        최근 조직공학 기술의 발달로 재해와 질병으로 인한 손상된 조직과 장기의 대체연구들 이 진행되고 있다. 장기 대체 연구의 핵심 요소 중 하나는 재건된 조직이나 장기가 혈관 망을 형성하여 host tissue로부터 양분과 산소의 전달이다. 본 연구는 조직공학 기법을 이용하여 장기 재건에 필수적인 혈관 재건을 위해 혈관을 구성하는 주세포인 endothelial cell을 체외에서 배양하는 것이다. Endothelial cell(EC)배양을 위해서는 세포지지체인 세 포외 기질(External cellular metrics, ECM)을 필요로 하기 때문에 ECM중에 대표적인 collagen과 gelatin을 사용하여 지지체에 따른 체외배양능을 비교하였다. 실험 동물로는 돼지 대동맥을 채취하여, 대동맥 속에 collagenase type I을 주입하고, 혈관의 입·출구를 봉합한 상태로 10분 간 37℃에서 처리하였다. 관류된 용액은 10% FBS가 함유된 기본배 양액(EGM-2 media)을 사용하여 2번 수세한 후 회수된 세포를 각각의 ECM이 처리된 dish위에서 배양 하였다. EC세포인지를 확인하기 위해서 EC표지 인자인 CD31과 vWF 항체의 발현을 flow cytometry로 확인 하였고, 회수된 세포에서 두 단백질이 모두 발현 되었다. ECM에 따른 EC의 세포 형태를 비교하였을 때 형태학적 차이는 없었다. Basement Membrane Extract위에서 calcein-AM으로 염색된 EC는 ECM의 종류와 상관 없이 2-6시간 사이에 Tube Formation을 보였다. 또한 endothelial cell의 표지 마크인 CD31, Flk1, vWF의 mRNA 발현양과 IHC에 의한 단백질 발현을 조사한 결과 collagen 지지체 위에서 배양된 endothelial cells에서 발현양이 더 높았다. 결론적으로 두 가지 ECM에서 모두 성공적으로 endothelial cell의 배양이 가능하지만 collagen위에서 배양된 endothelial cell이 더 우수한 maintenance능력을 가짐을 확인 할 수 있었다.

        As information-oriented industry has been developed and electronic devices has come to be smaller, lighter, multifunctional, and high speed, the components used to the devices need to be much high density and should have find pattern due to high integration. Also, diverse reliability problems happen as user environment is getting harsher. For this reasons, establishing and securing products and components reliability comes to key factor in company's competitiveness. It makes accelerated test important to check product reliability in fast way. Out of fine pattern failure modes, failure of Electrochemical Migration(ECM) is kind of degradation of insulation resistance by electro-chemical reaction, which it comes to be accelerated by biased voltage in high temperature and high humidity environment. In this thesis, the accelerated life test for failure caused by ECM on fine pattern substrate, 20/20μm pattern width/space applied by Semi Additive Process, was performed, and through this test, the investigation of failure mechanism and the life-time prediction evaluation under actual user environment was implemented. The result of accelerated test has been compared and estimated with life distribution and life stress relatively by using Minitab software and its acceleration rate was also tested. Through estimated weibull distribution, B10 life has been estimated under 95% confidence level of failure data happened in each test conditions. And the life in actual usage environment has been predicted by using generalized Eyring model considering temperature and humidity by developing Arrhenius reaction rate theory, and acceleration factors by test conditions have been calculated.

        본 연구는 온도에 따라 특성이 변하는 아스팔트 콘크리트의 파괴인성을 규명하기 위하여 기존의 유효균열 모델을 수정하는 연구를 다루고 있다. 본래의 ECM모델은 콘크리트와 같은 고체에 적용되도록 개발되어 재료의 포아슨 비를 고려하지 않는다. 하지만 아스팔트 콘크리트는 온도변화에 민감하여 온도에 따라 포아슨 비가 변화하므로 다양한 온도하에서 정확한 파괴 특성을 알기 위해서는 포아슨 비가 고려되어져야 한다. 3개의 개질아스팔트 결합재를 포함한 4가지 결합재를 사용하여 밀입도 아스팔트 혼합물을 제조하여 초기균열 보에 대한 3점 휨 시험을 $-5^{\circ}C$부터 $-35^{\circ}C$까지에서 수행하였다. 탄성계수, 휨강도 및 파괴인성을 시험을 통하여 구하였다. 시험결과 포아슨비가 고려되는 수정 ECM 공식을 사용하므로서 보다 정확한 값들을 얻을 수 있었다. 폴리머 개질 아스팔트 혼합물이 일반아스팔트 혼합물에 비하여 더 낮은 저온하에서 더 높은 강성과 파괴인성을 유지함을 알 수 있었다.

Paying special attention to the head-to-head relation between 1 and C in the two types of complements, it proposes that I-to-C movement is prerequisite for the selectional checking in control infinitives.

