        The purpose of this study was to investigate the complex ankle exercises on balance. 22 participants (male: 14, female: 8) with functional ankle instability were participated. Functional ankle instability was selected to be less than 24 points using the Cumberland ankle instability tool (CAIT) with people who had severe ankle sprain and then experiencing ankle giving way. A total of 20 minutes performed three times a week for four weeks with muscle strength and balance exercises. Muscle strengthening exercise was performed with Theraband, and balance exercise was performed with unstable support plates. Biodex balance systemⓇ was used to measure static and dynamic balance. The dynamic balance was selected in grade 2, 4, and 8. The static and dynamic balance (grade: 2, 4,and 8) balance was significantly decreased in anterior-posterior, and medial-lateral directions (p<.05). The instability was significantly increased after exercise (p<.05). These results suggest that complex exercises are beneficial to decreasing the functional ankle instability.

        The purpose of this study is to identify the effects of two trunk stability exercise types on the gait factors of stroke patients. We randomly divided 24 old elderly patients with hemiplegia, who were hospitalized due to stroke, into a two groups, each with its own six-week exercise program: one that used of a dynamic trunk stability exercise using with physio-balls(n=12) and a group of one that used a static trunk stability exercise using on mats(n=12). After measuring the participants gait ability a sin a pre-test, we again measured their ability again as in a posttest after two-for both types of six-week exercise programs for each group. The analysis of the data analysis showed that both ball and mat exercise programs significantly improved the participants’ gait velocity and stride length; cadence, however, was significantly changed only by the ball exercise program. In conclusion, both types of trunk stability exercise may be useful in improving the gait ability of stroke patients, and, in particular, the former can be used as an exercise method that effectively significantly affects more various other gait factors.

        The compressive strength and electrical resistance of pitch-based carbon fiber (CF) in cementitious materials are explored to determine the feasibility of its use as a functional material in construction. The most widely used CFs are manufactured from polyacrylonitrile (PAN-based CF). Alternatively, short CFs are obtained in an economical way using pitch as a precursor in a melt-blown process (pitch-based CF), which is cheaper and more eco-friendly method because this pitch-based CF is basically recycled from petroleum residue. In the construction field, PAN-based CFs in the form of fabric are used for rehabilitation purposes to reinforce concrete slabs and piers because of their high mechanical properties. However, studies have revealed that construction materials with pitch-based CF are not popular. This study explores the compressive strength and electrical resistances of a cement paste prism using pitch-based CF.

        To determine the medicinal properties of Pleurotus cornucopiae and Safflower Oil (PS), the osteoblast effect was investigated. PS can prevent estrogen deficiency-induced bone loss. It was taken the measurements of biochemical factors such as serum osteocalcin, alkaline phosphatase, IGF-1, and bone status. Forty adult female Sprague-Dawley rats were randomly assigned to four equal groups for 12 weeks of treatment: (1) sham operation (SHAM) + vehicle; (2) ovariectomy (OVX) + vehicle; (3) OVX + 17β-estradiol (25 μg/kg); (4) OVX + PS (300 mg/kg/d, PO). At the end of the experiment, bone turnover, and trabecular microarchitecture were assessed by micro-computed tomography (micro-CT), and histopathology, respectively. Ovariectomized rats experienced remarkable increases in global volumetric bone density, and trabecular microarchitecture deterioration. These OVX-induced pathological changes are reversible in that most of them could be mostly corrected upon 17β-estradiol treatment. PNS treatment significantly prevented bone mass loss and microarchitecture deterioration. Moreover, PS enhanced osteoblast activity but suppressed osteoclast turnover, as evidenced by decreased levels of serum C-terminal telopeptides of type I collagen and elevated levels of alkaline phosphatase. PS mitigates estrogen deficiency-induced deterioration of trabecular microarchitecture.

