UV and O3 are materials used in the water treatment process, and many studies have been reported to remove organic matters, contaminants, and microorganisms. In this study, we were investigated effects of Chirnomidae (Chironomus flaviplumus, Chironomus riparius), which are contamination indicator species to exposure UV and O3 for the survival rate, body color change and gene expression response. The survival rate of C. flaviplumus exposed to UV decreased to about 70% after 24 hours, and C. riparius about 50%. There was no change in the survival rate of C. flaviplumus exposed to O3, and C. riparius decreased to 95% after 10 minutes of exposure, but there was no change during the subsequent exposure time. In addition, UV and O3 exposure to the two species in body color faded in a time-dependent. In the HSP70 gene expression, C. riparius showed an increase in expression after UV exposure compared to the control group, and a significant difference was shown 12 hours after exposure (P<0.05). C. flaviplumus exposed to O3 showed a relatively low expression compared to the control group, and showed a significant difference at 10 minutes and 1 hour after exposure (P<0.05). These results reported the ecotoxicological effects on Chironomidae according to UV and O3 exposure. Therefore, the results of this study can be used as basic data to understand the effects of UV and O3, which are disinfectants used in water treatment plants, on Chirnomidae entering plants. Cadmium (Cd), a heavy metal found in the aquatic environment, accumulates in organisms through the food chain. In the study, we investigated the survival rates, measurement of body Cd levels, and expression analysis of the stress response genes (Heat shock protein 70: HSP70 and Heat shock protein 60: HSP60) and antioxidant enzyme Glutathione S-Transferases (GST) on benthic oligochaete worm Tubifex tubifex exposed three concentrations of Cd, to analyze the bioaccumulation and changes of stress gene expressions to exposure toxicity of the Cd-spiked sediment. Survival rates of T. tubifex exposed to the Cdspiked sediment were 93% at 0.4 mg kg-1 Cd, 96% at 1.87 mg kg-1 Cd, and 93% at 6.09 mg kg-1 Cd for 10 days. Cd concentration in the body of T. tubifex was higher than that in the sediment. After Cd exposures for 10 days, the body Cd levels were 18.4 mg kg-1, 13.06 mg kg-1, and 79.11 mg kg-1 at exposed three concentrations of Cd, respectively. Upregulation of HSP70 gene expression was observed at all concentrations of exposed Cd as a time-dependent manner, whereas transcriptional expression of the HSP60 gene increased as a timedependent manner in T. tubifex exposed to the relative high concentration (6.09 mg kg-1) of Cd. However, GST gene expression increased on day 1 at all concentrations after Cd exposures, and then downregulated until 10 days. These results indicate to ecotoxicological and molecular effects in benthic oligochaete worm T. tubifex to Cd-spiked sediment and provide the basic information for the utilization of environmental toxicity assessment using the T. tubifex as a aquatic pollution indicator species.
EU taxonomy requires to solve problems for safe management of radioactive waste and disposal of spent fuel, which is a precondition for growing demand for nuclear power plant. Currently, Korea manages about 18,000 tons of high-level radioactive waste at temporary storage facilities in nuclear power plant sites, but such temporary storage facilities are expected to become saturated sequentially from 2031. Therefore, it is necessary to secure a permanent disposal facility to safely treat high-level radioactive waste. In accordance with the second basic plan for high-level radioactive waste management in 2021, it is necessary to establish requirements for regulatory compliance for the site selection and site acquisition, investigation and evaluation, and construction for the establishment of a deep geological disposal facility. In this study, we analyzed the regulatory policies and cases of leading foreign countries related to deep geological disposal facilities for high-level radioactive waste disposal waste such as IAEA, USA, Sweden, and Finland using data analysis methodology. To analyze a large amount of textbased document data, text mining is applied as a major technology and a verification standard that secures validity and safety based on the regulatory laws described so far is developed to establish a regulatory base suitable for domestic deep geological disposal status. Based on the collected data, preprocessing and analysis with Python were performed. Keywords and their frequency were extracted from the data through keyword analysis. Through the measured frequency values, the contents of the objects and elements to be regulated in the statutory items were grasped. And through the frequency values of words co-occurring among different sections through the analysis of related words, the association was obtained, and the overall interpretation of the data was performed. The results of analyzing regulations of major foreign countries using text mining are visualized in charts and graphs. Word cloud can intuitively grasp the contents by extracting the main keywords of the contents of the regulations. Through the network connection graph, the relationship between related words can be visually structured to interpret data and identify the causal relationship between words. Based on the result data, it is possible to compare and analyze the factors to be supplemented by analyzing domestic nuclear safety case and regulations.
