        In order to examine the effects of four different light spectra (white, red, green, and blue) on the oocyte maturation, the change of reproductive parameters, via brain-pituitary-gonad (BPG) axis in grass puffer, were investigated. After exposure four different light spectra for 7 weeks, the abundance of gonadotropin-releasing hormone (GnRH) mRNA which is a type of seabream (sbGnRH) and two different subunit of gonadotropin hormones mRNAs, follicle-stimulating hormone (fshβ) mRNA and luteinizing hormone (lhβ) mRNA, were analyzed in the brain and pituitary. Histological analysis showed that the mature oocyte ratio in the green spectrum was higher than other light spectra-exposed groups. Gonadosomatic index (GSI) and oocyte developmental stage were also investigated in the gonad based on histological observations. GSI value with the presence of yolk stage oocytes was significantly increased in the green spectrum-exposed group when compared to that of the other light-exposed groups (white, red, and blue) (p˂0.05). The abundances of sbGnRH mRNA and fshβ mRNA in the green spectrum-exposed group were also significant higher than those of the other light spectra-exposed groups (p˂0.05). These results indicate that the maturation of oocyte in grass puffer can be accelerated by exposure to the spectrum of green. To better understand the molecular mechanism for the maturation of oocyte in grass puffer, further study examining the relationship between oocyte development and its related genes is required.

The sbGnRHmRNA and fshβ mRNA may play an important reproductive parameters role in the initiation of maturation of oocyte. To better understand the molecular mechanism for the maturation of oocytein grass puffer, further study examining the relationship between oocyte development and its related genes (e.g., sbGnRHmRNA and fshβmRNA) is required.

        This study carried out to examine the association between the haplotypes of mitochondrial control region (CR) and growth traits of two F1 progeny populations of the Red-spotted grouper. We found polymorphic patterns of the 133-bp repeat units in the CR. In the BBR01 population (70-days after hatching), a total of 1,091 F1 progeny were divided into three haplotypes (H01, H03 and H04). The significant differences were found in the levels of BL, BW and LWI (p<0.05). The F1 animals with the haplotype H01 had greater level of BL (50.815±4.586 mm) than those of H03 (47.270±6.486 mm) and H04 (47.179±6.278 mm). The H01 F1 fishes were heavier level of BW (2.270±0.559 g) than those of H03 (1.789±0.711 g) and H04 (1.797±0.706 g). In the BBR02 population (11-months after fertilization), three haplotypes H03, H04 and H05 were detected. The significant difference was found only in the BL values among three haplotypes (p<0.05). The F1 animals with the haplotype H03 had greater level of BL (19.22±2.000 cm) than those of H04 (18.64±1.964 cm) and H05 (18.86±1.512 cm). There were no significant differences in BW and LWI among haplotypes in the BBR02 population (p>0.05). These results suggested that the mitochondrial haplotypes may affect the growth traits during early developmental stage of the Red-spotted grouper. The marker-assisted selection system for broodstock animals may be helpful in improving performance traits for aquaculture of the Red-spotted grouper.

        In fish, light (photoperiod, intensity and spectra) is main regulator in many physiological actions including body growth. The present study was to investigate the effect of light spectra on the body growth in red spotted grouper Epinephelus akaara. Experiment performed from December, 2016 to March, 2017 and fish (n=200, body weight 6.9±0.1, total length 7.5±0.1) was reared under different light spectra (blue, green, red and white) for 15 week under 12 hour light and 12 hour dark photoperiod and natural water temperature (13.6~16.7℃). We examined the mRNA expression levels of growth hormone (GH) in the brain with pituitary and photoreceptors mRNA expression in the retina. Red spotted grouper of BW was no different in each for the four groups (light spectra of blue, green, red and white) to five week of rearing period, but BW of rearing blue light spectra group fish was significantly increased in ten week of rearing period. After that, in 15week of rearing period, BW of four group fish was no showed difference respectively. The total length (TL) of fish was no differed in four experimental groups under the experimental duration respectively. The gh mRNA was more highly expressed in rearing blue light spectra group fish than other experimental groups in 15 weeks. The short wavelength sensitive opsin (sws) mRNA was more increased in rearing blue light spectra group fish than other groups in 15 weeks. The middle wavelength sensitive opsin (mws) and rod opsin mRNA expression was no differed in four experimental groups under all experimental durations respectively. The long wavelength sensitive opsin (lws) mRNA expression was more increased in rearing red light spectra group fish than other groups in 15 weeks. Our present results suggest that somatic growth of red spotted grouper is induced under blue light condition under low water temperature duration. Also, these results are thought to be affected of the gh and sws mRNA expression blue wavelength. However, in this experiment we did not confirm the correlation between GH and photoreceptor. Therefore, further studies are needed to clarify the correlation of the light spectra and photoreceptor with body growth in red spotted grouper.

