With the significant increase in spent ion-exchange resin generation, to meet the requirements of Waste Acceptance Criteria (WAC) of the Wolsong disposal facility in Korea, blending is considered as a method for enhancing disposal options for intermediate level waste from nuclear reactors. A mass balance formula approach was used to enable blending process with an appropriate mixing ratio. As a result, it is estimated around 44.3% of high activity spent resins can be blended with the overall volume of low activity spent resins at a 1:7.18 conservative blending ratio. In contrast, the reduction of high activity spent resins is considered a positive solution in reducing the amount of spent resins stored. In an economic study, the blending process has been proven to lower the disposal cost by 10% compared to current APR1400 treatment. Prior to commencing use of this blending method in Korea, coordinated discussion, and safety and health assessment should be undertaken to investigate the feasibility of fitting this blending method to national policy as a means of waste predisposal processing and management in the future.

Salvia miltiorrhiza (Danshen) is perennial plant and commonly used in traditional Chinese medicine to treat numerous diseases like cardiovascular and cerebrovascular diseases, coronary heart disease, myocardial infarction, stroke and some viral diseases. Nevertheless, no study has been conducted on the antiviral properties of crude extract of Salvia miltiorrhiza against Influenza virus. In an attempt to identify new potential anti-influenza virus agents, 200 natural oriental herbal medicines were screened and we found that Salvia miltiorrhiza has a potential anti-influenza effect. Therefore, in this study, we investigated the protective effect of aqueous extract from Salvia miltiorrhiza against divergent influenza A subtypes using murine model of influenza A infection. Effective dose of aqueous extracts of Salvia miltiorrhiza in BALB/c mice displayed higher survival rate and lower lung viral titers when challenged with lethal doses of influenza A subtypes ({A/Aquatic bird/Korea/ W81/2005(H5N2)}, {A/PR/8/34(H1N1)}, {A/Aquatic bird/Korea/W44/2005(H7N3)} and {A/Chicken/ Korea/116/2004(H9N2)}). In vivo results exhibited that Salvia miltiorrhiza induced prophylactic effect in BALB/c mice against Influenza virus by disrupting viral replication or preventing viral infection by creating an antiviral state in the lungs. Taken together, the use of aqueous extracts of Salvia miltiorrhiza as an orally active antiviral agent, will be potential candidates for prophylactic treatments against Influenza A virus for humans and animals.

This study is to compare the effect of estrus synchronization and embryo transfer between Korean and Mongolian cattle. Embryos were collected from 9 donors housed in Asan city in South Chungcheong Province, South Korea. Embryos were collected 9 donors from Khushaat sum, Selenge province and Bayanchandmani sum, Tov province in Mongolia. Follicle Stimulating Hormone (FSH), Controlled Internal Drug Release (CIDR) and Prostaglandin (PG) were used for superovulation. Subsequently, Artificial Insemination (AI) was done for donor cow and embryo was collected after 7 and 8 days. Collected embryos were compared between Mongolian and Korean cattle. Finally, good quality and fresh embryos were transferred to 50 and 22 recipients of cows in Korea and Mongolia respectively. The findings show that Korean native cattle each donor cow produced on an average 16.9 embryos and, 10.9 embryos were found transferable. But in case of Mongolia the average production of embryos per donor cow was 8.6 embryos and, 6.2 embryos were found transferable. Embryo collection after 7 and 8 days was not difference in embryo production in Korea. But, in Mongolia embryo production after 8 days was found more efficient than the 7 days. Korean native recipient's cows (74.6%) and Mongolian recipient's cows (71.0%) respectively were found transferable ovarian stage. The result suggested that efficiency of embryo production from the superovulation method treated of Korean cow were higher than the Mongolian cow. The pregnancy rate of Korea native cattle was 72%, which was about 10% higher than that of Mongolia cattle.

