Background: The ability of adeno-associated viruses (AAVs) to transduce various cell types with minimal immune responses renders them prominent vectors for gene editing (GE), with different AAV serotypes exhibiting distinct transduction efficiencies due to their specific cellular tropism. However, detailed molecular processes of AAV infection and penetration, as well as the optimal serotype for specific purposes, remain poorly understood. Porcine models are widely used in research benefitting both human and livestock due to anatomical and physiological similarities to humans. Methods: Transduction efficiencies of 18 AAV serotypes (AAV1–9, 6.2, rh10, DJ, DJ/8, PHP.eB, PHP.S, 2-retro, 2-QuadYF, and 2.7m8) were evaluated in immortalized porcine lung epithelial cells (pLCsImt) and pulmonary alveolar macrophages 3D4/31 (PAMs 3D4/31). Results: We found AAV2, DJ, and 2.7m8 to be the most effective in both cell types. The highest enhanced green fluorescent protein expression of 52.46 ± 2.4% in pLCsImt and 64.08 ± 2.4% in PAMs 3D4/31 was observed for AAV2, while negligible transduction was observed for AAV4, rh10, DJ, PHP.eB, PHP.S, and 2-retro. AAV-DJ showed superior transduction efficiency in PK-15, as compared to AAV2 and 2.7m8. Results emphasize the cell type-specific nature of AAV serotype transduction efficiencies. Notably, AAV2 was most effective in both lung and macrophage cells, whereas AAV-DJ was more effective in renal cells. Conclusions: Our findings suggest that AAV2 was identified as the most efficient serotype for transducing pLCsImt and PAMs 3D4/31, compare to the PK-15 cells. Understanding cell type-specific preferences of AAV serotypes offer crucial insight for tailoring AAV vectors to specific tissue and optimizing genome editing strategies, with potential implications for the advancement of personalized medicine and development of treatments for human and livestock.
Haemaphysalis longicornis는 사람과 동물에게 여러 심각한 병원체를 전달하는 주요 매개체로, 한반도에 널리 분포하고 있다. H. longicornis는 Rickettsia spp., Borrelia spp., Francisella spp., Coxiella spp., 그리고 중증열성혈소판 감소증후군 바이러스 (SFTS virus) 등을 매개하는 것으로 알려져 있다. 국내에 서식하는 H. longicornis의 미생물 군집과 관련된 연구는 많이 진행되지 않은 것으로 확인되었다. 이 연구는 한반도 내 다양한 지역에서 채집된 H. longicornis의 미생물군집 다양성을 지역별, 성장 단계 및 성별에 따라 분석하였다. 2019년 6월부터 7월까지 질병관리청 권역별기후변화매개체감시거점센터 16개 지역에서 채집한 H. longicornis의 16S rRNA 유전자 V3-V4 영역을 PCR로 증폭 후 Illumina MiSeq 플랫폼으로 시퀀싱하였다. Qiime2를 활용한 미생물 다양성 분석을 통해 총 46개의 샘플에서 1,754,418개의 non-chimeric reads를 얻었으며, 평균 126개 의 operating taxonmic unit (OTU) 을 식별하여 총 1,398개의 OTU를 확인하였다. 대부분의 지역에서 Coxiella spp.가 우점종으로 나타났으며, 특히 Coxiella endosymbiont는 가장 높은 우점도를 보이며, Coxiella burnetii와 계통 발생 학적으로 유사한 것으로 확인되었다. 이 연구를 통해 분석된 결과는 각 지역의 H. longicornis 미생물군집 데이터 베이스 구축에 활용되었으며, 이를 통해 지역별 미생물군집의 특이성을 식별할 수 있게 하였다. 이는 한반도의 H. longicornis에 의한 질병 전파 연구와 이를 통한 공중보건 개선에 기여할 것으로 기대된다.
한강 3개 보 구간 수변부 11개 지점을 대상으로 2023년 총 2회 채집된 저서성 대형무척추동물은 총 100종 평균 426.0 개체/m2였다. 여주보에서 64종으로 가장 많이 출현하였고, 강천보에서 59종, 이포보에서 54종, 한강보상류 에서 41종, 한강보하류에서 32종이 확인되었다. 전체 평균 개체밀도는 강천보에서 944.1 개체/m2로 가장 많았고, 한강보상류에서 282.7 개체/m2, 여주보에서 251.9 개체/m2, 이포보에서 180.6 개체/m2, 한강보하류에서 172.9 개체 /m2 순으로 확인되었다. 한강 보 구간에서는 플라나리아류, 실지렁이, 깔따구류 등이 우점함. 보구간의 특성상 상대적으로 단순한 하상 및 깊은 수심에 적응성이 높은 실지렁이, 깔따구류의 우점은 일반적인 특성으로 볼 수 있으며 일시적으로 수변부에서 밀집된 플라나리아류의 채집이 이뤄져 우점된 결과를 보인 것으로 보임. 한강 보 구간 수변부 및 중앙부 조사지점별 군집분석을 실시한 결과 한강보 하류는 3개 보에 비해 우점도가 높고 다양도 와 풍부도가 낮았으며 전반적으로 상류부에 위치한 한강보상류 등의 지점에서 군집안정성이 확보되고 있는 것으로 보이며 이러한 점은 상대적으로 다양한 미소서식환경에 기인한 것으로 판단된다.
