In 1929, an American businessman wanted to put Mickey's image on a child's sketch pad, and since then, many animation companies have extended their work to commercial areas, promoting the advancement of their own industries. Since 1955, Disney has instilled its animated scenes, characters, and magic into the amusement parks in the United States, Japan, France, China's Shanghai and Hong Kong to build large-scale paradises. These parks contain several cartoon merchandise stores, in addition to catering and hotel services. Revenue from Disneyland theme parks currently comprise of 70% of Disney group's profits. Disney relies upon the influence of its own products and the development strategy of creative diversified merchandise. Disney is among the forefront of world-class entertainment media enterprises, and has become one of the hallmarks of the globalization of American culture.

Background : This project was conducted to determine proper seed harvest time and seed storage methods of Adenophora triphylla var. japonica Hara. Methods and Results : When Adenophora triphylla var. japonica Hara. transplanted this year, seeding can only be done the following year. Therefore, the seeds harvested in the interval of ten (10) days were investigated for seed harvest amount, germination rate, seed storage temperature and storing on container, between the period late August and mid-October. It is when the seed fructification started and ended. The seeds harvested from the first ten (10) days of October to mid October were 87.9% in whole seeds. The proper seed harvesting time was mid October which has a germination rate of 75% in whole seed harvest time. The lowest germination rate of 27.3% was at the germination temperature of 15℃. As the temperature rises, the germination rate increases. In 29℃, the germination rate was at 79% which is the highest; and from 30℃, the germination rate decreased to 71%. When the storage temperature was at 0℃ after storage of 5 months, the germination rate was at 83%. However, after storage of 12 months, it decreased by 7% that was 76%. When in 4℃, after storage of 5 months the germination rate was at 85%. However, after storage of 12 months it decreased by 8% which was 77%. In room temperature after storage of 5 months, germination rate was at 77%, but after storage of 12 months it decreased by 22% which was 55%. When seeds were stored in vinyl container, it was observed that the germination rate was at 86% after 5 months of storage, which was 9% higher than seed stored in paper which was 77%. After 12 months storage, seed in vinyl container had a germination rate of 71%, which was 3% higher than seed in paper at 68%. Conclusion : It is concluded that the proper time of seed gathering of Adenophora triphylla var. japonica Hara. is during the first ten (10) days of October to mid-October. When the long time storage is needed, below zero temperature with vinyl container is more appropriate.

Seashore Paspalum (Paspalum vaginatum Swartz) is a warm season grass and indigenous to tropical and subtropical regions of coastal areas worldwide. The species is used as feed for cattle and horses and has been very successful for golf courses worldwide. One of the most outstanding characteristics of seashore paspalum is its tolerance to saline soils compared to other warm season turfgrasses. The development of new seashore paspalum cultivars with improved traits could be facilitated through the application of biotechnological strategies. The purpose of this study was to product for herbicide resistant seashore paspalum using Arobacterium-mediated transformation and this study is the first report on transformation and herbicideresistant transgenic plants in seashore paspalum. Embryogenic calli were induced from the seeded variety of pseashore paspalum. Embryogenic calli were transformed with Agrobacterium tumefaciens strain EHA105 carrying the binary vector pCAMBIA3301 with two genes encoding gusA and bar. Transformed calli and plants were selected on medium containing 3 mg/l PPT. PCR detected the presence of the gusA and bar gene, indicating both genes are integrated into the genome of seashore paspalum. A chlorophenol red assay was used to confirm that the bar gene was expressed. By application of herbicide BASTA, the herbicide resistance in the transgenic seashore paspalum plants was confirmed.

Transient expression profiles for several chimeric β-glucuronidase (GUS) gene constructs were determined in microspore-derived embryos of wheat following microprojectile bombardment. The constructs analyzed consisted of the uidA (GUS) reporter gene driven by six different promoters [Cauliflower Mosaic Virus 35S (CaMV35S), Nopaline synthase (NOS), Mannopine synthase (MAS), Chlorella Mosaic Virus Adenin methyltransferase (AMT), maize Ubiquitin 1 (UBI1), and enhanced 35S (E35S)]. The total numbers of GUS blue spot were determined manually under a dissecting microscope after histochemical staining for GUS. Results suggest that the E35S promoter is the most active and UBI1 promoter is the second active in embryos or embryogenic calli derived from wheat microspore. In addition, by flurometric assay on GUS, the E35S promoter was the best. Therefore, both UBI1 and E35S promoter are suitable for constitutive expression of the gene of interest in microspore-derived embryos of wheat. This information describing promoter functionality in wheat will be important when designing gene constructs for traits modification and when choosing appropriate cultivars for improvement through gene transfer experiments.