        This study analyzed the mitochondrial DNA (mtDNA) sequences of the Red-spotted grouper, Epinephelus akaara (Perciformes, Serranidae), and used for construction of molecular phylogeny and for association between maternal haplotypes and phenotypic differences of F1 progeny. This study revealed phylogenetic position of the endangered red-spotted grouper, Epinephelus akaara (Perciformes, Serranidae) based on the nucleotide sequences of complete mt genome. Complete nucleotide sequences were determined from the mt genomes of two individuals of the red-spotted grouper caught in South Korea. The mitochondrial genome had 16,795 base pairs (bp) and 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a noncoding control region. The two mt genomes were highly homologous (99.71% similarity). The two mt genomes of E. akaara determined in this study were found in Clade I in the phylogenetic tree with those of E. awoara, E. fasciatomaculosus, E. sexfasciatus, E. diacanthus, E. sticus, and E. morio, suggesting that this may be helpful to understand phylogenetic position of Epinephelus species including red-spotted grouper. The genetic structure and phylogenetic relationship were investigated in the red-spotted grouper populations using the sequence polymorphisms of the cytochrome c oxidase subunit I(COI) gene and variable number of tandem repeats (VNTRs) of the control region (CR). A total of forty-one COIhaplotypes were found from 174 COIsequences from East Asia. The Jeju Island population (n=5) had four haplotypes, and the South Sea population (n=105) had twenty-five haplotypes. The Hong Kong population had nineteen haplotypes from fifty-nine COIsequences determined in this study. Among the COIhaplotypes, EAC_03 is commonly found in all populations (Jeju Island and South Sea of Korea, China, Hong Kong and Taiwan). In addition, there were four haplotypes (EAC_12, EAC_14, EAC_28 and EAC_35) also common among the populations tested in this study and collected from NCBI database. However, twenty haplotypes were specific in the Korean populations, and fifteen haplotypes were specific in the China and Hong Kong populations. The neighbor-joining (NJ) trees constructed from the phylogenetic analyses based on the polymorphisms of the COIhaplotypes showed the monophyletic branching pattern within the genus Epinephelus, indicating that the red spotted grouper populations had evolved from common maternal ancestors. Consequently, East Asian red-spotted grouper populations are maternally related at least in part, as well as sharing the same evolutionary history, and still affected by the East Asian ocean current (Kuroshio). From the haplotype analysis for mtDNA CR, we obtained VNTR polymor-phisms in all populations tested. We found five haplotypes for the CR VNTR patterns. The 133-bp repeat units were counted two to five. Using CR VNTR haplotypes, the statistical association was examined between mtDNA haplotypes and growth traits of aquafarming young fishes of the red-spotted grouper. A total of 386 F1 progeny, which were randomly selected from a progeny population produced by artificial insemination in the farm, were genotyped and statistically compared their body length (BL), body weights (BW) and length-weight indexes (LWI) at 11-months after hatching. There haplotypes H03, H04 and H05 were detected for CR in the parents and progeny populations. The significant difference was found in the BL values among three haplotypes (p<0.05). The F1 animals with haplotype H03 had freater level of BL (19.22±2.000 cm) than those of H04 (18.64±1.964 cm) and H05 (18.86±1.512 cm). There were no significant differences in BW and LWI among haplotypes (p<0.05). These results concluded that the maternal lineages affected the growth rates during early developmental stage in the red-spotted grouper. These findings suggested that the mitochondrial background of the fertilized eggs may play an important role in the early development, and the markerassisted selection system for broodstork animals may be helpful in improving performance traits for aquaculture industry as well as for conservation biology of the endangered red-spotted grouper. However, the results from the association analysis between haplotypes and phenotypes of F1 progeny (n=1,093) at 60-days after hatching showed that there were no significant difference (p>0.05). Consequently, the results of this study may be useful information for understanding the evolutionary relation with other species and may be good genetic markers for breeding management in the red-spotted grouper aquaculture system.