Since high-level radioactive wastes contain long-lived nuclides and emit high energy, they should be disposed of permanently through a deep geological disposal system. In Korea, the first (2016.07) and the second (2021.12) basic plans for the management of high-level disposal systems were proposed to select sites for deep geological disposal facilities and to implement business strategies. Leading countries such as Finland, Sweden and France have developed and applied safety cases to verify the safety of deep geological disposal systems. By examining the regulatory status of foreign leading countries, we analyze the safety cases ranging from the site selection stage of the deep geological disposal system to the securing of the permanent disposal system to the investigation, analysis, evaluation, design, construction, operation, and closure. Based on this analysis, we will develop safety case elements for long-term safety of deep geological disposal systems suitable for domestic situation. To systemically analyze data based on safety cases, we have established a database of deep geological disposal system regulations in leading foreign countries. Artificial intelligence text mining and data visualization techniques are used to provide database in dashboard form rather than simple lists of data items, which is a limitation of existing methods. This allows regulatory developers to understand information more quickly and intuitively and provide a convenient interface so that anyone can easily access the analyzed data and create meaningful information. Furthermore, based on the accumulated bigdata, the artificial intelligence learns and analyzes the information in the database through deep learning, and aims to derive a more accurate safety case. Based on these technologies, this study analyzed the legal systems, regulatory standards, and cases of major international leading countries and international organizations such as the United States, Sweden, Finland, Canada, Switzerland, and the IAEA to establish a database management system. To establish a safety regulation base suitable for the domestic deep geological disposal environment, the database is provided as data to refer to and apply systematic information management on regulatory standards and regulatory cases of overseas leading countries, and it is expected that it will play a key role as a forum for understanding and discussing the level of safety of deep geological disposal system among stakeholders.
Gut contents analysis is essential to predict the impact of organisms on food source changes due to variations of the habitat environment. Previous studies of gut content analysis have been conducted using traditional methods, such as visual observation. However, these studies are limited in analyzing food sources because of the digestive process in gut organ. DNA metabarcoding analysis is a useful method to analyze food sources by supplementing these limitations. We sampled marine fish of Pennahia argentata, Larimichthys polyactis, Crangon affinis, Loligo beka and Sepia officinalis from Gwangyang Bay and Yeosu fisheries market for analyzing gut contents by applying DNA metabarcoding analysis. 18S rRNA v9 primer was used for analyzing food source by DNA metabarcoding. Network and two-way clustering analyses characterized the relationship between organisms and food sources. As a result of comparing metabarcoding of gut contents for P. argentata between sampled from Gwangyang Bay and the fisheries market, fish and Copepoda were analyzed as common food sources. In addition, Decapoda and Copepoda were analyzed as common food sources for L. polyactis and C. affinis, respectively. Copepoda was analyzed as the primary food source for L. beka and S. officinalis. These study results demonstrated that gut contents analysis using DNA metabarcoding reflects diverse and detailed information of biological food sources in the aquatic environment. In addition, it will be possible to provide biological information in the gut to identify key food sources by applying it to the research on the food web in the ecosystem.