Bemisia tabaci is a species complex consisting of at least 40 species which are morphologically indistinguishable. Althoughgenetic diversity of B. tabaci have been studied in many countries, its information is lack in Bangladesh. The COI sequencewas determined from 91 samples collected in Bangladesh and the phylogenetic relationship was constructed. Four crypticspecies (Asia I, Asia II-1, Asia II-5 and Asia II-10) were present in different regions. While both Asia I and Asia II-1was widely distributed in all around the country, Asia II-5 was central and southern regions, and Asia II-10 was onlyin the central region. Otherwise, our results showed that Asia II-5 had the highest haplotype and nucleotide diversity.Genetic groups of B. tabaci in Bangladesh were most similar with those of Thailand, Vietnam, Pakistan, China, and Indiabut did not show any aggressive cryptic species such as MED and MEAM-1.

This study was conducted to investigate the effects of alpha-lipoic acid (aLA) as an antioxidant that decrease the reactive oxygen species (ROS) in bovine embryonic development. Slaughterhouse derived bovine immature oocytes were collected and 4 different concentrations (0, 5, 10 and 20 mM) of aLA was supplemented in bovine in Vitro maturation (IVM) medium. After 20 hrs of IVM, maturation rates, levels of ROS and glutathione (GSH), and further embryonic development after parthenogenetic activation (PA) and in Vitro fertilization (IVF) was investigated according to aLA concentrations. Maturation rate was significantly higher in 10 mM group than other groups (80.5% vs. 62.9, 73.9, 64.2%; P<0.05). In the levels of ROS and GSH in matured oocytes as an indicator of oocyte quality, significantly better results were shown in 5 and 10 mM groups compared with other 2 groups. After IVM, significantly higher rates of blastocyst formation were shown in 10 mM groups in both of PA (27.9% vs. 18.8, 22.3, 14.2%; P<0.05) and IVF (32.6% vs. 23.9, 27.3, 16.2%; P<0.05) embryos. In addition, significantly more cell total cell number and higher inner cell mass ratio in 10 mM PA and IVP blastocysts showed developmental competence in 10 uM groups. Therefore, based on the entire data from this study, using 10 μM of aLA confirmed to be the optimal concentration for bovine oocyte maturation and embryonic development.

The objectives of the present study were to investigate the effect of five chemical inhibitors on methane production of the pure cultures of methanogens. To examine the methanogenesis inhibition by 2-bromoethane sulfonic acid (BES), 3-bromopropane sulfonic acid (BPS), lumazine, propynoic acid and ethyl 2-butynoate, they were added into the pure cultures of Methanobacterium formicicum KOR-1 and KOR-12, Methanoculleus bourgensis KOR-2, Methanosarcina mazei KOR-3, KOR-7, KOR-9 and KOR-10, Methanosarcina vacuolata (DSM 1232), Methanobacterium bryantii (ATCC 33272), Methanosarcina mazei (ATCC BAA159) and Methanosarcina barkeri (ATCC 43240). Each methanogen was anaerobically incubated at 39℃ for 6 and 10 days, and then methane gas production was measured. All methanogens were sensitive to BES, lumazaine and ethyl 2-butynoate and as the addition levels of the chemicals increased, the methanogenesis was linearly reduced. BPS and propynoic acid failed to inhibit any of the methanogens. There were important differences among species of methanogens regarding their sensitivity to the different inhibitors. In general, the strains of Methanosarcina species were the most resistant to BES, lumazaine and ethyl 2-butynoate and the strains of Methanobacteriales order the least resistant. The different resistances to chemical inhibitors may be caused by different cell membrane structure, substrate utilization for methanogenesis and competition with other microorganisms for electron receptors. In conclusion, differences among methanogens habituating in anaerobic ecosystems, such as the rumen of ruminants regarding their resistance to chemical inhibitors should be considered when strategies of inhibition of methanogenesis are designed.

