The soybean cyst nematode (Heterodera glycines Inchinoe; SCN) is a devastating pest of soybean and is responsible for significant losses in yield. The use of resistant cultivars is the effective method to reduce or eliminate SCN damage. The objective of this research is to identify AFLP markers linked to the SCN resistant genes. Bulked genomic DNA was made from resistant and susceptible genotypes to SCN and a total of 19 primer combinations were used. About 31 fragments were detected per primer combination. The banding patterns were readily distinguished in resistant and susceptible bulked genotypes. Polymorphic fragments were detected between resistant and susceptible bulked genotypes in the primer combination of CGT/GGC, CAG/GTG and CTC/GAG. In primer combinations of CGT/GGC and CAG/GTG, bulked resistant genotype produced a polymorphic bands. However, in primer of CTC/GAG, bulked susceptible genotype produced a polymorphic fragments. Three AFLP markers identified as a polymorphic fragments between bulked genomic DNA were mapped in 85 F2 population. Among them, only two markers, CGT/GGC and CTC/GAG, was linked and was mapped. Broad application of AFLP marker would be possible for improving resistant cultivars to SCN.
This study was conducted to develop the clonal propagation technique through in vitro culture using by leaf sheath explants of Phalaenopsis grown in vitro. The highest frequency of protocorm-like body (PLB) formation was obtained when explants of leaf sheath were cultured on VW medium containing 30g/L sucrose, 500 mg/L activated charcoal, 150 mVL coconut water, 1 mg/L NAA, 1 mg/L 2ip and 2.5 g/L gelrite. The PLB formation rate of VW medium was highest followed by modified Hyponex medium, and lowest in MS medium. Plantlets induced from PLBs transferred to plastic pots including spagnum moss were well developed.
The 129 soybean genotypes were collected in 43 island locations from January to May 2001. Seeds of 129 genotypes collected were analyzed for crude protein and fatty acid composition contents. The crude protein content was averaged to 41.1 % and ranged from 37.4% to 44.4%. The average palmitic acid, stearic acid, oleic acid, linoleic acid and linolenic acid content were 12.0%, 4.0%, 23.2%, 55.5%, and 7.9%, and the ranges of those were 10.0% to 15.0%, 3.0% to 4.8%, 21.7% to 25.5%, 50.2% to 58.3% and 7.0% to 12.0%, respectively. Heritabiliries of palmitic acid, oleic acid, linoleic acid, and crude protein were higher, but that of stearic acid and linolenic acid were relatively lower. The genotypic correlation coefficients between crude protein and oleic acid showed highly positive correlation, but that of linoleic acid showed highly opsitive correlation, but that of linoleic acid showed highly negative correlation and also palmitic acid, stearic acid and linolenic acid showed negative correlation.
This experiment was conducted to investigate the effects of turf grass growth to seedling rates and bed soil types. The results of this experiment were summarized as follows; the more increasing the seedling rates, the plant height and leaf length were longer, but the number of leaf and number of branch were fewer. Incase of transplanting of turf grass seedling by rice transplanting machine, it might be considered that the proper alternative bed soil was sandy loam soil with regarding to the economic aspects. The maximum seedling rate of turf grass in the seedling tray for rice was 1,000 of seedling amount due to the nutrient competition with intensive seedling. As the results indicated, it might be recommended that the proper alternative bed soil was sandy loam soil with 1,000 of seedling rates in case of transplanting of turf grass seedling by rice transplanting machine.
Special objective of this study is to investigate morphological characteristics of seeds and effects of environmental factors such as light and temperature on their germination and seedling growth of Angelica acutilobu in Umbelliferae plants, using them as oriental medicine materials. Seed shape of Angelica acutilobu is ovate. Color of seed coats varied from yellow to dark brown. Seed size was 5.5 mm in length, 4.0mm in width. One thousand-seed weight of Angelica acutilobus was 3,210mg. Optimum temperatures for seed germination and seedling growth ranged from 20 to 25℃.
