        Effects of newly developed functional rice grains Goami (high-amylose rice) and Nokwon (green-kerneled rice) on the glucose metabolism and antioxidative defense system in C57BL/6N mice under high fat diet condition were investigated. Animals were randomly divided into five groups (n = 8) and given experimental diets for eight weeks: normal control diet (NC), high fat diet (HF), and high fat diet supplemented with white rice (HF-WR), Goami rice (HF-GR), and Nokwon rice (HF-NR). At the end of the experimental period, the HF group exhibited markedly higher blood glucose level, insulin concentration, plasma lipid peroxidation and lower hepatic glycogen concentration than that exhibited by NC group. However, diet supplementation of Goami and Nokwon suppressed the high fat diet-induced hyperglycemia and oxidative stress through inhibition of the glucose-regulating enzymes and enhancement of the antioxidant enzymes activities. The results illustrate that the new functional rice Goami and Nokwon may be useful in the development of functional foods with preventive effect against high fat diet-induced hyperglycemia and oxidative stress.

        The proteins from functional rice cultivars (Nogwonchalbyeo, Giant embryonic, Arhyangchalbyeo, and Goamibyeo) and general white rice were extracted and separated using two-dimensional (2D) gel electrophoresis. A wide variation in the molecular weight (MW) and pH range of the expressed proteins in rice samples were observed. The green-kerneled rice (Nogwonchalbyeo) exhibited proteins with MW of 9-57 kDa and appeared at a pH range of 4-7. The Giant embryonic contained proteins with MW of 31-63 kDa and a pH range of 5-6. The aromatic glutinous rice (Arhyangchalbyeo) showed proteins with MW of 24-28 and pH of 5.8-6.8. The high-amylose rice (Goamibyeo) exhibited proteins with MW of 3-63 and pH of 5.2-5.6. The identified proteins uniquely found and highly expressed in each cultivar may have a significant role on rice functionality. The results illustrate that the 2D gel electrophoresis is a valuable method in the determination of the protein expression profiles in functional rice grains and may be useful in the identification of specific marker proteins associated with the functional property of rice.

        고 기능성 쌀들의 장차 생활습관성질환 맞춤형 즉석밥의 활용 가능성을 검토하고자 고아미, 큰눈쌀, 백진주, 아랑향 찰 등 4품종의 쌀 및 쌀가루의 취반적성 및 조리성을 각각 검정하였다. 조 단백질, 조 지질 및 조섬유소 함량 등 영양 소 함량은 고아미가 가장 높은 수치를 나타내고 있었다. 백 진주와 고아미는 일반품종의 현미 및 백미와 비슷한 양상의 수분흡수율을 보이고, 아라향찰은 85℃에서 상대적으로 높 은 수분흡수율을 보였다. 또한 DSC parameter 중 호화열 (△H)은 고아미가 높은 수치를 나타내고 있었으며, 여러 종 류의 기능성 쌀 밥 중에서 관능적으로 가장 기호도가 높은 품종은 아랑향찰이었고, 큰눈쌀 및 백진주도 현미상태임에 도 불구하고 비교군 백미보다 높은 기호도를 나타내어, 즉 석밥을 개발을 위한 품종으로 적합하다고 사료된다.

        Glutamine synthetase (GS) is an enzyme that plays an essential role in the metabolism of nitrogen by catalyzing the condensation of glutamate and ammonia to form glutamine. Exposure of plants to cadmium (Cd) has been reported to decrease GS activity in maize, pea, bean, and rice. To better understand the function of the GS gene under Cd stress in rice, we constructed a recombinant pART vector carrying the GS gene under the control of the CaMV 35S promoter and OCS terminator and transformed using Agrobacterium tumefaciens. We then investigated GS overexpressing rice lines at the physiological and molecular levels under Cd toxicity. The GS activity along with mRNA expression were found higher in transgenic than in wild type plants. And this is validated by the low malondialdehyde contents observed 10 days after treatment. GS overexpression in rice resulted in the modulation of expression of enzymes responsible for membrane peroxidation, which may result in the sudden death of plants. Our results thus describe the features of a transgenic rice plants with enhanced tolerance to Cd toxicity.

        Aroma development in rice has been reported due to the lack of function of betaine aldehyde dehydrogenase gene (badh2) on rice chromosome 8. A lot of functional markers have been designed based on the InDels, such as 7bp deletion in exon 2, 803bp deletion in exon 4 and 5, 8bp deletion in exon 7, and 3bp insertion in exon 13. Although there were a lot of functional SNPs, other InDels have not been detected by a PCR-based marker. Here we developed a simple, co-dominant, functional cleaved amplified polymorphic sequence (CAPS) marker for fragrance trait based on 1bp insertion in exon 14. The developed marker showed a high efficiency in discriminating that special aromatic rice variety, and displayed perfect co-segregation with the trait of fragrance in the F2 population. This new marker developed in the present study would be useful in molecular breeding of fragrant rice varieties.