The objective of this experiment was to explore the effects of Roscovitine (Rosco) prior to in vitro maturation (IVM) of immature pig oocyte. Brilliant cresyl blue test has been used to select the good quality of oocyte. Specifically, the effects of Rosco exposure on nuclear and cytoplasmic maturation, diameter, intracellular glutathione (GSH) and reactive oxygen species (ROS), and embryonic development after parthenogenetic activation (PA) and somatic cell nuclear transfer (SCNT), and gene expression levels in SCNT embryos have been measured. Cumulus oocyte complexes (COCs) have been exposed in 75 μM of Rosco for 22 and 44 h. The COCs that were matured in the IVM for 44 h without Rosco used as control group. Diameter of matured porcine oocytes 44 h culture with Rosco was significantly lower than 22 h culture with Rosco and control groups. GSH was higher in control group than 22 h and 44 h with Rosco but reduction of ROS in 22 h than 44 h with Rosco. In PA, exposure with Rosco 44 h oocytes group has been significantly lower than 22 h and control group in rates of maturation, cleavage and blastocyst formation. Similarly, in SCNT embryos rates of maturation, cleavage and formation of blastocyst have been also significantly lower in 44 h Rosco treated group than other two groups. SCNT embryos treated with Rosco 22 h showed greater expression levels of POU5F1, DPPA2 and NDP52Il mRNA compared with other two groups. Our results demonstrate that Rosco treatment with 22 h prior to IVM improves the development competence of porcine oocyte.

Cut roses often have a short vase life due to water stress and ethylene damage under unfavorable postharvest conditions. In this study, we investigated the effect of various pretreatment solutions on the vase life and postharvest quality of the cut rose cultivar ‘Jinny’ (Rosa hybrida L.). Cut roses were pretreated with eight different preservative solutions for 10 hours: aluminum sulfate (AS), Chrysal (CHR), FloraLife (FLR), lysosome (LYS), MS-1 (MS1), MS-2 (MS2), silver n itrate ( SN), a nd s ilver t hiosulfate ( STS). We found that pretreatment with all solutions except LYS prolonged the vase life and improved the postharvest quality of the cut roses. Among these, STS was the most effective pretreatment solution, significantly extending the vase life from 11.8 days (control) to 19.9 days, retaining the initial fresh weight and a positive water balance for longer, and inhibiting microbial growth in the vase. STS also enhanced the water uptake rate, and maintained the high chlorophyll and soluble sucrose contents in the leaves of the cut rose flowers. In addition, we found that MS1 and MS2, which are natural plant extracts, had strong antimicrobial effects and consequently prolonged the vase life of cut roses by more than 4 days compared with the control. Therefore, MS1 and MS2 can be considered as alternative preservatives in the cut flower industry.

This study was conducted to establish the optimal chemical post-activation conditions in porcine embryonic development after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT) using 4 different chemical compositions (cytochalasin B (CB), cyclohexamide (CHX), demecolcine (DC), 6-dimethylaminopurine (DMAP). Porcine embryos were produced by PA and SCNT and then, cultured for post-activation with CB (7.5 μg/mL), CB (7.5 μg/mL) + CHX (10 μg/mL), CB (7.5 μg/mL) +DC (0.4 μg/mL), and CB (7.5 μg/mL) + DMAP (2 mM). In PA embryonic development, cleavage rates have been significantly higher in CB group (94.7%) and CB+DMAP group (94.1%) than that of CB+CHX and CB+DC group (88.1 and 84.3%, respectively). There have been no significant differences in blastocyst formation rates among the four groups. In cell number of blastocyst was shown in CB group (42.3%) significantly higher than CB+CHX and CB+DC group (40.6 and 40.6%, respectively). In SCNT embryonic development, CB+DMAP group (89.7%) significant differences were found on embryo cleavage rates when compared with other three groups. Blastocyst formation rates in CB+DMAP group (26.9%) were significantly higher when compared with CB, CB+CHX, and CB+DC groups (25.5, 20.2, and 22.1%, respectively). In blastocyst cell number, CB+DMAP group (41.4%) was found higher significant difference compared with other three groups. Additionally, we have investigated survivin expression in early development stages of porcine SCNT embryos for more confirmation. Our results establish that CB group and CB+DMAP group for 4 h during post-activation improves pre-implantation improvement of PA and SCNT embryos.