        The goal of this paper is to examine a rather interesting paradigm involving mixed clausal coordination, where different types of clauses (finite CPs, ECM, and control infinitives) are coordinated. It will be shown that the paradigm in question has consequences for several phenomena; in particular it sheds light on the controversial issue of the proper analysis of PRO as well as the issue of whether the SpecvP where the subject is base-generated is higher or lower than the SpecvP which serves as the landing site of object shift.

        Adipogenesis is a primary energy valancing response in physiological status and critical in embryo development. One of the essential factors for initiation and maintaining of adipogenesis is the composition of extracellular matrix. Previously, we confirmed the effects of diphlorethohydroxycarmalol (DPHC), an extract of Ishige okamurae, on the antiobesity effects and ECM stability in adipose tissue. In vitro model for adipogenesis study, 3T3-L1, a precursor cell type of adipocyte, and the adipose-tissue derived stem cell (ADSC) can be used. Usually the induction period for adipocyte is shorter in 3T3-L1 than in ADSCs. However, so far, the difference of the expression patterns of ECM components in 3T3-L1 and ADSCs, and the effects of DPHC are not much known. We induced differentiation of 3T3-L1 and ADSCs into adipocyte with or without DPHC (0, 0.4, 2, 10, 50 μg/mL) and confirmed the adipogenesis with adipogenic markers (PPAR-γ, LDL). After then, the levels of collagen type 1 alpha 1 (Col1a1), collagen type 3 alpha 1 (Col3a1), collagen type 4 (Col4), collagen type 6 (Col6), Elastin (Eln) and microfibrillar associated protein 5 (Mfap5) were analyzed with real-time RT-PCR. During early adipogenesis of ADSC, the expression levels of Col1, Col3, Col6, and Mfap5 mRNA were decreased but Col4 and Eln mRNA were increased. In the matured adipocyte, the expression levels of Col1, Col3, Col4, Mfap5 mRNA were decreased but not Eln. In the case of early differentiation of 3T3-L1, the expression levels of Col1, Col3, Eln mRNA were decreased but the expression levels of Col6 and Mfap5 were increased. In matured adipocyte of 3T3-L1, the expression levels of Col1, Col3, Eln, Mfap5 mRNA were increase but the expression level of Col6 mRNA was decreased. The expression levels of Col4, Eln mRNA were suppressed by 50 mg/mL DPHC treatment during early adipogenic period of ADSC. On the other hand in 3T3-L1, the expression levels of Col3 and Col6 mRNA were not changed by the DPHC treatment during early induction period. In the matured adipocytes derived from ADSC, Col1 mRNA levels was not decreased by the treatment of 50 mg/mL DPHC. Col4 mRNA levels was not increased by DPHC treatment. In the case of matured adipocytes derived from 3T3-L1, DPHC suppressed the increase of Col1, Col3, Col6 mRNA expression and the expression of Col4 and Eln mRNA was decreased. In summary, these data show that expression levels of each ECM component types are dramatically changed with some common patterns in two cell types, and the treatment of DPHC can modify the expression patterns of some ECM components in each cell types. It is suggested that one of the reason of antiadipogenic effect of DPHC may be the ECM modification.

        Exceptional Case Marking (ECM henceforth) refers to Case marking of the embedded subject by the matrix verb. The construction in which ECM occurs is called ECM construction. As its name implies, it is exceptional in that two elements in different local domains are involved. This paper aims at answering several questions concerning the ECM construction in Korean: (A) Why is there such a construction as the ECM construction? (B) Does the subject of the complement clause raise to the matrix clause? (C) Why is ECM only available in the /w construction, but not in other constructions? (D) What is the categorial status of the ko construction? I make a proposal that ECM is a syntactic realization of pragmatic force `Focus`. Thus, the complement subject raises into the Spec of the matrix vP to be assigned INT reading as discussed in Chomsky (2001) in OS languages. The movement is possible across the ko construction that is assumed to be a CP because Case assignment via the p(ostpositional) complementizer requires the complement subject to raise into the Spec of CP to meet Spec/Head agreement. Other clausal complements in Korean do not allow ECM since Case transmission is not available and nominal CPs do not allow movement into its Spec.

        Korean ECM constructions seem to give rise to many interesting problems with the minimalist program. Of these problems, we are mainly concerned with the following: (A) How is the ECMed NP/DP assigned accusative Case despite the fact that the clause containing it is supposed to be finite? (B) How can the ECMed NP/DP be affected by the accusative Case assigner though finite clauses are phases? Accessing answers to these problems, we will find out that some unknown properties of the embedded predicates in Korean ECM constructions are involved in the transitivity restriction, and that there is a derivational mechanism for determining whether given movement is A or A`.

        This paper accounts for the Case assignment of ECM verbs within the GB theory and the minimalist theory. The Case assignment of ECM verbs is accounted for on the "exceptional" level: that is, it is different from the other constructions within the GB theory. The Case assignment of believe verbs is consistently accounted for, like the Case assignment of all the constructions within the minimalist theory. Therefore, as a result from this paper, it comes to see that the minimalist theory is more explanatory adequacy than the GB theory in respect of the idea of grammar pursueing the minimum of grammar theory.