        Oocyte is the central factor in the bi-directional communication axis in the ovarian follicles. It controls the cumulus or granulosa cells to perform functions which are beneficial for its own development via secreting paracrine growth factors, including GDF9 and BMP15. The aim of this study was to investigate whether the recombinant GDF9 and BMP15 are able to promote meiotic resumption and cumulus expansion of canine COCs during IVM, as well as to demonstrate the actions of GDF9 and BMP15 in regulating the expression of connexin transcripts in the ovarian granulosa cells. As results, GDF9 and BMP15 significantly improved the meiotic resumption rate and cumulus expansion by activating ERK1/2 signaling. Treatments with GDF9 significantly improved the expression of CyclinB1 but inhibited the expression of Cx43 transcripts. In addition, cumulus expansion genes (MAPK1, Ptgs2, Tnfaip6 and Ptx3) were differentially improved by GDF9 and BMP15. In the ovarian granulosa cells, GDF9 suppressed the expression of Cx43 transcripts by binding ALK4/5/7 receptors and activation Smad2/3 signaling, whereas, BMP15 stimulated the expression of Cx43 transcripts by binding ALK2/3/6 receptors and activating Smad1/5/8 signaling. In conclusion, by regulating functions of granulosa/cumulus cells, oocyte has the potential to enhance the growth and maturation of itself.

        Somatic cell nuclear transfer (SCNT) is the technique which generates embryos by transferring diploid nucleus into an enucleated oocyte, it has produced specific animals successfully in a variety of species. However, the developmental capacity of SCNT embryos is still relatively lower than that of embryos produced in vivo. Oocyte is a kind of lipid rich cells, its quality limits the efficiency of embryo production. L-carnitine is a co-enzyme facilitating the transportation of long chain fatty acids across the inner mitochondria membrane where fatty acids are used for generating adenosine triphosphate (ATP) via beta-oxidation. It also has antioxidant actions which may protect mitochondrial membranes and DNA against damage induced by reactive oxygen species (ROS). Whether L-carnitine is functional in bovine SCNT embryos are unknown. Therefore, the objective of this study was to examine the effects of L-carnitine on oocyte maturation and developmental competence of subsequent SCNT embryos. L-carnitine was supplemented during IVM, then intracellular ROS and GSH levels, mitochondrial activity, gene expression of COCs were analyzed at the end of IVM. SCNT embryos were produced subsequently, apoptosis detection and gene expression evaluation were performed in blastocysts. In the results, treatments with 1.5 mM and 3 mM L-carnitine significantly improved maturation rates (P<0.05). Treatments with 3 mM L-carnitine effectively induced improvement in nuclear maturation, intracellular GSH levels and mitochondrial activity, as well as a reduction in intracellular ROS levels (P<0.05). mRNA levels of CPT1A, ACAA1, ACAA2, AREG, EREG, SOD1, GPX4, GLUT1 and CDC2 transcripts were effectively up-regulated by 3 mM L-carnitine treatments (P < 0.05). Similarly, 3mM L-carnitine induced an increase in blastocyst developmental rates and an improvement in blastocyst quality (P<0.05). Our study indicates that L-carnitine treatment during IVM improves oocyte nuclear maturation and subsequent SCNT embryo development.

        The mesenchymal stem cells (MSC) has been investigated as a source of stem cell therapy to replace and treat damaged cells. Human endometrial epithelial and stromal cells was isolated from hysterectomy tissue and the direct evidence of stem/progenitor cells in the human endometrium was identified. Endometrium derived stem cells (EnMSCs) are known to have a high proliferative ability, genetic stability, lack of tumorigenicity and low immnunogenicity during long-term cultivation. Here, we aimed to identify MSC in canine endometrium and characterize its potential to differentiate into decidua cells. EnMSCs were isolated from thrown-away spayed uterus of adult canine depending on their estrus cycle, and identified by flow cytometry, immunocytochemistry and flow cytometry with MSC specific markers. We then characterized the ability of EnMSCs by the doubling-time analysis, colony-forming units and MSC differentiation assays. Isolated EnMSCs expressed stem cell specific genes (Sox2, Oct4, Nanog, MCAM, Endoglin, Susd2 and IGTB) and MSC surface markers (CD90, CD44 and CD117). EnMSCs are also differentiated into adipogenic, osteogenic and chondrogenic cells morphologically under modified conditions with the expression of lineage specific genetic markers. EnMSCs showed higher proliferation ability than canine amniotic fluid derived MSCs which were used as a positive control. EnMSCs were cultured at low density (10, 20, cells/cm2) and initiated to form small colonies of loosely-arranged cells and gradually formed large colonies of densely-packed cells which underwent self-renewal with high proliferative potential which is similar to the clonogenicity feature of human endometrium-derived stem cells. EnMSCs were then induced to differentiate into decidua cells with 0.5 mM dbcAMP. After 14 days, EnMSCs changed their morphology into the elongated and rounded shape. The induced decidual cells expressed PRL and IGFBP1 which are typically expressed in decidua cells. In conclusion, we successfully isolated and characterized MSC in the canine endometrium which differentiated into decidua cells. These results showed that endometrium may be a promising source of stem cells, and furthermore raise the possibility of canine EnMSCs as a novel hypothetical decidualisation model of infertility associated with decidualisation insufficiency and implantation failure.

