        The purpose of this study is to measure the effect of change in heel height on lower extremities activity of young women on high-heeled shoes that young women prefer from more kinetic and realistic perspective as this study changes the degree of slope on a treadmill. The study subjects are 15 young and healthy women who do not have any external injuries or problem with walking and understand the purpose of this study clearly. They wore three different height of heels(1cm, 7cm, 12cm) and walked on a treadmill at a constant speed of 3km/h. EMG value of four muscles (anterior tibial muscle, gastrocnemius muscle, straight muscle of thigh, and biceps muscle of thigh) were collected when walking and the change according to the height of heels were analyzed using one-way ANOVA. Multiple comparison analysis on anterior tibial muscle and heel height showed that the group with 12cm heel showed significantly high muscle activation compared to the groups with 1cm and 7cm heels. The result of this study can be used for various perspectives from inferring and mediating problems caused by wearing high heels on different ground slopes for a long time

        Cloning or somatic cell nuclear transfer (SCNT) using adult somatic cell to derive cloned embryos is a promising new technology with potential applications in both agriculture and regenerative medicine. Mammalian embryos derived by nuclear transfer are capable of development to the blastocyst stage with a relatively high efficiency of 30~ 50%. However, in full-time development, usually only 2% of NT embryos can result in live births due to abnormalities in placenta formation. In SCNT embryos, the donor cell nucleus is epigenetically reprogrammed by oocyte cytoplasm during development. Incomplete reprogramming of the donor cell genome is considered a major reason for low cloning efficiency. Aberrant epigenetic modifications include DNA methylation, histone modification and X-chromosome-inactivation. Due to a lack of basic knowledge regarding the embryos following nuclear transfer, the success rate of cloning is low. Therefore, elucidation of the molecular mechanism of SCNT embryo development will be of great value for further research. MicroRNAs (microRNA) are single-strand RNA molecules of about 19 23 nucleotides in length, which regulate gene expression by imperfect base pairing with target mRNA, subsequently guiding mRNA cleavage or translational repression. Since the first discovery and functional annotation in 1993 of the small RNA, lin-4 and let-7, which are involved in developmental timing and gene regulation during C. elegans larval development, microRNAs have received scientific attention. Now hundreds of microRNAs have been identified in various multicellular organisms, and many microRNAs have been shown to be evolutionarily conserved. The roles proposed for this novel class of tiny RNA molecules are diverse. They are likely to be involved in developmental timing, differentiation, cell proliferation, signaling pathways, apoptosis, metabolism, heterochromatin formation, genome rearrangement, brain development and carcinogenesis. Currently (2006- present) we are working to determine the role of microRNAs on the epigenetic regulation of fertilized and cloned embryo development. The general hypothesis of our research is that genetic and epigenetic factors regulate the development of preimplantation mammalian embryos, and aberrant modulations in cloned embryos are causes of abnormal development and low success rate of cloned embryos.

        This study was performed to evaluate the contact toxicity and residual toxicity of the 26 commercially registered insecticides against cigarette beetle, Lasioderma serricorne and maize weevil, Sitophilus zeamais adult. Among 26 insecticides, seven insecticides (chlorpyrifos, chlorpyrifos-methyl, fenitrothion, fenthion, phenthoate, cypermethrin, α-cyhalothrin) against L. serricorne adult and five insecticides (chlorpyrifos, chlorpyrifos-methyl, fenitrothion, fenthion, phenthoate) against S. zeamais adult showed perfect mortality. LC50 (ppm) values of those selected insecticides were appeared lowest value on α-cyhalothrin (1.46 ppm) against L. serricorne adult and chlorpyrifos-methyl (0.64 ppm) against S. zeamais adult, respectively. However, chlorpyrifos-methyl still remain high toxicity until 90th days after treatment against both L. serricorne and S. zeamais adults. From above results, it will be useful information to select insecticides effective against L. serricorne and S. zeamais adults.

        The effects of electron beam irradiation on development and reproduction of Spodoptera litura were examined. Eggs, larvae, pupae, and adults were irradiated at target doses of 30, 50, 100, 150, 200, and 250 Gy. When eggs were irradiated with 100 Gy, egg hatching was perfectly inhibited. When irradiated to the larvae, pupation was inhibited at 100 Gy and larval period was delayed. When irradiated to the pupae, emergence was inhibited at 100 Gy and above. When irradiated to the adults, longevity and fecundity did not show any differences. However, egg hatching was strongly decreased at 100 Gy and above. Also, electron beam irradiation was not induced the instantaneous death of S. litura. The levels of DNA damage in S. litura adults were evaluated using the alkaline comet assay. Our results indicate that electron beam irradiation increased levels of DNA damage. These results suggest that electron beam irradiation induced abnormal development and reproduction by DNA damage in S. litura.

