        Organic waste generated by small and medium-sized (S&M-sized) metal decontamination in NPP decommissioning. To lower the concentration of these organic substances for a level acceptable at the disposal site, the project of “Development of Treatment Process of Organic Decontamination Liquid Wastes from Decommissioning of Nuclear Power Plants” is being carried out. The conditioning and treatment process of organic liquid waste was designed. Also, the literature was investigated to make simulated organic liquid waste, and the composition of these waste was analyzed and compared. As the decontamination agent, organic acids such as EDTA, oxalic acid, citric acid are used. The sum of the concentrations of these organic materials was set to a maximum value of 1,000 ppm. The major metal ions of the decontamination liquid waste estimated are 59Fe, 51Cr, 54Mn, 63Ni, and the concentrations are respectively 527, 163, 161, 159 ppm. Additional major metal ions are 60Co, 58Co, 137Cs. 58Co is replaced by 60Co because it has the same chemical properties as 60Co. Unlike the HLW, the contamination level of S&M-sized metal in primary system was quite low, so 60Co is set to 2,000 Bq/g. Considering the contribution of fission and gamma ray dose constant, 137Cs was estimated to 360 Bq/g. Also, suspended solids of decontamination liquid waste were set at 500 ppm. Under these assumptions, the simulated organic liquid waste was made, and then organic substances and metal ions were analyzed with TOC analyzer and ICP-OES. The TOC analysis value was expected to 392 ppm in consideration of the equivalent organic quantity. the test result was 302 ppm. Some of organics appears to have been decomposed by acid. The values of metal ions (Fe3+, Cr3+, Mn2+, Ni2+) analyzed by ICP-OES are 139, 4, 152, 158 ppm, respectively. A large amount of Cr3+ and Fe3+ were expected to exist as ions, but they existed in the form of suspended solid. Mn2+ and Ni2+ came out similar to the expected values. The designed conditioning and treatment process is largely divided into pretreatment, conditioning, and decomposition processes. After collecting in the primary liquid waste storage tank, large particulate impurities and suspensions are removed through a pretreatment process. In the conditioning process, treated liquid waste passes through UF/RO membrane system, and pure water is discharged to the environment after monitoring. Concentrated water is decomposed in the electrochemical catalyst decomposition process, then this water secondarily passes through the RO membrane system and then discharged to the environment after monitoring. Through an additional experiment, the conditioning and treatment process will be verified.

        꽃송이버섯은 우리나라에서 2000년대 초반부터 재배를 시작하였고, 국내에서는 전라북도농업기술원이 육성한 ‘너울’이 2016년에 최초로 등록되어 있다. 그러나, 꽃송이버섯에 대한 재배매뉴얼이 없어 여전히 농가마다 자실체 생산성의 차이가 큰 실정이다. 이에 꽃송이버섯 ‘너울’의 발이조건을 구명하고 안정생산 기술을 개발하고자 배지 pH, 배지함수율, 종균접종량 조건에 따른 연구를 수행한 결과, 재배일수, 수확률과 자실체 중량을 고려한 연중 병당 수 량은 pH 3.8 처리구가 363.6 g으로 가장 높게 나타났으나 pH 3.6 조건에서는 수량이 189.5 g으로 급감하는 점, pH 4.0 조건이 배양일수가 짧고 pH 3.8 조건과 비교하여 재배일수가 같으며, 자실체 중량 차이의 유의성이 인정되지 않는 점, 농가에서 활용 시 정밀한 pH 조절이 어려운 점을 고려하여 pH 3.9±1로 조절하여 재배해야 안정생산이 가능할 것으로 판단된다. 아울러 배지함수율을 65%로 조절하고, 배지부피의 4%에 해당하는 액체종균을 접종 후 배양실 내 습도조건을 50% 이하로 설정하면 연중 병당 341.8 g을 생산할 것으로 판단된다.

        In this study, extraction of uranium(VI) from an aqueous nitric acid solution was investigated using tri-n-butyl phosphate (TBP) as an extractant in an ionic liquid, 1-alkyl-3-methylimidazolium bis (trifluoromethylsulfonyl)imide ([Cnmim][Tf2N]). The distribution ratio of U(VI) in 1.1 M TBP/[Cnmim][Tf2N] was significantly high when the concentration of nitric acid was low. The value of the distribution ratio decreased as the concentration of the nitric acid increased at lower acidities, and then increased with a nitric acid concentration of up to 8 M. This can be attributed to the different extraction mechanisms of U(VI) based on nitric acid concentrations. Thus, a cation exchange at low acidity levels and an ion-pair extraction at high acidity levels were suggested as the extraction mechanism of U(VI) in the TBP/[Cnmim][Tf2N] system.