        본 연구는 벼에서 양성자 펌프의 활성을 증가시키는 것으로 알려진 오옥실 결합단백질 ABP57 유전자의 과발현에 따른 분자생물학적 특성을 분석해 작물 분자육종을 위한 기초자료로 활용하고자 수행하였다.ABP57 유전자 과발현 형질전환체와 대비 품종 낙동벼의 뿌리조직에서 유전자 발현 변화를 비교 분석한 결과는 다음과 같다. 두 식물체간에 발현량이 2배 이상 차이를 보인 유전자 수는 29,389개 중 총 148개였으며, 그 가운데 65개는 발현량 증가를 보였다. ABP57 유전자 과발현 형질전환체에서 발현량 변화를 보인 유전자들의 생물적 기능을 분석하기 위해 유전자 온톨로지 분석을 수행한 결과 각 부문별로 생물적 작용 부문에 67개 세부항, 세포 구성요소 부문에 59개 세부항, 분자기능 부문에 29개 세부항에 각각 영향을 미치는 것으로 나타났다. ABP57 유전자 과발현에 따라 세 개 부문 모두에서 공통적으로 나타난 영향은 H+-ATPases의 활성 증가, 여기에 필요한 에너지원 관련 화합물(ATPs, purines) 생합성 및 대사관련 활동 증가, 이온의 막투과 및 운반체 활동 증가 등이 확인되어 Kim et al.(2001)의 선행 연구결과와 일치하는 경향이었다. 그러나, 엽록체의 일부 구성요소나 G-protein 결합 단백질 신호전달경로, cytokine 신호전달 경로 등과 관련된 단백질 활성 감소는 식물 생육에 부정적 영향을 미칠 것으로 추정된다.따라서, 향후 작물의 효율적 분자육종을 위해서는 개별 유전자의 명확한 기능 분석과 더불어 유전자 상호작용에 대한 네트워크 분석 등에 대한 연구개발 노력과 함께 형질전환 식물체에 대한 생리적 특성, 표현형, 농업형질 특성 등에 대한 효과적 분석을 위해 전통육종과의 지속적 교류와 협력이 반드시 필요할 것이다.

        Environmentally inflicted stress (abiotic stress) such as high drought stress could be limiting the plant productivity. The mechanism of drought stress signaling in plant related with anti-apoptosis has not yet been full described. Understanding drought stress signaling is key to producing drought-tolerant plant. In this study we recently have identified Oryza sativa genes related abiotic stress water deficit. Abiotic stress related genes were screened from Oryza sativa cDNA library and identified gene by yeast functional screening. The yeast expression showed that they east cell grow well on SD-galactose-Leu-Ura-. The screening of over than 7000 clones from Oryza sativa cDNA libraries has been identified. 28 clones that survived following BAX-expression on inducible galactose medium. R12H780 clones confirmed protein prediction like putative senescence-associated-protein. This gene contains an open reading frame (ORF) of 108 amino acids. Transcription of R12H780 was induced in response to drought stresses, RT-PCR analysis showed transcript level in plant strongly detected in earliest time of drought stress treatment. Yeast transformed with R12H780 gene displayed markedly improved tolerance to PEG treatment, and high salinity in comparison to the control yeast (vector only). The results indicate R12H780 expression represents a new type of drought stress related gene with anti-apoptotic in Oryza sativa and endows tolerance to several types abiotic stress.

        Rice seed storage proteins (SSPs) are accumulated in storage organelles of the endosperm during seed maturation. The SSPs from the rice seeds consist of glutelins as a major SSP, and prolamins and globulins comprise about the rest 20 % of the SSPs. To improve the nutritional quality of rice seeds or processing properties of rice flour, we are attempting to change the composition of the SSPs in rice seeds. For this purpose, we generated many transgenic rice plants, which show the altered levels of the SSPs, by using the RNA interference (RNAi). Accumulation of glutelins was 76% reduced in the GluA-RNAi lines. The Pro-RNAi lines revealed the reduced levels of prolamins to 36%. The protein level of globulins was 61% reduced in the Glb-RNAi lines. Interestingly, an obvious reduction of glutelins, prolamins, and globulins was not examined in the GluA:Pro:Glb-RNAi lines. This suggests that a reduction of a few SSPs could be compensated by the increases of other SSPs at the protein levels. We are also attempting to generate transgenic rice plants expressing both a high-molecular-weight (HMW) glutelin subunit and a low-molecular-weight (LMW) glutelin subunit. These manipulations of rice SSPs might be an important contribution on improving the functional properties of rice seeds.

        Rapid extension of genomic database leads to the remarkable advance of functional genomics. This study proposes a novel methodology of functional analysis using 5-methyltrytophan (5 MT) mutant together with their 2-DE analysis and public microarray database. A total of 24 proteins was changed in 5 MT mutant and four remarkably different expressed proteins were identified. Among them, three spots were converted to Affymetrix probe. A total of 155 microarray samples from Gene Expression Omnibus (GEO) in NCBI was retrieved and followed by constructing gene co-expression networks over a broad range of biological issues through Self-Organising Tree Algorithm. Three co-expressing gene clusters were retrieved and each functional categorization with differential expression pattern was exhibited from 5 MT resistance mutant rice. It was indicated new co-expression networks in the mutant. This study suggests that on investigating possibility which correspond 2-DE to microarray database with their full potential.