        The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is one of the most important pest species devastating many horticultural, ornamental crops and fruit trees. Difficulty in managing this mite is largely attributed to its ability to develop resistance to many acaricides. Development of 3,700 folds resistance to etoxazole was found in the population of T. urticae collected from rose greenhouses in Buyeo, Chungnam Province in August 2000. This population has been selected for eleven years with etoxazole (over 500 times), and increased over 5,000,000 fold in resistance as compared with susceptible strain (S). Etoxazole-resistant strain was shown to be maternally inherited. The objective of this study was to determine whether resistance of T. urticae to etoxazole was linked with point mutations in the mitochondrial gene. DNA sequencing of cytochrome c oxidase subunit I (COX1), COX2, COX3, cytochrome b (CYTB), NADH dehydrogenase subunit 1 (ND1), ND2, ND3, ND4, ND5, and ND6 were analyzed by comparing two isogenic etoxazole-susceptible (EtoS) and etoxazole-resistant (EtoR) strains. As a result, all genes revealed no point mutations between the two strains.

        An ambrosia beetle, Platypus koryoensis, is a vector of Raffaelea quercimongolicae that is known to cause Korean Oak Wilt (KOW), one of the serious threats to forest healthy in Korea. To manage P. koryoensis properly, it is necessary to clarify flight period of the adult. This experiment was conducted to elucidate the relationship between temperature and the flight period based on field observation in three forests consisted of Quercus mongolica from 2007 to 2009 except winter season. Date of flight period for 50% (FP50) was estimated by the cumulative Weibull distribution model based on cumulative proportion of the adult density and air temperature. Relationship between site temperature and the date of FP50 of P. koryoensis was the most significant when temperatures below 6.5℃ were excluded, suggesting lower threshold temperature for the flight period based on the site temperature. The pooling cumulative proportion of flight period against degree days was well described by the degree-day model, which has explanatory power for the 89% of year and site variation in the flight period and predicted accurately the flight pattern in 2011.

        From April 2010 to the present, seasonal occurrence of Riptortus pedestris was investigated. Five locations for population density and migration route tracking were the same as last year (two sites of Mt. Yangseong, Jujung-dong, O-Chang, and the campus of Chungbuk National University(CBNU)). In Mt. Yangseong, separated the farmland (80m) and forest (200 and 300m) by altitude, population density in 2010 was showed high density at mid-June, mid-Aug., and late-Oct., but population density in 2011 was showed high density at late-April, late-July and mid-Aug. In O-Chang and Jujung-dong, population density in 2010 was captured the adult from June to Aug. in the farmland, and from Sept. to Nov. in the forest. Total captured number of R. pedestris were high in farmland than forest. In campus of CBNU, the adult was captured from mid-Jul. to late-Aug. It showed the difference in the population density by its environmental conditions. Attraction effect of pheromone lure trap with soybeans were good than that of pheromone lure trap only. To track the migration of R. pedestris, we painted the fluorescent paint with different color. Being painted 500 R. pedestris were released and recaptured 10 and 20 days after release. Among released 500 R. pedestris at asl 300m on Yangseong site A and B in 25 Apr. were recaptured one at 300 and 80 m at site A, and site B was recaptured one marked R. pedestris adult at 200 m. In 25 May, release at 200 m in site A was recaptured only 80 m with 2 and site B was recaptured 200m with 8 and 80m with 1. Released in 17 Jul. at 80 m, marked R. pedestris adults were recaptured only 80 m with 5 and 2, respectively. It means that R. pedestris adults seem to migrate from the forest to the farmland in this season, but the data from Aug. to Oct. in 2010 was different.

        The black-veined white, Aporia crataegi (Lepidoptera: Papilionoidea), is nearly extinct in South Korea, although substantial numbers of dried specimens are available. One of the common practices for such species is to launch re-introduction program after proper amount of genetic information are analyzed from donor and donee populations. In this study, we sequenced complete mitochondrial genome (mitogenome) of A. crataegi to design species-specific primers for subsequent population works and to further understand the mitogenome evolution in lepodiopteran Papilionoidea. The 15,140-bp long A. crataegi mitogenome that has typical sets of 37 genes is smallest among true butterfly species with overall slightly smaller size in genes and regions throughout the genome. Arrangement of the genome is identical to those of other lepidopteran mitogenomes, in which tRNA cluster located between the A+T-rich region and ND2 gene is translocated into tRNAMet, tRNAIle, and tRNAGln from ancestral arrangement, tRNAIle, and tRNAGln, tRNAMet. The A/T content of the genome at 81.3% is the highest in Pieridae, but lower than that of lycaenid species (81.7% ~ 82.7%) The high A/T content in the genome is also reflected in codon usage, accounting for 41.69% of A/T-composed codons (TTA, ATT, TTT, and ATA). Unlikely the diversified or modified usage of anticodon for tRNASer(AGN) the species of Pieridae including A. crataegi all unanimously have GCT that has been hypothesized as ancestral for Lepidoptera. A total of 111 bp of non-coding sequences are dispersed in 13 regions, ranging in size from 1–49 bp. Among them relatively longer ones (≥ 16 bp) all have relatively higher sequence identity to other regions of the genome, suggesting partial duplication of the sequences during A. crataegi evolution. As has been reported in some species of Lepidoptera, the A. crataegi A+T-region also has typically found conserved sequences (e.g., poly-T stretch, ATAGA motif, ATTTA element, microsatellite-like A/T sequence, and poly-A stretch) and one tRNA-like sequence, and this feature was commonly found in true butterfly species.

