This study was carried out to find a useful mushroom at Chungnam Agricultural Research And Extention Service. Twenty materials used were collected from domestic and exotic area. These races were compared bontanical characteristics to leading varieties by PCR-RAPD methods. Mycelial growth temperature of Chongpung and Myongwol were at 20 to 25 ℃ and 25 to 30 ℃ at PDA medium, respectively mycelial growth of these varieties were similiar at pH 6.5 to 7.5. In case of mushroom cultivation temperature ranges, Chongpung was at 5 to 26℃ and Myongwol was at 7 to 28℃, but the optimum temperature range of these were appeared at 15 to 19℃. Culture temperature of these was 23℃ and period of mycelial culture was needed 23 to 24 days under 850cc/pp, while was needed 11 to 12 days at waste cotton medium. Cap color of these at first inducing mushroom was all dark blue, but at late growing stages Chongpung was shown as grey, and Myongwol was shown as dark grey. Yield of Chongpung was appeared as 46kg/3.3m2 and that of Myongwol was 41kg /3.3m2, while Chunchu No2 as check was 40kg/3.3m2. Results from PDA medium and PCR-RAPD analysis two of these were different from others.
The recycling method of enokitake cultural waste and the potentiality of second flush for enokitake were determined, because this fungus is not as prolific as the more commonly cultivated white rot fungi in the conversion of sawdust to mycelial mass. The mycelial growth of F. velutipes on several substrates, variously treated with rice bran was promoted at ratios of 10-20% (w/w) on all substrates, but suppressed at above ratios, although some difference was there. The mycelial densities generally increased correlated to the supplementation contents of rice bran. It could be concluded that F. velutipes preferred mild acidic to acidic conditions for mycelial growth, considering that the mycelial growth rate was highest on waste of pH 6.01, treated with 0.1 % Ca(OH)2 and on populus mixed waste of pH 6.02, non treated. The ranges of substrate bulk densities, which was pertinent for mycelial linear growth were from B.D. (g/cc) 0.17 to 0.23 on waste and populus mixed waste all. The pertinent contents of rice bran supplementation in bottle cultivation was from 20 to 30% on waste and 20% on populus mixed waste, considering the requried duration for pinheading and fruiting yields. Standard bulk density for filling and utilizing the waste and populus mixed waste for commercial f. velutipes cultivation were B.D.(g/cc) 0.19 ~ 0.23, and 0.23~ 0.25, which could be conversed to 510~ 540g/900m1 and 520~ 570g/900m1, respectively, The second flush of F. velutipes was tried and the re-inoculation by sawdust and liquid spawn showed somewhat good results, indicating the potentiality of second crop and suggesting further research for it.
Many ascomycetes were collected at Mt.Unjang, Pyonsan penisula national park, Mt.Moak Chonbuk provincial park, Daea-ri hebarium and Mt.Whaam from August 1996 to October 1999. They were identified. According to the resulting, Lanzia echinophila, Discina parma, Hypomyces auranticus, Helotium versicolor, Cordyceps militaris f. albino, Isaria sinclairii are newly to Korea. They were designed Korean common names by author.
The medicinal constituents contained in Korean and Chinese Angelicae dahuricae Radix were compared by confirming their qualities. From the extracts of Angelicae dahuricae Radix, oxypeucedanin, imperatorin, and alloisoimperatorin etc. furanocoumarin derivatives were identified by GC/MS analysis. Through GC/FID analysis, the furanocoumarin derivatives content of Baizi cultivated in Korea was more than that cultivated in China, except for one cultivated at Ankuk province, and so confirmed to possess, on the whole, good quality medicinal constituents by content, as compared with the ones cultivated in China.
Embryogenic callus cultures of Korean native Seosanjong of ginger(Zingiber of officinale Rosc.) were induced through stem explants taken from in vitro shoot-tip cultures. Among the four concentrations of 2,4-D tested in Murashige and Skoog medium, 0.5 and 1 mg/L of 2,4-D was most effective in inducing embryogenic callus. Leaf explants did not express any new morphogenetic response in all 2,4-D concentrations tested. Plantlets transferred to hormone-free MS medium were developed and successfully acclimatized under greenhouse.
Callus cultures from leaf explants of Gypsophila paniculata L. cv. 'Bristol Fairy' have been tested their growth and morphogenic capacity on Murashige and Skoog medium supplemented with 0.l, 0.5, 1 and 3 mg/L 2,4-D. The frequency of callus formation ranged from 43.3% to 100%. The optimal 2,4-D concentration for promoting callus formation and growth was 0.5 to 3 ㎎/L. 4.2∼ 5.6% of adventitious roots were obtained with the use of 0.1 and 0.5 mg/L 2,4-D. Calli grown well on 1.0 mg/L 2,4-D was the heaviest among the calli grown in various concentrations.
This examination of vegetation is conducted from June to Oct. 1999 through on the spot survey, centering on the habitat of Abies koreana around Imgeolryeong, Jangteomok, and Jeseokbong in Mt Jiri and Yeongsil path and Seongpanak path in Mt. Halla. Species composition tables of the Abies koreana community by altitude and slope in Mt. Jiri and Mt. Halla are made based on the community composition tables examined in quadrat. The Abies koreana-Saso quelpaertensis community in a west slope of Mt. Halla is found that Abies koreana of 4-6m in height forms subtree layer and that of 8-9m in height the tree layer and it reflects a difference between community structure by slope and main composition species. While the tree layer of Abies koreana community is 12-14 in height around the area of 1,290-1,560m above the sea of Imgeolryong and path from Baekmudong to Jangteomok, it is 8-12m in height in the area of 1,680-1,780m above the sea of path from Jangteomok to Jeseokbong. It means that community structure depends on area and attitude. This study finds out that commonly appearing composition species similar to coverage and presence are Lepisorus thunberianus, Quercus mongolica, Solidago virga-aurea var. asiatica, Ligularia fischeri, Carex lanceolata, Clintonia udensis, Magnolia sieboldii, Betula ermani, Acer pseudo-sieboldianum. As soil environment of surveyed area, pH of surveyed spots is similar in Mt. Jiri and Mt. Halla, electrical conductivity is higher in Mt Jiri, and contents of organic matter is relatively higher in a west slope of Mt. Jiri and Mt. Halla and lower in their south and west slopes. Contents of organic matter and total nitrogen show that the area of 1,550m above the sea in a west slope of Mt. Halla is some higher. For pH and contents of total nitrogen and organic matter, Mt. Halla is higher than Mt. Jiri but for electrical conductivity, Mt. Jiri is relatively higher than Mt. Halla.
This study was carried out obtain basic information for growth characteristics by different age of Dendropanax morbifera in southern part of Korea, such as at Sunchon, Haenam, Gangjin, Wando. The pH of soil in cultivation area was 5.2 to 5.6 and organic matter was very high up to 10.6%. P2O5, content of soil in cultivation area was 35.3 to 42.1 mg/1, C.E.S was 13.9 to 14.4 me/100g, and moisture rate was 19.2 to 21.1 %. The flowering of Dendropanax morbifera began from 6~7 years old tree. The flowering date was at 10th of July at Wando. The growth characteristics of 12 years old tree was 929.5cm in stem height, 134.1mm in stem diameter, and 15 years old tree was 1,117.9cm in stem height, 160.8mm in stem diameter. The number of leaf was 13.9 at five years old tree, and the petioles length was 12.6 at five years old tree. Xylem sap can be had at more than 10 years old tree with good growth more than 10 cm stem diameter.r.
