To produce an intestinal immunomodulatory beverage containing Centella asiatica extract (CAE), three types of CAE-added beverage prototypes were prepared, and their immunomodulatory activities and marker compounds were analyzed. As a result of the cytotoxicity assessment, all the beverages did not show significant toxicity compared to the control group. Next, the immunomodulatory activities of the beverage prototype were evaluated using the inflammatory model of IL-1β-induced intestinal epithelial cell line. All the samples significantly reduced the production of IL-6, IL-8, and MCP-1 in a CAE concentration-dependent manner. In addition, CAE-added beverages inhibited NO, IL-6, and IL-12 production in LPS-induced RAW 264.7 cells. When the major triterpenoids, as marker compounds for the production of CAE-added beverages, were analyzed by HPLC-DAD, only asiaticoside was detected beyond the limit of quantification, while madecassoside, madecassic acid, and asiatic acid were not detected. The amounts of asiaticoside in CAE-added beverage prototypes were confirmed in No. 1 (19.39 μg/mL), 2 (19.25 μg/mL), and 3 (19.98 μg/mL). In conclusion, the results of this study suggested that CAE-added beverage prototypes induced immunomodulatory effects in the intestinal inflammatory cell line models and asiaticoside could be used as a marker compound for CAE-added beverage production.
Centella asiatica (C. asiatica) has been widely used in food, cosmetics, and pharmaceutical industry as a functional material. In a previous study, we have investigated not only pharmacological effects such as antioxidative and anti-inflammatory effects, but also analyzed various functional ingredients. In this study, triterpenoids were analyzed using HPLC-DAD to determine marker compounds among functional ingredients. When triterpenoids were analyzed, asiaticoside from C. asiatica was determined as an optimal marker compound. Next, specificity, linearity, limited of detection (LOD), limited of quantification (LOQ), precision, accuracy, and range were evaluated using HPLC-DAD to determine asiaticoside contents in C. asiatica juice and extracts. The specificity was elucidated by chromatogram and retention time using an established analytical method. The coefficient of correlation obtained was 0.9996. LOD was 4.99 μg/mL and LOQ was 15.12 μg/mL. Intra- and inter-day precision of asiaticoside were determined to be 0.48~1.68% and 0.08~1.09%, respectively. Furthermore, the recovery rate of asiaticoside was 98.88% and the analytical range of Field-70E was determined to be 0.625~10 mg/mL. As a results of evaluating ABTS, DPPH, and FRAP antioxidative effect, Field-70E showed potent antioxidant activities. Results of this study could be used as basic data for quality standardization of C. astiatica juice and extracts.
Recently, as the internet usage is increasing, accordingly generated text data is also increasing. Because this text data on the internet includes users’ comments, the text data on the Internet can help you get users’ opinion more efficiently and effectively. The topic of text mining has been actively studied recently, but it primarily focuses on either the content analysis or various improving techniques mostly for the performance of target mining algorithms. The objective of this study is to propose a novel method of analyzing the user’s requirements by utilizing the text-mining technique. To complement the existing survey techniques, this study seeks to present priorities together with efficient extraction of customer requirements from the text data. This study seeks to identify users’ requirements, derive the priorities of requirements, and identify the detailed causes of high-priority requirements. The implications of this study are as follows. First, this study tried to overcome the limitations of traditional investigations such as surveys and VOCs through text mining of online text data. Second, decision makers can derive users’ requirements and prioritize without having to analyze numerous text data manually. Third, user priorities can be derived on a quantitative basis.
The objective of this study was to investigate the non-thermal sterilization effect of methods such as high hydrostatic pressure (HHP) and UV-irradiation (specifically with regard to the storage stability and shelf-life of carrot juice. The microbial counts of the non-sterilized product increased from 5.51 to 7.34 log CFU/mL up to the fifth day, and then decreased to 5.46 log CFU/mL at six days. UV-irradiation was increased from 2.37 to 4.92 log CFU/mL. HHP was maintained under the 3 log CFU/mL. The pH of UV-irradiation and HHP was maintained withing the range of 6.29~6.30 and 6.20~6.22 during storage. However, the pH arising from non-sterilization decreased from 6.31 to 4.49. The color of non-sterilization changed significantly during storage, but UV-irradiation and HHP were similar during storage. The β- carotene content of non-sterilization was noted to have decreased from 269.45 to 65.19 μg/mL during storage. The UV- irradiation and HHP decreased from 263.46 and 268.35~281.16 μg/mL to 243.42 and 244.09~269.29 μg/mL, respectively. In conclusion, these findings suggest that HHP can be used for the pasteurization, or sterilization, of carrot juice and the optimal condition is two minutes.
치매는 보통 경미한 건망증으로 시작되지만 시간이 경과하면 인간을 황폐화시키는 진행성 질환으로 현재까지 원인은 70여 가지 이상으로 알려져 있다. 경증치매는 사회생활이나 직업상의 능력이 비록 상실되더라도 아직 독립적인생활은 영위할 수 있고, 적절한 개인위생을 유 지하며, 비교적 온전한 판단력을 보유하고 있는 상태이다. 이는 보호와 지원으로 독립된 생활의 가능과 진행을 늦출 수도 있기 때문에 다양 한 서비스와 치료가 필요한 시점이다. 이에 본 연구는 경증치매 노인들에게 집단 원예치료 프로그램을 적용하여 심리정서, 사회적 적응 효 과를 알아보는데 그 목적이 있다. 연구대상은 B시의 N사회종합복지관과 H요양병원을 이용하는 경증치매노인들 26명을 대상으로 두 집 단 유사 실험설계를 적용하여 2016년 9월부터 12월까지 주1회 60분씩 총 12회기에 걸쳐 실시하였다. 연구도구는 임은애(2010)에 의해 개 발된 원예치료 평가지표(HTEI: Horticultural therapy evaluation indices)의 4가지 척도 중 심리‧정서적 척도(PES: Psycho-emotional), 사회 적 척도(SS: Social scale)를 사용하였다. 자료분석은 SPSS for windows 18.0을 사용하여 통계 처리하였으며, 평균과 표준편차를 산출하였 고, 각 측정 시기별 변화를 알아보기 위해 반복측정 분산분석을 실시하였다. 연구 결과 심리·정서적, 사회적 기능의 모든 면에서 각 집단별 로 통계적 유의한 차이를 나타내었다.