        This study investigated the effect of different photoperiods (14L: 10D, 12L:12D and 10L:14D) on the gonadal development and GtH mRNA expression in the pituitary of damselfish. The results showed that gonadosomatic index (GSI) was significantly lower in shot photoperiod (10L:14D), in comparison with other photoperiodic group during the spawning season. After 60 days treatment, histological analysis of gonad tissue showed that the gonad of 10L:14D and 12L: 12D treatment groups were resting phase with spermatogonia and perinucleolus stage oocytes but the gonad of 14L:10D treatment group was still ripe phase with spermatozoa and mature stage oocyte. The FSHβ and LHβ mRNA expression in pituitary drastically decreased shot photoperiod treatment from July (spawning period). These results suggest that photoperiod is considered to be the most effective environmental factor in controlling the reproductive cycle of damselfish.

        In this study, we investigated the characteristics of CCK-producing cells and mucus-secreting goblet cells with respect to stomach fish and stomachless fish of the Gobiidae in order to provide a basis for understanding the digestive physiology. Hairychin goby (Sagamia geneionema), which is stomachless fish, the numbers of mucus-secreting goblet cells is highest in the posterior intestine portion (P<0.05), while CCK-producing cells are scattered throughout the intestine. Gluttonous goby (Chasmichthys gulosus), which is stomach fish, mucus-secreting goblet cells are most abundant in the mid intestine portion (P<0.05), whereas CCK-producing cells are observed only in the anterior and mid intestine portion. Trident goby (Tridentiger obscurus) which is stomach fish, mucus-secreting goblet cells were most abundant in the mid intestine portion (P<0.05). CCK-producing cells are found in the anterior and mid intestine portion. Giurine goby, Rhinogobius giurinus which is also stomach fish, the largest number of mucus-secreting goblet cells showed in anterior intestine portion except for esophagus (P<0.05). CCK-producing cells are present only in the anterior and mid intestine portion. In S. geneionema, digestive action occurs in the posterior intestine portion to protect and functions to activate digestion. In contrast, in C. gulosus, T. obscurus and R. giurinus, their digestive action occurs in the anterior and mid intestine portion to protect and functions to activate digestion. Further studies of the modes of food ingestion by these fish, the contents of their digestive tracts, and the staining characteristics of the goblet cells need to be carried out.

        In fish, light (photoperiod, intensity and spectra) is main regulator in many physiological actions including growth. We investigate the effect of light spectra on the somatic growth and growth-related gene expression in the rearing tiger puffer. Fish was reared under different light spectra (blue, green and red) for 8 weeks. Fish body weight and total length were promoted when reared under green light condition than red light condition. Expression of somatostatins (ss1 and ss2) in brain were showed higher expression under red light condition than green light condition. The ss3 mRNA was observed only higher expression in blue light condition. Expression of growth hormone (gh) in pituitary was detected no different levels between experimental groups. However, the fish of green light condition group was showed more high weight gain and feed efficiency than other light condition groups. Our present results suggest that somatic growth of tiger puffer is induced under green light condition because of inhibiting ss mRNA expression in brain by effect of green wavelength.