Recently, with the development of genetic technology, interest in environmental DNA (eDNA) to study biodiversity according to molecular biological approaches is increasing. Environmental DNA has many advantages over traditional research methods for biological communities distributed in the environment but highly depends on the established base sequence database. This study conducted a comprehensive analysis of the habitat status and classification at the genus level, which is mainly used in eDNA (12S rRNA, 16S rRNA, 18S rRNA, COI, and CYTB), focusing on Korean registration taxon groups (phytoplankton, zooplankton, macroinvertebrates, and fish). As a result, phytoplankton and zooplankton showed the highest taxa proportion in 18S rRNA, and macroinvertebrates observed the highest ratio in the nucleotide sequence database in COI. In fish, all genes except 18S rRNA showed a high taxon ratio. Based on the Korean registration taxon group, the gene construction of the top 20 genera according to bio density observed that most of the phytoplankton were registered in 18S rRNA, and the most significant number of COI nucleotide sequences were established in macroinvertebrates. In addition, it was confirmed that there is a nucleotide sequence for the top 20 genera in 12S rRNA, 16S rRNA, and CYTB in fish. These results provided comprehensive information on the genes suitable for eDNA research for each taxon group.
The number of dismantled nuclear facilities is increasing globally. Dismantling of nuclear facilities generates large amount of waste such as concrete, soil, and metal. Concrete waste accounts for 70% of the total amount of waste. Since hundreds of thousansds of tons of concrete waste generated, securing technology of reduction and recycling of waste is emerging as a very important issue. The objective of this study is to synthesize geopolymer using inorganic materials from cement fine powder in concrete waste. Dismantled concrete waste contains a large amount of calcium silicate hydrate(C-S-H), Ca(OH)2, SiO2, etc., which is an inorganic material required for the synthesis of geopolymer. SiO2 affects the compressive strength of the geopolymer and Ca(OH)2 affects the curing rate. A high concentration of alkali solution is used as an alkali activator, and alkali activator is necessary for the polymerzation reaction of metakaolinite. The experiment consists of three steps. The first step is to react with concrete waste and hydrochloric acid to extract ions. In the solid after filtration, SiO2 and Al2O3 are composed of 84.10%. It can be used instead of commercial SiO2 required for the synthesis of geopolymer. The second step is to add NaOH up to pH 10, impurities can be removed to extract Ca(OH)2 with high purity. The final step is to add NaOH up to pH 13, and Ca(OH)2 extraction. The alkali solution generated after the last reaction can be recycled into an alkali activator during the synthesis of the geopolymer. If dismantled concrete waste is recycled during geopolymer synthesized, the volume reduction rate of dismantled concrete waste is more than 50%. If you put the radioactive waste in the recycled solidification materials synthesis from concrete waste by dismantling of nuclear facilities, it is possible to reduce the amount of waste generated and disposal costs.
Kori-1, the nuclear power plants in South Korea, first started operation in April 1978 and was suspended permanently in 2017. The saturation rate time of spent nuclear fuel generated by major nuclear power plants operating in Korea are getting closer. If we fail to dispose spent nuclear fuel, which is equivalent to high-level radioactive waste, the nuclear power plants will have to be shutdown. High-level radioactive waste is permanently disposed through a deep geological disposal system because it contains long-term half-life nuclides and emits high energy. To select the deep geological disposal site and construct the disposal facilities, it is necessary to establish appropriate regulatory policies accordingly. The status of database construction in OECD-NEA, NRC, SITEX, and IAEA, which provides safety regulations for deep geological disposal system, stipulates each requirement for dismantling nuclear power plants. However, details such as specific figures are not specified, and guidelines for the disposal of high-level radioactive wastes are not clearly distinguished. In Korea, the CYPRUS program, an integrated database system, has been developed to support comprehensive performance evaluation for high-level waste disposal. However, due to several difficult situations, maintenance and upgrades have not been performed, so the research results exist only in the form of raw data and the new research results have not been reflected. Other than that, there is no preemptive basis for regulating the deep geological disposal system. With real-time database, we can develop a regulatory system for the domestic deep disposal system by systematically analyzing the regulatory condition and regulatory case data of international organizations and foreign leading countries. The database system processed and stored primary data collected from nuclear safety reports and other related data. In addition, we used relational database and designed table to maximize time and space efficiency. It is provided in the form of a web service so that multiple users can easily find the data they want at the same time. Based on these technologies, this study established a database system by analyzing the legal systems, regulatory standards, and cases of major foreign leading countries such as Sweden, Finland, the United States, and Japan. This database aims to organize data for each safety case component and further prepare a safety regulatory framework for each stage of development of disposal facilities suitable for the domestic environment.