        A bacterium was isolated from feces of Allomyrina dichotoma larva that feeds on well-rotted sawdust of woods. The isolate was identified as a Gram-positive and spore-forming stain. We named the isolate as Bacillus amyloliquefaciens on the basis of morphological and biochemical properties as well as 16S rRNA gene sequences. The culture of bacteria beneficially increased root and shoot growth of tomato, pepper and cucumber plants compared to the distilled water control. In addition, the bacterial culture strongly inhibited the mycelial growth of several fungal phytopathogens. The drop collapse assay with this culture showed a surfactant activity that is a major indicator for the selection of biocontrol agent. Also, a bacterium has ability of wastewater treatment. These data demonstrate the potential application of B. amyloliquefaciens as a biocontrol agent.

        Poly(vinyl chloride)-g-poly(oxyethylene methacrylate) (PVC-g-POEM) 가지형 공중합체를 원자전달라디칼 중합을 통해 합성하여 전기변색소자의 전해질에 적용하였다. 가소화된 고분자 전해질은 가소제로서 propylene carbonate (PC)/ethylene carbonate (EC) 혼합물을 도입하여 제조하였으며, Lithium tetrafluoroborate (LiBF4), lithium perchlorate (LiCIO4), lithium iodide (LiI) and lithium bistrifluoromethanesulfonimide (LiTFSI)를 사용하여 염의 종류에 따른 영향을 조사하였다. 광각 x-선 산란(WAXS)과 시차주사 열량법(DSC) 측정 결과 고분자 전해질의 구조와 유리전이온도(Tg)가 변하였고, 이는 POEM 내의 에테르의 산소와 리튬염 사이의 상호작용으로 인해 변했다는 것을 FT-IR 분광법을 통하여 확인하였다. 투과전자현미경(TEM) 측정 결과 PVC-g-POEM 가지형 공중합체의 미세상분리 구조가 PC/EC와 리튬염의 도입에도 변하지 않는 것을 관찰하였다. 가소화된 고분자 전해질은 poly(3-hexylthiophene) (P3HT) 전도성 고분자를 이용한 전기변색소자에 적용되었다.

        원자전달 라디칼 중합(ATRP)에 의해 poly(vinyl chloride) (PVC) 주사슬과 poly(styrene sulfonic acid) (PSSA) 곁사슬로 되어있는 양쪽성 PVC-g-PSSA 가지형 공중합체를 합성하였다. PVC-g-PSSA 가지형 공중합체 고분자를 템플레이트로 사용하고 졸겔법을 적용하여, 결정성 아타네제상의 미세기공 이산화티타튬 필름을 제조하였다. TiO2 전구체인 TTIP를 친수성인 PSSA 영역과 선택적으로 작용시켜 TiO2 메조기공 필름을 성장하였으며, 이를 주사전자 현미경 (SEM)과 엑스레이회절 (XRD)분석을 통해 분석하였다. 스핀코팅 횟수와 P25 도입에 따른 염료감응 태양전지 성능을 체계적으로 분석하였다. 그 결과 준고체 고분자 전해질을 이용하였을 때, 100 mW/㎠ 조건에서 에너지 변환 효율이 2.7%에 이르렀다.

        Anthocyanins are naturally occuring phytochemicals and the main components of the coloring of plants, flowers and fruits. They are known to elicit antioxidative, anti-inflammatory and cancer preventive activity. In this study, we investigated anthocyanins in black / yellow soybean seedcoats using different methods of detection - thin layer chromatography (TLC), capillary zone electrophoresis (CZE) and HPLC analysis. The anthocyanins in soybean seedcoats were extracted by five independent methods of extraction and the aglycons (anthocyanidins) of the corresponding anthocyanins were prepared by acid mediated hydrolysis. The anthocyanin / anthocyanidin in black soybean seedcoat showed characteristic TLC mobility, CZE electrophoretic retention and HPLC migration time while little of anthocyanins were detected from yellow soybean seedcoat. The extracted anthocyanins showed pH dependent retention time in CZE and spectral change in UV-Vis spectrum. HPLC analysis of the hydrolyzed extract of black soybean seedcoat identified the presence of four anthocyanidins. The major anthocyanin in black soybean seedcoat was cyanin (cyanidin-3-O-glucoside), with the relative order of anthocyanidin in cyanidin > delphinidin > petunidin > pelargonidin.