        This study investigated structure and function of the reproductive system in Aplysia kurodai by means of anatomical, histological, and histochemical observation. Reproductive system of this species is consisted of ovotestis, small hermaphroditic duct, ampulla, accessory genital mass and large hermaphroditic duct. The ovotestis is composed of a large number of follicles, and both oocytes and spermatocytes matured in the same follicle. The small hermaphroditic duct is a single tube and contains a swelling, the ampulla, which functions as a storage organ for endogenous sperm and an oviduct. The accessory genital mass is connected to both the small and large hermaphroditic duct, and consisted of three glands: albumen, membrane (winding) and mucus gland. The albumen gland is consisted of granular cells producing basophilic and neutral mucopolysaccharides. The membrane and mucus gland are consisted of granular cells producing acidophilc and sulfated mucopolysaccharides. The large hermaphroditic duct is a single tubular gonoduct linking the accessory genital mass to the common genital aperture but is consisted of two parallel compartments. Internally, these two compartments are incompletely divided by internal septum or fold, which are called as the red hemiduct and white hemiduct, respectively. The red hemiduct functions as an oviduct and the white hemiduct functions as a copulatory duct. The reproductive system of A. kurodai is externally comprised a single tube, i.e., monaulic type. However, internal structure of duct is incompletely divided into oviduct and copulatory duct, i.e., the oodiaulic type.

        Light characteristics are very specific in the aquatic environment. Fish vision and different light spectra perception are related to each species’ natural habit. Light is one of the main environmental conditions and can be easily manipulated in artificial rearing settings. Mucus-secreting goblet cells are the main regulators of digestion. In this study, we established whether the light spectrum (natural condition, full spectrum: green, 520 nm; red, 590 nm, and blue, 480 nm) influences growth performance and digestive activity related to mucus-secreting goblet cell activity in order to develop a good management protocol and optimal rearing system for nursery stage of Epinephelus akaara. For each light spectrum, fish (11.5 ± 0.2 g in mean initial body weight, 9.0 ± 0.1 cm mean initial total length) were reared 16 weeks under a flow-through system and fed commercial pellet diets twice daily. At the end of the experiment, the final body weights differed among the fish reared under different light spectra. The highest growth performance value and feed efficiency were observed in fish reared under the green light condition. Mucus-secreting goblet cell activity was significantly higher in the fish under green light condition than in the fish under the natural, red, and blue light conditions. Rearing of E. akaara under the green light condition had positive effects on fish growth performance and digestion. We recommend that the appropriate light spectrum for nursery stage of E. akaara is the green light condition from the perspective of growth performance and the synergistic effects of mucus-secreting goblet cells. However, longer light treatment periods are needed in future investigations to clarify the effects of light spectrum on each growing stage of E. akaara.

        As a preliminary investigation into the effect of environmental factors control for gonadal development, we examined the involvement of photoperiod and water temperature in ovarian development of Epinephelus. akaara. For the induction of sexual maturation, E. akaara reared in recirculating aquaculture system (RAS). During November 2013, the photoperiod and water temperature was adjusted to 12L:12D and 18℃, respectively. In the photo-thermal treatment group, every 3 weeks daylight was increased as follows a 13L:11D and 14L:10D, and control group was maintained under natural condition. After 9 weeks, water temperature was increased 23℃ in photo-thermal treatment group. The sampled fish every 3 weeks revealed increase in gonadosomatic index (GSI; 5.18±1.38), oocyte diameter and vitellogenic oocytes (423.9±36.1 ㎛) were observed in gonads 12 weeks under photo-thermal treatment group. However, ovarian development was maintained immature stage in control group. In this environmental factors manipulation trial, seventy one of the 95 females (578.4 ± 25.4 g in mean body weight, 31.0 ± 0.5 cm mean total length) treated with HCG injection (doses 500 IU/kg BW) were induced ovulation by artificial stripping. The total volume of ovulated eggs were 3,470 ml and the total volume of fertilized eggs was 3,295 ml. The fertilization rate and hatching rate were 95% and 98%, respectively. These results suggest that the photoperiod as well as water temperature are major environmental factors in triggering the gonadal development of E. akaara.

