In vitro proliferation system was achieved by using nodal segment excised from greenhouse grown juvenile stock plants of Alnus japonica. Stem explants were cultured on MS medium supplemented with different plant growth regulators of cytokinin and/or their combinations. The most effective cytokinin source was the combination of zeatin 2.0 mg/L and TDZ 0.05 mg/L producing the average number of shoots (16.8 ± 3.6). In addition, healthy roots were formed after small clumps of shoots were transferred to half strength of MS medium containing IBA 0.02 mg/L with optimal rooting capacity. Soil acclimatization was successfully conducted in cell tray containing artificially mixed soil with 92 % survival rate.

Allium wakegi was cultured shoot tip in the condition of light culture. The Allium wakegi added plant growth regulator was observed of plant regeneration and bulblet formation. Callus Induction and growing rate was the best of 78% when added alone 2,4-D 0.5mg/L. In the formation of shoot, its regeneration rate was 96% when added BA 0.5mg/L in the light culture condition. When BA 0.5mg/L and NAA 0.5mg/L mixed and BA 0.5 mg/L and NAA 1.0mg/L mixed, the rates were 99% and 97% respectively, and these conditions were suitable for forming shoot. In the formation of roots, when added NAA 2.0mg/L in the light culture condition, the regeneration rate was 90.6 % and the roots were abnormal. When added NAA 1.0mg/L, the rate was 82 % and the highest. In the formation of bulbs, when BA 05mg/L and NAA 1.0mg/L mixed, the root generantion and its size in the bulbs was the best compare to other treatment experiments.

This study was conducted to evaluate the effect of mecoprop-wp concentrations on weed control, growth characters and yield in flax, after 7cm and 14cm of plant height it to the field. The major weeds were Alopecurus aequalis var. a murensis (kom) ohwi, Persicaria hydropiper spach, Lamium amplexicaule L, Stellaria alsine Grimm var. undulata ohwi, Eleusine indica(L.) Gaertn. Flax yield were increased somewhat more with 250g/10a mecoprop-wp at 7cm of plant height than the other treatment and by hand weeding treatment of mecoprop-wp, 250g/10a at 7cm of plant height was slightly harmful for the flax with recommended concentration, On the other hand, all treatments were harmful in the double dosage level.

The purposes of this study were to clarify of floristic composition and the landscape structure on the habitats of three Thalictrum plants, which were distributed in Daejeon, Korea. Floristic composition of habitats was organized by living in flatten open space needed a lot of sunlight and humidity. Landscape ecological structure of habitats was defined by soils of loamy skeletal mesic, slope between 25 to 30˚ Vegetation rating was 75 percent of woody coverage at less than 450 m above the sea levels, respectively. Distribution patterns of Thalictrum plants in Daejeon were presented on the 1 km X 1 km grid map.

Root chicory (Cichorium intybus L. var. sativus) is potential alternative medicinal and sugar crop which accumulates a high amount of linear polyfructan, inulin in its roots. A problem in root production is that over-wintered stock plants often flower. Once the plant becomes reproductive, stem elongation and root growth slows and floral buds arise from every node, rendering the plants useless for propagation. The objectives of this research was to examine the effectiveness of manipulating environmental factors containing photoperiod, temperature and number of leaf states. The experiment was performed in growth chamber to create two photoperiods (8 h, and 16 h) with three temperature regimes (5℃/3℃, 10℃/8℃ and 15℃/13℃ day/night temperature) for a total of six treatments on three type of true-leaf stage of plant. Data of bolting rate, shoot and root length, shoot and fresh weight was invetigated in each treatments. This is the first report on changes in bolting rate and shoots and roots production during a whole growing season and differences in the effect of cold and photoperiod treatment depending on the true-leaf stage of plant.

Bacterial grain rot of rice caused by Burkholderia glumae was examined between weather condition and disease incidence. From 1998 to 2000, average disease incidence was 3.0 % without difference in survey regions. However, it was related closely to amount of rainfall that disease incidence higher in 1998 and 2000 to 3.0 % and 3.6 % respectively than 2.3 in 1999 relatively small volum of rainfall season.

To demonstrate the importance of transformation efficiency in independent event, molecular and cytogenetic analysis were conducted with genomic DNA and chromosome of transgenic plants produced by Agrobacterium tumefeciens LBA4404 (pSBM-PPGN: gusA and bar). Selection ratios of putative transgenic calli were similar in independent experiments, however, transformation efficiencies were critically influenced by the type of regeneration media. MSRK5SS-Pr regeneration mediun, which contains 5 mgL-1 kinetin, 2% (w/v) sucrose in combination with 3% (w/v) sorbitol, and 500 mgL-1 proline, was efficient to produce transgenic plant of rice from putative transgenic callus in the presence of L-phosphinotricin (PPT). With MSRK5SS-Pr medium, transformation efficincies of Nagdongbyeo were significantly enhanced from 3.7% to 6.3% in independent callus lines arid from 7.3% to 19.7% in plants produced, respectively. Stable integration and expression of bar gene were confirmed by basta herbicide assay, PCR amplification and Southern blotting of bar gene, and fluorescence in situ hybridization (FISH) analysis using pSBM-PPGN as a probe. In Southern blot analysis, diverse band patterns were observed in total 44 transgenic plants regenerated from 20 independent PPT resistant calli showing from one to five copies of T-DNA segments, however, the transformants obtained from one callus line showed the same copy numbers with the same fractionized band patterns.