This article explores how the concepts of masculinity and socialisation influence the decision-making process young males undergo when purchasing and using typically feminine related products (Beauregard, 2007). We use socialisation theory to understand the varying differences in the progression and perceptions of males today with regard to masculinity and skincare products. The article utilises a scenario exercise to cross reference their attitudes to male skin care brands and we propose that male attitudes to skincare and their socialised background are inherently linked. The result show six latent themes driven by theory-led coding (Kotler, 2000; Otnes and McGarth, 2001; Kraft and Weber, 2012). The ‘act’ of masculinity and the ‘adoption of femininity’ explain the influences in early socialisation. Meanwhile, ‘efficiency’ (or practicality), ‘help’, ‘trust’ and ‘past experience’ are additional influencers in the decision-making process. Specifically, males with predominantly female skincare users in the home embrace skincare related products, whereas males who were brought up to ‘act’ like men, particularly without a father presence in the home, are more likely to adhere to the masculine stereotype and avoid using such products. Finally, males with a strong father figure in the home are found to differentiate from their fathers, either by avoiding the same brands or choosing not to use the products at all. These behaviours (social, psychological and personal) are the external factors which ultimately affect their final purchase choice. Overall, the article offers an extended understanding on these external influences that drive the purchase decision of this under-researcher consumer segment.