        Sweetpotato whitefly, Bemisia tabaci (Gennadius) (Hemiptera: Aleyrodidae), is a serious pest of many economically important crops. The insect has developed resistance to chemical insecticides. Therefore, the development of microbial agent is necessary. Among the several entomopathogenic fungi, Lecanicillium lecanii Btab01 which has high insecticidal activity was carried out this experiments. To develop mass culture, we subcultured L. lecanii Btab01 on PDA, TSA, SDA+Y, RA and GSA media at 25℃ incubator to select the optimal solid culture medium. Hyphal growth was measured every 3 or 4 days. L. lecanii Btab01 grew fastest in RA, followed GSA, SDA+Y, PDA and TSA. L. lecanii Btab01 was cultured on PDB, TSB, SDB+Y, RB, GSB media at 25℃, 180rpm shaking incubator to select the optimal liquid medium. Spore germination was measured by spread plate method every 12 or 24 hours. Spore germination appeared 7.8×108 CFU/ml after 4 days in RB, followed GSB (5.5×108 CFU/ml), SDB+Y (2.7×108 CFU/ml), TSB (1.7×108 CFU/ml) and PDB (0.6×108 CFU/ml).

        본 연구는 체외성숙된 돼지난포란을 액상정액으로 수정시 수정시간과 배양배지가 난포란의 발달에 미치는 영향을 조사하기 위하여 실시하였다. 정자농후정액 (30∼60 ml)을 채취하여 실온에서 2시간 정도 서서히 냉각시킨 후, 정액을 15 ml 튜브에 담아 800×g로 10분간 원심분리하였다. 상층액은 버리고 하부의 정자는 5 ml LEN 희석액으로 1×10/sup 9/ 전자/ml가 되도록 재희석하였다. 희석된 정액은 4℃ 냉장고에 보존하였다. 미성숙 난모세포의 성숙에 사용된 배지는 26.19 mM sodium bicarbonate, 0.9 mM sodium pyruvate, 10 ㎍/ml insulin, 2 ㎍/ml vitamin B/sub 12/, 25 mM HEPES, 10 ㎍/ml bovine apotransferrin, 150 μM cysteamine, 10 IU/ml PMSG, 10 IU/ml PMSG, 10 IU/ml EGF, 0.4% BSA, 75 ㎍/ml sodium penicillin G, 50 ㎍/ml streptomycin sulfate그리고 10% pFF를 첨가한 TCM-199 배지였다. 22시간 성숙 배양한 후 난모세포는 cysteamine과 hormone들을 배제한 후 38.5℃, 5% CO₂ incubator에서 22시간 더 성숙시켰다. 성숙된 난모세포는 채취 후 2일간 4℃에 보존된 액상정액으로 수정되었다. 난모세포는 500 μl mTBM 수정 배지에서 1×10/sup 6/ 정자/ml의 농도로 1, 3, 6 그리고 9시간 동안 수정시켰다. 그 후 난모세포는 500 μl NCSU-23, Hopes buffered NCSU-23, PZM-3 그리고 PZM-4 배양배지에 옮겨서 6, 48 그리고 144시간을 더 배양하였다. 정자침투율, 웅성전핵형성율 그리고 난모세포의 난할율은 6 및 9시간 수정시간에서 1 및 3시간 수정시간 보다 높았다. 6시간 수정시 배반포형성율 (33.6%)은 1, 3 그리고 9시간 수정시 배반포형성율 (11.4, 23.0 그리고 29.6%) 보다 높았다. 배반포의 평균세포수는 6, 9, 3 그리고 1시간 수정시 각각 32.9, 27.6, 26.3 그리고 24.4개 였다. 분할된 난모세포의 배반포형성율 그리고 배반포의 평균세포수는 NCSU-23, PZM-3 그리고 PZM-4 배양배지보다 HEPES buffered NCSU-23 배양배지가 우수하였다. 결론적으로 4℃ 보존 돼지액상정액은 체외성숙된 돼지 난모세포의 체외수정에 사용될 수 있음이 입증되었다. 또한 체외성숙된 돼지 난모세포는 500 μl mTBM 수정배지에서 1×10/sup 6/ 정자/ml로 6시간 공배양시키는 것이 바람직하며, HEPES buffered NCSU-23 배양배지에서 배양하는 것이 좋다는 결과를 얻었다.

        Shoot tips of chinese yam (Dioscorea opposita Thunb.) were cultured on MS medium containing 0.5 mg/L BA to produce micro-tubers in vitro. To stimulate the formation of shoots and micro-tubers, and produce large micro-tubers, the sections of micro-tubers were cultured on MS media with BA and IAA. The shoot multiplication, and the micro-tuber formation and growth were very effective on the media containing 2.0 mg/L BA and 0.5~1.0 mg/L IAA. Sucrose added to MS medium with 2.0 mg/L BA and 0.5 mg/L IAA to stimulate more micro-tuber growth. The medium added 50 g/L sucrose was very effective in the increase of plant fresh weight and micro-tuber growth. After 4 weeks' culture of micro-tuber sections on the medium with 2.0 mg/L BA, 0.5 mg/L IAA and 50 g/L sucrose, the liquid media were added into the same vessels. The micro-tuber growth was stimulated remarkably by the addition of liquid medium. The addition of 25 ml liquid medium containing 10 g/L activated charcoal, 3x MS salts and 250 g/L sucrose was the most effective in micro-tuber growth.