        We investigated the change mRNA expression of GtHs subunits (FSHβ, LHβ) in the pituitary, androgen receptor (AR), estrogen receptor (ERα) in gonad and histological observation of gonads in longthooth grouper Epinephelus bruneus by treatment Femara, an aromatase inhibitor (AI). Longtooth grouper (body weight 408±43.1 g; one year) cultured in Future Aquaculture Research Center, NFRDI were used in the experiments. The experiment was conducted for 12 weeks from 21 August 2013. Fish received intramuscular injection of AI at 5 mg/g BW dose in three times every 3 weeks. Fish were sampled pituitary and gonads at 3, 6, 12 weeks post-injection (n=50). The mRNA levels of FSH-β, LH-β in pituitary and AR, ERα mRNA in gonad were evaluated using qRT-PCR and qPCR. The histological change of gonads observed on light microscope. The gonads of control group contained most perinucleolus oocyte. At 3 to 6 weeks post-injection, the gonads of AI-treated group contained a few degenerated oocytes, spermatogonia and spermatocytes. At 12 weeks post-injection, gonads contained spermatids undergoing spermatogenesis. From 6 to 12 weeks post-injection, the expression level of GtHs subunits mRNA in pituitary was significantly higher than control group. The expression level of AR mRNA in gonad was higher than control group from 3 to 12 weeks post-injection. The expression level of ERα mRNA in gonad was lower than control group from 6 to 12 weeks post-injection. These results suggest that immature longtooth grouper with AI treatment induced masculinization via change of GtH subunits in pituitary, AR and ERα mRNA in gonad.

        This study was investigated spawning behavior, structure of egg masses and egg development in Aplysia kurodai inhabiting the coastal waters of Jeju Island, Korea. The mating and courtship behavior of A. kurodai occurred in the form of unilateral copulating with chain formation. In chain copulation, only the first animal acted as a female; the second and succeeding animals acted as males (sperm donors) to the animals in front and as females to the animals behind. The fertilized eggs were packaged in capsules that are embedded in jelly to form a cylindrical string called an egg masses. The number of capsule per cm of the egg masses was 55 to 60 capsules and each capsule within the egg masses held 15 to 25 eggs. After spawning, the egg masses were bright yellow or orange in color. This egg masses color not changed until embryos developed into trochophore stage. Thereafter, as embryo developed from trochophore stage to veliger stage the egg masses color became brownish. The fertilized eggs were spherical, with a diameter of approximately 80±1 μm at spawning. At 5 to 6 days after spawning, the embryo developed into trochophore stage and began to rotate within the egg capsule. In the trochophore stage, the precursor of the velum, called the prototroch or prevelum, developed. At 10 days after spawning, the prevelum is transformed into the velum, and the trochophore developed into veliger stage. Between 10 to 15 days after spawning, the veligers broke out of the egg capsule, and hatched as free-swimming larvae.

        The fertilized eggs of E. septemfasciatus are spherical and transparent with buoyancy at 790 to 890 μm (average 821.8±2.0 μm) in diameter with 170 to 230 μm oil globules (average 192.9±0.93 μm). Hatching began approximately 46 and 35 hours after fertilization at 22.0℃ and 25.0℃ water temperature, respectively. The average total length of newly hatched larvae was 1.75±0.03 mm. Most of the yolk and oil globules were absorbed within 3 to 4 days after hatching. The larvae reached 2.48 to 2.72 mm in total length, and their mouths and anuses opened at 3 to 4 days after hatching. In this time, the mouth diameters of the larvae were 0.209 to 0.238 mm. The larvae reached 3.24 to 4.15 mm in total length at 11 to 17 days after hatching, and began to metamorphose at the time the second dorsal and pelvic spines appeared and elongated. The abdominal cavity was densely lined with melanophores. The larvae reached 5.12 mm in total length at 24 days after hatching.