The New Product Development (NPD) in the textile industry is peculiar, as it follows constraints that are particular to the fashion business. A qualitative study comprising 45 interviews were conducted in a South American textile industry and results showcase that fashion forecast, pressure for newness, and cost reduction are relevant matters. Introduction The textile industry follows a particular cycle for New Product Development (NPD) as it answers the demand from the fashion industry. Fashion supply chain is fragmented, as the production process is fragmented with companies playing different roles in the production cycle. There are companies that uses the raw material to produce yarns that will be transformed into fabrics, which then it is transformed into clothes, through process that may be a single sewing to complex process that demands intermediate process such as industrial washes. These clothes are sold to consumers through various selling channels, such as stores, retailers, e-commerce, and even door-to-door. Usually one company plays only one of these roles in the supply chain, what makes the production plan to be fragmented amongst several different companies in the supply chain (Şen, 2008). After globalization, this supply chain became even more complex as each chain may be located in different parts of the world, due to production costs and optimization. This scenario results in a supply chain that demands several planning skills, that works, sometimes, with extensive production cycles. (Thomassey, 2014). But, as Christopher, Lowson, & Peck (2004) noted, the supply chain in the fashion business must be agile to respond to changes in demand from consumers, which often occurs, and it can be also very complex as clothing is a way for consumers to either differentiate or fit in society, and the rules to be in or out of society patterns changes fast (Cholachatpinyo, Fletcher, Padgett, & Crocker, 2002a, 2002b). Due to this complexity and richness of information, the production cycle in fashion business has been extensively studied in the literature (Alexander & Contreras, 2016; Aung & Sha, 2016; Oxborrow & Brindley, 2014) but authors often use a wide perspective to grasp an overview of the supply chain they are studying, using market data and information, or gathering general information from companies that are present in the fashion supply chain. Authors focused their analysis in the supply chain itself, and we could not find studies that focused on the perspective of the product development task in the challenging context of fashion business. There are some characteristics from the fashion business that differentiate New Product Development (NPD) from a classical Business to Business (B2B) scenario. In a brief description of the fashion business production system, we see that it starts with the production of raw materials that varies from natural fibers (e.g. cotton, silk, linen) or artificial fibers (e.g. polyester, nylon, elastane) that are then spinning into threads that may contain one or more different fibers. These threads are then transformed into fabrics through several different weaving techniques, and then it may go through some finishing treatments (e.g. dying, washing), and then it goes to garment manufacturing where garment makers produce clothes for their own brands or for third-parties brands. These clothes are then sold to consumers through various channels (e.g. retail stores, electronic stores). If we analyze this brief description we can see that fashion may influence in all parts of the production cycle, including the demand for raw materials: if denim products are in vogue, then there is a growing demand for cotton products, if sportswear is in vogue, then the demand improves for artificial fibers, and so on. One other thing that should be noted is that usually each part of the production cycle is performed by a different company: the most common scenario we see in the textile industry includes companies that produce only threads, and sell these threads to textile companies that produce fabrics to sell to garment makers that produces clothes to sell to brands to sell them to consumers. There are some companies that integrates two or more parts of the production cycles but they are the exception, not the rule. This fragmented production process combined with the fashion influence result in long production cycles: a designer’s idea may take from three to eighteen months to be available to consumers, depending on the production channels assessed by the designer. These characteristics showcase how NPD in the fashion business may be complex and therefore it should be investigated further in the literature, and this is the objective of this study. In order to do that, we researched the specific literature in NPD that is vastly discussed in literature as it is responsible for creating, managing and releasing products that are both appealing to customers and profitable to the company. (Brown & Eisenhardt, 1995). Understanding what makes an NP succeed in the market has been on the marketing agenda for a long time, including the fashion business (Alexander & Contreras, 2016; Clarke, 2012; Thompson & Haytko, 1997). One interesting point of view is to understand the predecessors of NP success in the market. Henard & Szymanski (2001) conducted a meta-analysis of 60 studies focused on the antecedents of NP success and proposed a list of 24 predictors of NP performance, which they grouped into four categories: (i) product characteristics, (ii) firm strategy characteristics, (iii) firm process characteristics and (iv) marketplace characteristics. We selected the most relevant predecessors discussed by Henard & Szymanski (2001) and Song & Xie (2000) to use as a basis for a script for interviews with NPD professionals. Methodology We conducted in-depth interviews with 45 professionals from different areas that participate in, or are impacted by, NPD process in a South American textile industry. From now on we will call this company “Southex”. These interviews offered a deep understanding of the perspective of NPD professionals, both insiders (professional that work in the NPD team) or outsiders (professional from departments that work as an internal supplier or client for the NPD team). Southex is one of the biggest textile producers in the world, with producing units in Latin America, commercial offices in Europe, Asia, and America. Sales are over U$ 1 bi/year, and the company employed more than 8,000 people in 2012, when this study was performed. Their main market is Latin America where fashion seasons are in opposition to the ones in the North part of the globe, home of the fashion trendsetters for global macro trends (Europe, USA, and Japan). This leads to a one-year postponement of fashion trends, so when something is released as a fashion trend for summer in Europe, it will be considered a trend for summer one year later in Latin America. Southex NPD team had 12 exclusively dedicated professionals as of 2012. Southex launches products in the market with the concept of collections, when the company releases a group of NPs in the market. Every year, the company releases two collections in the market. It usually takes up to six months for NPD team to complete and launch a new collection. Research into fashion trends, commercial needs and technological improvements available in the market are the main sources the company uses to develop NP. Prior to launching a product in the market, the NPD team needs to test its production process in order to guarantee that the product can be produced on a large scale at competitive costs. Every collection is launched as a sample of fabrics that will be produced 4-6 months in the future. If a collection is released in January, the products that Southex is launching will start to be produced in June. So their clients, mainly garment makers and fashion brands, need to coordinate their production chain to start receiving these fabrics after June. Considering the production cycle in these garment makers, it is estimated that the clothing produced with the fabric launched by Southex in January will be delivered to retail stores (and then available to the consumer) in November, almost one year after Southex has launched the fabric in the market. In order to gather a deep perspective of all matters related to NPD we interview 45 professionals, totaling over 1,600 hours of recorded interviews. 12 insiders, comprising 100% of the team available at that moment, and 33 outsiders from departments such as Marketing, Sales, and Technical Support. Their positions ranged from analysts to directors.. All interviews followed a predefined script of topics. Interviews were transcribed and sent to the corresponding professional for validation, during which they could ask for adjustments. 12 professionals asked for changes in the transcription and we used the changed and approved text, together with the 33 unchanged texts in this analysis. Results and Discussion All texts were then codified according to content, which involved identifying similar topics and applying codes to facilitate analysis of the interviews. Using the predecessors of NP success has facilitated our understanding of the matters related to NPD at Southex. By formatting the presentation of information gathered in the interviews with the predecessors we were able to see the main issues found in NPD. In total, there were 421 mentions codified. After analyzing the codification of the interviews, we can see that Cross function is the main topic addressed by respondents. This is due to the relationship that the flow of NPD activities in the company. As we can see, there were several issues related to the relationship of NP team with the outsiders and that communication amongst insiders and outsiders needs to be improved. There are some context-specific content that appeared as relevant in the analysis of these interviews: insiders are worried with the pressure they feel to forecast fashion preference for products. They say that fashion industry is constantly demanding new technologies, new products, and this has been creating a pressure to accelerate product development they think it is not healthy to the system. Also there is a general worry (insiders and outsiders) with the pressure for price reduction in basic products. Some products are considered basic and are included for several collections (interviewees mentioned that some products are included in more than 15 successive collections) and for these products instead of demanding for innovation, there is a demand for price reduction which leads outsiders to perform tests to substitute materials or changes in the production to reduce costs without prior knowledge from NPD team, which causes several problems in the organization. It is important to point out that even though we used only one company in this study, it was a complete and deep perspective, as we were able to collect high quality data with all 45 professionals that are directly or indirectly related to NPD in the company. Also, as Southex works as a thread and fabric producer, it showed us an insider perspective about the textile supply chain in the fashion business. Even though the exploratory study presented the reality of a firm in an extensive and very detailed way, it may not reflect the full complexity of other organizations in the market; thus, caution is advised if applied in a market that differs deeply from the scenario presented in this study. However, it is also important to remember that this is a qualitative approach where generalization is not expected, and the attention for details is important.

