This study was conducted to develop the clonal propagation technique through in vitro culture using basal stem explants in Phalaenopsis hybrid grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when basal stem explants were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 ml/L coconut water, 1 mg/L NAA, 5 mg/L 2iP and 2.5g/L gel rite. PLBs transferred to Hyponex medium were regenerated to plantlets. Plantlets transferred to plastic pots containing spagnum moss were developed and successfully acclimatized under greenhouse. The flower was bloomingly opened in plants regenerated from basal stem explants. The flower was not different from both mother plant and plant induced through clonal propagation of Phalaenopsis hybrid.
The objective of this study is to investigate morphological characteristics of seeds and effects of environmental factors such as light and temperature on the germination and seedling growth of Adenophora triphylla var. japonica, Codonopsis lanceolata, Platycodon glandiflorum, Which belongs to Campanulaceae and useful for on oriental medicine materials. Seed shape of Codonopsis lanceolata is Elliptic. Platycodon glandiflorum is Oblong. Color of seed coats varied from yellow to dark brown. Seed size of Adenophora triphylla var. japonica is 1.37mm in length, 0.69mm in width. One thousand-seed weight of Adenophora triphylla var. japonica is 258mg. Seed size of Codonopsis lanceolata is 4.45mm in length, 2.11mm in width. One thousand-seed weight of Codonopsis lanoceolata is 1,278mg. Seed size of Platycodon glandiflorum is 2.31mm in length, 1.10mm in width. One thousand-seed weight of Platycodon glandiflorum is 988mg. Optimum temperatures for seed germination and seedling growth range from 20 to 25℃.
Colored apiculus, awn, and long empty glume are indicators of wildness and are usually eliminated during rice domestication. Genetic analysis was conducted to clarify the inheritance patterns of awn, apiculus color, and long empty glume in Korean rice collection. Based on individual characterization of F2 progenies derived from crosses between parents with colorless and purple apiculus, two (3 colored: 1 colorless) or three dominant genes (9 purple: 3 red: 4 colorless) are estimated as controlling this character by simultaneous complementary action. Different inheritance systems were detected between S237 and S245 of 'Shareibyeo' which belong to the weedy type. To determine the genes responsible in awning and long empty glume characters, the inheritance of landrace varieties of rice ('Naengdo' and 'Yuna') was investigated. In the crosses of awned land race and awnless cultivar, three dominant genes are supposed to control the awning genetic system by 63 awned: 1 awnless individual. As for long empty glume, one recessive gene, g-l on the chromosome 4, was the one controlling the segregation ratio of 3 normal empty: 1 long empty glume. By analyzing the Korean rice collection, the inheritance systems of these wild characters may lead to a better understanding of rice domestication in the future.
This study was carried out to obtain basic information for possibility of oral vaccine in carrot using Agrobacteruim -mediated transformation system. The epitope region of porcine epidemic diarrhea virus (PEDV) spike gene which is classified as a member of the Coronaviridae and causes an acute enteritis in pigs was successfully expressed in carrot (Daucus carota) using the Agrobacterium-mediated transformation system. Hypocotyl segments of in vitro germinated plantlets were infected with Agrobacteriun tumefaciens LBA 4404 harboring PEDV spike gene. Embryogenic callus (EC) was induced on MS selection medium with 1 mg/L 2,4-D, 50 mg/L kanamycin and 300 mg/L cefotaxime after 45 days of culture. Subcultured ECs on MS selection medium without 2,4-D were converted to somatic embryos (SE) of various stage; globular, heart and torpedo stage. Putative transgenic embryos were selected on MS medium with 50 mg/L kanamycin and 300 mg/L cefotaxime. Regenerated plantlets from transformed SE were induced on MS medium containing 50 mg/L kanamycin after 30 days of culture. Genomic PCR confirmed the integration of PEDV spike gene into nuclear genome of carrot and northern blot analysis demonstrated the expression of PEDV spike gene in transgenic carrot.
