        Enhanced green fluoresce protein gene (egfp) was expressed in Beauveria bassiana ERL836 based on the Agrobacterium tumefaciens-mediated transformation (AtMT) method in this study. The ERL836 transformants were generated with pCambia-egfp binary vector. Ten transformants were randomly selected and analyzed for the T-DNA insertion and gene expression. The results revealed that 60% of the fungal putative transformants were inserted by the T-DNA fragment. Of these transformants, 33.33% (2 transformants) expressed the egfp gene. The egfp transformants showed strong green fluorescence with different expression levels. The results of this study could provide a reference for foreign protein expression in B. bassiana by using the AtMT method.

        It has been known that endophyte (Neotyphodium coenophialum) is beneficial to tall fescue (Schedonorus phoenix Scop.) because the mutualistic endophyte is able to confers tolerance against abiotic and biotic stresses to tall fescue. However, this fungal endophyte produces toxic alkaloid resulting in negative effects on animal performance. Recently, Non-toxic endophyte have been developed and inserted into tall fescue to avoid detrimental effect on animal but remaining positive influence on tall fescue. In order to keep this beneficial impact, it is essential to have endophyte infected tall fescue through vertical transmission from maternal plants to seeds. Little research has been carried out on endophyte transmission. To get basic information related to endophyte transmission, experiment was conducted to examine the effect of plant genotype on endophyte transmission under water stresses. Overall endophyte concentration in seeds was higher than that in panicles and endophyte concentration in seeds and panicles relied on plant. This study revealed that drought is not a critical component to control the endophyte transmission from maternal plants to seeds. Plant genotype is an important factor controlling the endophyte transmission from plant to seed.

        To date, chemical managements of plant bacterial diseases are complicated by limitations on use of antibiotics in agriculture, antibiotic resistance development, and limited efficacy of alternative control agents. In this study, thus, we performed screening of eco-friendly farming material (notice no. 2-4-064) containing tannic acid as new antibacterial-activity against 7 plant bacterial pathogens: Pectobacterium carotovorum subsp. carotovorum (Pcc), Ralstonia solanacearum (Rs), Acidovorax avenae subsp. citrulli (Aac), Xanthomonas cirti pv. citri (Xcc), Erwinia pyrifoliae (Ep), Clavibacter michiganensis subsp. michiganensis (Cmm), and Streptomycesscabies (Sc), Initial screening of antibacterial effects of eco-friendly farming material was performed using the paper disk diffusion method and co-cultivation in broth culture. Tannic acid based eco-friendly farming material showed inhibitory effect against Pcc, Rs, Aac, Xcc, Cmm, and Ss, whereas it did not show inhibition zone against Ep. However, when it tested by co-cultivation in broth culture, it showed strong inhibition effect against all pathogens that declined growth curve compared to bacterial pathogen only. Interestingly, when we observed morphological changes on those pathogens by SEM, cell morphologies of 7 pathogens were slightly changed that seem to be malfunction in their cell envelope.

        Although several adverse effects have been increased in recent years, synthetic agro-chemicals have been widely used to control diseases on paprika. This research was conducted to isolate and to characterize the antagonistic microorganism to control major paprika diseases, gray mold rot, fruit and stem rot, phytophthora blight, sclerotium rot, and wilt disease. Analysis of the fatty acid and analysis of the 16S rDNA gene sequence revealed that YKJ2 isolated in this research belongs to a group of Burkholderia cepacia. Specially, 16S rDNA gene sequence of YKJ2 showed 99% of sequence similarity with B. cepacia. Observation through the optical microscope revealed that YKJ2 was effective on suppression of the spore germination and the hyphal growth of pathogens. YKJ2 treatment on pathogens induced marked morphological changes like hyphal swelling and degradation of cell wall. In the case of phytophthora blight, the zoosporangium formation was restrained. On the basis of the results of this study, we propose that an antagonistic microorganism, B. cepacia, found in this study naming as “B. cepacia strain YKJ2” and has great potential as one of biological control agents against major diseases of paprika.
