        Respiratory distress resulting from a neuromuscular disorder is often misdiagnosed and not managed properly until development of severe respiratory failure. Amyotrophic lateral sclerosis (ALS) is a rare but archetypical motor neuron disease. Most cases manifest as weakness, muscular atrophy, and progressive paralysis of the limbs. Late stage of the disease is characterized by respiratory failure due to respiratory muscle weakness. In a few cases, 3% of patients, respiratory failure is the initial presentation, preceding other symptoms. We report on a case of ALS accompanied by acute respiratory failure and weaning failure, with a review of the literature.

        Genomic DNAs were extracted from the turtle leg (Pollicipes mitella, 1798) population of Tongyeong, Yeosu and Manjaedo located in the southern sea of Korea. The turtle leg population from Tongyeong (0.929) exhibited higher bandsharing values than did turtle leg from Manjaedo (0.852). The higher fragment sizes (>1,200 bp) are much more observed in the Yeosu population. The number of unique loci to each population and number of shared loci by the three populations, generated by PCR using 7 primers in the turtle leg (P. mitella) population of Tongyeong, Yeosu and Manjaedo. Genetic distances among different individuals of the Tongyeong population of the turtle leg (lane 1-07), Yeosu population of the turtle leg (lane 08-14) and Manjaedo population of the turtle leg (lane 15-21), respectively, were generated using the CLASSIFICATION option in Systat version 10 according to the bandsharing values and similarity matrix. The dendrogram, obtained by the seven decamer primers, indicated three genetic clusters: cluster 1 (TONGYEONG 01-TONGYEONG 07), cluster 2 (YEOSU 08-YEOSU 14), and cluster 3 (MANJEDO 15-MANJEDO 21). Tongyeong population could be evidently discriminated with the other two Yeosu and Manjaedo populations among three populations. The longest genetic distance (0.305) was found to exist between individuals’ no. 02 of the Tongyeong population and no. 13 of the Yeosu population. It seems to the authors that this is a result of a high degree of inbreeding in narrow region for a long while.

        The turtle leg (Pollicipes mitella), which belongs to the family Pollicipes, is widely distributed in the rock along the shore areas of the southern sea, Jeju island and Ulleungdo island of Korean Peninsula. Generally, the size, type, stripe pattern, and color of this species vary in accordance with water depth, turbidity, nutrition, growth period, water temperature, and other environmental aspects. Genomic DNAs were isolated from 21 individuals of three turtle leg (P. mitella) populations of Tongyeong, Yeosu and Manjaedo located in the southern sea of the Korean peninsula. After incubation, we added 300 l of 3 M NaCl, and gently pipetted for a few minutes. 600 l of chloroform was then added to the mixture and inverted (no phenol). The DNA pellets were then incubation-dried for more than 10 hours, maintained at -40℃ until analysis, then dissolved in the ultra-pure water. The concentration of the extracted genomic DNA was measured by its absorbance ratio at 260 nm, with a spectrophotometer (Beckman DU 600 series, UK). Seven primers were exposed to generate the unique loci to each population and number of shared loci by the three populations of turtle leg which could be clearly scored. The hierarchical clustering tree was analyzed by the similarity matrices to generate a dendrogram using pc-package program Systat version 10. As regards average bandsharing value (BS) results, the higher fragment sizes (>1,200 bp) are much more observed in the Yeosu population. The number of unique loci to each population and number of shared loci by the three populations generated by PCR using 7 decamer primers in the turtle leg (P. mitella) population of Tongyeong, Yeosu and Manjaedo. Genetic distances among different individuals of the Tongyeong population of the turtle leg (lane 01~07), Yeosu population of the turtle leg (lane 08~14) and Manjaedo population of the turtle leg (lane 15~21), respectively, were generated using the CLASSIFICATION option in Systat version 10 according to the bandsharing values and similarity matrix.         The seven selected primers OPA-02, OPA-04, OPA-18, OPD-07, OPD-08, OPD-15 and OPD-16 were used to generate unique shared loci to each species and shared loci by the two species. The hierarchical dendrogram indicates three main branches: cluster 1 (RORETZI 01~RORETZI 11) and cluster 2 (HILGENDORF 12~HILGENDORF 22) from two geographic populations of ascidians, Halocynthia roretzi and H. hilgendorfi. The shortest genetic distance displaying significant molecular difference was between individuals' HILGENDORF no. 14~HILGENDORF no. 19 (genetic distance =0.008). Ultimately, individual no. 02 of the RORETZI ascidian was most distantly related to HILGENDORF no. 21 (genetic distance=0.781). These results demonstrate that the H. roretzi population is genetically different from the H. hilgendorfi population. From what has been said above, the potential of PCR analysis to identify diagnostic markers for the identification of two ascidian populations has been demonstrated. Generally speaking, using a variety of decamer primers, this PCR method has been applied to identify specific markers particular to line, species and geographical population, as well as genetic diversity/polymorphism in diverse species of organisms.

        'Angelicae Pubescentis Radix' (APR) is an important oriental medical preparation. In Korea, Aralia continentalis has been recognized as the source plant of APR. Aralia cordata, which is difficult to distinguish from A. continentalis, and Heracleum moellendorffii, which is frequently used in lieu of A. continentalis, are traded in Korean herbal markets. In contrast, in China, Angelica pubescens is recognized as the source plant of APR. In this study, we devised a method not only to discriminate A. contientalis from A. cordata, but also to discriminate both A. contientalis and A. cordata from H. moellendorffii and A. pubescens. Based on the discrepancy in the sequences of specific regions of ITS, we designed a Cont F/ Cont R primer set to amplify a 173 bp PCR band that appears only in A. continentalis. Additionally, we designed an Ara F/ Ara R primer set to amplify a 278 bp PCR band that appears in both A. continentalis and A. cordata. Using these primer sets and the ST R primer to confirm the PCR amplification results, we developed a simple multiplex PCR method for differentiating A. continentalis from A. cordata and to concurrently differentiate both A. continentalis and A. cordata from other APR herbs.

        이 연구에서는 자기결정이론에 근거한 확인규제 모형을 토대로 운동이나 신체활동 자체에 관심이 없는 학생들에게 인지적-행동적 참여를 촉진시키기 위한 전략으로 자율성지지 접근의 당위성 제시를 활용하여 그 효과를 검증하고자 하였다. 그 결과, 중재집단 내 학생(남학생, n=17)들이 통재집단 내 학생들(남학생, n=20)에 비해 자율성지지 환경인식, 확인규제, 의도에서 향상을 보였으며, 무동기에서는 소폭의 감소를 보였다. 특히, 지각된 자율성지지와 확인 규제에서 집단 간의 유의한 차이를 보였다. 더불어, 당위성 제시의 효과에 대한 사회적 타당도를 확보하기 위해 집중 그룹 인터뷰를 실시한 결과 중재집단 내 학생들은 첫째, 신체활동에 대한 긍정적인 인식변화, 둘째, 상호 간의 긍정적인 사회적 관계의 형성, 그리고 셋째, 개인차에 따른 미래 신체활동에 대한 의도를 나타냈다. 이러한 결과를 토대로 현장적용 가능성과 향후 연구과제에 대한 방향을 제시하고자 한다.