The objective of this study was to establish the effect of post-activation treatment with cytoskeletal regulators of CB, CB+CHX, CB+DC, CB+6’DMAP on embryonic development of pig oocytes after parthenogenesis (PA) and somatic cell nuclear transfer (SCNT). PA and SCNT embryos were produced by using in vitro matured pig oocytes and treated for 4 h after electric activation with cytochalasins B (7.5 μg/ml), CB+cycloheximide (10 μg/ml), CB+demecolcine (0.4 μg/ml), and CB+2mM 6-Dimethylaminopurine. Post-activation treatment of PA oocytes with CB, CB+CHX, CB+DC and CB+6’DMAP no significant differences were found in embryo cleavage (83.2~91.5%), mean cell number of blastocysts (40.6~ 42.3% cells/blastocyst) but significantly (P<0.05) differences blastocyst formation (28.6~36.4%). When PA oocytes were treated with CB, CB+CHX, CB+DC, CB+6’DMAP, blastocyst formation was significantly (P<0.05) improved by CB (36.6%) compared to CB+CHX (30.9%), CB+DC (28.6%) and CB+6’DMAP (35.2%). In SCNT, was not significantly (P<0.05) increased by post-activation treatment with CB+CHX (81.3%), CB+DC (83.9%) and CB+6’DMAP (90.0%) compared to CB (84.5%) on embryo cleavage, blastocyst formation (19.1%~23.6%) and blastocyst cell number (39.6~41.4% cells/blastocyst) also were not influenced. But increased tendency in CB+6’DMAP. In addition, we investigated survivin expression in porcine SCNT embryos during the early developmental stages. The levels of survivin mRNA in 2-4 cell stage SCNT embryos were significantly higher 6’DMAP treated group than other treatment groups of SCNT embryos. These observations suggested that 2-4 cell cleaving embryos at have high developmental competence, and which may be influenced by survivin expression in porcine SCNT embryos. Our results demonstrate that post-activation treatment with CB, CB+CHX, CB+DC, CB+6’DMAP improves pre-implantation development of SCNT embryos and the stimulating effect of cytoskeletal modifiers on embryonic development is differentially shown depending on the origin (PA or SCNT) of embryos in pigs.