The fall armyworm, Spodoptera frugiperda, has developed extremely high levels of resistance to chlorantraniliprole and other classes of insecticides in the field. As microRNAs (miRNAs) play important roles in various biological processes through gene regulation. we examined the miRNA profile of S. frugiperda in response to Chlorantraniliprole, Indoxacarb and Thiamethoxam. Transcriptome analysis showed significant changes in the abundance of some miRNAs after treatment of S. frugiperda larvae with LC20 concentrations of three insecticides. A total of 197 miRNAs were systematically identified from S. frugiperda, and 16, 9, 2 miRNAs were differentially expressed after treatments of three insecticides. Importantly, three miRNAs were significantly downregulated and three were upregulated by RT-qPCR after treatment the LC50 of three insecticides with S. frugiperda larvae. Microinjection of agomirs of these six miRNAs into S. frugiperda larvae resulted in significant changes in mortality rates when exposed to three insecticides. Additionally, we also screened potential target genes for some of differentially expressed miRNAs, which may play important roles in insecticide resistance development. These findings provide valuable insights into the molecular mechanisms of insecticide resistance and underscore the potential of miRNAs as targets for the development of novel pest control strategies in S. frugiperda.
Honey bees are crucial pollinators for agricultural and natural ecosystems, but are experiencing heavy mortality in Korea due to a complex suite of factors. Extreme winter losses of honey bee colonies are a major threat to beekeeping but the combinations of factors underlying colony loss remain debatable. Finding solutions involves knowing the factors associated with high loss rates. To investigate whether loss rates are related to Varroa control and climate condition, we surveyed beekeepers in korea after wintering (2021–2022 to 2022–2023). The results show an average colony loss rate of 46%(2022) and 17%(2023), but over 40% colony loss before wintering at 2022. Beekeepers attempt to manage their honey bee colonies in ways that optimize colony health. Disentangling the impact of management from other variables affecting colony health is complicated by the diversity of practices used and difficulties handling typically complex and incomplete observational datasets. We propose a method to 1) Varroa mite population Control by several methods , and 2) Many nursing bee put in hive before wintering.
This study was conducted to determine the appropriate seeding dates by verifying the difference in winter survival and productivity of alfalfa according to fall sowing dates in the central area of South Korea. The experiment was conducted for 2 years (2020 and 2021) at the field in the Department of Animal Resources Development, NIAS located in Cheonan. Sowing dates started from September 18 to November 8 with 10 days of intervals during 2020 and 2021; SO1 (September 18), SO2 (September 28), SO3 (October 8), SO4 (October 18), SO5 (October 28), and SO6 (November 8). After sowing, the winter survival rate was measured in the spring of the following year, and the dry matter yield was measured by harvesting at 10% flowering and harvesting five times a year. SO6 failed to winter survival, and SO5 also had a lower winter survival rate than SO1~4 (p<0.05). The average annual dry matter yield of alfalfa linearly decreased with delaying sowing dates (p<0.05). The feed value did not differ in the same year by delaying the sowing date in the same year. These results suggest that sowing date should be started before October 18 to increase winter survival and productivity of alfalfa in the central area of South Korea.
Technosignature, previously known as SETI(search for extraterrestrial intelligence), is the scientic evidence of past or present extraterrestrial civilizations. Since NRAO's Project Ozma was performed in 1960, most of the noticeable technosignature searches have been done by radio telescopes, hoping to find strong and narrow bandwidth signals that cannot be explained by known natural processes. Recently, the Breakthrough Listen project has opened a new opportunity for technosignature by utilizing both optical telescopes, radio telescopes, and next-generation radio telescope arrays. In this review, mainly based on NASA Technosignatures Workshop (2018), we review the current trends of technosignature surveys, as well as other possible methods for detecting technosignature. Also, we suggest what the Korean community could contribute the technosignature research, including the new SETI project with Korea VLBI Network (KVN).
Since the farside of the moon is a place to avoid arti cial radio frequency interference (RFI) created byhuman civilization, it is a most suitable place for searching technosignature, which are signs of technolog-ical civilization in the universe, in the radio band. The RFI is a factor that makes the study of searchingtechnosignature quite complicated because it is di cult to distinguish between technological signals pro-duced by human and extraterrestrial civilizations. In this paper, we review why the farside of the moon isthe best place to detect technosignature and also introduce radio observatories on the farside of the moonthat have been proposed in radio astronomy. The SETI (Search for Extraterrestrial Intelligence) projecton the farside of the moon is expected to be one of the main candidates for international collaborationresearch topics on lunar surface observatory.
The main goal of the Korean VLBI Network Calibrator Survey (KVNCS) is to expand the VLBI calibrators catalog for KVN, KaVA(KVN and VERA Array), EAVN(East-Asian VLBINetwork), and other extended regions. The secondKVNCS (KVNCS2) aimed to detect VLBI fringes of new candidates for calibrators in the K band. Out of the 1533 sources whose single-dish flux density in the K band was measured with KVN telescopes (Lee et al. 2017), 556 sources were observed with KVN in the K band. KVNCS2 confirmed the detection of VLBI fringes of 424 calibrator candidates over a single baseline. All detected sources had a high Signal-to-Noise Ratio (SNR) of >25. Finally, KVNCS2 confirmed 347 new candidates as VLBI calibrators in the K band, resulting in a 5% increase in the sky coverage compared to previous studies. The spatial distribution was quasi-uniform across the observable region (Dec. > −32.5◦). In addition, the possibility as calibrator candidates for the detected sources was checked, using an analysis of the flux-flux relationship. Ultimately, the KVNCS catalog will not only become the VLBI calibrator list but is also useful as a database of compact radio sources for astronomical studies.