        This study examines the effects on fertilization rate (FR), hatching rate (HR), and normal individual rate after artificial fertilization using frozen thawed sperm according to the cryoprotectant (DMSO) concentration and the period of cryopreserved sperm of longtooth grouper, Epinephelus bruneus. Performing artificial fertilization using frozen-thawed sperm, after freezing the sperm at different DMSO concentration of 5.0%, 7.5%, 10.0% respectively, FR were (DMSO 5.0%: 99.5±0.8%, DMSO 7.5%: 99.5±0.7%, and DMSO 10.0%: 99.6±0.6%). The results are not significantly different from the control fresh sperm (100%). HR also (DMSO 5.0%: 96.2±2.3%, DMSO 7.5%: 95.3±3.6%, 10.0%: 96.6±1.8%) were not significantly different in each group. The normal individual rate after hatching using with control fresh sperm (98.4%±0.5) and DMSO concentration level of 5.0% (97.8±0.1%) were not significantly different. However, with 7.5% (97.2±0.6%) and 10.0% DMSO concentrations (95.9±0.2%) are lower than the normal individual rate after hatching observed in the control and 5.0% DMSO. Performing artificial fertilization using frozen-thawed sperm at different frozen period (2 days, 2 years, and 3 years), 10% DMSO FR and HR of 3 years (FR; 66.8±1.8%, HR: 82.0±12.9%) and 2 years (FR; 78.5±14.8%, HR: 79.3±0.6%) cryopreserved sperm were lower than control (FR; 100%, HR: 91.1±3.6%) and 2 days cryopreserved sperm (FR; 99.6±0.6%, HR: 96.6±1.8%). These results suggest suitable DMSO concentration ranges of cryopreservation sperm for E. bruneus is 5 to 10% and with 2 to 3 years cryopreservation period, cryopreservation sperm can be useful for seed production.

        To effects of sex maturation in olive flounder by regulating long photoperiod, gonadal development and GTH mRNA expression in the pituitary were investigated. Photoperiod was treated natural photoperiod and long photoperiod (15L:9D) conditions from September 2011 to March 2012. The results showed that natural photoperiodic group showed a higher gonadosomatic index (GSI) than long photoperiodic group during the spawning season (March 2012). The histological analysis of ovarian tissue showed that natural photoperiod group of ovaries contained vitellogenic oocytes, but long photoperiod group of ovaries mainly contained perinucleolus staged oocyte and oil-drop staged oocytes. The FSH mRNA of olive flounder, under natural photoperiod group, showed a significantly higher expression but no significant difference under long photoperiod group. The LHβ mRNA showed a significantly higher expression only under natural photoperiod group. These results may suggest that long photoperiodic information regulates secretion of pituitary FSH and LH and maintain early growing stage of gonadal development in this species.

        To understand the sex reversal characteristics in the longtooth grouper (Epinephelus bruneus), this study examined the sex reversal and artificial masculinization of wild caught E. bruneus reared in indoor rearing tank after a 17α-methyltestosterone injection. To domesticate a broodstock, 64 wild caught E. bruneus, between 47.0 to 110.0 cm in total length and from 1.5 to 21.4 kg in body weight, were reared in indoor rearing tank (4.0 to 5.0 m wide, and 2.5 to 3.0 m depth) for four years. Seven specimens showed sex reversal from female to male during indoor rearing condition, whose total length and body weights were from 63.0 to 99.0 cm and from 4.4 to 13.2 kg, respectively. After inducing artificial masculinization in 14 female E. bruneus with a 17α-methyltestosterone (2.0 mg/kg BW) implants for 3 years, spermiation occurred in 9 specimens (total length: 54.0 to 68.0 cm, body weight: 2.3 to 4.3 kg). Among the female to male sex reversals, two specimens returned back to being female, whose body weights were 2.8 kg (initially 2.6 kg) and 2.7 kg (initially 2.3 kg). Therefore, this study suggested that E. bruneus (> 3.0 kg) was more effective in masculinizing by 17α-methyltestosterone implants.