Introduction Porcine embryonic stem cells (pESCs) derived from cloned embryos might be a useful animal model in biomedical research, however, establishment of cloned pESCs is difficult by its incomplete nuclear reprogramming. Here, we report the improved development competence of porcine cloned embryos by vitamin C (VC) supplement to establish the pESCs. Materials and Methods Slaughterhouse-derived oocytes were in vitro matured for 44h and parthenogenetic and cloned embryos were produced using matured oocytes. Both of embryos were cultured for 6 days in PZM-5 media and development rates were examined. Four different concentration of VC (0, 25, 50, 100, and 200 μg/ml) was supplemented in IVM and IVC media and preimplantation developments in the 5 groups were compared in both of embryos Results and Discussion In the cleavage rates of IVM group, significantly higher rate was shown in 50 mg/ml group than other groups (84.5 ± 0.6% vs. 69.8 ± 5.5, 75.7 ± 1.8, 80.4 ± 0.2, 72.4 ± 0.1%; P<0.05), respectively. Significantly higher rates of blastocyst development also were shown in 50 mg/ml group than other groups (27.0 ± 2.0% vs. 20.4 ± 1.4, 22.1 ± 1.3, 23.7 ± 1.2, 19.6 ± 1.3%; P<0.05), respectively. In the cleavage rate of IVC group, non-significantly different with each group (84.0 ± 1.3, 86.7 ± 1.0, 88.4 ± 1.4, 76.7 ± 3.0, 64.6 ± 4.4; P<0.05). In the blastocyst rate of IVC group, significantly higher rate was shown in 25mg/ml and 50 mg/ml group than other groups (22.3 ± 1.7, 23.8 ± 1.7% vs. 19.1 ± 1.3, 15.9 ± 1.0, 5.8 ± 1.5%; P<0.05) In conclusion, supplement of 50μg/ml of VC in IVM and IVC media enhanced the development of porcine parthenogenetic embryos and these results will be a helpful information in the development of porcine cloned embryos and derivation of its embryonic stem cells.

Introduction The objective of this study was to investigate the effects of staining of porcine cumulus-oocytes complexes (COCs) by brilliant cresyl blue (BCB) test prior to in vitro maturation may be used to select developmentally competent oocytes. Furthermore, milrinone can be used to promote developmental competence of porcine embryos produced during parthenogenesis (PA) and somatic cell nuclear transfer (SCNT). Materials and Methods Slaughterhouse-derived porcine cumulus-oocyte-complexes (COCs) were exposed to BCB and treated oocytes divided into BCB+ (colored cytoplasm), BCB- (colorless cytoplasm) groups. After division into 2 groups, intracellular glutathione (GSH) and reactive oxygen species (ROS) of matured oocytes were compared. And, preimplantation development of PA and SCNT embryos were also compared between 2 groups. BCB- oocytes were exposed to milrinone with different concentrations (0, 50, 75, and 100μM) for 6 h prior to IVM for further development of embryos. Results and Discussion GSH was higher in BCB+ group than BCB- group whereas ROS was lower in BCB+ than BCBgroup. In parthenogenetic embryos, BCB+ oocytes group was significantly higher on maturation (87.5 vs 80.6, 71.3%), cleavage (88.6 vs 82.9, 76.3%), and blastocyst formation rates (34.3 vs 27.8, 25.3%) than control and BCB- oocytes groups, respectively. Moreover, ratio of ICM:TE cells were higher in BCB+ oocytes group (30.3% vs. 28.6, 26.4%, respectively) than other groups. In cloned embryos, the significant higher blastocyst formation rates were shown BCB+ groups (30.6% vs. 26.0, 20.1%) than BCB- groups. To improve the cytoplasmic maturation in BCB- oocytes, 4 different concentrations of milrinone (0, 50, 75, and 100μM) were supplemented in the IVM media for 6 h. BCB- oocytes supplemented with 75μM milrinone showed the significantly higher rates of blastocyst formation than other groups. Our results demonstrate that staining of porcine oocytes with BCB before IVM may be used for selection of good quality oocytes and milrinone supplementation can be used to improve embryo developmental competence of porcine embryos.
