화색변형 화훼 형질전환체 개발을 위한 유전자를 개발하고자 클레마티스로부터 청색소 발현 유전자를 동정하여 ClF3 ′5 ′H로 명명하였다. 아그로박테리움 공동배양 경유 형질전환기술을 이용 하여 페튜니아 Dream’s Red 품종에 ClF3 ′5 ′H 유전자를 도입하였 고, 선발배지에서 재분화한 식물체 51개체를 PCR분석하여 49개체로 ClF3 ′5 ′H 유전자의 도입을 확인하였다. 또한 22개체의 서던분석 을 통해 ClF3 ′5 ′H 유전자가 1~3개 copy가 도입되었음을 확인하였 고, 노던분석을 통해 형질전환체에서의 ClF3 ′5 ′H 유전자의 정상 적인 발현을 확인하였다. ClF3 ′5 ′H 유전자가 도입된 페튜니아 형질전환 식물체 7개체가 개화할 때 ClF3 ′5 ′H 유전자가 도입된 페튜니아아의 꽃잎 주요색이 적색에서 진적색으로, 화관 내부색이 적보라색에서 진적보라색 또는 자주색 계역로, 암술머리색이 연두색에서 자주색 계열로, 꽃잎 가장자리 모양이 물결모양으로 변형되는 것을 확인하였다. 또한 실시간 정량 유전자 분석을 통해 형질전환 7개체에서의 표현형적 변화가 ClF3 ′5 ′H 유전자의 발현에 의한 것임을 입증하였다.

나리에 있어 Cucumber mosaic virus(CMV) 바이러스 저항 성 육종을 위해 주요 모본 및 부본으로 이용한 우수 유전자원 에 대해 바이러스 저항성 검정을 실시하였다. 바이러스 접종 원은 CMV-Taean으로 농업과학원에서 분양 받아 담배에 증식 하여 접종하였다. 바이러스 접종 식물체는 조직배양을 이용 하여 ‘Connecticut King’, ‘Party Diamond’, ‘Prato’, ‘Casa Blanca’, ‘Mona’, ‘Dublin’, ‘Rodrigo’, ‘Brunello’, ‘Avocado’ 품 종과 ‘FA08-17’ 계통에 대해 바이러스 무병주를 생산하여 실 험에 이용하였다. 바이러스 접종 효율을 높이기 위하여 접종 횟수를 1회와 2회로 나누어 실시하였으며 인산염 버퍼의 pH 를 6.9, 7.2, 7.5로 달리하여 접종하였다. 2회 접종, 인산염의 pH 6.9에서 접종 성공률이 높았다. ‘Casa Blanca’, ‘Avocado’ 품종의 잎에서 약한 모자이크와 노란색의 mottle 증상이 관측 되었으며 ‘Brunello’, ‘Rodrigo’, ‘FA08-17’에서 Hypersensitive response like 반응이 관찰되었다. ‘Brunello’, ‘Rodrigo’ 품종 에서 RT-PCR 검정 결과 바이러스가 검출되지 않았다. Reverse transcription-polymerase chain reaction(RT-PCR) 결과 바이 러스가 검출되었던 ‘Connecticut King’, ‘Prato’, ‘Casa Blanca’ 의 경우 Enzyme-linked immunosorbent assay(ELISA) 흡광도 가 낮게 나타났다. RT-PCR과 ELISA 검사 결과 잎과 구근에 서 CMV 검출이 다르게 나타남에 따라 나리에 있을 것으로 추측되며 바이러스 감염과 연관된 TOM1, OBE2, DND1 유 전자를 대상으로 발현 분석을 하였으나 유의한 결과를 얻지 못하였다.

A new standard rose cultivar ‘Green Star’ was bred from the cross between pink color standard cultivar ‘Pastel move’ and red color standard cultivar ‘Vital’ at the National Institute of Horticulture & Herbal Science. The cross was made in 2008 and ‘Green Star’ was finally selected in 2013 after investigating characteristics for three years from 2011 to 2013. ‘Green Star’, a light yellow standard cultivar grows vigorously and has good and big shape. The major characteristics of this cultivar are 122.0 stems/㎡/year in yield, 77.0±8.0 cm in length of cut flower, 12.1±0.4 cm in flower diameter, 37.7±4.2 in petal number, and 11.0±1.4 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer’s preference of this cultivar is relatively higher than that of control cultivar, ‘Moonlight’.

A new light purple rose cultivar ‘Violetta’ was bred from the cross between light purple standard cultivar ‘Ocean Song’ and yellow standard cultivar ‘golden gate’ at the National Institute of Horticulture & Herbal Science. The cross was made in 2010 and ‘Violetta’ was finally selected in 2013 after investigating characteristics for three years from 2011 to 2013. ‘Violetta’, a light purple color standard cultivar grows vigorously and has good shape and good vase life. The major characteristics of this cultivar are 167.1 stems/㎡/year in yield, 77.9±6.0 cm in length of cut flower, 12.8±0.3 cm in flower diameter, 55.5±4.9 in petal number, and 13.1±1.6 days in vase life. This cultivar can be propagated by both cutting and grafting. The consumer’s preference of this cultivar is relatively higher than that of control cultivar, ‘Aqua’.
