        This experiment was conducted to study whether priming with deep sea water results in enhancement of seed germination and to identify the optimum concentration of the priming solution, and duration of priming using sweet pepper (Cv. California wonder), rice (Cv. Ilpum) and ginseng seed. Sweet pepper and rice seeds were primed with 5 various concentrations (5%, 10%, 15%, 20% and 30%) for deep sea water for 48 hours, 24 hours and 12 hours at 25℃ and ginseng seeds in 5%, 10%, 15%, 20%, 25% and 30%, and 2,4,6, and 8 electrical conductivity (EC) which were made by desalinating deep sea water. Priming in deep sea water (DSW) improved the early and final germination percentage, mean germinal on rate, emergence percentage and root and shoot length, compared with plain water, KNO3 and without priming treatments. In sweet pepper, 24 hours priming with 5 percentage DSW significantly improved the early germination percentage and radical length. It has also improved the mean germination and emergence days and early emergence percentage, compared with KNO3 and control. Whereas, in rice, 48 hours priming with 10 percent DSW significantly improved the early germination percentage, plumule emergence percentage, root length and shoot height. Hence the best seed priming treatment on sweet pepper and Rice are 24 hours with 5 percentage DSW and 48 hours with 10 percentage DSW, respectively, whereas in ginseng, priming with EC4, EC8 and 25% DSW had shown better germination.

        화성벼와 O. minuta 간 여교잡을 통해 반복친인 화성벼와 출수기, 간장 등 여러 작물학적 특성에서 뚜렷한 차이를 보이는 염색체단편 치환계통 "WH79006"을 육성하였는데 화성벼와 WH79006의 차이는 WH79006에 이입된 O. minuta 염색체 단편에 의한 것이라고 할 수 있다. WH79006이 화성벼의 유전적 배경에 O. minuta의 어느 염색체 단편을 가지고 있는지를 검정하기 위해 SSR마커를 이용하였다. 벼 염색체에 골고루 분포한 294개의 SSR 마커를 검정한 결과 최소한 28개의 이입 단편을 검정하였는데 이들은 2번 염색체를 제외한 모든 염색체에 분포하였으며 전체 길이는 약 168cm이었다. 간장 관여 QTL을 탐색하기 위해 화성벼/WH79006 조합의 75개 F2 개체를 육성, 간장을 조사하였다. QTL분석 결과 6번 염색체의 RM225부근에서 간장에 관여하는 QTL cl6가 탐지되었다. cl6는 전체 표현형변이의 9.6~% 를 설명하였으며 O. minute의 대립유전자가 간장을 크게 하는 방향으로 작용하였다. 이 결과는 O. minuta가 포복형임에도 불구하고 간장 조절 유전자들 중에는 간장을 크게 하는 방향으로 작용하는 유전자가 있음을 제시하고 있으며, 이 유전자들은 앞으로 QTL 분석을 통해 그 작용을 밝힐 예정이다. 추후교잡 집단을 이용 O. minuta 특이적 단편들이 어느 형질과 관련이 있는지를 밝힐 예정이다.

        To demonstrate the importance of transformation efficiency in independent event, molecular and cytogenetic analysis were conducted with genomic DNA and chromosome of transgenic plants produced by Agrobacterium tumefeciens LBA4404 (pSBM-PPGN: gusA and bar). Selection ratios of putative transgenic calli were similar in independent experiments, however, transformation efficiencies were critically influenced by the type of regeneration media. MSRK5SS-Pr regeneration mediun, which contains 5 mgL-1 kinetin, 2% (w/v) sucrose in combination with 3% (w/v) sorbitol, and 500 mgL-1 proline, was efficient to produce transgenic plant of rice from putative transgenic callus in the presence of L-phosphinotricin (PPT). With MSRK5SS-Pr medium, transformation efficincies of Nagdongbyeo were significantly enhanced from 3.7% to 6.3% in independent callus lines arid from 7.3% to 19.7% in plants produced, respectively. Stable integration and expression of bar gene were confirmed by basta herbicide assay, PCR amplification and Southern blotting of bar gene, and fluorescence in situ hybridization (FISH) analysis using pSBM-PPGN as a probe. In Southern blot analysis, diverse band patterns were observed in total 44 transgenic plants regenerated from 20 independent PPT resistant calli showing from one to five copies of T-DNA segments, however, the transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.
