection rate was 0.22. According to the monitoring result of malaria vector mosquitoes, early summer (June-July) could be an appropriate time for a malaria elimination campaign until September.

        Dengue is the most important arboviral human infection worldwide. Approximately 50-100 million cases occur annually and more than 2.5 billion people are at risk of infection. This virus is possibility of overseas inflow to be high in Korea. We collected mosquitoes in Jeju, Busan, Gunsan, and Incheon using BG Sentinel trap from April to August in 2016. Collected mosquitoes were conducted virus detection using real-time PCR method. A total of 15 species comprising 7 genera were identified and 3,696 female mosquitoes collected. The most dominant species ratio (SR) was 49.4% (Culex pippins complex) followed by 21.8% (Aedes albopictus), 13.6% (Ae. vexans) and Ochlerotatus dorsalis (4.8%). Dengue virus was not detected in any mosquitoes until now.

        West Nile Fever is one of most serious disease spreading throughout world since 1990. West Nile Virus (WNV) is transmitted by infected mosquitoes. The possibility of overseas inflow of this virus seems to be high in Korea. To know the WNV infection of mosquitoes, we collected mosquitoes in Jeju, Busan, Gunsan, and Incheon using CDC light trap and BG Sentinel trap from April to August in 2016. Collected mosquitoes were conducted virus detection using real-time PCR method. A total of 22,632 mosquitoes representing 9 genera and 18 species were collected. The most dominant species was Culex pippins complex (36%) followed by Aedes vexans (13%), Ae. albopictus (10%) and Ochlerotatus koreicus (10%). WNV was not detected in any mosquitoes until now.

        The Asian tiger mosquito, Aedes albopictus, is considered as potential vector of Zika virus in Republic of Korea (ROK). Vector control during mosquito season is one of critical factors for decline of viral transmission. Total 14 oversea travel-associated Zika cases by mosquito bite were reported throughout ROK from January to September 2016 and vector control and monitoring at surroundings of patient’s residences was carried out during three weeks after confirmation of the virus. Although population density rates of Ae. albopictus were remarkably various according to ecological surroundings, population density of Ae. albopictus near forest was higher than urban. All captured Aedes mosquitoes were used for detection of three flavivirus, Zika, Dengue and Chikunguya, using RT-PCR and any virus was not detected. Population density of Ae. albopictus decreased > 65% on average after vector control and in one area > 95% of population density decreased. Our data might reveal that vector monitoring and control at surroundings near residences of oversea travel-associated Zika patients might effectively prevent viral transmission by mosquito bite and naturalization of the virus in ROK.

        We investigated the distribution of ixodid ticks in severe fever with thrombocytopenia syndrome (SFTS) case reported areas using flagging method from May to November 2014 in Korea. We collected 13,471 ixodid ticks with 2 genera and 3 species. Among them, Genus Haemaphysalis and Ixodes accounted for 99.9% (13,455) and 0.1% (16), respectively. Genus Haemaphysalis consisted of 2 species, H. longicornis (96.2%) and H. flava (3.7%). I. nipponensis was only collected in Genus Ixodes. H. longicornis was the dominant species (over 90%) in most study areas. Ixodid ticks were evenly distributed at various environmental sites, such as residences, hills, grassfields, fields, graves, copses, other sites. The further study on SFTSV infection in ixodid ticks will be needed.

        There are over 3,000 different species of mosquitoes throughout the world; currently 56 species are reported in Korea. Mosquitoes can act as vectors for many disease-causing viruses and parasites during blood sucking. Various species of mosquitoes are estimated to transmit various types of disease to more than 700 million people annually in Africa, South America, Central America, Mexico, Russia, and much of Asia, with millions of resultant deaths. At least two million people annually die of these diseases, and the morbidity rates are many times higher still. To prevent the spread of diseases, KNIH (Korea National Institute of Health) used three categorized methods; identifying or taxonomic analysis of mosquitoes, detecting virus caring mosquitoes, and detecting malaria from Anopheline mosquitoes. We have proved that taxonomic analysis using DNA barcording method (COI gene) is useful to complement identification of mosquito species. In detecting virus, we have reported Cx. orientalis as a new potential Japanese encephalitis virus (JEV) vector. We also reported that the Anopheline mosquito species composition and Plasmodium vivax infection rates in malaria hot spot in Korea.

        Aedes albopicuts, known as asian tiger mosquito or forest mosquito, is characterized by its black and white strips on legs and body. Ae. albopictus is an important vector mosquito for the transmission of many viral pathogens such as dengue fever and chikungunya fever. Unlike other mosquito, Ae. albopictus attacks people mainly during the daytime in the forest. To evaluate the diel activity of Ae. albopictus, three times mosquito collecting were conducted using BG Sentinel trap with dry ice in bamboo forest in Damyang-gun, Jeoullanam-do, in South Korea from August to September, 2009. Captured mosquitoes were counted and released every one hour during 25 hours experiment time. The result showed that Ae. albopictus activity began with sun rising at approximately 09:00, with peak in the early evening between 16:00 to 19:00, and ended with sunset at 21:00. Among the several factors affecting on the mosquito activity, light intensity seems to be a main factor. Especially, if the light intensity is over some threshold, it negatively effects on the activity of Ae. albopictus.