        We collected four species of Osmia spp. consisting of O. cornifrons, O. pedicornis, O. taurus and O. satoi in Yeongwol, Bonghwa and Yechon locations. O. pedicornis was the dominant species, which accounts for 59.6% to 76.4% in the three locations. The sex ratio of Osmia spp. were from 1:1 to 1: 2.8 in O. cornifrons and from 1:2.8 to 1: 5.5 in O. pedicornis. The sex ratio of O. cornifrons and O. pedicornis was different in 3 locations. The female and male cocoons of O. cornifrons were the long elliptical-shaped and grey-brown color. The female and male cocoons of O. pedicornis were the long elliptical-shaped and polished dark-brown color. In both species of O. cornifrons and O. pedicornis, the cocoon size of the females was significantly larger than that of the male. Between those two species, the latter had a relatively bigger size than the former.

        In the age of sexual maturity of queen, mating occurred at 6.7% immediately after eclosion and it was the highest as 85.0% at 10 days after emergence. However, a remarkable decrease was occurred at 20 days after the emergence. With regard to oviposition rate, the highest rate was observed as 81.3-81.8% at 6 to 10 days of eclosion. The rate of colony foundation and progeny-queen production were the highest as 43.8% and 37.5%, respectively, at 8 days of eclosion. In the age of sexual maturity of male, mating rate was as high as 38.3% immediately after the eclosion, the highest as 80.0% at 25 days of eclosion, and 76.6% at 8 days of eclosion. The oviposition rate was the highest as 76.9% at 6 days of eclosion, and decreased to 75.0% at 8 days and 72.7% at 10 days of eclosion. Summarized, our results indicate that sexual maturity for mating of B. terrestris is most favorable 6-8 days after eclosion for queen and 6-10 days after for male.

        Ethyl formate (EF) and phosphine (PH3) is an alternative fumigant to methyl bromide (MB). The egg, nymph, and adult stages of Frankliniella occidentalis in asparagus were examined for the fumigation activity of EF and PH3. The eggs of F. occidentalis were more tolerance than other stages (adults and nymphs) in both EF and PH3. When the EF was treated for 4 hrs at 5℃ and 20℃, the eggs of F. occidentalis were LCT99=98.70 mg/L and LCT99=61.13 mg/L, and adults were LCT99=11.50 mg/L and LCT99=3.18 mg/L, respectively. However, at 5℃ and 20℃, the eggs were LCT99=83.76 mg/L and LCT99=53.6 mg/L, and adults were LCT99=4.58 mg/L and LCT99=3.44 mg/L, respectively for 4 hrs treatment of PH3. The PH3 was not any external phytotoxic damages at the maximum dose (4 mg/L), but the EF caused significant phytotoxicity in asparagus.