CYPRUS is a web-based waste disposal research comprehensive information management program developed by the Korea Atomic Energy Research Institute over three years from 2004. This program is stored as existing quality assurance documents and data, and the research results can be viewed at any time. In addition, it helps to perform all series of tasks related to the safety evaluation study of the repository in accordance with the quality assurance system. In the future, it is necessary to improve the user convenience by clarifying the relationship between FEP and scenarios and upgrading output functions such as visualization and automatic report generation. This purpose of this study is to research and develop the advanced program of CYPRUS. This study is based on building FEP, DIM and scenario databases. It is necessary to develop an algorithm to analyze and visualize the FEP, DIM and scenario relationship. This project is an integrated information processing platform for DB management and visualization considering user convenience. The first development goal is to build long-term evolutionary FEP, DIM, and scenarios as a database. The linkage by FEP item was designed in consideration of convenience by using a mixed delimiter of letters and numbers. This design provides information on detailed interactions and impacts between FEP items. Scenario data lists a series of events and characteristic change information for performance evaluation in chronological order. In addition, it includes information on FEP occurrence and mutual nutrition by period, and information on whether or not the repository performance is satisfied by item. The second development goal is to realize the relationship analysis and visualization function of FEP and scenario based on network analysis technique. Based on DIM, this function analyzes and visualizes interactions between FEPs in the same way as PID, RES, etc. In addition, this function analyzes FEP and DIM using network analysis technique and visualizes it as a diagram. The developed platform will be used to construct and visualize the FEP DB covering research results in various disposal research fields, to analyze and visualize the relationship between core FEP and scenarios, and finally to construct scenarios and calculation cases that are the evaluation target of the comprehensive performance evaluation model. In addition, it is expected to support the knowledge exchange of experts based on the FEP and scenario integrated information processing platform, and to utilize the platform itself as a part of the knowledge transfer system for knowledge preservation.
An estuary is a water ecosystem with a high abundance of the species diversity, due to a variety of complex physicochemical factors of the area where freshwater and ocean mixed. The identification of Corbicula species in the estuary environments is difficult because of various morphological characteristics. In this study, we provide taxonomic information on Corbicula species with taxonomic difficulties using morphological and genetic analysis. This study was conducted on clams from the Seomjin River-Gwangyang Bay, one of the major production area of marsh clam in Korea. As a result, we characterized Cytocrome C Oxidase subunit I (COI) sequences of the Corbicula. The 636 bp nucleotide sequences of COI have 98% homology among Corbicula species collected from 2 sites of Seomjin River-Gwangyang Bay. The phylogenetic analysis with 17 species of Corbicula indicated that most of the species collected from Seomjin River-Gwangyang Bay were brackish water clam (Corbicula japonica), and only one Asian clam (Corbicula fluminea). The evolutionary distance between C. japonica and C. fluminea was less than 0.003. Therefore, it was confirmed that C. japonica is phylogenetically closely related to C. fluminea. In 9 species of Cyrenidae, phylogenetic tree was classified into three lineages. These results will be used as an important data for an identification of clam species by providing genetic information for Corbicula species with a morphological diversity. The fish community in the Seomjin River-Seomjin River Estuary-Gwangyang Bay coast continuum was investigated three times from March 2019 to October 2019. The collected species at the eight sites during the survey period were 49 species belonging to 31 families, including two endangered species. According to Bray-Curtis similarities, observations were divided into four groups based on the fish community composition; two groups (group 1, 2) and two uncategorized groups (group 3, 4). ANOSIM based on spatial and temporal groupings indicated that the spatial differences in fish communities (R=0.398, P=0.001) were relatively more important than the temporal differences (analysis of similarities, R=0.273, P=0.002). In particular, there were significant differences between groups 1 and 2 (analysis of similarities, R=0.556, P=0.001), and similarity percentage analysis revealed that Argyrosomus argentatus (9.4%), Favonigobius gymnauchen (6.9%) and Konosirus punctatus (5.9%) contributed to these differences of fish assemblages for each group. The fish fauna distributed in the Seomjin River-Gwangyang Bay ecosystem were spatially divided and the number of species and number of individuals showed seasonal differences. This study could be a basis for understanding changes in the fish community and implementing conservation and management strategies on major species within a continuous environment of the river-estuary-ocean continuum.