Several wild plant species are known to contain biologically active substances that are allelopathic to weed species as well as antioxidant to foods. Plant extracts or residues from leaves of 4 species, Achyranthes japonica (speedwell), Cucumis sativus (Cucumber), Trifolium repens (white clover), and Vicia angustifolia (narrowleaf vetch) were bioassayed against Medicago sativa (alfalfa) or Echinochloa crus-galli (barnyard grass) to determine their allelopathic effects, and used for measurement of antioxidant activities. The aqueous extracts applied on filter paper significantly inhibited root growth of alfalfa. Aqueous extracts or residues from V. angustifolia showed the most inhibitory effect on alfalfa or barnyard grass seedling growth and followed by A. japonica and T. repens. Oxidative stability by Rancimat method, antioxidant activity by TBA (2-thiobarbituric acid) method and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging activity for the ground samples or methanol extracts were the greatest in V. angustifolia, although were less than those of commonly used antioxidants, BHT (butylated hydroxytoluene) and ascorbic acid. These results suggest that the wild plant species had potent allelopathic and antioxidant activities, and that their activities differed depending on plant species.

This study was conducted to investigate the relationships between yearly variations of weather elements and productivity in rice. In addition, coefficients of correlation among yield and yield components were used to find out the relationships between weather elements and productivity. Coefficients of variance (C.V.) of air temperature mean was considerable with 25%, but the variation by duration of sunshine was small in May. Culm length and number of spikes were great with c.v. of 21.5, 16.4%, respectively. Coefficients of correlation between temperatures of cultivation in May and yield were positive correlations. Coefficients of correlation between precipitation and sunshine of cultivation period from Oct. to May and yield were negative correlations. Coefficients of correlation amount the culm length, number of spikes, 1,000 grains wt. of seed, and yield were positively significant at the level of 1 %, respectively.

The establishment of an efficient protocol for plant regeneration and micropropagation from leaf explant cultures of Chicory, Cichorium intybus L. var. sativus. is reported. Callus formation rate appeared 100% from explant in all growth regulators, but calli formed in the prensence of naphthaleneacetic acid (NAA) were appeared very compact and non-embryogenic state. The regenerated shoots were obtained from leaf explant cultures on solid MS medium containing different concentrations of cytokinins and auxin. The highest number of shoots (5.7) per explant and shoot growth (2.8cm) was obtained on MS medium containing 0.1 mg BAP L-1 and 0.1 mg NAA L-1 . Indole acetic acid was the most suitable auxin for root formation among three auxins tested. 2,4-D had no effect on shoot and root formation.

We used an amplified fragment length polymorphism (AFLP) analysis, a novel PCR-based technique, to differentiate Xanthomonas oryzae pv. oryzae (Xoo) of Korean races. The 6 strains of Xoo K1, K2, K3 races were tested with 81 AFLP primer combinations to identify the best selective primers. The primer combinations were selected according to their reproducibility, number of polymorphic bands and polymorphism detected among Xoo strains. 18 strains of Xoo K1, K2 and K3 races were analyzed with the selected combinations of primer set. Some primer combinations (Eco R I +1 / Mse I+1) could differentiate Xoo of Korean races that were not distinguished by other fingerprinting analysis. Thus AFLP fingerprinting permitted very fine discrimination among different races.

Lettuce (Lactuca sativa L.) is known to contain water-soluble substances that are biologically active. Aqueous or methanol extracts and residues from leaves of lettuce plants were assayed to determine their allelopathic effects, and the causative allelochemicals from fractions were quantified by means of HPLC analysis and bioassayed. Extracts from oven-dried leaf samples were more phytotoxic than those from freeze-dried samples. Leaf extracts of 40 g L-1 were completely inhibitory on root growth of alfalfa (Medicago sativa L.), while root growths of barley (Hordeum vulgare L.) and soybean (Glycine max L.) were less sensitive. Early seedling growth of both alfalfa and barnyard grass (Echinochloa crus-galli) was significantly reduced by methanol leaf extracts. The major allelopathic substances analyzed by HPLC were coumarin, trans-cinnamic acid, o-coumaric acid, p-coumaric acid and chlorogenic acid. Of them p-coumaric acid was found as the greatest amount (8.9 mg 100 g-1 ) in the EtOAc fraction; only coumarin was found in all the fractions. Hexane and EtOAc fractions of L. sativa reduced alfalfa root growth more than did BuOH and water fractions. These results suggest that lettuce had potent herbicidal activity, and that its activity differed depending on type and amount of causative compounds by fraction.