        The intracellular endosymbiont bacterium Wolbachia is currently considered the most abundant in arthropods and has also been isolated from nematodes, amphipods, isopods, mites and spiders. Recently, Wolbachia-based research was focused on the control of disease vector-population, such as several mosquitoes such as genus Aedes and Anopheles which cause dengue fever and malaria, respectively. For the analysis of regional difference between vector mosquito Aedes albopictus and Wolbachia, we selected different regions and collected Ae. albopictus which were distinguished with mountain chain and waterway. Whole genomic DNA were extracted from collected specimens with 9 regions. PCR analysis and sequencing were accomplished in each specimen for Wolbachia detection and identification using WSP gene. As a results, almost mosquitoes were infected with two strain of Wolbachia both wAlbA and wAlbB. However, regional separation of vector mosquitoes, wAlbA strain of Wolbachia were showed more than 98% sequence similarity. In this study, we first reported that Wolbachia infection and type of Wolbachia in Korea and endosymbiont Wolbachia was showed highly sequence homologies.

        Severe fever with thrombocytopenia syndrome (SFTS) is an emerging disease, causing high fever, thromobocytopenia, leukopenia, gastrointestinal simptoms (vomiting, diarrhea), hemorrhage and multiorgan dysfunction. It was known that Haemaphysalis longicornis (H. longicornis), a major tick in Korea, is the principal vector for severe fever with thrombocytopenia syndrome virus (SFTSV). Firstly, to investigate the existence and prevalence of SFTSV in the country, we collected the ticks in 9 provinces in South Korea and detected SFTSV from H. longicornis ticks from 2011 to 2012. And we also investigated SFTSV infection in ticks collected from healthy humans without SFTS symptom and their residential areas. From the former study, we collected 13,053 ticks, and H. longicornis (90.8%, 11,856/13,053) was the most abundant among them. The minimum infection rate (MIR) of SFTSV in H. longicornis was 0.5% (59 pools). From the later study, totally 422 ticks of 8 species were collected from humans and at their neighborhood throughout the nation from May to October, 2013 and SFTSVs were detected from 12 pools (5.7% MIR) by RT-PCR. Finally, we diagnosed 35 patients by detecting SFTSV gene with their sera, and isolated 26 SFTSVs among them in 2013. These findings indicate that SFTSVs are prevalent and SFTS is probably endemic in Korea.

        As a part of disease vector and possible overseas inflow caused by global warming monitoring around airport and port area, the distribution of domestic mosquitoes was investigated using BG-sentinel trap. Mosquitoes were collected during 5-days per each area in early September, using 10 traps in Cheongju (airport) and 15 traps in Pyeongtaek (port). The numbers of mosquitoes collected in Cheongju and Pyeongtaek area were 1,762 including 10 species and 1,042 including 11 species respectively. In Cheongju, Aedes albopictus (657 individuals, 37.3%) and Culex pipiens complex (415 individuals, 23.6%) were dominant species; while in Pyeongtaek, Cx. pipiens complex (806 individuals, 77.4%) was dominant followed by Anopheles spp. (90 individuals, 8.6%) and Ochlerotatus koreicus (79 individuals, 7.6%). Similar species were collected in both Choengju and Pyeongtaek area. Although most species collected in both Cheongju and Pyeongtaek area were the same, Cx. inatomii was only collected in Pyeongtaek. In case of the Genus Aedes, more number of Ae. albopictus was collected compared to Oc. koreicus in Cheongju; while Oc. koreicus was more in Pyeongtaek. In case of the genus Culex, the individuals belonging to the vishinui group were collected such as Cx. bitaeniorhynchus, Cx. tritaeniorhynchus and Cx. orientalis. This study may provide basic information for efficient prevention of vector mosquito and potential overseas inflow of diseases, also we would try to expand different area in Korea.

        The intracellular endosymbiont bacterium Wolbachia is currently considered the most abundant in arthropods. Many Wolbachia manipulate host reproductive systems, it lead to male-killing, cytoplasmic incompatibility, parthenogenesis and feminization of genetic males with large impact on host ecology and evolution in arthropods. In this study, we investigated the distribution of Wolbachia infection in Ae. albopictus according to geographical distribution. We selected 9 areas and collected Ae. albopictus, these areas were distinguished with mountain chain and waterway. Whole genomic DNA were extracted from collected specimens and PCR analysis were accomplished in each specimen using Wolbachia specific primers such as 16S and WSP genes. As a results, we collected 730 Ae. albopictus at least 30 specimens in each area. Wolbachia infection rate showed different patterns between geographical region. Cheonbook, Cheonnam and Yeongdong showed 100% infection rate followed by Jeju (97%), Chungchung (88%), and Gyungnam (85%) with two Wolbachia marker genes. Our results indicated that Ae. albopictus are commonly infected with Wolbachia, it is possible that Wolbachia may act as endosymbiont in Ae. albopictus regardless of geographical region. Although, low infection rate of Wolbachia, we need continuous survey for the evaluation of Wolbachia strain within Ae. albopictus as a vector of dengue fever.