As the number of aging nuclear power plants increases, the market for dismantling nuclear power plants is growing rapidly. About 40% of the cost of dismantling nuclear power plants is the waste treatment cost incurred during the dismantling process, of which concrete waste accounts for a significant portion of the total waste. Securing technology for reducing and recycling concrete waste is very important not only in terms of economy but also in terms of environment. The objective is to synthesize geopolymer using inorganic materials from cement fine powder in concrete waste. Cement fine powder in concrete waste has a large amount of inorganic elements necessary for filing materials for radioactive waste treatment such as CaO and SiO2. In particular, Ca(OH)2 is synthesized by extracting Ca2+ from concrete waste. It can be used as an alkali activator to synthesize geopolymer. The mortar from crushed concrete was used as a source of calcium. The first step is to react with concrete waste and hydrochloric acid to extract ions. The second step is to react with NaOH and synthesize Ca(OH)2. The product was divided into two stages according to the reaction method and order. The first and second products were washed and dried, and then XRD and XRF were performed. The second product was matched only Ca(OH)2 and CaCO3 at the XRD peak. In the case of XRF, it was analyzed to have a purity of 67.80–78.73%. Synthesis of geopolymer by recycling materials extracted from concrete waste can reduce disposal costs and improve the utilization rate of disposal sites.
A pump-type eDNA filtering system that can control voltage and hydraulic pressure respectively has been developed, and applied a filter case that can filter out without damaging the filter. The filtering performance of the developed system was evaluated by comparing the eDNA concentration with the conventional vacuum-pressured filtering method at the catchment conduit intake reservoir. The developed system was divided into a voltage control (manual pump system) method and a pressure control (automatic pump system) method, and the pressure was measured during filtering and the pressure change of each system was compared. The voltage control method started with 65 [KPa] at the beginning of the filtering, and as the filtering time elapsed, the amount of filtrate accumulated in the filter increased, so the pressure gradually increased. As a result of controlling the pressure control method to maintain a constant pressure according to the designed algorithm, there was a difference in the width of the hydraulic pressure fluctuation during the filtering process according to the feedback time of the hydraulic pressure sensor, and it was confirmed that the pressure was converged to the target pressure. The filtering performance of the developed system was confirmed by measuring the eDNA concentration and comparing the voltage control method and the hydraulic control method with the control group. The voltage control method obtained similar results to the control group, but the hydraulic control method showed lower results than the control group. It is considered that the low eDNA concentration in the hydraulic control method is due to the large pressure deviation during filtering and maintaining a constant pressure during the filtering process. Therefore, rather than maintaining a constant pressure during filtering, it was confirmed that a voltage control method in which the pressure is gradually increased as the filtrate increases with the lapse of filtering time is suitable for collecting eDNA. As a result of comparing the average concentration of eDNA in lentic zone and lotic zone as a control group, it was found to be 96.2 [ng μL-1] and 88.4 [ng μL-1l], respectively. The result of comparing the average concentration of eDNA by the pump method was also high in the lentic zone sample as 90.7 [ng μL-1] and 74.8 [ng μL-1] in the lentic zone and the lotic zone, respectively. The high eDNA concentration in the lentic zone is thought to be due to the influence of microorganisms including the remaining eDNA.