        Entomopathogenic fungi, widely distributed in nature, have been developed as biopesticides of agricultural pests. However, little is known about isolation method for obtaining a variety of highly virulent strains from the soil. In this work, the previously established method using mealworm (larvae of Tenebrio molitor) as a baiting system was used to efficiently isolate fungal strains from soil, and the isolated fungal library in Korea was established. To isolate entomopathogenic fungi, soil samples from 10 regions of Korea such as Iksan, Imsil, Gunsan, Yeosu, Suncheon, Gwangyang, Jinju, Hamyang, Boseong and Jangseong were collected. Of the collected soils, 21 and 34 isolates of entomopathogenic fungi were isolated from Iksan and Gunsan, respectively. The isolates were classified as genera of Metarhizium, Beauveria and Isaria. This work can suggest an effective insect pathogenic fungal isolation method

        Entomopathogenic fungi have been known as promising candidates in biological control. However, a large number of conidia with higher stability should be produced for industrialization. In this work investigated the possibility of several grains for the production of thermotolerant Isaria javanica and I. fumosoroseus conidia. The fungal conidia were produced on ten grains for 14 days in Petri dish conditions. Of the ten granular substrates, millet, perilla seed and barley showed the high conidial production, 15.7 ± 8.4, 17.3 ± 5.2 and 13.5 ± 1.9 × 109 conida/g in I. javanica and 15.9 ± 0.6, 15.0 ± 0.5 and 15.0 ± 0.7 ×109 conida/g in I. fumosoroseus, respectively. However, the two strains showed relatively low thermal stability regardless of substrates, when exposed to 45℃ for 1, 2 and 3 hours. This work suggests a possible substrate for conidial production of entomopathogenic fungi with low cost.

        Agrobacterium tumefaciens-mediated transformation (AtMT) has been widely used for generation of fungal transformants and recently applied to Beauveria bassiana. In this study to comprehend how the AtMT promoter influences on the expression of selection marker (hygromycin B resistance gene; hph), two different Ti-Plasmids were constructed: pCeg (gpdA promoter-based) and pCambia-egfp (CaMV 35S promoter-based). Putative transformants were subjected to the PCR, RT-PCR and qRT-PCR to inspect the T-DNA insertion rate and gene expression level. In conclusion, more than 80% of the colonies succeeded in AtMT transformation and the hph expression level of AtMT/pCeg colonies was higher than that of AtMT/pCambia-egfp colonies. This result can provide useful information on the AtMT of B. bassiana, especially antibiotics susceptibility and promoter-dependant expression level.

        Entomopathogenic fungi are expected to play a role as great biological control agents in the global bio-pesticide market in the future. The first step is the collection of fungal isolates and it should be a platform for the development of highly effective biopesticides. In this work, we constructed a fungal library using a mealworm pathogenecity-based fungal collection method and further characterized some isolates with high virulence. A phylogenetic tree was generated, and of the isolates 17 isolates’ biological features were characterized, such as morphology, virulence against several insect, stability of conidia for heat, production of biologically active materials, such as enzymes. This work reports an attractive entomopathogenic fungal library including the information of effective isolates in pest management.

        Many turfgrass pests, Ectinohoplia rufipes, Exomala orientalis and Popillia quadriguttata, cause serious damage to grass fields in Korea. In this study, control of turfgrass pests using entomopathogenic fungi were investigated using Protaetia brevitarsis seulensis as an alternative insect. In a preliminary experiment, some Metathizium sp. showed ca. 60% virulence after 7 days exposure to the entomopathogenic fungi. Compared to the contact-exposure, overall a spray treatment showed lower control efficacy against P. brevitarsis seulensis. Furthermore, mortality of various developmental stages of P. brevitarsis seulensis was investigated under laboratory conditions. This work suggests that when treated with fungi mortality of P. brevitarsis seulensis was closely related with developmental stage. These results provide a basic information for the control of P. brevitarsis seulensis using entomopathogenic fungi

        Entomopathogenic fungi are potential pesticides as biological control agents. However, many previous studies have focused on basic researches to control pest using insect-pathogenic fungi. Herein, the molecular characteristics of insect defense mechanisms against fungal pathogens and fungal pathogenicity to insects were investigated. The whole genome of Beauveria bassiana (Bb) JEF-007 was sequenced (36,538,394 bp) using Pacbio sequencing. Based on this data, the transcriptomes of bean bug and fungus were analyzed using RNA-seq. In the transcriptome analyses, six and five genes were validated to be involved in the fungal pathogenicity and insect defense, respectively. This work can provide an understanding of the insect-pathogen interactions