In this study, an on-site survey was conducted around Gasado island at Jodo-myeon, Jindo-gun, Jeonnam during July 2 to 25, 2002. The naturalized plants investigated in the surveyed sites were revealed 15 families and 39 species, including Althaea rosea and Oxalis articulata, which are ornamental plants. Only 10 species appeared in Imjado island among the surveyed sites the surveyed sites and Imjado Island, including Elymus sibiricus, Lolium perenne, Chenopodium album, Oxalis articulata, Oxalis corymbosa, Oenothera odorata, Xanthium strumarium, Bidens frondosa, Ambrosia artemisiaefolia var. elatior, and Taraxacum officinale. 6 species were distributed only in Gasado island, Jindo-gun, which included include Festuca myuros, Chenopodium ambrosioides, Amorpha fruiticosa, Malva neglecta, Calendula arvensis, and Carthamus tinctorius. 18 species were showed in Soheuksan island.; Yeosu area, 29 species; Imja Island in Sinan-gun, 34 species, and Gasado Island in Jindo-gun revealed 39 species. It is believed that the species of naturalized plants have been expanded compared to the other areas because of close location to the inland and large population fastiy. The Urbanization Index(UI) of this area was found to be 19.5. The naturalized plant communites in these surveyed sites were classified into Rumex crispus, Elymus sibiricus, and Lolium perenne community according to the vegetation table. Furthermore, they formed a community around areas that received great artificial influence from human dwellings or roads due to the characteristics of species distribution
The objectives of this study, an analysis of the variation for leaf and fruit characteristics among the selected ten populations of Sorbus commixta Hedl. could be used for the conservation of gene resources and could provide information to superior trees selection. The results obtained from this study can be summarized as follows; Approximately, the Mt. Sungin population at Ulleung island showed larger values in overall characteristics and populations. On the other hand, Mt. Halla population at Jeju island showed the smaller values of the overall characteristics and populations. ANOV A tests showed that there were statistically significant differences in all leaf characteristics among the populations as well as individual trees within populations. But, for fruit characteristics, differences were statistically significant only among the populations. Cluster analysis using single linkage method based on leaf and fruit characteristics showed that ten selected populations of S. commixta in Korea could be clustered into three groups. Group I is Mt. Sungin at Ulleung island, Group II is Mt. Halla at Jeju island, and Group III comprises Osan, Mt. Kaji, Mt. Duckyoo, Mt. Balwang, Mt. Sobaek, Mt. O-dae, Mt. Jiri, and Mt. Taebaek. The selection level based on major agronomic traits, which are the Number of Fruit per Fruiting Lateral(NFL) over 50, and Fruit Length(FL) and Width(FW) over 10 mm, and Weight of 100 Fruit(WFI00) over 66 g, was applied on 100 sample trees, and five trees were selected. The selection effects from selected trees in NFL, FL, FW, and WF100 were evaluated as 132%, 151 %, 142%, and 264% compared to the mean of those 100 sample trees, respectively. Especially, Ulleung 2 showed excellent values that NFL and WFI00 were 95, and 69 g, respectively, suggesting a promising new cultivar for larger fruit and high productivity.
This study was conducted to investigate the relationships between yearly variations of meteorological elements and yearly variations of productivity in rice. In addition, correlation coefficients among yield and yield components were used to find out the relationships between meteorological elements and productivity. Yearly variation of the mean air temperature in May was large with coefficients of variation(C.V.) of 25.0%, but the variation of the duration of sunshine in May were relative small. No. of panicles per hill and 1,000 grains wt. of brown rice were great with C.V. of 21.1, 19.7%, respectively, brown rice yield show more or less C.V. of 5.5% and milled rice show still less variation. Correlation coefficients between temperatures in period of cultivation from May and yield were positive correlations. Correlation Coefficients between precipitation in period of cultivation from Sep. to Oct. and yield are positive correlations. Correlation coefficients amount the panicle length, no. of panicles, no. of spikelets, ratio of ripened grains, 1,000 grains wt. of brown rice, milled rice yield, brown rice yield and milled rice yield were positively significant at the level of 1 %, respectively.
Allium wakegi Araki was grown at plant spacings of 5, 10, 15, and 20 cm to determine the effect of planting density on the growth and yield. Allium wakegi Araki plants grown at the 5 cm plant spacing had the lowest bulb diameter and bulb weight, while plants at the lowest density (20 cm spacing) had the highest bulb diameter, bulb number, bulb weight and fresh weight. In general, plants grown at narrower spacings produced significantly smaller bulb diameter and bulb weight, but resulted in the highest yields and plants per hectare and lower fresh weights per plant.
Germination efficiency, such as days to germination and conditions for the hybrid seeds, was tested after hybrid seeds were sown on the medium. Seeds were germinated from 67 to 126 days after seeding in all cross combinations, and germination condition was different among hybrid combinations. There was big difference on days to germination based on the different media, that is, days to germination in the hyponex medium was shorter than that in the Knudson C medium. Mutants such as MMS and NaN3 were used to cause mutation. Germination occurred with 0.1 % NaN3 and MMS. However, even though germination occurred in other treatments (0.01 % and 0.2% of NaN3 and MMS), brownish phenomenon was intense, or did not proceed and got worse after 4-5 months of seeding. In addition, it was performed to choose appropriate medium for the growth of Cymbidium rhizome whose media adaptability is more different than other orchids. Different concentration of NAA and Kinetin was used. As a result, C. goeringii hybrid showed difference for the concentration of NAA and Kinetin combination.
Many higher fungi were collected at Mt.Jiri, Mt.Pukhan, Mt.Moak from June 2002 to October 2002.They were identified. According to the result Clitocybe phyllophila, Omphalina wyniae, Mycena diosma, Cortinarius haasii and Cortinarius allutus were newly to Korea. They were designed Korean common names by author.
Effects of shaking, medium consistency and anther density on polyhaploid production in two wheat cultivars, Pavon and Chris, were studied using a modified 85D12 medium. Pavon produced more calli in shaking and more albino plants tban Chris. However, Chris produced threefold more green plants than Pavon in non-shaking treatment. More calli and green plants were derived from non-shaking treatment than those from shaking treatment. Anthers were cultured on both liquid and semi-solid 85D12 media, using two anther densities, 48 and 96 anthers per plate. Although Pavon generally produced more calli and albino plants than Chris, Chris produced more green plants than Pavon. More green plants were derived from semi-solid medium than those from liquid medium. A factor that may affect plant regeneration from anthers is the length of time on initiation medium. Most of the calli for both genotypes were transferred during the first two time periods. Fertility, as measured by seed set, was determined for all surviving regenerated plants. About 24% of Chris and Pavon anther-derived green plants in the experiment of medium consistency and anther density produced seed.