This study was conducted to investigate the relationships between yearly variations of climatic elements and yearly variations of productivity in sesame. In addition, correlation coefficients among yield and yield components were estimated. The data of yield and yield components were investigated for 10 years from 1992 to 2001. The meteorological data gathered at the Yeosu Weather Station for the same period were used to find out the relationships between climatic elements and productivity. Yearly variation of the amount of precipitation in July and September were large with coefficients of variation(c.v.) of 64.59, 92.47%, respectively, but the variation of the average temperature in June and August were relative small. Yield and plant height greatly with c. v. of 26.24, 23.41 %, respectively, 1, 000 grain weights show more or less c.v. of 3.83% and length capsule setting show still less variation. Correlation coefficients between maximun temperature in period of cultivation(from June to September) and yield are positively significant at the level of 5.1 %, respectively. Correlation coefficients amount the plant height, length capsule setting, number of capsules per plant, weight of 1, 000 grains and seed yield were positively significant at the level of 1 %, respectively. Simple linear regression equations by the least square method are estimated for number of capsules per plant(Y1) and the maximun temperature in August(X) as Y1=10.1255+0.1725X, and for yield(Y2) and the maximun temperature in August(X) as Y2=21.6151 + 1.3724X.

It is effective in removing dangerous for an unborn child and suppressing pain. It is also good as a detoxification and in treating cold. In the long history of Korea, dyeing has been applied for a means representing the grace of natural and inner esthetic consciousness of man. Vegetable dyes give us such great benefits, diversified color, but no pollution. And ramie fabric has distinctive features such as beautiful brilliance, elegance, and strong durability. So, it is regarded as a special product of Korea traditionally. These studies were carried out to treat with acetate iron, dichloride copper and alum with a mordant to ramie fabric. The ramie fabric was died with Perilla frutescens Herba extract. The results of experiment showed as follows: First, the chromophoric degree was the highest in acetate iron but not distinction in another mordants. Second, the light colorfastness was the highest in non treated and dichloride copper, but alum was the lowest. Third, the discoloration was alum and dichloride copper showed first grade in washing colorfastness. Abrasion colorfastness was not significant in this test. According the previous results, Perilla frutescens Herba has an efficiency in removing skin disorders and suppressing pain. So it considered that Perilla frutescens Herba can be applied effectively to theraphy of detoxification and in treating cold.

The objective of this study was to understand the conservation of gene resources and provide information for mass selection' of winter bud characters among the selected populations of Kalopanax septemlobus Koidz using analysis of variance(ANOVA) tests. The obtained results are shown below; 1. Ten populations of K. septemlobus were selected for the study of the variation of winter bud characters in Korea. The results of the analysis of variance(ANOVA) tests shows that there were statistically significant differences in all of the winter bud characters among those populations. 2. Correlation analysis shows that width between Height and DBH(Diameter at breast height) characters have negative relationship with all of the characters, as ABL(Apical branch length), ABW(Apical branch width), AWBL(Apical branch winter bud length), AWBW(Apical branch winter bud width), ABT(Apical branch No. of thorns), ABLB(Apical branch No. of lateral bud) and LBL(Lateral branch length), LBW(Lateral branch width), LBT(Lateral branch No. of thorns), LBLB(Lateral branch No. of lateral bud). 3. The result of principal component analysis(PCA) for winter buds showed that the first principal components(PC' s) to the fourth principal component explains about 78% of the total variation. The first principal component(PC) was correlated with AWBW, LWBW, and LBL and the ratio of ABL/ABW and LBL/LBW out of 16 winter bud characters. The second principal component correlated with ABL, ABW, ABLB, LWBL(Lateral branch winter bud length), and LBW and the ratio of AWBL/AWBW. The third principal component correlated with ABL, ABW, LWBL, LBL, and the ratio of LBL/LBW. The fourth principal component correlated with LBL and the ratio of LWBL/LWBW(Lateral branch winter bud width), LBL/LBW. Therefore, these characters were important to analysis of the variation for winter bud characters among selected populations of K. septemlobus in Korea. 4. Cluster analysis using the average linkage method based on 10 selected populations for the 16 winter bud characters of K. septemlobus in Korea showed a clustering into two groups by level of distance 1.1(Fig. 3). As can be seen in Fig. 3, Group I consisted of three areas(Mt. Sori, Mt. Balwang and Mt. Worak) and Group Ⅱ contisted of seven areas(Suwon, Mt. Chuwang, Mt. Kyeryong, Mt. Kaji, Mt. Jiri, Muan, and Mt. Halla). The result of cluster analysis for winter bud characters corresponded well with principal component analysis, as is shown in Fig. 2.
To develop optimal storage method of root of Zingiberis Rhizoma, which has been grown as major cultural herbal medicine materials, root of Zingiberis Rhizoma, was stored for 10 months with different packing materials and sealing methods. The loss in dry weight as influenced by packing materials and sealing methods was the lowest at vacuum packing and followed by complete sealing methods with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between packing materials but was significantly different between sealing conditions. Conclusionally, vacuum packing and complete sealing with polyethylene film appears to be optimal for storage of Zingiberis Rhizoma.