깔따구 (Diptera: Chironomidae)는 저서성 대형무척추동 물로 환경 및 수질 변화에 민감한 영향을 받는 중요한 환경 지표생물이다. 이러한 깔따구 유충이 정수장에서 본 연구에서는 제주 정수처리장에서 발견된 깔따구 유충의 종을 분류하기 위해 형태 사진 및 COI (cytochrome c oxidase subunit I) Primer로 증폭시킨 DNA의 염기서열을 계통수 분석을 통해 분석을 실시하였다. 정수장 내 수도꼭지와 소화관 등에서 채집된 17개체는 둥근깃깔따구 (O. tamarutilus) 14개체, 타마긴털깔따구 (P. tammaater) 3개체 총 2종으로 확인되었다. 각 깔따구 종의 형태적 특징은 두부, 하순기절, 대악, 안테나, 발톱의 형태적 특징의 분류기로 종 동정하였다. NCBI Genbank에 등록된 깔따구 19종 COI 염기서열을 기반으로 본 연구에서 조사된 17개체의 계통진화적 분석 결과 채집된 깔따구 COI 염기서열이 둥근깃깔따구 (O. tamarutilus)와 타 마긴털깔따구 (P. tammaater) 2종으로 각각 계통군 (clade)를 이루는 것이 확인되었다. 이러한 결과는 정수장 환경별 발견 되는 깔따구 유충의 다양성의 확인과 형태적-유전적 종 동정 분류정보를 바탕으로 수환경 관리 및 평가를 위한 기반 정보로 활용될 것이다.
Two waste forms, namely cement and geopolymer, were investigated and tested in this study to solidify the corrosive sludge generated from the surface and precipitates of the tubes of steam generators in nuclear power plants. The compressive strength of the cement waste form cured for 28 days was inversely proportional to waste loading (24.4 MPa for 0wt% to 2.7 MPa for 60wt%). The corrosive sludge absorbed the free water in the hydration reaction to decrease the cementation reaction. When the corrosive sludge waste loading increased to 60wt%, the cement waste form showed decreased compressive strength (2.7 MPa), which did not satisfy the acceptance criteria of the repository (3.45 MPa). Meanwhile, the compressive strength of the geopolymer waste form cured for 7 days was proportional to waste loading (23.6 MPa for 0wt% to 31.9 MPa for 40wt%). The corrosive sludge absorbed the free water in the geopolymer when the water content decreased, such that a compact geopolymer structure could be obtained. Consequently, the geopolymer waste forms generally showed higher compressive strengths than cement waste forms.
오존은 수돗물 정수장에서 이용되는 소독 물질로 미세오염 물질들을 비롯해서 박테리아나 병원성 미생물체를 효과적으로 제거하는 것으로 많은 연구가 보고되어 있다. 본 연 구에서는 실내 사육 중인 붉은 체색을 지닌 Glyptotendipes tokunagai를 대상으로 서로 다른 농도의 오존 노출에 따른 영향을 파악하기 위해 치사율, 체색 변화와 heat shock protein 70 (HSP70) 유전자 발현을 측정하였다. 오존에 노출 된 G. tokunagai에서 농도-시간 의존적으로 치사율 증가가 관찰되었다. 또한 체색 변화는 오존 농도에 따라 붉은색의 체색이 체절마다 엷어지며 탈색되고 경직되는 현상이 보였다. HSP70 유전자 발현은 저농도인 0.2~0.5 ppm에서 노출 10분과 20분에 유의한 수준으로 높게 나타났으나 (P<0.05), 30 분 노출 후에는 발현량이 감소하는 경향을 보였다. 생리적으로 저산소층에 대해 적응능력이 뛰어난 깔따구 경우에도 오존은 매우 강력한 치사 효과를 유발하여 30분 노출 후 경직과 헤모글로빈 파괴로 인한 탈색이 유발되는 것을 보여주었다. 따라서 본 결과는 수돗물 정수장에서 병원성 미생물을 제거하는 데 사용되는 오존이 수생물에 주는 영향성을 파악하는 기초자료로서 활용될 수 있을 것이다.
